Measurement Uncertainty for Analytical Method of Hyaluronic Acid Used as a Dietary Supplement (식이보충제로 사용되는 히알루론산의 분석법에 대한 불확도 산정)

  • This study aimed to estimate the measurement uncertainty related to determination of hyaluronic acid used as a dietary supplement by high-performance liquid chromatography. According to the guidelines of the Association of Official Analytical Chemists, the analytical processes for determination of hyaluronic acid were performed. And the measurement uncertainty obtained during the analytical processes were expressed in accordance with mathematical/ statistical guidances of GUM (Guide to the Expression of Uncertainty in Measurement) & EURACHEM (Focus for Analytical Chemistry in Europe) for the analytical operations. For the uncertainty in measurement produced based on this analytical method, the expanded uncertainty was calculated by using the relative standard uncertainty between analytical results and sources of uncertainty in measurement (sample weight, final volume, extraction volume, standard solution, matrix and instrument etc). In the results of 95% confidence interval, it was calculated that the uncertainty in measurement was $57.75{\pm}8.76{\mu}g/kg$ (k=2.0). Therefore, it showed that the measurement uncertainty obtained by this analytical method influences on 15.2% of the contents of hyaluronic acid as the analytical results.

  • Klebsiella sp. L-10 was treated with physical and chemical mutagens, and one of the NTG mutant which increased hyaluronic acid complex yield 2.5 folds was selected. The yield of hyaluronic acid complex from Klebsiella sp. L-10 NTG 50 mutant reached maximum level I the YPD medium containing 0.1% yeast extract, 3% Bacto-tryptone, 3% dextrose, each 30mM of $K_2HPO_4$ and $KH_2PO_4$ (pH 6.0-6.5) with shaking culture at $37^{\circ}C$ for 24 hrs, and 2900mg of hyaluronic acid complex per litre of culture was produced under the above condition.

  • Hyaluronic acid (HA) is a natural glycosaminoglycan and is used widely in the pharmaceutical field. Lipoic acid (LA) helps the regeneration of exogenous and endogenous antioxidants such as Vitamin C and Vitamin E as well as glutathione. It also acts as antioxidant indirectly. Hydrophilic HA as a biodegradable and biocompatible polymer was conjugated with hydrophobic LA as an antioxidant to form the graft copolymer. The carboxyl group of HA was modified by adipic acid dihydrazide (ADH). The synthesis of HA-g-LA graft copolymers was characterized by FT-IR, $^1H$-NMR spectroscopy. The conjugates could form the self-assembled nanoparticles in aqueous solution. The particle size and critical aggregation concentration were verified to use the nanoparticle as a carrier fur the hydrophobic material.

  • Hyaluronic acid and chitosan-based poly(ethylene oxide) (HA-PEO and Chitosan-PEO) hydrogels have been employed as unique biomedical polymeric materials with properties such as bioactivity from polysaccharide, biocompatibility of HA and chitosan as well as PEO and control release of bioactive molecules from the hydrogel itself. We here examine in situ hydrogels based on hyaluronic acid and chitosan in terms of their synthesis, mechanical properties, morphologies and in vitro cellular interactions on their surface and inside. In vivo bone regeneration of HA-PEO and Chitosan-PEO hydrogels was compared with in mouse model.

  • 현재, 미용식품으로 판매되고 있는 대부분의 제품은 피부를 구성하는 콜라겐과 hyaluronic acid 등이 배합되어 있으며, 이러한 것들은 피부에 윤기를 주는 것을 목적으로 한다. 그러나 이들 성분은 고분자물질이기 때문에 섭취시 고분자물 그 자체로 피부로 이행될 것으로는 생각되지 않는다. 일반적으로 콜라겐이나 hyaluronic acid는 소화효소에 의해 저분자의 아미노산이나 펩타이드, 당질로 분해되고 흡수되어 각 장기나 피부조직에서 콜라겐이나 hyaluronic acid로 재생된다고 알려져 있다. 피부조직에서의 재생은 진피 내의 피부선유아세포에서 일어난다. 따라서, 피부섬유아세포의 작용을 활성화시키는 것이 아름다운 피부를 가지는데 중요하다.

Basic rheology of dermal filler

  • Dermal injectables are the most popular material for facial enhancement, including volumizing and wrinkle correction. However, although hyaluronic acid is the most common component of dermal injectables, the character of hyaluronic acid products is quite variable. These materials can be described in terms of their viscoelastic properties using four parameters. In this article, the author would like to describe these properties in detail. Although many physicians consider this material to be convoluted, thoroughly understanding the characteristics of various injectables enables us to tailor procedures accordingly and to enhance the final results.

Hyaluronic Acid Filler Injection Guided by Doppler Ultrasound

  • Doppler ultrasound can be used to detect almost all arteries of the face before injecting the hyaluronic acid (HA) filler. The relatively more dangerous sites of filler injection are the glabellar wrinkle, forehead, temple, nose, and nasolabial fold area, and it is recommended to map the vasculature of these areas by Doppler ultrasound before performing filler injection. The Doppler ultrasound detection method is included as a video. Internal carotid arterial branches, the supratrochlear, supraorbital, and dorsal nasal arteries, and external carotid arterial branches, the superficial temporal and facial arteries, are very important arteries when injecting HA filler; thus, Doppler ultrasound detection is recommended.

  • Treatment of osteoarthritis of the ankle joint is similar to that of any other large joint and includes conservative and surgical treatments. Surgical option in severe osteoarthritis is joint fusion or joint replacement, whereas conservative treatment is limited and includes mainly ankle supports, physical therapy, and oral medication. Hyaluronic acid was discovered in 1934 and now has been widely used in the knee and shoulder joints. We reviewed the articles about an intra-articular hyaluronic acid injection in the treatment of osteoarthritis of the ankle joint.

  • To develop hyaluronic acid (HA)-based anticancer agent carrier, hyaluronic acid was chemically modified with the hydrophobic group of deoxycholic acid(DA). The physicochemical properties of the deoxycholic acid-conjugated HA (HADA) were investigated by using $^1H$ NMR, FTIR spectrophotometer and TEM. Paclitaxel (Tx)-loaded HADA nanoparticles were prepared by a dialysis method. The loading efficiency of drug and drug contents of Tx-loaded HADA nanoparticles (HADA-Tx) were measured by HPLC. The anticancer activity of HADA-Tx was investigated by its cytotoxicity against KB cell in vitro. The HADA-Tx was shown to have the superior potential for the anticancer drug delivery.

Anti Photoaging Effects of Hyaluronic Acid following Oral Administration in SKH-1 Hairless Mice (SKH-1 Hairless Mice에서의 히알루론산 경구 투여를 통한 피부 광노화 개선 효과)

  • Photoaging is the main extrinsic aging factor that is induced due to UVB. Many studies have revealed that application of hyaluronic acid to the skin is effective in healing photoaging. However, the effect of hyaluronic acid through oral administration is unclear. The aim of this research was to investigate the effect on skin photoaging after oral administration of hyaluronic acid. During a ten week study, hyaluronic acid was fed to hairless mice, which were illuminated by UV radiation. After ten weeks, wrinkle indicator and histological changes were determined. Compared with the control group, the sample group had a decrease in wrinkle depth, thickness, and number. Especially, the HA 160 mg/kg group had a similar value of wrinkle depth ($19.44{\pm}0.75{\mu}m$), number ($654.00{\pm}98.34$), and thickness ($1.35{\pm}0.08mm$), when compared with the RA (retinoic acid) group ($19.28{\pm}0.95{\mu}m$, $653.57{\pm}83.54$, and $1.34{\pm}0.07mm$ respectively). Also, the treated group showed improved elastosis and decreased collagen degradation resulting from UV irradiation. Thus, we can conclude that hyaluronic acid has a positive effect in improving skin photoaging induced by UV radiation. Hyaluronic acid therefore has the potential to be an ingredient in skin health function foods.
