Chemopreventive Effect of Protein Extract of Asterina pectinifera in HT-29 Human Colon Adenocarcinoma Cells

  • We investigated the effect of protein extract of Asterina pectinifera on the activity of 4 enzymes that may playa role in adenocarcinoma of the colon: quinone reductase (QR), glutathione Stransferase (GST), ornithine decarboxylase (ODC), and cyclooxygenase (COX)-2. QR and GST activity increased in HT-29 human colon adenocarcinoma cells increased that had been exposed to 4 concentrations of the protein extract (80, 160, 200, and $240{\mu}g/mL$). Additionally, 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced ODC activity decreased significantly in cells exposed to the extract in concentrations of $160{\mu}g/mL$ (p<0.05), $200{\mu}g/mL$ (p<0.005), and $240{\mu}g/mL$ (p<0.005). TPA-induced COX-2 activity also decreased in cells exposed to extract concentrations of 10, 20, 40, and $60{\mu}g/mL$. COX-2 expression was also inhibited in cells exposed to this extract. These results suggest that this protein extract of A pectinifera has chemopreventive activity in HT-29 human colon adenocarcinoma cells, and therefore, may have the potential to function as a chemopreventive agent in human colorectal cancer.

Probiotics Inhibit Lipopolysaccharide-Induced Interleukin-8 Secretion from Intestinal Epithelial Cells

  • It has been suggested that probiotics could be useful for the prevention of symptomatic relapse in patients with inflammatory bowel disease (IBD). Interleukin (IL)-8 has been well recognized as one of the pro-inflammatory cytokines that could trigger inflammation and epithelial barrier dysfunction. In this study, the anti-inflammatory effects of probiotics were investigated using a human epithelial cell line (HT-29). Probiotics from infant feces and kimchi were tested for their cytotoxicity and effects on adhesion to epithelial cells. The present results show that seven strains could form 70 % adhesion on HT-29. The probiotics used in this study did not affect HT-29 cell viability. To screen anti-inflammatory lactic acid bacteria, HT-29 cells were pretreated with live and heat-killed probiotics, and lipopolysaccharide (LPS) ($1{\mu}g/mL$) was then added to stimulate the cells. The cell culture supernatant was then used to measure IL-8 secretion by ELISA, and the cell pellet was used to determine IL-8 and toll-like receptor (TLR-4) mRNA expression levels by RT-PCR. Some probiotics (KJP421, KDK411, SRK414, E4191, KY21, and KY210) exhibited anti-inflammatory effects through the repression of IL-8 secretion from HT-29 cells. In particular, Lactobacillus salivarius E4191, originating from Egyptian infant feces, not only decreased IL-8 mRNA expression, but also decreased TLR-4 expression. These results indicate that Lactobacillus salivarius E4191 may have a protective effect in intestinal epithelial cells.

붉은 양파 분말의 화학성분 및 생리활성 (Chemical Components and Biological Activities of Red Onion Powder)

  • We investigated the chemical components of red onion powder dried using the low temperature vacuum method and the inhibitory effects of solvent extracts of the dried red onion powder on the growth of HT-1080 human fibrosarcoma and HT-29 human colon cancer cells and $H_2O_2$-induced oxidative stress. The moisture content of the dried red onion powder was 17.95%, while the vitamin C content was 96 mg/100 g and the total phenols content was 39.1 mg/mL. The inhibitory effects of acetone with methylene chloride (A+M) and methanol (MeOH) extracts of the red onion powder on the growth of HT-1080 and HT-29 cancer cells increased in a dose dependent manner (p<0.05). The inhibitory effect was greater on the growth of HT-29 cells, while the A+M extracts had a higher inhibitory effect than the MeOH extracts. Treatment with the hexane, 85% aq. methanol, butanol and water fractions of the extract led to significant inhibition of the growth of both cancer cell lines (p<0.05). Among the fractions, the hexane and 85% aq. methanol fractions showed a greater inhibitory effect. To determine the protective effect on $H_2O_2$-induced oxidative stress, a DCFH-DA (dichlorodihydrofluorescin diacetate) assay was conducted. All fractions, including the crude extracts of dried red onion, appeared to lead to a significant reduction in the levels of intracellular reactive oxygen species (ROS), and these reductions occurred in a dose dependent fashion (p<0.05). Among the fractions, the 85% methanol fraction showed the greatest protective effect on the production of lipid peroxides.

금속단백분해효소의 활성 저해를 통한 삼귀고의탕의 전이억제 효과 (Anti-metastatic Effect of Samguikoeui-Tang Via Inhibition of Matrix Metalloproteinases Activities)

  • This study was performed to examine the anti-metastatic effect of ethanol extract of Samguikoeui-Tang (SGKE), a formula consisting of four oriental herbs, in highly-metastatic HT1080 human fibrosarcoma cells. SGKE significantly inhibited the adhesion of HT1080 cells to matrigel at nontoxic concentrations in a dose-dependent manner, while it did not exert cytotoxicity against HT1080 cells up to the concentration of 100 ${\mu}g$/ml. Also, SGKE depressed the activity of matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) by gelatin zymography. However, SGKE did not affect the mRNA expression of MMP-2 and TIMP-2, an inhibitor of MMP-2, by RT-PCR analysis. In addition, the effect of SGKE on HT1080 cell invasion was determined using Boyden chamber assay. SGKE suppressed the invasion of HT1080 cells in a dose-dependent manner. Taken together, these results suggest that SGKE has an anti-metastatic effect via inhibition of MMP-2 and -9 activities.

  • Purpose: The purpose of this study was to investigate the effect of electroacupuncture on pain threshold and 5-HT synthesis in raphe magnus nucleus of dog brain stem following nociception induced by 1% carageenan. Method: Twenty male dogs were randomly divided into two groups, a control group and an electroacupuncture group with the following parameters: ST36, LI11 and SP9 with 120Hz and 0.5mA. The dogs were sacrificed at 30, 60, 90, 120, 150 and 180 min after nociception using 1% carageenan injection into the plantar side of the dog's left leg. The pain threshold was recorded by an Analgesia-meter. After making brain slice sections, the tissues were immunostained with 5-HT antisera (1:2,500). Results: The pain threshold for the electroacupuncture group was higher than for the control group. The number of 5-HT immunoreactive neurons in the electroacupuncture group was greater than that of the control group. Differences between the two groups regarding the number of 5-HT immunoreactive neurons at 30 min and 60 min (p<0.05), at 120 min and 150 min (p<0.01), and at 8 days (p<0.05) were significant. Conclusion: The increases in pain threshold and number of 5-HT immunoreactive neurons in the raphe magnus nucleus of dog brain stem showed that electroacupuncture can attenuate pain transduction through the descending tract.

오미자 열수추출물의 대장암세포 증식억제 효과 (Effects on Hot Water Extract of Schizandra chinensis on Colon Cancer)

  • 본 실험에서는 대장암 세포 HT-29의 증식을 억제하고 세포 사멸을 유도하는 천연소재 발굴을 목적으로, 오미자(Schizandra chinensis Baillon) 열수 추출물을 이용하여 인체대장암 세포 HT-29의증식에 미치는 영향을 확인하였다. 오미자 열수 추출물이 HT-29 대장암 세포의 apoptosis 유도 효과 및 기전에 미치는 영향을 분자생물학적 방법으로 실험하여 다음과 같은 결론을 얻었다. MTT assay를 통해 인체 대장암세포 HT-29는 오미자 시료농도 0, 1.0, 2.0, 4.0 mg/mL에서 암세포 사멸농도가 각각 0%, 10%, 70%, 88%를 나타내었다. 대장암세포에 오미자 추출물을 처리하고 cell cycle 분석 결과, 시료농도 의존적으로 sub-G1기가 증가하였고, G0/G1기는 감소되는 것을 통해, apoptosis가 일어나 세포 증식을 저해하는 것으로 확인되었다. 대장암세포 핵의 형태학적 변화를 보면, 오미자 추출물 처리 시 농도 의존적으로 세포수가 감소되는 것이 뚜렷이 관찰되었고 cell shrinking, chromatin condensation등 apototic body 등과 같은 형태학적 변화들이 뚜렷하게 관찰되었다. RT- PCR을 통한 유전자 발현은, 오미자 추출물 농도 의존적으로 p53 유전자 발현이 증가되는 것을 통해 대장암세포의 증식억제를 확인할 수 있었다. In vitro 실험에서 오미자 열수추출물이 대장암세포의 성장을 저해하는 효과가 있음을 확인하였으며, 오미자 추출물에 함유된 활성 본체 규명 및 apoptosis를 유도하는 작용기작에 관한 연구가 수행중이다.

Pharmacological Properties of CDBT in Hypoxia-induced Neuronal Cell Injury and Their Underlying Mechanisms

  • Objectives: This study aimed to reveal the pharmacological properties of the newly prescribed herbal mixture, Chenmadansamgamibokhap-tang(CDBT), against hypoxia-induced neuronal cell injury (especially mouse hippocampal neuronal cell line, HT-22 cells) and their corresponding mechanisms. Methods: A cell-based in vitro experiment, in which a hypoxia condition induced neuronal cell death, was performed. Various concentrations of the CDBT were pre-treated to the HT-22 cells for 4 h before 18 h in the hypoxia chamber. The glial cell BV-2 cells were stimulated with $IFN{\gamma}$ and LSP to produce inflammatory cytokines and reactive oxygen species. When the neuronal HT-22 cells were treated with this culture solution, the drug efficacy against neuronal cell death was examined. Results: CDBT showed cytotoxicity in the normal condition of HT-22 cells at a dose of $125{\mu}g/mL$ and showed a protective effect against hypoxia-induced neuronal cell death at a dose of $31.3{\mu}g/mL$. CDBT prevented hypoxia-induced neuronal cell death in a dose-dependent manner in the HT-22 cells by regulating $HIF1{\alpha}$ and cell death signaling. CDBT prevented neuronal cell death signals and DNA fragmentation due to the hypoxia condition. CDBT significantly reduced cellular oxidation, cell death signals, and caspase-3 activities due to microglial cell activations. Moreover, CDBT significantly ameliorated LPS-induced BV-2 cell activation and evoked cellular oxidation through the recovery of redox homeostasis. Conclusions: CDBT cam be considered as a vital therapeutic agent against neuronal cell deaths. Further studies are required to reveal the other functions of CDBT in vivo or in the clinical field.

겨울과 봄철의 CO2 시비 토마토 온실에서 온도에 따른 CO2 농도 구명 (Determination of Carbon Dioxide Concentration in CO2 Supplemental Greenhouse for Tomato Cultivation during Winter and Spring Seasons)

  • 본 연구는 겨울과 봄철의 CO2 시비 토마토 온실에서 낮과 밤의 온도 변화에 따른 CO2 농도 변화를 구명하고 계산된 CO2 공급량을 비교하고자 수행하였다. 온실 내 CO2 농도는 2022년과 2023년의 겨울철 실험 온실에서 생육단계가 LAI 2.0인 대표적인 날들 중 누적광량 차이로 인한 평균 온도가 다른 날을 선정하여 분석하였다. 평균 온도 18℃ 이상의 경우(고온, HT), CO2 공급 시간대(10:00-13:00) 이후 13, 14, 15, 16시에 각각 31.7, 26.8, 23.8, 22.4℃일 때 CO2 농도는 459, 299, 275, 239µmol·mol-1이었고 평균 온도 18℃ 이하의 경우(저온, LT), 22.0, 18.9, 15.0, 13.7℃일 때 각각 500, 368, 366, 364µmol·mol-1으로 HT에서 CO2 감소량이 LT보다 유의적으로 높았다. 야간 CO2 농도는 HT의 경우 오후 6시(346µmol·mol-1) 부터 서서히 증가되어 오전 3시(454µmol·mol-1)에 가장 높았으며 9시간 동안 약 100µmol·mol-1이 증가되어 시간당 약 11µmol·mol-1이 증가되었고(토마토 240개체, 엽면적 330m2), LT에서는 그 증가폭이 낮았다. 봄철 CO2 시비 온실에서 CO2 시비 개시 직전(오전 7:30)에 측정된 CO2 농도는 야간온도가 15℃ 이상의 경우(HT)는 15℃ 이하의 경우(LT)보다 3-4배 높은 것으로 나타났으며 그날의 계산된 공급 CO2 시비량도 HT에서 적었다. 본 연구결과에서 야간 CO2 농도는 재배 시기에 따른 온도의 영향을 받으며, 야간에 증가된 CO2는 광합성 작용의 주요 공급원이 된다는 것을 알 수 있었다. 이를 고려한 CO2 공급 모델식 개발이 필요한 것으로 판단된다.

  • 6-Nitroquipazine has higher binding affinity for SERT than other selective serotonin reuptake inhibitors. We have prepared 6-nitroquipazine based rhenium complexes which would lead to the development of potential SPECT imaging agents with $^{99m}Tc$ for 5-HT transporter.
