많은 연구에서 돼지의 난포란 혹은 배아가 낮은 온도에 대해 민감한 것은 세포질 내에 함유된 지방구와 관련이 있다고 하였으며, 이런 세포질내의 지방구를 제거함으로 동결능력이 향상된다고 하였다. 이렇게 지방구가 제거된 돼지의 난포란과 배아를 효과적으로 동결보존하기 위해서는 새로운 개념의 동결보존 기술이 필요하다고 하겠다. 따라서, 본 연구는 미성숙 단계에서 지방이 제거된 돼지 난포란의 유리화 동결에 적합한 방법을 찾기 위하여 straw를 이용한 유리화 동결법과 electron microscopic grid(EM grid)를 사용한 유리화 동결법을 실시하여 효율적인 동결방법을 검토하였다.
본 연구는 돼지 미성숙 난포란의 유리화 동결액에 노출시 독성과 삼투압 스트레스가 가장 적은 유리화 동결액을 조사하기 위하여 실시하였으며, 그 결과를 요약하면 다음과 같다. 미성숙 난포란을 EFS(40% ethylene glycol +18% ficoll +0.3 M sucrose) 용액, ES(5.5 M ethylene glycol +1.0 M sucrose) 용액, GE(10% glycerol +20% ethylene glycol) 용액에 각각 노출하고 대조구로 아무 처리하지 않은 난포란을 체외성숙하여 관찰한 체외성숙율은 ES 용액과 대조구 에서 EFS 용액과 GE 용액보다 유의적으로 높았으며(P<0.05), 유리화 동결액의 독성에 의한 난포란의 퇴화율은 대조구, ES 용액, EFS 용액, GE 용액 순으로 유의적으로 낮은 퇴화율을 나타내었다(P<0.05). 미성숙 난포란을 EFS 용액, ES용액, GE용액에 각각 노출하고 대조구로 아무 처리하지 않은 난포란을 체외성숙, 체외수정하여 관찰한 정상 수정율은 대조구가 세 종류의 유리화 동결액보다 유의적으로 높았고(P<0.05), 유리화 동결액 간에는 유의적인 차이가 없었다. 다정자 침입율과 난자당 침입한 평균 정자수는 모든 처리구간에 유의적인 차이가 없었다. 미성숙 난포란을 EFS 용액, ES 용액, GE 용액에 각각 노출하고 대조구로 아무 처리하지 않은 난포란을 체외성숙, 체외수정, 체외 배양하여 관찰한 수정 3 일째의 분할율은 ES 용액과 대조구가 EFS 용액과 GE 용액보다 유의적으로 높았으며(P<0.05), 수정 7일째의 배반포 형성율은 모든 처리구간에 유의적인 차이가 없었다. 배양기간 동안 나타난 퇴화율은 대조구가 세 가지 유리화 동결액에 비해 유의적으로 낮았고(P<0.05), ES 용액은 EFS 용액과 GE 용액보다 유의적으로 낮은 퇴화율을 나타내었다(P<0.05).
The objective of this study was to examine the effect of in vitro maturation (IVM) medium, cytochalasin B (CB) treatment during intracytoplasmic sperm injection (ICSI), and electric activation on in vitro development ICSI-derived embryos in pigs. Immature pig oocytes were matured in vitro in medium 199 (M199) or porcine zygote medium (PZM)-3 that were supplemented with porcine follicular fluid, cysteine, pyruvate, EGF, insulin, and hormones for the first 22 h and then further cultured in hormone-free medium for an additional 21~22 h. The aim of present study was to investigate regulatory mechanism of alpha-linolenic acid (ALA) during in vitro maturation (IVM) on nuclear and cytoplasmic maturation of porcine oocytes. Basically, immature cumulus-oocyte complexes (COCs) were incubated for 22 h in IVM-I to which hormone was added, and then further incubated for 22 h in IVM-II without hormone. As a result, relative cumulus expansion was increased at 22 h after IVM and it was enhanced by treatment of ALA compared with control group (p < 0.05). During IVM process within 22 h, cAMP level in oocytes was decreased at 6 h (p < 0.05) and it was recovered at 12 h in ALA-treated group, while oocytes in control group recovered cAMP level at 22 h. In cumulus cells, it was reduced in all time point (p < 0.05) and ALA did not affect. Treatment of ALA enhanced metaphase-I (MI) and MII population of oocytes compared with oocytes in control group at 22 and 44 h, respectively (p < 0.05). Intracellular GSH levels in ALA group was increased at 22 and 44 h after IVM (p < 0.05), whereas it was increased in control group at 44 h after IVM (p < 0.05). In particular, the GSH in ALA-treated oocytes during 22 h of IVM was higher than control group at 22 h (p < 0.05). Lipid amount in oocytes from ALA group was higher than control group (p < 0.05). Treatment of ALA did not influence to absorption of glucose from medium. Cleavage and blastocyst formation of ALA-treated oocytes were enhanced compared with control group (p < 0.05). These findings suggest that supplementation of ALA could improve oocyte maturation and development competence through increasing GSH synthesis, lipid storage, and regulation of cAMP accumulation during early 22 h of IVM, and these might be mediated by cumulus expansion.
The objective of this study was to examine the effect of in vitro culture media on embryonic development of in vitro-matured (IVM) oocytes after parthenogenetic activation (PA) in pigs. Immature pig oocytes were matured in TCM-199 supplemented with porcine follicular fluid, cysteine, pyruvate, EGF, insulin, and hormones for the first 22 h and then further cultured in hormone-free medium for an additional 22~26 h. IVM oocytes were activated by electric pulses and cultured in porcine zygote medium-3 (PZM-3) and North Carolina State University-23 supplemented with essential and non-essential amino acids (NCSU-23aa). These media were further modified by supplementing 2.77 mM myo-inositol, 0.34 mM trisodium citrate, and $10{\mu}M$${\beta}$-mercaptoethanol (designated as mPZM-3 and mNCSU-23aa, respectively). Culture of PA embryos in mPZM-3 significantly increased development to the blastocyst stage than culture in NCSU-23aa (36.2% vs. 24.8%, p<0.05). Modified PZM-3 showed a significantly higher blastocyst formation than NCSU-23aa in both groups of embryos that were activated at 44 h and 48 h of IVM (51.0% vs. 35.5% and 49.0% vs. 34.2% in oocytes activated at 44 h and 48 h of IVM, respectively). Irrespective of the follicle diameter where oocytes were collected, embryonic development to the blastocyst stage was increased (p<0.05) by the culture in mPZM-3 compared to culture in NCSU-23aa (25.9% vs. 34.2% and 32.9% vs. 44.8% in embryos derived from small and medium size follicles, respectively). Our results demonstrated that culture media had significant effect on preimplantation development PA embryos and that mPZM-3 was superior to mNCSU-23 in supporting development to the blastocyst stage in pigs. This beneficial effect of mPZM-3 on embryonic development was not impaired by other factors such as time of oocyte activation and origin of immature oocytes (small and medium size follicles).
The objective of this study was to examine the effect of thymidine treatment during $in$$vitro$ maturation (IVM) of porcine follicular oocytes on blastocyst development. Porcine oocytes were treated with thymidine (10 mM, 20 mM and 30 mM) for 2 or 6 hr in the preiods of IVM I and/or II. The survival rates of the blastocysts in the 6 hr treatment groups of 10 mM and 20 mM during IVM I period were significantly higher than those of control group ($p$<0.05). However, the survival rate of the blastocysts in the 2 hr treatment group of 20 mM during IVM II period was significantly higher than control group ($p$<0.05). Furthermore, the survival rate of the blastocysts in the 6 hr treatment group of 30 mM during IVM II period was significantly lower than control group ($p$<0.05). Consistent with the previous result, blastocyst development of both IVM I and II treatment group was also showed as similar pattern. Total and apoptotic cell numbers of blastocysts derived from thymidine treated porcine oocytes were examined by using Tunel assay. The results showed that there was no significant differences in total cell number of blastocysts between thymidine treated and untreated groups. However, apoptosis-positive cells in the thymidine treated group (6 hr IVM I) were significantly lower than those of other groups ($p$<0.05). Taken together, these results indicate that high quality oocytes were selected by DNA synthesis mechanism according to high concentration thymidine treatment during porcine oocyte maturation. Therefore, we concluded that presumptive selected oocytes by thymidine treatment during maturation periods improved the further embryo development and embryonic quality of IVF embryos by decreasing the incidence of apoptosis in preimplantation porcine embryos.
체외수정의 성공률에 있어서 배란되기 직전의 충분히 성숙된 난자를 채취하는 것이 중요하며, 최근에는 질 벽을 통하여 초음파를 이용하는 방법을 많이 사용하고 있다. 난자 채취 시술 시 정맥 마취제를 투여하는데 최근에는 수술회복이 빠르고 오심이나 구토 등의 부작용이 적은 Propofol(2,6-disoprooylphenol)과 Thiopental sodium을 사용한다. 이들 마취제가 마취 투여 시간과 양에 따라 난자의 성숙률과 수정률, 발생률에 어떠한 영향을 미치는지 조사하고자 하였다. 돼지에서 추출한 성숙 전 단계의 난자를 마취제인 Propofo1 및 Thiopental sodium의 다양한 농도와 시간에 노출시킨 후 난자 성숙를을 조사하였으며 돼지 정자와 체외수정시킨 후 수정률과 발생률을 관찰하였다. 또한, 수정 단계 없이 단독 발생하는 처녀생식에 대한 영향도 조사하였다. Propofol에 단시간 노출된 난자는 대조군과 큰 차이가 없었으나 고농도에서 장시간 노출 시 성숙률이 현저히 높게 나타났다. 그러나 수정률은 높은 농도에 장시간 노출할 경우 대조군보다 낮아졌는데, 이와 같은 일시적인 난자 성숙은 Propofol이 처녀생식 유발물질이기 때문으로 보여진다. Thiopental sodium은 저농도 단시간 처리 시 난자의 성숙률과 수정률을 모두 감소시켰다. 이상의 결과로 볼 때, 각 마취제의 최적농도와 투여시간을 결정하여 이러한 영향을 최소화시키는 것이 체외수정의 성공률과 밀접한 관계가 있는 것으로 생각된다.
The objective of this study was to determine the effect of fructose that was supplemented to a chemically defined in vitro maturation (IVM) medium on oocyte maturation and embryonic development after parthenogenesis in pigs. The base medium for in vitro maturation (IVM) was porcine zygote medium (PZM) that was supplemented with 0.05% (w/v) polyvinyl alcohol (PVA) or 10% (v/v) porcine follicular fluid (pFF). In the first experiment, when immature pig oocytes were matured in a chemically defined medium that was supplemented with 5.5 mM glucose or with 1.5, 3.0 and 5.5 mM fructose, 3.0 mM fructose resulted in a higher nuclear maturation (91.5%) than 1.5 and 5.5 mM fructose (81.9 and 81.9%, respectively) but showed a similar result with 5.5 mM glucose (94.2%). However, there was no significant differences among groups in the embryo cleavage (89.4-92.4%), blastocyst formation (37.5-41.1%), and mean cell number of blastocyst (30.8-34.2 cells). Fructose at the concentration of 3.0 mM (1.08 pixels/oocyte) resulted in a higher intra-oocyte glutathione (GSH) content than 1.5 and 5.5 mM fructose (1.00 and 0.87 pixels/oocytes, respectively) while the cumulus cell expansion was not influenced. In the second experiment, effect of individual and combined supplementation of a chemically defined maturation medium with 5.5 mM glucose or 3.0 mM fructose was examined. No significant effect was found in the nuclear maturation (86.3-92.6%). Embryo cleavage was significantly increased by the combined supplementation with glucose and fructose (95.2%) compared to that with 3.0 mM fructose only (85.7%) while blastocyst formation (37.3-42.8%) and embryonic cell number (33.3-34.1 cells) were not altered. Effect of supplementation of pFF-containing medium with glucose and fructose + glucose was examined in the third experiment. No significant effect by the supplementation with glucose and fructose or glucose alone was observed in the nuclear maturation of oocytes (90.7-94.1%) and blastocyst formation (51.0-56.5%). Our results demonstrate that 3.0 mM fructose was comparable to 5.5 mM glucose in supporting in vitro oocyte maturation and embryonic development after parthenogenesis and could be used as an alternative energy source to glucose for in vitro maturation of pig oocytes.
Embryonic genome activation (EGA) is the first major transition that occurs after fertilization, and entails a dramatic reprogramming of gene expression that is essential for continued development. Although it has been suggested that EGA in porcine embryos starts at the four-cell stage, recent evidence indicates that EGA may commence even earlier; however, the molecular details of EGA remain incompletely understood. The RNA polymerase II of eukaryotes transcribes mRNAs and most small nuclear RNAs. The largest subunit of RNA polymerase II can become phosphorylated in the C-terminal domain. The unphosphorylated form of the RNA polymerase II largest subunit C-terminal domain (IIa) plays a role in initiation of transcription, and the phosphorylated form (IIo) is required for transcriptional elongation and mRNA splicing. In the present study, we explored the nuclear translocation, nuclear localization, and phosphorylation dynamics of the RNA polymerase II C-terminal domain in immature pig oocytes, mature oocytes, two-, four-, and eight-cell embryos, and the morula and blastocyst. To this end, we used antibodies specific for the IIa and IIo forms of RNA polymerase II to stain the proteins. Unphosphorylated RNA polymerase II stained strongly in the nuclei of germinal vesicle oocytes, whereas the phosphorylated form of the enzyme was confined to the chromatin of prophase I oocytes. After fertilization, both unphosphorylated and phosphorylated RNA polymerase II began to accumulate in the nuclei of early stage one-cell embryos, and this pattern was maintained through to the blastocyst stage. The results suggest that both porcine oocytes and early embryos are transcriptionally competent, and that transcription of embryonic genes during the first three cell cycles parallels expression of phosphorylated RNA polymerase II.

In this study, we examined the effectiveness of in vitro fertilization of porcine immature oocytes on the embryo development of blastocysts or hatched blastocysts and the number of cells according to the in vitro fertilization conditions. In the in vitro fertilization of in vitro matured porcine oocytes, there were no significant differences between treatment groups regarding fertilization rate, blastocyst rate, and embryo development of hatched blastocysts according to the storage periods of liquid sperm of 24, 48, and 72 hours. The embryo development rate of hatched blastocysts after the fertilization according to different spermatozoa concentrations ($0.4{\times}10^5$, $1.2{\times}10^5$, and $3.6{\times}10^5$ cells/ml) showed the highest rate in the group with a spermatozoa concentration of $1.2{\times}10^5$ cells/ml; in particular, this rate was significantly higher than that in the $0.4{\times}10^5$ cells/ml group (p<0.05). The total number of blastocysts cells as well as trophectoderms (TE) that developed in each treatment group were also significantly higher in the $1.2{\times}10^5$ cells/ml group than in any other groups (p<0.05). In contrast, the embryo development rate of blastocysts according to different co-incubation periods of sperm and oocyte (1, 3, and 6 hr) was high in the 6-hour group; in particular, the rate was significantly higher than that of the I-hour group (p<0.05). Furthermore, the total number of oocytes cells and TEs that developed was significantly higher in the 6-hour group than any other group (p<0.05). In this study, the most effective treatment conditions for porcine embryo development and high cell number were found to be as follows: a sperm storage period of less than 72 hours, a spermatozoa concentration of $1.2{\times}10^5$ cells/ml, and a 6-hour co-incubation period for sperm and ooocyte.
