  • DNA methylation is one of the major epigenetic regulations of gene expression. The DNA methylation patterns are dramatically changed during gametogenesis and embryogenesis, and especially, it has been known that embryonic stem cells show a distinct methylation pattern. In this study, we examined the methylation patterns of imprinting genes, H19, Igf2r, and Snrpn, in stem cells induced from fertilized embryo (fES) and somatic cell nuclear transferred embryo (ntES). The methylation pattern of H19 gene in both fES and ntES were similar. However, the methylation patterns of Igf2r and Snrpn in ntES (hypermethylated) were slightly different from fES cells.

Effects of Refeeding with a Protein-Free Diets Supplemented with Various Essential Amino Acids on the Plasma Insulin-Like Growth Factor-I Concentration in Fasting Young Chickens

  • The effect of refeeding with various single essential amino acids on the recovery of plasma insulin-like growth factor-I (IGF-I) concentration in fasted young chickens was examined. Young chickens (29 days of age) were divided into 15 experimental groups. Chickens in one group were fed on the commercial diet ad libitum for 4 days. The remaining 56 chickens in 14 experimental groups were fasted. After 2 days of fasting, 52 chicks in 13 fasted groups were refed with one of the following experimental diets for 2 days. Eleven experimental diets were protein-free diets supplemented with one of 11 essential amino acids (Arg, Gly, His, Ileu, Leu, Met, Phe, Lys, Thr, Trp, Val). The remaining 2 experimental diets were a protein-free diet containing 11 essential amino acids and a protein-free diet not supplemented with amino acids. Birds in the remaining fasted group continued to be fasted for 2 days. Fasting for 2 days markedly reduced plasma IGF-I concentration. When fasted chickens were refed the protein-free diet containing either Gly alone or all essential amino acids, plasma IGF-I concentration was recovered to the level similar to that of fed chickens. Protein-free diet alone, however, failed to restore the reduced IGF-I concentration in plasma. Body weight loss modulated by feeding with protein-free diets supplemented with various single essential amino acids was associated with changes in plasma IGF-I concentrations. We concluded that body weight loss by feeding with a protein-free diet was lower than that of fasted chickens and that body weight loss associated with the decrease in plasma IGF-I concentration was modulated by feeding with protein-free diets containing various single essential amino acids.

Relationships between Methionine Supply, Nitrogen Retention and Plasma Insulin-like Growth Factor-I in Growing Sheep Nourished by Total Intragastric Infusions

  • Four 4-month old Charolais${\times}$Dorset male sheep (initial liveweight $25.0{\pm}1.1\;kg$), fitted with rumen and abomasal fistulas and nourished by total intragastric infusions, were used to study the relationships between methionine (Met) supply, nitrogen (N) retention and plasma insulin-like growth factor-I (IGF-I). Four graded levels of Met, i.e. 0 g/16 g N, 1.76 g/16 g N, 3.52 g/16 g N and 7.04 g/16 g N, were infused into abomasums as experimental treatments. The sheep and treatments were allocated in a $4{\times}3$ incomplete Latin square design (Yudon square design). The experiment lasted 3 periods and each period was 10 days. Quadratic correlations were found between Met level (x, g/16 g N) and N retention (y, g/d): y = $-0.03x^2$+0.41x+2.62, $r^2$ = 0.66, n = 12, p = 0.008, and between methionine level (x, g/16 g N) and plasma IGF-I concentration (y, ng/ml): y = $0.80x^2$-4.53x+190.24, $r^2$ = 0.51, n = 12, p = 0.009. No significant correlation was found between plasma IGF-I (x, ng/ml) and N retention (y, g/d) (p>0.05). It was concluded that Met level had a significant influence on N retention and plasma IGF-I concentration whereas IGF-I might not be an important mediator in the regulation of N metabolism by Met in growing sheep nourished by total intragastric infusions.

  • This study was carried out to investigate safety evaluation of IGEs separated and refined from bovine milk and commercial recombinant human IGFs. In order to evaluate toxicity of these samples, acute toxicity test and short term toxicity test were investigated with IGF-I separated and refined from colostrum and commercial recombinant human IGF-I from R&D systems company. for acute toxicity test, we selected recombinant human IGF-I from R&D systems company and establish one control group and three dose-level groups(0, 10, 20 and 50 $\mu\textrm{g}$ per rat). We have intravenously injected tail of rats with selected sample once. After 20 days, pathological cellular tissue analyses were investigated with liver, kidney and spleen of 12 rats in all test groups. However, Morbid tissue and abnormal statistical results were not discovered in all cellular tissues. For short term toxicity test, we selected IGF-I separated and refined from colostrum and establish one control group and three dose-level groups(0, 5, 10 and 15 $\mu\textrm{g}$/day per rat). Rats were orally injected with selected sample once a day during two weeks. After short term toxicity test period, Pathological cellular tissue analyses were investigate with liver, kidney and spleen of 12 rats in all test groups. However, Morbid tissue and abnormal statistical results were not discovered in all cellular tissues. These results suggest that IGF-I treated groups show no significant toxicological findings with changes of body weight, food consumption, water consumption, and pathological findings compared with control groups.

Characterization of Insulin-like Growth Factor-free Interaction between Insulin-like Growth Factor Binding Protein 3 and Acid Labile Subunit Expressed from Xenopus Oocytes

  • The acid-labile subunit (ALS) is known to interact with the IGF binding protein (IGFBP) in the presence of insulin-like growth factors (IGFs). Studies, however, indicate that ALS forms a doublet with IGFBP3, independent of IGFs. To characterize the structural domain required for the IGF-free ALS-IGFBP3 interaction, seven recombinant human IGFBP3 mutants were generated: three deletion mutants and four site-specific mutants that had altering N-terminal regions of IGFBP3. ALS and IGFBP3 mRNAs were co-injected into Xenopus oocytes, and their products were cross-linked and immunoprecipitated using antisera against ALS or IGFBP3. Among the deletion mutants, the mutant of D40 (deleted in 11-40th amino acids) exerted no effect in the interaction with ALS, while D60 (${\Delta}11$-60) demonstrated a moderate reduction. D88 (${\Delta}11$-88), however, showed a significant decrease. In the case of site-specific mutants, the mutation that alterated the IGF binding site (codons 56 or 80) exerted a significant reduction in the interaction, whereas codons 72 or 87 showed no significant change in the interaction with ALS. The stability of the ALS-IGFBP3 interaction was analyzed according to a time-dependent mode. Consistent with the binding study, mutants on the IGF binding sites (56 or 80) consistently show a weakness in the ALS-IGFBP3 interaction when compared to the mutants that covered the non-IGF binding sites (72 or 87). This study suggests that the N-terminal of IGFBP3, especially the IGF binding site, plays an important role in interacting with ALS as well as in stabilizing the dual complex, independent of IGFs.

Interaction Between Acid-Labile Subunit and Insulin-like Growth Factor Binding Protein 3 Expressed in Xenopus Oocytes

  • The acid-bible subunit (ALS) associates with the insulinlike growth factor (IGF)-I or II, and the IGF binding protein-3 (IGFBP-3) in order to form a 150-kD complex in the circulation. This complex may regulate the serum IGFs by restricting them in the vascular system and promoting their endocrine actions. Little is known about how ALS binds to IGFBP3, which connects the IGFs to ALS. Xenopus oocyte was utilized to study the function of ALS in assembling IGFs into the ternary complexes. Xenopus oocyte was shown to correctly translate in vitro transcribed mRNAs of ALS and IGFBP3. IGFBP3 and ALS mRNAs were injected in a mixture, and their products were immunoprecipitated by antisera against ALS and IGFBP3. Contrary to traditional reports that ALS interacts only with IGF-bound IGFBP3, this study shows that ALS is capable of forming a binary complex with IGFBP3 in the absence of IGF When cross-linked by disuccinimidyl suberate, the band that represents the ALS-IGFBP3 complex was evident on the PAGE. IGFBP3 movement was monitored according to the distribution between the hemispheres. Following a localized translation in the vegetal hemisphere, IGFBP3 remained in the vegetal half in the presence of ALS. However, the mutant IGFBP3 freely diffused into the animal half, despite the presence of ALS, which is different from the wild type IGFBP3. This study, therefore, suggests that ALS may play an important role in sequestering IGFBP3 polypeptides via the intermolecular aggregation. Studies using this heterologous model will lead to a better understanding of the IGFBP3 and ALS that assemble into the ternary structure and circulate the IGF system.

  • Psoriasis is a chronic skin disease characterized by angiogenesis. It has been reported that growth factor as vascular endothelial growth factor(VEGF) and insulin like growth factor(IGF) II are overexpressed in psoriatic epidermis. To investigate the inhibitory effects of IGF-II induced VEGF and HIF-1${\alpha}$ expression by RR extracts, we performed MTS assay, western blots using HaCaT cells. RR extracts significantly reduced IGF-II induced HIF 1${\alpha}$ protein level via MAPK pathway in HaCaT cells. Also, RR extracts inhibited IGF-II induced VEGF mRNA and protein expression levels in the HaCaT keratinocytes. These results suggest that inhibition of HIF-1${\alpha}$ and VEGF expressions by RR extracts contributes to the anti angiogenic effects.

Correlation Analysis of Serum Hormones and Components in Hanwoo during Slaughtering Process (한우 도축시 혈청 호르몬과 혈액 성분의 상관 관계 분석)

  • Concentration of hormones and blood components at the last fatting stage was changed before slaughter in Hanwoo steers and bulls. Two months before slaughter and shipment, concentration of cortisol and creatinine was increased, but that of calcium was decreased. Concentration of insulin growth factor-1 (IGF-1) was decreased after shipment, and inorganic phosphorus (IP) was decreased at slaughter. It is unclear that changes of concentration in between 2 months before slaughter and shipment were either caused by aging or stresses (abstinence, environmental change, blood drawing, and shipment). Changes of blood concentration between shipment and slaughter may be accounted for overall responses from abstinence, shipment, and unfamiliar environment. A positive correlation between 2 months before slaughter and before shipment was detected for IGF-1, total protein (TP), albumin, creatinine, high density lipoprotein cholesterol (HDLC), and globulin in steers, and creatinine and globulin in bulls. A positive correlation between 2 month before slaughter and slaughter was detected for IGF-1, blood urea nitrogen (BUN), IP and HDLC in steers, and creatinine in bulls. A positive correlation between before shipment and slaughter was detected for testosterone, IGF-1, creatinine, triglyceride, HDLC and globulin in steers, and TP, creatinine, HDLC and globulin in bulls.

Effect of Recombinant Bovine Somatotropin Injections on Deer Stag Plasma Insulin-like Growth Factor-I Profiles (rBST 투여가 숫사슴의 혈액 내 IGF-I 함량에 미치는 영향)

  • This study was conducted to elucidate the influence of recombinant bovine somatotropin (rBST) on the plasma IGF-I profiles in deer stags. Slow release rBST (250mg each) preparations were injected subcutaneously during the antler's non-growing period and growing period. It was observed that significant increases in the plasma IGF-I concentrations were occurred right after injections of the rBST irrespective of antler's non-growing period or growing period. When the rBST preparations were injected two times with one week interval during the antler's non-growing period, the mean level of IGF-I was found to be 2,920.7ng I mL, indicating that the IGF-I concentration increased up to 8.4-folds compared with a control. After two weeks from second shot, however, the IGF-I levels decreased to the level of the control. Similar results were also obtained with both a Sika and a Red deer during the antler's growing period. Increases in the IGF-I level of up to 5.6-folds with the Sika deer and 4.3-folds with the Red deer were found. Based on the present study, it is likely that the rBST may be used as a possible candidate for an antler's growth stimulating agent.

Effects of Intra-duodenal Infusion of Limiting Amino Acids on Plasma Insulin-like Growth Factor I, Growth Hormone, Insulin and Liver Insulin-like Growth Factor I mRNA in Growing Goat Fed a Maize Stover-based Diet

  • The effects of intra-duodenal infusion of methionine (Met), lysine (Lys) and leucine (Leu) on dry matter intake (DMI), the concentrations of insulin-like growth factor I (IGF-I), growth hormone (GH) and insulin in plasma, and liver IGF-I mRNA level were investigated in two experiments for Liuyang Black growing wether goats. In Experiment 1, three goats ($10.0{\pm}0.1$ kg) were fitted with ruminal, proximal duodenal and terminal ileal fistulaes to determine the infusion amounts of Met, Lys and Leu at the duodenum according to essential amino acid flows into the duodenum and their apparent digestibility. The infusion amounts were 0.77 g/d, 0.91 g/d and 0.58 g/d respectively. In Experiment 2, 4 groups of goats (($10.0{\pm}0.2$ kg) for each group, were cannulated at the duodenum, and were infused with a mixture of Met, Lys and Leu (Control), or mixtures with 21% Met, Lys or Leu replaced with glutamate respectively on a nitrogenous basis. The replacement of 21% Met, Lys or Leu with glutamate did not affect intakes of maize stover, concentrate or both (p>0.05) when compared with the control. The replacement of 21% Met or Lys significantly (p<0.05) reduced plasma GH, insulin and IGF-I concentrations and liver IGF-I mRNA level. The replacement of 21% Leu with glutamate reduced (p<0.05) plasma IGF-I concentration only, but not plasma insulin and GH, as well as liver IGF-I mRNA level (p>0.05). The close relationships between supplying Met and Lys in the lumen of the duodenum and plasma IGF-I, GH and insulin concentrations, as well as liver IGF-I mRNA level in this study indicate that the effects of the limiting amino acids on nutrition of animals are likely intermediated via their effects on these hormones, and these hormone profiles could be used as intermediate markers for the limiting order of amino acids.
