한국산 여뀌속 Persicaria절(마디풀과)의 핵 리보오솜 ITS 염기서열 변이 (Variation of nuclear ribosomal ITS sequences of Polygonum section Persicaria (Polygonaceae) in Korea)

  • 본 연구에서는 한국산 여뀌속 Persicaria절 분류군들에 대학 핵 리보오솜 (nrDNA)의 ITS 염기서열 분석을 수행하여 본 절 분류군들의 유연관계를 추정하고자 하였다, 본 연구결과 본 절 분류군의 nrDNA ITS 구간 염기서열은 본 절 분류군의 분류학적 타당성, 한계 및 계급설정 및 분류군 간의 계통적 유연관계를 추정하는 데 있어 유용한 것으로 나타났다. ITS 염기서열 분석결과 얻어진 neighbor-joining tree에서 본 절 한국산 분류군들은 크게 P.amphibium과 나머지 분류군들을 포함하는 ground의 2개의 계열로 구분되었다. 나머지 분류군들을 포함하는 두번째 계열은 다시 (1) P. lapathifolium, (2) P. persicaria와 P. viscoferum, (3) P. orientale 및 P. viscosum, (4) P. japonicum 및 (5) P. longisetum, P. erecto-minus var. koreense, P. caespitosum var. laxiflorum, P. hydropiper, P. pubescence, P. tinctorium, P. foliosum, P. trigonocarpum을 포함하는 group으로 세분되었다. 이러한 결과는 과거 형태적 식별형질에 의해 제안되었던 본 절 분류군간의 유연관계와 근본적으로 일치하였다.

Chemosystematics and Molecular Phylogeny of a New Bioflocculant-Producing Aspergillus Strain Isolated from Korean Soil

  • The ubiquinone and G+C contents of the bioflocculant-producing fungus, a new Aspergillus strain, were detennined using high-perfonnance liquid chromatography. The internal transcribed spacers 1 and 2 (ITS1 and ITS2), and the 5.8S ribosomal DNA (rDNA) of the strain were amplified and sequenced. The strain contained ubiquinone-l0($H_2$)as a major quinone and the G+C content was 49 mol%. A phylogenetic analysis of the ITS regions indicated that the strain belonged to the genus Aspergillus according to its previously classified morphological characteristics. Based on a sequence homology search, the strain was most closely related to Petromyces muricatus (anamorph, A. muricatus; accession number, AJ005674). The sequence of a new Aspergillus strain in ITS1 and ITS2, and 5.8S rDNA showed 97% homology to P. muricatus. Therefore, the strain is believed to be a new bioflocculant-producing Aspergillus strain.

ITS 염기서열 분석 및 CAPS를 이용한 조이시아 속(Zoysia) 들잔디와 갯잔디의 구별 (Molecular Identification of Zoysia japonica and Zoysia sinica (Zoysia Species) Based on ITS Sequence Analyses and CAPS)

  • Zoysia 속 잔디는 학교운동장 및 공원, 골프장, 스포츠경기장과 같이 다양한 장소에 식재되고 있는 중요한 잔디이다. 해안가에서 자생하는 Zoysia 속 들잔디와 갯잔디는 외부 형태적 특성이 유사하여 외부 형태적 분류 뿐 만 아니라 분자생물학적 분류도 필요하다. 본 연구에서는 nrDNA-ITS(Internal Transcribed Spacer)의 DNA 바코드 분석을 통해서 자생하는 들잔디와 갯잔디의 분자생물학적 신속한 분류체계를 확립하고자 하였다. 이를 위해 난지형 잔디인 Zoysia 속 들잔디(Z. japonica) 및 갯잔디(Z. sinica)와 한지형 대표 잔디인 크리핑 벤트그라스(A. stolonifera) 및 켄터키 블루그라스(P. pratensis)의 nrDNA-ITS 염기서열을 확보하였다. 확보된 들잔디및 갯잔디, 크리핑 벤트그라스, 켄터키 블루그라스의 ITS 염기서열 전체 구간은 각 686bp와 687bp, 683bp, 681bp으로 확인되었으며, nrDNA-ITS 내부 염기서열구간 분석 결과, ITS1의 크기는 248-249bp, ITS2는 270̵-274bp, 5.8S rDNA는 163-164bp의 차이로, 각 4종의 잔디가 ITS 염기서열을 이용하여 식별되었다. 특히, 들잔디와 갯잔디 nrDNA-ITS 염기서열은 19 염기(2.8%) 차이를 나타냈으며, ITS1과 ITS2의 G + C 함량은 55.4-63.3% 임을 확인하였다. 이러한 들잔디와 갯잔디의 ITS 염기서열 차이를 바탕으로 CAPS 마커로 전환하여 대조구 및 수집된 자생 Zoysia 속 잔디 영양체 62개체를 분석한 결과, 외부형태학적 분류법으로 들잔디 개체, 갯잔디 개체로 동정되었지만, ITSCAPS 마커를 이용한 분자생물학적 분류법으로 들잔디 36개체와 갯잔디 22개체 뿐만 아니라 들잔디와 갯잔디간의 자연교배종 4개체도 식별하였다. 이상의 결과에서 들잔디와 갯잔디는 ITS 염기서열 및 ITS 기반 CAPS를 통하여 식별할 수 있을 것으로 판단된다.

Modification of ERα by UFM1 Increases Its Stability and Transactivity for Breast Cancer Development

  • The post-translational modification (e.g., phosphorylation) of estrogen receptor α (ERα) plays a role in controlling the expression and subcellular localization of ERα as well as its sensitivity to hormone response. Here, we show that ERα is also modified by UFM1 and this modification (ufmylation) plays a crucial role in promoting the stability and transactivity of ERα, which in turn promotes breast cancer development. The elevation of ufmylation via the knockdown of UFSP2 (the UFM1-deconjugating enzyme in humans) dramatically increases ERα stability by inhibiting ubiquitination. In contrast, ERα stability is decreased by the prevention of ufmylation via the silencing of UBA5 (the UFM1-activating E1 enzyme). Lys171 and Lys180 of ERα were identified as the major UFM1 acceptor sites, and the replacement of both Lys residues by Arg (2KR mutation) markedly reduced ERα stability. Moreover, the 2KR mutation abrogated the 17β-estradiol-induced transactivity of ERα and the expression of its downstream target genes, including pS2, cyclin D1, and c-Myc; this indicates that ERα ufmylation is required for its transactivation function. In addition, the 2KR mutation prevented anchorage-independent colony formation by MCF7 cells. Most notably, the expression of UFM1 and its conjugating machinery (i.e., UBA5, UFC1, UFL1, and UFBP1) were dramatically upregulated in ERα-positive breast cancer cell lines and tissues. Collectively, these findings implicate a critical role attributed to ERα ufmylation in breast cancer development by ameliorating its stability and transactivity.

Observations on Fragmentation Pathway of Farinomalein and its Isomers by Structural Investigation Using LC-MS/MS

  • Farinomalein is a maleimide-bearing compound well known for its anti-fungal activity. In the present study, synthesis of farinomalein is achieved via Stobbe condensation followed by Haval-Argade contrathermodynamic rearrangement. Kinetically driven Stobbe condensation followed by condensation with beta-alanine reveals formation of two isomers of farinomalein. This article describes application of LC-MS/MS in structure elucidation of farinomalein 1 and its isomers 2 and 3 encountered in its synthesis. The proposed distinct fragmentation pathway is supported by rational organic reaction mechanism. These fragmentation pathways are significant for analytical method development of farinomalein in near future. The structures of farinomalein 1 and its isomers 2 and 3 have been assigned undisputedly.

Secondary Structure and Phylogenetic Implications of ITS2 in the Genus Tricholoma

  • The internal transcribed spacer (ITS) region in the genus Tricholoma was analyzed, including for its primary nucleotide sequence and secondary structural characterization. The secondary structures of the ITS2 region in the genus Tricholoma were identified for use in bioinformatic processes to study molecular evolution and compare secondary structures. Ten newly sequenced ITS regions were added to the analysis and submitted to the GenBank database. The resulting structure from a minimum energy algorithm indicated the four-domain model, as previously suggested by others. The conserved secondary structure of the ITS2 sequences of the genus Tricholoma exhibited certain unique features, including pyrimidine tracts in the loops of domain A and a complete structure containing four domains, with motifs identified in other ITS2 secondary structures. A phylogenetic tree was derived from sequence alignment based on the secondary structures. From the resulting maximum parsimonious tree, it was found that the species in the genus Tricholoma had evolved monophyletically and were composed of four groups, as supported by the bootstrapping values and pileus color.

ITS 염기서열을 이용한 한국산 참취속 식물의 유연관계분석 (Phylogenetic Analysis of Korean Native Aster Plants Based on Internal Transcribed Spacer (ITS) Sequences)

  • 본 연구는 국내산 참취속 식물의 ITS(internal transcribed spacer) 염기서열을 결정하고 이들을 이용하여 $Kalimeris$, $Gymnaster$, $Heteropappus$ 속 식물들간의 유연관계를 분석하고자 수행되었다. 섬쑥부쟁이($A.$ $glehnii$) 등을 포함한 11종의 참취속 식물의 ITS 염기서열을 결정하였으며 유전자 은행에 등록된 $Kalimeris$ 속 식물의 ITS 염기서열을 포함하여 군집분석을 실시하였다. ITS1의 길이는 249-253bp로, ITS2의 길이는 181-217bp로 나타났으며 G + C 함량은 47-54%의 변이를 나타내었다. 본 연구에서 다룬 16종의 18개체간 염기 치환율은 한 site당 ITS1의 경우는 9%, ITS2는 10%로 나타나 ITS1과 2 부위간의 차이가 거의 없는 것으로 나타났다. 또한 협의의 참취속($Aster$ sensu strict) 식물과 $Kalimeris$ 속 식물의 치환율을 비교한 결과 ITS1과 ITS2 지역 모두 $Kalimeris$ 속 식물이 낮게 나타났다. 계통분석결과, 갯개미취는 군외군으로부터 가장 먼저 분지하였으며, 일본산 $A.$ $bellidiastrum$이 나머지 분류군들을 위한 자매군으로 유집되었다. 참취는 다른 참취속 식물들과 분리되어 독립적인 분계조를 형성하였다. 갯쑥부쟁이($A.$ $hispidus$)를 제외한 $Kalimeris$ 속 식물은 91%의 높은 지지율을 가지고 광의의 참취속내로 유집되었으며 $Gymnaster$$Heteropappus$ 속 식물 역시 광의의 참취속으로 유집되었다. 이러한 연구결과는 $Kalimeris$, $Gymnaster$$Heteropappus$로 세분되었던 참취속을 ITS 염기서열을 바탕으로 통합될 수 있을 것으로 판단된다.

Cultural Conditions for Mycelial Growth and Molecular Phylogenetic Relationship in Different Wild Strains of Schizophyllum commune

  • The common split-gilled mushroom, Schizophyllum commune is found throughout the world on woody plants. This study was initiated to evaluate conditions for favorable vegetative growth and to determine molecular phylogenetic relationship in twelve different strains of S. commune. A suitable temperature for mycelial growth was obtained at $30^{\circ}C$. This mushroom grew well in acidic conditions and pH 5 was the most favorable. Hamada, glucose peptone, Hennerberg, potato dextrose agar and yeast malt extract were favorable media for growing mycelia, while Lilly and glucose tryptone were unfavorable. Dextrin was the best and lactose was the less effective carbon source. The most suitable nitrogen sources were calcium nitrate, glycine, and potassium nitrate, whereas ammonium phosphate and histidine were the least effective for the mycelial growth of S. commune. The genetic diversity of each strain was investigated in order to identify them. The internal transcribed spacer (ITS) regions of rDNA were amplified using PCR. The size of the ITS1 and ITS2 regions of rDNA from the different strains varied from 129 to 143 bp and 241 to 243 bp, respectively. The sequence of ITS1 was more variable than that of ITS2, while the 5.8S sequences were identical. A phylogenetic tree of the ITS region sequences indicated that the selected strains were classified into three clusters. The reciprocal homologies of the ITS region sequences ranged from 99 to 100%. The strains were also analyzed by random amplification of polymorphic DNA (RAPD) with 20 arbitrary primers. Twelve primers efficiently amplified the genomic DNA. The number of amplified bands varied depending on the primers used or the strains tested. The average number of polymorphic bands observed per primer was 4.5. The size of polymorphic fragments was obtained in the range of 0.2 to 2.3 kb. These results indicate that the RAPD technique is well suited for detecting the genetic diversity in the S. commune strains tested.

국도상의 지능형교통시스템의 효과성 분석에 관한 연구 (A Study on the Effectiveness of Intelligent Transport Systems on National Highways)

  • 현재 지능형교통시스템(Intelligent Transport Systems, ITS)은 교통시설에 대한 다양한 정보와 자료를 수집하여 가공할 수 있는 첨단교통시스템으로서 사회적 관심이 커지고 있으며, 고속국도 뿐 아니라 일반국도에도 ITS 인프라 구축이 활발히 진행되고 있다. 하지만 ITS 설치에 대한 명확한 효과 분석이 어려워 이에 대한 계량화된 방법론이 필요한 실정이다. 이에 본 연구에서는 익산지방국토관리청에서 관할하고 있는 국도 ITS 사업의 효과분석을 위해 경제성분석을 실시하였다. 분석기간을 10년, 할인률을 5.5%로 가정하였으며 효과척도로는 통행시간 절감, $CO_2$ 배출 절감, 연료 절감, 도로전광표지(Variable Message Sign, VMS) 정보의 가치를 선정하여 각 항목에 대한 계량화 방안을 마련하였다. 경제성 분석 결과 익산지방국토관리청의 ITS 구축사업은 편익/비용 비율이 1.20, 내부 수익률이 12.4%, 순 현재 가치가 14.8억원으로 나타나 ITS 구축이 경제성이 있는 것으로 나타났다.

  • This study was undertaken to investigate the effects of ITS and EGF on embryonic development of in vitro matured and fertilized bovine oocytes in culture medium su, pp.ementing with or without calf serum. When fertilized oocytes were cultured in TCM-199 containing 0, 0.1, 0.5 and 1.0\ulcorner/ml ITS with 5% calf serum, the rates of development to blastocyst stage and the cell number of blastocysts were not significantly different among all treatments. And also monolayer of cumulus/granulosa cells prepared in containing calf serum and ITS were no beneficial effects of embryonic development. On the other hand, when EGF was su, pp.imented to TCM-199 containing calf serum or calf serum free, embryonic development rates(24.0 2.8% to 29.2 1.7% or 8% to 9%) and cell number of blastocysts(p<0.05) were significantly increased compared with EGF-free(22.1 2.1 or 1.0%, p<0.05). But when fertilized oocytes were cultured with cumulus/granurosa cells in TCM-199 containing EGF and calf serum, the rate of embryos development to the blastocyst stage and cell number of blastocysts were not significantly different compared with EGF-free and any concentrations. These results showed that ITS and EGF was not improved the development of bovine embryo in vitro matured and in vitro fertilized with calf serum and/or monolayer of cumulus/granulosa cells.
