소핵시험은 세포분열 단계 중 간기 세포의 세포질 내 소핵 유무를 조사함으로써 유전독성을 평가하는 시험법이다. 최근 화장품 안전성 평가에 동물실험을 금지하거나 최소화하려는 노력이 확산되고 있어 유전독성 평가에 있어서도 기존의 동물실험이 아닌 새로운 in vitro 시험법이 요구되고 있다. 본 연구에서는 3차원 배양인공피부모델인 KeraSkin$^{TM}$을 이용하여 도포 처치된 물질의 유전독성을 평가하였다. 2종의 유전독성물질인 mitomycin C (MMC)와 methyl methanesulfonate (MMS)는 농도 의존적으로 세포독성과 소핵 형성이 유도된 반면, 대조물질인 4-nitrophenol (4-NP)와 trichloroethylene (TCE)에서는 농도 의존적으로 세포독성은 관찰되었으나 소핵은 형성되지 않았다. 따라서 인공피부모델을 이용한 소핵시험이 화장품과 같은 피부적용물질의 in vitro 유전독성 평가에 유용할 것으로 사료된다.
Photosensitized peroxidation of membrane lipids has been implicated in skin pathologies such as phototoxicity and premature aging. We have previously reported that syriacusin compounds isolated from Hibiscus Syriacus inhibited lipid peroxidation. Here, we investigated whether syriacusins could be effective inhibitor to skin aging using ultraviolet-irradiated human dermal fibroblast cells (HDFCs). Syriacusins A, B, and C inhibit the activity of human neutrophil elastase (HNE), a serine protease to degrade extracellular matrix (ECM) proteins including elastin, with $IC_{50}s$ of 8.0, 5.2, and $6.1\;{\mu}M$, respectively. No changes in cell viability were detected by syriacusins A and B in UV-B ($10\;mJ/cm^2$) irradiated HDFCs. Matrix metallo-proteinase (MMP)-1 expression in HDFCs was increased by UV-B irradiation. MMP-1 expression in UV-B irradiated HDFCs was decreased by $10\;{\mu}M$ and $20\;{\mu}M$ syriacusin A to 50% and 20% of untreated control, respectively. Syriacusin B treated with $20\;{\mu}M$ reduced MMP-1 expression in UV-B irradiated HDFCs to 60% of untreated control. Syriacusin A also inhibited MMP-2 expression accompanying the increase of type-I pro-collagen in UV-B irradiated HDFCs. These results demonstrate that syriacusin A could be a more effective compound to inhibit skin aging caused by UV irradiation. It suggests that syriacusins A and B might be developed as possible agents to treat or prevent skin aging.

Skin aging is the most readily observable process involved in human aging. Ultraviolet B (UVB) radiation causes photo-oxidation via generation of reactive oxygen species (ROS), thereby damaging the nucleus and cytoplasm of skin cells and ultimately leading to cell death. Recent studies have shown that high levels of solar UVB irradiation induce the synthesis of matrix metalloproteinases (MMPs) in skin fibroblasts, causing photo-aging and tumor progression. The MMP family is involved in the breakdown of extracellular matrix in normal physiological processes such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes such as arthritis and metastasis. We investigated the effect of diphlorethohydroxycarmalol (DPHC) against damage induced by UVB radiation in human skin keratinocytes. In UVB-irradiated cells, DPHC significantly reduced expression of MMP mRNA and protein, as well as activation of MMPs. Furthermore, DPHC reduced phosphorylation of ERK and JNK, which act upstream of c-Fos and c-Jun, respectively; consequently, DPHC inhibited the expression of c-Fos and c-Jun, which are key components of activator protein-1 (AP-1, up-regulator of MMPs). Additionally, DPHC abolished the DNA-binding activity of AP-1, and thereby prevented AP-1-mediated transcriptional activation. These data demonstrate that by inactivating ERK and JNK, DPHC inhibits induction of MMPs triggered by UVB radiation.
국내의 주름개선 혹은 미백 효능을 평가하기 위한 인체시험 방법은 식약청 가이드라인에 근거하여 시행 되어왔으며, 인체시험에서 육안평가 및 기기평가 결과에 대해 시험군과 대조군 간의 효과를 비교하기 위해서 unpaired t-test를 주로 이용하였고, 시술 전후의 효과를 비교하기 위해서 paired t-test를 이용하였다. 설문평가 결과에 대해서는 빈도분석을 이용한 기술통계법이 이용되고 있다. 미국 및 유럽의 임상 평가기관에서도 이와 유사한 시험법 및 통계분석 방법을 이용하고 있다. 그러나 동일 개체에 대하여 처치를 반복 적용하여 얻은 자료는 서로 관련성이 높아 이를 감안한 분석법을 적용해야 한다. 따라서 본 연구에서는 화장품 분야에서는 처음으로 기능성 화장품 중 주름 개선 및 미백 효능 시험의 육안평가 및 기기평가 자료에 대해 repeated measures ANCOVA (RM ANCOVA)와 repeated measures ANOVA (RM ANOVA)를 적용하여 통계 방법의 타당성 여부를 검증함으로서 현재의 인체시험 방법에 적합한 새로운 통계분석 방법을 제시하였다.

In spite of frequent usage of primary human foreskin keratinocytes (HFKs) in the study of skin biology, senescence-induced block-age of in vitro proliferation has been a big hurdle for their effective utilization. In order to overcome this passage limitation, we first isolated ten HFK lines from circumcision patients and successfully immortalized four of them via a retroviral transduction of high-risk human papillomavirus (HPV) E6 and E7 oncogenes. We confirmed expression of a keratinocyte marker protein, keratin 14 and two viral oncoproteins in these immortalized HFKs. We also observed their robust responsiveness to various exogenous stimuli, which was evidenced by increased mRNA expression of epithelial differentiation markers and pro-inflammatory genes in response to three reactive chemicals. In addition, their applicability to cytotoxicity assessment turned out to be comparable to that of HaCaT cells. Finally, we confirmed their differentiation capacity by construction of well-stratified three dimensional skin cultures. These newly established immortalized HFKs will be valuable tools not only for generation of in vitro skin disease models but also for prediction of potential toxicities of various cosmetic chemicals.
A pressure sensor that mimics the sensing ability of human skin has emerged as high-profile technology because it shows remarkable applications in numerous fields such as robotics, human health monitoring, and artificial prosthetics. Whereas recent pressure sensors have achieved high sensitivity similar to that of human skin, they still show limited detection bandwidth. Moreover, once these e-skin are fabricated, their sensitivity and stiffness are fixed; therefore, they can be used for only limited applications. Our study proposes a new adaptive pressure sensor built with uniform gallium microdroplet-elastomer composite. Based on the phase transition of gallium microdroplets, the proposed sensor undergoes mode transformation, enabling it to have a higher sensitivity and wider detection bandwidth compared with those of human skin. In addition, we succeeded in extending a single adaptive pressure sensor to sensor arrays based on its high uniformity, reproducibility, and large-scale manufacturability. Finally, we designed an adaptive e-skin with the sensor array and demonstrated its applications on health monitoring tasks including blood pulse and body weight measurements.
Objectives : This study was designed to investigate the effects of alcoholic fermentation extract from ovary and rind of pear on human skin. Methods : We investigated the effects of samples on human skin in terms of clinical trial. 32 of healthy volunteers, who lived in mokpo participated in clinical trial. Effects of sample on hygroscopicity and water-holding capacity were investigated. Sebum, moisture, scabrousness and desquamation were also investigated. Facial massages were conducted 8 times in each volunteers. Results : Single treatment group showed elevated levels of hygroscopicity (Normal : 106.4$\pm$2.1%, vs Pear : 114.1$\pm$6.2) and water-holding capacity (Normal : 87.1$\pm$4.4%, vs Pear : 96.2$\pm$1.3) on forearm of 16 volunteers respectively. In addition, massage group using sample showed elevated moisture (Control : 3.8$\pm$5.8%, vs Sample : 15.2$\pm$4.7%) and lowered scabrousness levels (Control : -12.3$\pm$4.4%, vs Sample : -22.0$\pm$5.2%) significantly. However, the levels of sebum did not affected by sample. Conclusions : These result suggest that alcoholic fermentation extract from ovary and rind of pear has moisturizing effect on human skin.
The purpose of this study was to identify Chinese university women's appearance management characteristics and clothing purchasing behavior. A self-administered questionnaire was used. Research period was 15th - 30th March. Frequencies, percent, factor analysis, cluster analysis, ANOVA, chi-square test were done. The results of study were: 1. The factors of appearance management characteristics were food management, skin care, weight control & tooth correction, nail & hair care, and use of identity kits. The factors of clothing selection criteria were outlook & appropriateness, management & service, fashion & brand, and individuality & price, and factors of information resources were marketer oriented & neutral and consumer oriented & store. 2. Chinese university women were segmented into positive appearance management group, food & skin management group, nail care & identity kits using group, and appearance management stagnated group. Positive appearance management group selected their clothing to pursuit individuality & relaxation, fashion, and conformity, considered fashion or brands more, used various information sources. Food & skin management group purchased their clothing for conformity. Nail care & identity kits using group considered management & service of clothing more, utilized consumer oriented & store information more. 3. Positive appearance management group were younger, and spent more pocket money and for their appearances. Food & skin management group were older, but nail care & identity kits using group were older. Appearance stagnated group spent less pocket money and appearance management.

LB00013, a newly developed recombinant human Interferon $\beta$, was tested for primary eye and skin irritation in male New Zealand White rabbits. In the primary eye irritation test, 0.1 ml of a solution of LB00013 was instilled into the eye. In the rinsing group, the eye was washed with water at 30 seconds after instillation. No reaction was produced at the cornea, iris and conjunctivae by LB00013. In the primary skin irritation test, LB00013 was applied to the back of rabbits for 24 hours. Primary irritation index was "0" in test and control sites of all animals. Thus LB00013 was evaluated as a non-irritant on the basis of the criteria of Draize et al. (1944).l. (1944).
This study was performed to investigate the effects of food habits and life style on the skin health of young Korean females in their late teens and twenties. The study focused on facial skin and used a self-administered questionnaire survey, an examination of skin type, and pH tests. More than half of the females had combination skin type or pH level higher than 5.8 which is regarded as normal. The variables that turned out to significantly (p<0.05) effect their facial skin types were body mass index (BMI), the type of residence, the amount of water intake, whether or not they intake drinks, the frequency and amount of alcohol drinking, favorite foods as side dishes at meals, intake frequency of fruit, the dairy, seaweed, and fast food, the frequency of washing their faces per day, the method of how they used water during face washing, whether or not they had professional skin care, and whether or not they wore color make up. The variables of the females' age, BMI, the amount of alcohol drinking, the frequency and duration of exercise, the controlling methods of stress, their favorite foods as side dishes at meals, and the intake frequency of soy products or animal foods significantly affected their facial pH levels (p<0.05). The results of this study indicate that some food habits and life styles are significantly related with facial skin health. More importantly BMI and favorite foods as side dishes during daily food intake could have remarkable effects on facial skin health.
