헛개나무 열수 추출물의 알코올 분해 및 간 보호 효과

  • The water extracts of Hovenia dulcis Thunb. showed significant hepatoptotective activity against D-galactosamine/LPS and $CCl_4-toxicity$. The extracts significantly inhibited the elevation of serum ALT, AST and LDH levels. The concentration of blood alcohol were decreased from 2hr after injected in rats with Hovenia dulcis extracts.

Inhibitory Activity on Angiotensin Converting Enzyme and Antioxidant Activity of Hovenia dulcis Thunb. Cortex Extract (헛개나무의 Angiotensin 전환 효소 저해 및 항산화 활성)

  • To develop a new functional materials, angiotensin converting enzyme (ACE) inhibitory activity, antioxidant effect and total phenolic content of Hovenia dulcis Thunb. cortex were evaluated. Methanol and water extract of H. dulcis inhibited ACE by 81% and 76%, respectively, at the concentration of $4,000\;{\mu}g\;m{\ell}^{-1}$ which were similar level with that (85%) of commercial peptide-type ACE inhibitor. Superoxide radical scavenging activity of two extracts $(99.5%{\sim}99.9%)$ were stronger than that (69%) of ascorbic acid at the final concentration of $200\;{\mu}g\;m{\ell}^{-1}$. Among the solvent fractions, ether and ethylacetate fraction showed also potent scavenging activities (91% and 85%) for superoxide radical. Inhibitory activities of two extracts on oxidation of human low density lipoprotein (LDL) which were similar with that of ${\alpha}-tocopherol$, were higher than 80% at the concentration of $50\;{\mu}g\;m{\ell}^{-1}$. Total phenol contents of methanol and water extracts were 7.2% and 3.6%, respectively, and that of ethylacetate showed the highest value as 60.8% among the solvent fractions. Therefore, it has been suggested that H. dulcis cortex could be a effective anti-hypertention and antioxidant resource to develope a new functional material.

Laxative Activity of the Hot-Water Extract Mixture of Hovenia dulcis Thunb. and Phyllostachys pubescens Mazel in Chronic Constipation Model SD Rats

  • This study examined the laxative effects of hot-water extracts of Hovenia dulcis Thunb. (HD), Phyllostachys pubescens Mazel (PM), and a 2:8 mixture of both (HP) in two chronic constipation models. For the loperamide-induced constipation model, animals were divided into an untreated group, negative control group (loperamide 4 mg/kg), positive control group (bisacodyl 4 mg/kg) group, and six treatment groups (HP 100 or 400, HD 50 or 100, and PM 100 or 400 mg/kg). For the low-fiber diet-induced constipation model, animals were divided into an untreated group (normal diet), negative control group (low-fiber diet), positive control group (Agio granule, 620 mg/kg), and the same treatment groups. Fecal number, weight, fecal water content, and intestinal transit ratio were higher in the groups treated with HP, HD, and PM than in the groups treated with loperamide or low-fiber diet. Thickness of colon mucosa and muscle layers were increased in the treated groups. Colon tension increased in the HP groups, and [Ca2+]i measurements using fura-2 as an indicator showed that HP inhibits ATP-mediated Ca2+ influx in IEC-18 cells. These results showed that the HP mixture has laxative activity by increased mucin secretion and inducing contractile activity and relaxation. It may be a useful therapeutic strategy for ameliorating in chronic constipation.

Effect of Amur Long-tailed Goral on the Germination of Hovenia dulcis Thunb. (산양이 헛개나무 종자의 발아에 미치는 영향)

  • Amur's long-tailed goral(Naemorhedus Caudatus) winter scats inhabitants of Wolaksan National Park were surveyed between 2006 and 2009. The scats included seeds of Hovenia dulcis and we confirmed germination of seed from the scats in the spring. 600 dulcis seeds goral scats were collected in the same location and period and 600 normal seeds were also observed in order to study the effect of goral on germination of dulcis. All of the seed(totally 1,200) were cultured in the same condition. As a result, germination rates of seeds from the goral scats and normal seeds were 32.5% and 0.8% respectively. So we confirmed that dulcis seeds in goral habitats germinate approximately 40 times more than without goral.

The Effects of Hovenia dulcis Fruit Hot Water Extracts on Anti-fatigue and Improvement of the Exercise Performance in SD Rats (랫드를 이용한 헛개나무과병 열수추출물 투여가 운동에 의한 피로회복과 운동수행능력에 미치는 기전 규명)

  • The present study investigated the effects of Hovenia Dulcis (HD) fruit extract powder on the improvement of physical activity, especially exercise capacity. Forty mice were divided into 4 groups including normal controls, negative controls, 100 (HD-100) and 200 (HD-200) mg/kg HD fruit extract powder groups for 5-times exercises using treadmill. Normal control did not performed treadmill running but others did 5-times for 10 days. HD fruit extract powders were administrated orally one-times per day for 10 days before treadmill exercise and normal and negative controls were fed with excipient water. After 5-times exercise, blood biochemical analysis showed that aspartate aminotransferase (AST), lactate dehydrogenase (LDH) and creatine phosphokinase (CK) activities and blood lactate concentration were statistically increased in negative controls than in normal controls. They were decreased in HD fruit extract powder groups, compared with negative controls. These results were considered as the effects of HD fruit extract powders on reduced tissue damages during exercise. Other measured indices did not reveal remarkable differences. All together, these results suggest that HD fruit extract powders may enhance the exercise performance by recovering the exercise-fatigue via blood lactate concentration by reducing blood LDH activity and via reduction of blood CK and AST activity.

Hotwater Extract of Hovenia dulcis Peduncle Improves Exercise Performance and Anti-fatigue Effect in Mice (헛개나무 열매 열수추출물 투여에 의한 생쥐의 지구력 운동 수행 능력 향상 및 피로개선 효과)

  • The aim of this study was to evaluate the effect of hot water extract of peduncle obtained from Hovenia dulcis Thunb (HD) which is commercially developed for the protective effect on the alcoholic hepatotoxicity, on the endurance capacity for weight loaded forced swimming mice. The swimming times to exhaustion in mice fed 100 and 200 mg/kg HD for 2 weeks were prolonged 3.6 and 3.7 fold, and for 4 weeks 1.9 and 2.7 fold compared with each vehicle control ($42.8{\pm}20.5$ min and $67.7{\pm}47.8$ min, for 2 and 4 weeks), respectively. Blood biochemical parameters for ALT, AST, creatinine and BUN were not significantly different between from HD fed or control mice. Although HD fed mice swam over 2 fold longer time than vehicle control mice at 4 weeks, blood biomarkers of physical fatigue such as glucose, triglyceride and free fatty acid, lactate were not significant different and even tended to ameliorate. Hepatic lipid peroxidation and SOD activity did not significantly change in HD fed- and vehicle control exhausted swimming mice at 2 or 4 weeks. However, catalase activity in HD-fed mice was significantly increased in a dose-dependent manner compared with vehicle control mice. The present study indicates that HD improved physical fatigue and exercise performance in mice. Therefore, it has a potential for the pharmacological effect of anti-fatigue.

Neuroprotective and Free Radical Scavenging Activities of Phenolic Compounds from Hovenia dulcis

  • The EtOAc-soluble fraction from a methanolic extract of Hovenia dulcis Thunb. exhibited neuroprotective activity against glutamate-induced neurotoxicity in mouse hippocampal HT22 cells. The neuroprotective activity-guided isolation resulted in 8 phenolic compounds (1-8), such as vanillic acid (1), ferulic acid (2), 3,5-dihydroxystilbene (3), (+)-aromadendrin (4), methyl vanillate (5), (-)-catechin (6), 2,3,4-trihydrobenzoic acid (7), and (+)-afzelechin (8). Among these, compounds 6 and 8 had a neuroprotective effect on the glutamate-induced neurotoxicity in HT22 cells. Furthermore, compound 6 had a DPPH free radical scavenging effect with an $IC_{50}$ value of $57.7{\mu}M$, and a superoxide anion radical scavenging effect with an $IC_{50}$ value of $8.0{\mu}M$. Both compounds 6 and 8 had ABTS cation radical scavenging effects with $IC_{50}$ values of $7.8{\mu}M\;and\;23.7${\mu}M$, respectively. These results suggest that compounds 6 and 8 could be neuroprotectants owing to their free radical scavenging activities.

Comparison of Hepatic Detoxification activity and reducing Serum Alcohol concentration of Hovenia dulsis $T_{HUNB}$ and Alnus japonica Steud. (헛개나무와 오리나무 추출물의 간 해독작용 및 체내 알콜 분해능 비교)

  • It was found that the level of alcohol concentration in both mouse and human sera can be significantly decreased up to 42% by oral administration of the mixtures of the extracts of Hovenia dulcis THUNB and Alnus japonica Steud. A single treatment of extract from Hovenia dulcis reduced the serum alcohol concentration to 32%, compared to 13% in treating the extract of Alnus japonica. Similar patterns were observed in enhancing alcohol dehydrogenase (ADH) and glutathion-S-transferase (GST) activity in the liver. The inhibition of cathepsin activity was also greatly reduced by administrating the mixture of both extracts : however, the extract of Alnus japonica did not affect the acitivity of cathepsin. It was concluded that the mixture of both extracts had synergic effect on reducing serum alcohol concentration and improving the detoxification process due to alcohol administration in the liver.

Callus formation and multiple shoot induction of Hovenia dulcis Thunb. (헛개나무의 캘러스 형성 및 multiple shoot 유기)

  • Loaves, stems, cotyledons, and roots of Hovenia dulcis Thunb grown in test tube were cultured on media containing different concentrations of single or combined growth regulators. In MS media containing 2mg/ι BA, the shoot formation rate was 95.5% and it was the highest frequency of shoot formation. MS media showed most efficiency in the shoot formation at 0.01mg/ι TDZ for the callus formation, but the color of callus changed to brown at a higher concentration of TDZ. Callus formation was 89.% at 0.5mg/ 2.4-D, but IAA, IBA, and NAA were not effective on the formation of callus. Calli were formed only on wound area when IAA, IBA, and NAA were added into MS media. Combined growth regulators (BA + auxin) were more effective in roots and nodes than leaves and cotyledons on the formation of shoot. More than 97% of shoot formation was obtained on MS media containing BA and auxin. For the production of multiple shoot, nodes of Hovenia dulcis were used and effect of growth regulators on the formation of multiple shoot was evaluated on MS media. Highest shoots (5.3) of Hovenia dulcis were induced on MS media supplied with 0.1mg/ι BA and 0.1mg/ι NAA, and an average of 6.4 shoots per explant were obtained in 1/2 MS media containing same concentration and growth regulators. An average of 7 shoots per explant after 4 weeks of culture from nodes of Hovenia dulcis was produced on a woody plant medium(WPM) containing 0.1mg/ι BA and 0.1mg/ι NAA. Shoot length was 6.0 cm in average.
