저출력레이저조사가 섬유아세포와 면역세포의 증식 및 유전자발현에 미치는 영향 (Effects of Low Level Laser on the Proliferation and Gene Expression of Fibroblasts and Immune Cells)

  • The growth and synthetic activities of fibroblasts are regulated by cytokines and growth factors derived from activated inflammatory cells. Stimulatory effect of low level laser (LLL) radiation on wound healing seems to be in part due to direct stimulatory action on cell proliferation and synthetic activities of fibroblasts. Also indirect stimulatory effect on the fibroblast function through inflammatory or immune cells is another possible mechanism of biostimulatory action of LLL. This study was performed to determine the growth rate of human gingival fibroblasts obtained biopsy and culture, fibroblast cell line, and immune cell line by using $[^3H]-$ thymidine incorporation test. And gene expression pattern was also analyzed by using the DNA probe such as Hsp70, IL-1$\beta$, MIP-1$\alpha$ and actin cDNA. Proliferation rate of gingival fibroblast was increased by LLL irradiation, but no more effect was added by LPS or IL-1$\beta$ pretreatment Enhanced Hsp70 gene expression was found from gingival fibroblasts and fibroblast cell line COS by LLL irradiation., which was not more increased by LPS or IL-1$\beta$ pretreatment. LLL-irradiated promyelcytic cell line HL-60 and macrophage cell line RAW264.7 showed significant stimulatory effect of proliferation rate when compared with respective control. However there were no changes in growth rate of other immune cell tested in this study, such as B cell line WR19n.l and 230, helper T cell line Jurkat and Hut78, cytolytic T cell line CTLL-r8. By LLL-irradiation Hsp70 gene expression was increased in RAW246.7 and HL-60, not in CTLL-R8. And IL-1$\beta$ and MIP-1$\alpha$ gene expression were induced only from LLL-irradiated RAW264.7. These results led us to presume that LLL radiation may affect to the immune cells, especially to macrophage, through which it might promote wound healing process.

Immune-Enhancing Effects of Crude Polysaccharides from Korean Ginseng Berries on Spleens of Mice with Cyclophosphamide-Induced Immunosuppression

  • Panax ginseng C. A. Meyer is well known as traditional herbal medicine, and ginseng berries are known to exhibit potential immune-enhancing functions. However, little is known about the in vivo immunomodulatory activity of Korean ginseng berries. In this study, crude Korean ginseng berries polysaccharides (GBP) were isolated and their immunomodulatory activities were investigated using cyclophosphamide (CY)-induced immunosuppressive BALB/c mice. In CY-treated mice, oral administration of GBP (50-500 mg/kg BW) remarkably increased their spleen sizes and spleen indices and activated NK cell activities. GBP also resulted in the proliferation of splenic lymphocytes (coordinating with ConA: plant mitogen which is known to stimulate T-cell or LPS: endotoxin which binds receptor complex in B cells to promote the secretion of pro-inflammatory cytokines) in a dose-dependent manner. In addition, GBP significantly stimulated mRNA expression levels of immune-associated genes including interleukin-1β (IL-1β), IL-2, IL-4, IL-6, tumor necrosis factor-α (TNF-α), interferon-γ (IFN-γ), toll-like receptor 4 (TLR-4), and cyclooxygenase-2 (COX-2) in CY-treated mice. These results indicate that GBP is involved in immune effects against CY-induced immunosuppression. Thus, GBP could be developed as an immunomodulation agent for medicinal or functional food application.

Antimicrobial Peptides in Innate Immunity against Mycobacteria

  • Antimicrobial peptides/proteins are ancient and naturally-occurring antibiotics in innate immune responses in a variety of organisms. Additionally, these peptides have been recognized as important signaling molecules in regulation of both innate and adaptive immunity. During mycobacterial infection, antimicrobial peptides including cathelicidin, defensin, and hepcidin have antimicrobial activities against mycobacteria, making them promising candidates for future drug development. Additionally, antimicrobial peptides act as immunomodulators in infectious and inflammatory conditions. Multiple crucial functions of cathelicidins in antimycobacterial immune defense have been characterized not only in terms of direct killing of mycobacteria but also as innate immune regulators, i.e., in secretion of cytokines and chemokines, and mediating autophagy activation. Defensin families are also important during mycobacterial infection and contribute to antimycobacterial defense and inhibition of mycobacterial growth both in vitro and in vivo. Hepcidin, although its role in mycobacterial infection has not yet been characterized, exerts antimycobacterial effects in activated macrophages. The present review focuses on recent efforts to elucidate the roles of host defense peptides in innate immunity to mycobacteria.

황기 부탄올 분획물이 생쥐의 세포성 면역기능에 미치는 영향 (Effects of the Butanol Fraction of Astragali Radix on the Cellular Immune Function in Mice)

  • Effects of the butanl fraction of Astragali Radix (BFAR) on the cellular and nonspecific immune responses were investigated in ICR mice. Mice were divided into 4 groups and BFAR at doses of 5, 25 and 125 mg/kg u ere administered orally to mice daily for 3 weeks, and the normal animals were given vehicle. The results of this study are summarized as follows; the relative weight of thymus was greatly increased by BFAR treatment, compared with that in mormal mice. However, the body weight gain was not affected. Delayed-type hypersensitivity (DTH) reaction to sheep red blood cells (SRBC) for cellular immunity was significantly enhanced by BFAR treatment, compared with those in normal mice. In these mice, BFAR also dose-dependently increased activities of phagocyte and natural killer (NK) cells as well as the number of leukocyte resulted from nonspecific immunity Thus, these results demonstrate that BFAF treatment results in a significant increase in both cellular and nonspecific immune responses to antigen in concentrations that enhance humoral immune function.

잣버섯 균사체로부터 분리한 수용성 단백다당체 Lepidan의 면역 증가 작용 (Enhancement of Immune Responses by a Water Soluble Proteoglycan, Lepidan from Lentinus lepideus)

  • In this study, we investigated the immunopotent activities of lepidan, a water soluble proteoglycan from Lentinus lepideus. To examine whether lepidan may affect nonspecific immune responses, we determined the effect on the production of nitric oxide (NO). Lepidan effectively increased the NO production in IFN-${\gamma}$ and LPS triggered RAW 264.7 cells. To further demonstrate the evidence that lepidan activates various immune cells, we determined the alkaline phosphatase activity, plaque-forming cell number and secretion of interleukine-4 (IL-4) and granulocyte/macrophage-colony stimulating factor (GM-CSF) after lepidan treatment in murine splenocytes. The results showed that lepidan increased alkaline phosphatase activity and the number of plaque-forming cells, which indicates that lepidan can lead B lymphocytes to late stage of differentiation. Also, when the splenocytes were cultured with lepidan for 48 hr, the level of IL-4 and GM-CSF in the supernatant dramatically increased. Taken together, these data suggest that lepidan is a biological response modulator that is able to activate immune responses.

추출 방법에 따른 육미지황탕의 항염증 작용 평가 (Evaluation of Anti-Inflammatory Effects of Yukmijihwangtang and Individual Drug Substances Based on the Extraction Methods)

  • Objectives: This study was performed to develop therapeutic prescription that is more significant than existing ones through extraction method and formulation changes. Methods: Yukmijihwangtang(YMJHT) was extracted in 80% ethanol, and their relative anti-oxidant activities as well as anti-inflammatory effects through immune modulation were measured. Results: Both water and ethanol extracted YMJHT showed does-dependent DPPH elimination activities. ROS inhibition activity was greater in water extracted YMJHT except for Moutan Cortex. NO inhibition assay results indicated that all groups showed higher NO inhibition activities in RAW 264.7 cells in dose dependent manner. Water extracted group showed higher NO inhibition activity than that of ethanol extracted group. TNF-${\alpha}$ secretion inhibition assay using RAW 264.7 cells, water extracted YMJHT showed higher activity than ethanol extracts. Growth rate of spleen cells was greater in all tested groups, with higher rate in YMJHT-EtOH than YMJHT-DW. Suppression of gene expression of IFN-r in spleen cells stimulated by Con A was higher in YMJHT-EtOH than YMJHT-DW. Suppression of gene expression of IL-10 in spleen cells stimulated by Con A was highest in YMJHT-DW with 40%. Suppression of gene expression of IL-4 in spleen cells stimulated by Con A were significant with 90% or higher in all groups and that of IL-12p35 were also higher than 90% in all cases. Conclusions: From the results, it shows that YMJHT has anti-inflammatory effects through immune modulation. However, the difference between YMJHT-EtOH and YMJHT-DW was not that significant. Further studies are needed to find out effective extraction methods of herbal medicine.

Effects of Acanthopanax sessiliflorus on Immune Cells such as Thymocytes, Splenocytes and Macrophages in Mice

  • Objective : Immune potentiation including activation of T cells, B cells, macrophages, and dendritic cells is known to play a key role in prevention and treatment of patients with cancer. In this study, we investigated the effects of Acanthopanax sessiliflorus (AR) on the immune system, especially on thymocytes, splenocytes, and macrophages. Methods : We investigated the effects of AR on proliferation of splenocytes in normal mice, and the effects on proliferation of splenocytes and thymocytes in tumor-bearing mice. In addition, the effect of AR on NO production using macrophages was investigated. Results : Treatment with AR accelerated proliferation of splenocytes in vitro. AR also accelerated thymocyte proliferation, but did not affect splenocytes proliferation in normal mice. In contrast, AR accelerated proliferation of splenocytes and thymocytes significantly in tumor bearing mice. In addition, NO production level from macrophages was elevated by treatment with AR. Conclusion : These results demonstrate that AR has anti-cancer activities and related mechanisms are involved in immune potentiation such as acceleration of immune cell proliferation and elevation of NO production level in macrophages. In addition, we also demonstrate the possibilities of AR as complementary and alternative medicine to standard anti-cancer drugs.

병동형 호스피스 환자를 위한 원예요법의 개발 및 수면의 질과 면역기능에 미치는 효과 (Development and Effects of Horticulture Therapy on Quality of Sleep and Immune Function in Patients in Hospice Units)

  • Purpose: The study was done to examine the development and effects of horticulture therapy on quality of sleep and immune function in patients in hospice units. Methods: The participants in this study were hospice patients in D hospital in D city. Thirty hospice patients were assigned to the experimental group, thirty to the control group. Data were collected from April 29 to July 26, 2009. The horticulture therapy program consisted of indoor and outdoor horticultural activities. The horticulture therapy was conducted for 30 minutes, 6 times a week for 3 weeks (a total 18 times). Measures were quality of sleep, and immune function by serum T-cell, NK-cell count. Data were analyzed using descriptive statistics, chi-square test and t-test with SPSS/WIN 13.0 version. Results: Patients in the experimental group receiving horticulture therapy had a significant difference in changes in the quality of sleep compared to the control group. There were also a significant difference in changes in the immune function (serum T cell and serum NK cell) between the experimental group and control group. Conclusion: The study results indicate that horticulture therapy developed for hospice patients is an effective, palliative intervention program to improve the quality of sleep and immune function of hospice patients.

팔손이 용매별 추출물의 항암 및 면역 활성 탐색 (Screening of Anticancer and Immune Activities by the Extracts of Fatsia japonica Decne. et Planch.)

  • 국내에서 자생하는 팔손이 나무의 각 부위를 이용하여 에탄올과 물 용매를 이용하여 각각 $65^{\circ}C$$65^{\circ}C$로 12시간 동안 2회 반복 추출하였으며 부위에 따른 항암 및 면역 활성의 변화를 측정하는 실험을 하였다. 실험결과 부위에 따른 항암 활성 증진 효과 실험 중 근피와 에탄올 용매 추출물이 가장 높게 나타나는 것을 확인 할 수 있었다. 면역세포의 생육활성 측정결과에서는 근피 부위가 높은 활성도를 나타내어 다른 부위에 비해 $40{\sim}60%$ 정도의 높은 활성을 나타내었다 이는 두릅나무과 수종들의 특징인 근피의 특이 활성을 나타내는 예인데 이에 반해 목부의 활성을 살펴보면 농도의 증가에 따라 활성도가 증가하기는 하지만 저농도의 시료에서는 100% 미만의 활성도를 나타냄으로서 면역 세포에 독성을 나타내는 것을 확인할 수 있었다. 면역세포의 활성과 관련하여 Cytokine의 분비량과의 상관관계를 확인 할 수 있었는데 면역세포의 생육활성이 가장 높은 때에 IL-6와 $TNF-{\alpha}$의 Cytokine의 분비량이 최대치인 $110{\sim}120$ $pg/m{\ell}$를 나타내었다. 이로서 앞선 결과들에 따라 팔손이 나무의 부위별 항암 및 면역 활성 실험에서 다른 두릅나무들과 수종들과 같이 근피의 높은 활성도를 확인할 수 있었으며 특히 가장 활성이 좋은 근피는 가장 낮은 활성을 나타내는 목부의 추출물과 비교하여 최대 40% 이상의 높은 활성도를 관찰할 수 있었다. 이로서 근피의 추출물이 다른 부위와 비교하여 높은 활성을 나타내는 것은 두릅나무과 수종들의 대표적인 특징 성분인 사포닌이 다른 부위에 비해 많이 함유되어 있는 것으로 사료되어지며 이는 후에 분획 및 물질 분석 실험을 통하여 확인되어져야 할 사항으로 사료되어진다.

오미자의 전통발효에 의한 면역활성 증진 (Enhancement of Immune Activities of Kadsura Japonica Dunal. through Conventional Fermentation Process)

  • 여러 가지 추출조건을 통해 얻은 오미자 추출물과 전통발효공정을 통해 얻은 오미자 발효액의 면역증진 활성을 비교하였다. 오미자 발효액이 B cell과 T cell에서 시료를 첨가하지 않은 대조군과 비교하여 각각 30%와 22%이상의 수치를 나타내며 가장 좋은 활성을 나타내었다. B, T cell의 cytokine 분비량 측정을 통한 실험에서도 오미자의 전통발효액이 생육증진과 유의적으로 가장 많은 분비량을 나타내었다. 대식세포를 이용한 NO$^-$ 생성능 측정에서 오미자 발효액 및 추출물의 활성이 LPS와 비교해 비슷하거나 높은 것을 확인하였다. 또한 시료를 LPS와 같이 첨가하였을 때 시료만을 첨가했을 때보다 높은 활성을 나타내어 농도에 따른 유의적 활성 증가가 이루어질 것으로 사료된다. NK cell의 활성측정에서 모든 시료 조건에서 6일 동안 생육이 증가하였으며 오미자 발효액을 첨가한 것이 아무것도 첨가하지 않은 대조군과 비교하여 약 1.4배의 생육도 증진을 나타내어 1.2배의 생육도 증진을 보인 100$^{\circ}$C에서 초음파를 병행한 물 추출물보다 높은 활성을 나타내었다. HPLC를 이용한 추출물들의 성분 분석에서 전통발효 공정을 통해 추출공정에서 확인할 수 없었던 특정 성분의 생성 및 기존 성분의 용출 증진이 이루어지는 것을 확인하였고, 이에 따라 면역활성의 증진이 이루어지는 것으로 추정할 수 있었다. 이상의 결과에서 오미자는 면역활성을 가지는 성분을 함유하고 있으며 전통발효 공정을 통해 생성되는 대사산물이 면역실험에서 유효활성 증진을 나타내는 것을 확인하였다. 이는 전통발효 공정을 통해 생성된 성분의 활성으로 세포독성이 감소하고 세포활성이 증가하기 때문인 것으로 생각된다. 이상의결과는 기존에 면역활성 증진 효과를 나타내는 초음파병행 공정 보다 높은 증진 효과로 전통발효 공정 및 활성성분에 대한 다각적인 연구가 필요할 것으로 사료된다.
