Genetic diversity and genetic differentiation in six natural populations of Abies holophylla Max were investigated using ISSR marker system. From 6 ISSR primers, the average percentage of polymorphic loci was 85.6%, and the average expected heterozygosity ($H_e$) was 0.288. From the result of AMOVA, 94.4% of total genetic variation came from the differences among individuals within populations, and 5.6% was caused by those of among-populations. On the basis of Bayesian inference, genetic differentiation (${\theta}^{II}$ and $G_{ST}$) and inbreeding coefficient for all populations were 0.045, 0.038, and 0.509, respectively. The correlation between genetic distance and geographical distance was highly significant at the Mental's test (r = 0.74, P < 0.05). Six populations divided into two groups according to the results of UPGMA and PCA. One group included Namwon, Cheongdo and Mungyeong population. The other was Inje, Hongcheon and Pyeongchang population. Also, in Bayesian clustering analysis, 6 populations were divided into two clusters. But Cheongdo population was assigned into the other cluster unlike those of UPGMA or PCA. Taking the regions based on the results of the cluster analysis into consideration of AMOVA, 3.9% of genetic variation came from the regional difference. The dendrogram from UPGMA could provide the most genetically reasonable explanation for the distribution of Abies holophylla populations in South Korea.

Genetic diversity among Thaumatococcus daniellii populations in the southwestern region of Nigeria were assessed using morphometric and molecular markers to determine the population structure and existing genetic relationship for its improvement, conservation and sustainable utilisation. Populations from five locations in each of the six states were used for the study. Morphometric data were collected on folia characters and analysed for variability. Genome DNA was isolated from the plant leaf and amplified by polymerase chain reaction with inter-simple sequence repeat markers (ISSR) to determine the allelic polymorphism, marker effectiveness and genetic relationship of the population. The results showed significant variations in petiole length and leaf dimensions of the populations within and across the states. These morphometric traits are the major parameters that delimit the populations and they correlated significantly at P≤0.05. Analysis of the electrophoregram showed that the ISSR markers are effective for the diversity study. A total of 136 loci were amplified with an average of 7.16 loci per marker, 63.2% of the loci were polymorphic. The Principal Coordinate Analysis revealed that seven factors accounted for 81.6% of the variation and the dendrogram separated the populations into two major groups at a genetic distance of 10 (about 90% similarity) with sub-groups and clusters. Most populations within the state had a high degree of similarity, nonetheless, strong genetic relationship exists among populations from different states. The close relationship between populations across the states suggests a common progenitor, which are likely separated by ecological or geographical isolation mechanisms.
The in vitro organogenesis is one of important issues in plant embryology, and somaclonal variations are existing in calli and/or regenerants induced from a process of the organogenesis with in vitro circumstances. In this study, expressions of organogenesis-related genes were evaluated and genetic stability of regenerants derived from the process of in vitro organogenesis were measured using ISSR markers in Imperata cylindrica 'Rubra', Poaceae. The expressions of organogenesis-related genes were detected all of regenerants at the process of the organogenesis. All ISSR markers produced with an average of 71 bands per in vitro-cultured regenerants, and the scorable bands were varied from two to eight with an average of 5.14 bands per a primer. The polymorphism rates of the in vitro regenerants were higher than that of mother plants (1.4%), showing 4.1% (pot-cultured regenerants), 4.3% (field-cultured regenerants), 4.2% (in vitro-cultured regenerants), 5.6% (calli with green shoots) and 1.4% (calli), respectively. The genetic similarity matrix (GSM) among all accessions ranged from 0.747 to 1.0 with a mean of 0.868. GSM of the regenerants showed differences (from 0.972 to 1.00) compared with that of mother plants (0.991). According to the clustering analysis, two independent groups were divided into; the one is mother plants and regenerants cultured at room and open field, the other is regenerants cultured in vitro. The results give a new insight for understanding the dynamics of organogenesis in monocot plant.

Agastache rugosa, a member of the mint family (Labiatae), is a perennial herb widely distributed in East Asian countries. It is used in traditional medicine for the treatment of cholera, vomiting, and miasma. This study assessed the genetic diversity and population structures on 65 accessions of Korean mint A. rugosa germplasm based on inter simple sequence repeat (ISSR) markers. The selected nine ISSR primers produced reproducible polymorphic banding patterns. In total, 126 bands were scored; 119 (94.4%) were polymorphic. The number of bands generated per primer varied from 7 to 18. A minimum of seven bands was generated by primer 874, while a maximum of 18 bands was generated by the primer 844. Six primers (815, 826, 835, 844, 868, and 874) generated 100% polymorphic bands. This was supported by other parameters such as total gene diversity ($H_T$) values, which ranged from 0.112 to 0.330 with a mean of 0.218. The effective number of alleles ($N_E$) ranged from 1.174 to 1.486 with a mean value of 1.351. Nei's genetic diversity (H) mean value was 0.218, and Shannon's information index (I) mean value was 0.343. The high values for total gene diversity, effective number of alleles, Nei's genetic diversity, and Shannon's information index indicated substantial variations within the population. Cluster analysis showed characteristic grouping, which is not in accordance with their geographical affiliation. The implications of the results of this study in developing a strategy for the conservation and breeding of A. rugosa and other medicinal plant germplasm are discussed.
This study was carried out to address an efficient in vitro regeneration system from seed-derived callus of Phragmites communis, and to evaluate genetic variations of the regenerants using ISSR markers. Shoot regeneration via calli was greatly influenced by N6 medium compared with MS medium, and plant regeneration frequency was 90% in N6 supplemented with BA 0.25 mg/L and BA 0.5 mg/L. According to ISSR analysis of the thirty regenerants, out of 94 loci detected overall, 16 were identified to be polymorphic with a rate (PR) of 17.0%. The mean gene diversity (h) of different in vitro condition was 0.03 and ranged from 0.008 for N6 with BA 5 mg/L, to 0.040 for MS with IAA 0.1 mg/L+kinetin 2 mg/L. The results indicate that the regenerants have a low genetic variation, and ISSR analysis is effective to detect genetic variation of regenerants.

This study analyzed 96 individuals in 6 populations using ISSR marker to investigate the genetic diversity of Salicornia herbacea populations. The total of 49 PCR amplification bands was observed in 6 ISSR primers, and 30 of them had genetic polymorphisms. The Shannon's information index (I) and gene diversity index (h), which indicate the genetic diversity of the Salicornia herbacea populations, were 0.382 and 0.249, respectively. The genetic diversity according to the population size was lowest with 0.092 (I) and 0.058 (h) in $0.1m{\times}0.1m$ and highest with 0.338 (I) and 0.227 (h) in $25m{\times}25m$, which was suitable for the furtherance of the high population with high genetic diversity. The UPGMA dendrogram based on Nei's genetic distance did not show a significant correlation with the distance between the Salicornia herbacea population. The results indicate that the Salicornia herbacea habiting in the restricted environment should have an area over a certain size to ensure the formation of a population with genetic diversity.

A simple and reliable cryo technique using desiccation and slow freezing of winter dormant buds was employed for 238 core collection of mulberry germplasm collected from diverse geographical regions and maintained under tropical conditions in the ex situ field gene bank to develop long-term biodiversity conservation for ensuring sustainable utilization of these valuable resources. Desiccation and freezing tolerance of bud grafts and excised shoot apices in the axillary buds of different Morus species under in vivo and in vitro condition indicated species-specific variation and most of the wild Morus species were found sensitive. In vitro regeneration and cryopreservation($-196^{\circ}C$) protocols using differentiated bud meristem like axillary winter dormant buds were worked out for a wide range of Morus species, land races, wild and cultivated varieties. Successful cryopreservation of mulberry winter dormant buds of different accessions belonging to M. indica, M. alba, M. latifolia, M. cathayana, M. laevigata, M. nigra, M. australis, M. bombycis, M. sinensis, M multicaulis and M. rotundiloba was achieved. Among wild species Morus tiliaefolia, and M. serrata showed moderate recovery after cryopreservation. Survival rates did not alter after three years of cryopreservation of different Morus species. ISSR markers were used to ascertain the genetic stability of cryopreserved mulberry, which showed no difference detected among the plantlets regenerated from frozen apices in comparison to the non-frozen material.
The genetic diversity and structure of eight populations of Coreanomecon hylomeconoides Nakai, an endemic Korean plant, were investigated using 50 ISSR loci from eight primers. The average percentage of polymorphic loci was 47.3%. The Shannon's index (SI=0.218) and gene diversity (h=0.142) were relatively lower than those of other long-lived perennials. The Sancheong (SI=0.233, h=0153), Gwangyang (SI=0.263, h=0.171), and Suncheon (SI=0.241, h=0.159) populations showed greater genetic diversity than the Namhae and Gwangju populations, which are on the edge of the distribution. Analysis of molecular variance (AMOVA) showed that 18% of the total variation could be attributed to differences among populations, and 82% to differences within populations, indicating moderate gene flow among adjacent populations. These results were supported by value of Nm (2.184). The UPGMA conducted using the genetic distance and Bayesian cluster analysis showed a remarkable geographic trend structured into east and west regions. Overall, the results indicate that the Sancheong and Gwangyang populations, which had a large population size and higher degree of genetic diversity, should be the focus of in situ conservation.
The morphological characteristics and genetic relationships among 32 germplasms of Zanthoxylum schinifolium and Zanthoxylum piperitum collected from two farms in Korea were investigated. The traits with the most variability were seed color, leaf size, and spine size. The intraspecific polymorphism of Z. schinifolium and Z. piperitum was 96.5% and 60.3%, respectively. The genetic diversity and Shannon’s information index values ranged from 0.11 to 0.33 and 0.19 to 0.50, with average values of 0.26 and 0.42, respectively. Two ISSR primers (UBC861 and UBC862) were able to distinguish the different species. The genetic similarity matrix (GSM) revealed variability among the accessions ranging from 0.116 to 0.816. The intraspecific GSM for Z. schinifolium and Z. piperitum was 0.177-0.780 and 0.250-0.816, respectively. The GSM findings indicate that Z. schinifolium and Z. piperitum accessions have high genetic diversity and possess germplasms qualifying as good genetic resources for cross breeding. The clustering analysis separated Z. schinifolium and Z. piperitum into independent groups, and all accessions could be classified into three categories. Z. Schinifolium var. nermis belonged to independent groups. Comparison of the clusters based on morphological analysis with those based on ISSR data resulted in an unclear pattern of division among the accessions. The study findings indicate that Z. schinifolium and Z. piperitum accessions have genetic diversity, and ISSR markers were useful for identifying Z. schinifolium and Z. piperitum.
Genus Hemerocallis is a herbaceous species and some species among their taxa are very important herbal medicines. We evaluated representative samples of the eight taxa in Korea with inter simple sequence repeats (ISSR) markers to estimate phylogenetic relationships within taxa of this genus. The studied taxa were Hemerocallis fulva L., H. fulva for. kwanso, H. dumortieri Morren, H. coreana Nakai, H. hongdoensis M.G.Chung & S.S.Kang, H. middendorffi Trautv. et Mayer, H. thunbergii Baker, H. minor Miller. In addition, we investigated the genetic variation and structure of Korean populations of these taxa. The mean genetic diversity was 0.098 across species, varying from 0.068 to 0.123. A low level of genetic variation was found in populations of Hemerocallis species. Specially, gene diversity for H. minor was maintained the highest among genus Hemerocallis. An indirect estimate of the number of migrants per generation (Nm=0.218) indicated that gene flow was not extensive among Korean populations of Hemerocallis species. The phylogenic tree showed distinct three clades. One includes H. fulva, H. fulva for. kwanso and H. middendorffi. Another includes three Hemerocallis species, H. dumortieri, H. thunbergii and H. minor. The H. coreana and H. hongdoensis were shown as the sister group to the second clades. Although the size of sampling was not large enough for eight Korean Hemerocallis species, the analyses of ISSRs will certainly provide an enhanced view on the phylogeny of species.
