The biochemical components of Namcheondlggae, Miryangdlkkae 25, Boradlggae and Ipdlkkae 1 were measured. The samples were extracted with hot water, 60% acetone or 80% ethanol for screening physiological activity. The crude protein content (4.36%) was found in the Miryangdlkkae 25 and calcium content (497.5 mg%) was found in the Namcheondlggae among the tested 4 perilla loaves. Fructose was 30.86 mg% in the Namcheondlggae and free amino acids at all perilla leaves was detected seventeen. In Boradlggae, glutamic acid and alanine were 25.37 and 11.91 mg%. Totally nine non-volatile organic acids were also detected and the contents of malic acid and glutaric acid were 28.34 and 14.57 mg% in Boradlggae. The Miryangdlkkae 25 had the highest vitamin C amount which was 113.24 mg%. Angiotensin converting enzyme (ACE) inhibition activity of 60% acetone extract of Miryangdlkkae 25 was 39.20% when added as addition of 200 ppm level and xanthine oxidase inhibition activity of 80% ethanol extract of Boradlggae was 46.71%. Electron denoting activity of 60% acetone extract from Namcheohndlggae was the strongest inhibition activity as 98.19% when 200 ppm level of the sample extracts were added.
For screening antimutagenic effects, the effects of 95 medicinal plants on the mutagenicity of aflatoxin $B_1$$(AFB_1)$ and benzo(a)pyrene [B(a)P] were investigated using the SOS chromotest with Escherichia coli PQ37. The mutagenicity induced by $AFB_1$ or B(a)P was reduced over 26% by 2 kinds and 8 kinds of medicinal plant, respectively. Eight plants (Bupleurum falcatum, Corydalis ternata, Gasfrodia elata, Ostericum koreanum, Pinellia ternatia, Poncirus trifoliata, Prunus armeniaca and Rehmannia glutinosa) were also shown to have inhibitory effects on both $AFB_1$ and B(a)P. The mutagenicity induced by $AFB_1$ or B(a)P was increased over 20% by 46 kinds and 2 kinds, respectively, and 8 medicinal plants (Chrysanthemum indicum, Cinnamomum cassia, Cyperus rotundus, Morus bombycis, Patrinia scabiosaefolia, Petasites japonicus, Polygonum multiflorium, Thyja orientalis) increased significantly the mutagenicity of both mutagens. However the 8 plants themself did not show the mutagenicity in SOS Chromotest with S-9 mix alone. This result suggests that the above 8 plants may have the co-mutagenic activities. In two bacterial mutation system, SOS Chromotest and Ames test, the mutagenic or antimutagenic activities of some medicinal plants wire similar except Ostricum koreanum, Eugenia caryophyllata and Scutellaria baicalensis.
This study was performed to investigate the ameliorating effect of a hangover beverage mixture (SBJ) that contains Dendropanax morbifera Lev. and several medicinal plant extracts, on hepatoprotection and alcohol-metabolizing enzymes in alcohol-induced hangover in both in vitro and in vivo models. In human hepatoma cell line, HepG2, 300 mM of ethanol-induced hepatotoxicity was significantly improved by pretreatment of SBJ by dose-dependent manner. In the in vivo study, administration of alcohol to rats raised to the concentration of blood alcohol and lactate dehydrogenase (LDH). Blood alcohol and LDH levels in SBJ-treated rats significantly decreased at 0.5 h and 8 h after acute ethanol administration (40%, 4.6 g/kg body weight) as compared to alcohol-treated rats. Hepatic alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) activity were significantly higher in SBJ-treated rats than in alcohol-treated rats. SBJ supplementation reduced formation of malondialdehyde (MDA), and inhibited reductions of hepatic superoxide dismutase (SOD), hepatic glutathione (GSH), glutathione-S-transferase (GST), glutathione reductase (GR) and glutathione peroxidase (GPx) levels, compared with rats administered alcohol. Plasma catalase (CAT), aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels showed unaltered resulted in all experimental groups compared with the control group. These results suggest that SBJ exhibit hepatoprotective properties by enhancing ADH, ALDH activity and stimulating the antioxidant defense system in alcohol-induced hangover.
To develop natural intermediate flavoring substances, optimal hydrolysis conditions and taste compounds for two step enzyme hydrolysate(TSEH) using Grass puffer(Takifugu niphobles) were investigated. The optimal conditions for TSEH method were revealed in temperature at $55^{\circ}C$ 3 hour digestion with Alcalase (pH 7.5) at the 1st step and 2 hour at $45^{\circ}C$ digestion with Flavourzyme(pH 6.0~6.5) at the 2nd step. TSEH method was superior to hot-water extraction on the aspect of yield, nitrogen contents and organoleptic taste quality such as umami and control of bitter taste formation. In taste active-components in Glass puffer TSEH, total free amino acid content was 4,502 mg%, major free amino acids were Pro, Leu, Lys, Hypro, Tau, Arg, Phe, Ala, Glu and Val in ordor. As for nucleotides, IMP(372 mg%) was the principal component and also contents of TMAO, total creatinine, and betaine were 43, 278 and 41 mg% in Glass puffer TSEH, respectively. The major inorganic ions in TSEH were Na(949 mg%), K(222 mg%), Cl(1,180 mg%) and $PO_4$(1,081 mg%).
The leaves of Selaginella tamariscina were used for the treatment of many diseases in traditional medicine. In the study, antioxidant, anti-inflammatory, and wound healing activities of the hot-water extract(STW) and 80%ethanol extract(STE) obtained from S. tamariscina were evaluated. As a result, the polyphenol content of STW and STE were 38.108±0.766 mg/g and 17.927±1.064 mg/g, respectively. The DPPH and ABTS radical scavenging activities with the IC50 values of the STW were over 2 times lower than that of the STE. In the MTT assay, RAW264.7 cell viability of two extracts was decreased by about 6% at 1 mg/mL, whereas for HaCaT cell viability increased by 18% at 50 ㎍/mL. In addition, STW and STE suppressed the production of nitric oxide(NO), Tumor-necrosis(TNF)-. COX-2 and PGE2 in lipopolysaccharide(LPS) induced RAW264.7 cells. Furthermore, the STE showed wound healing effect through the promotion of skin cell migration in TNF- stimulated human keratinocytes. These results indicated that the STW and STE have the potential to be used as a new cosmetic active ingredients in skin care.
The leave of Polygonum tinctorium (LPT) have been used for centuries as a traditional medicine and as a food ingredient and natural dye. The aim of the current study was to develop high-value added products using LPT. Hot water extract (HWE) and ethanol extract (EE) of LPT were prepared, respectively, and their bioactivity was evaluated. The extraction ratio for the HWE was 27.6%, which was two-fold higher than that of the EE. The contents of total polyphenol in the HWE and total sugar in the EE were 51.2 mg/g and 297.8 mg/g, respectively. The total flavonoid and reducing sugar contents were similar in the extracts, irrespective of the extraction solvent. The HWE did not show antimicrobial activity in a disc-diffusion assay, but the EE showed strong growth inhibition against gram-positive bacteria. The EE exhibited stronger DPPH and ABTS radical scavenging activities and reducing power than those of the HWE. The HWE was particularly effective as a scavenger of nitrite ($RC_{50}$ of $6.0{\mu}g/ml$). In an antithrombosis activity assay, the EE showed significant anticoagulation activity as determined by an extended blood coagulation time (thrombin time, prothrombin time, and activated partial thromboplastin time), in addition to platelet aggregation activity. The HWE also showed platelet aggregation inhibitory activity. This report provides the first evidence of antithrombosis activities of LPT. Our results suggest that LPT has potential as a new antioxidant and antithrombosis agent.

In this study, antioxidant efficacy was analyzed to verify the value of use as an antioxidant-related functional cosmetic material using Cheongtaejeon tea extract. Cheongtaejeon tea was extracted, concentrated, and freeze-dried with hot water and 70% ethanol as a solvent to prepare samples, and then ABTS+ radical scavenging activity, electron donating ability, SOD-like ability, reducing power, ferric reducing antioxidant power (FRAP), total phenolic, and flavonoid contents were analyzed. In the ABTS+ radical scavenging activity experiment, both the Cheongtaejeon hydrothermal extract (CTW) and the Cheongtaejeon 70% ethanol extract (CTE) showed erasure activity of more than 98% at a concentration of 1,000 ㎍/mL, and in the electron donating analysis experiment, CTW and CTE, respectively, 42.20% and 78.82%. As a result of SOD-like activity measurement, activity of 39.73% and 67.39% of CTW and CTE, respectively, was confirmed. In the case of FRAP and reducing power experiments, both CTW and CTE showed high effects, and as a result of analyzing the total phenol and flavonoid contents, both CTW and CTE showed high contents. Based on the results of the experiment, the Cheongtaejeon tea extract is expected to have a highly valuable as a functional cosmetics material related to antioxidants.
This study was conducted to investigate the effect of addition levels of coffee and green tea by products extract including polyphenols through hot water extraction on rumen fermentation. The treatment groups consisted of coffee extract (CO), green tea extract (GR) and mixed extract (MIX), and the addition level was 10 µL, 20 µL and 30 µL of three levels. The experiment consisted of a total of 10 experimental groups including the control group, and a full factorial design was used. The effect of polyphenol addition in coffee and green tea by-products was analyzed through main and interaction effect of statistical analysis. The total polyphenol content of the extracts was 106.15, 79.10 and 185.25 ㎍ GAE/g DM for coffee by-product, green tea by-product and mixture, respectively. Total gas production was significantly lower in the treatment groups than in the control (114.00 mL/gDM) (p<0.05). Methane emission tended to decrease as the polyphenol addition level increased. Moreover, the MIX showed the lowest methane emission when 30 µL was added (p<0.05). Volatile fatty acids showed a significant difference compared to the treatment group as a control (98.06 mM) (p<0.05), but there was no change according to the level of polyphenols. As a result of the main effect and interaction, it is thought that the effect on methane reduction and improvement of rumen fermentation in MIX20 can be expected. In a series of studies, the addition of 20 µL of a blended extract of coffee and green tea by-products is thought to reduce methane to levels that do not inhibit rumen fermentation.
The purpose of this study was to verify the antifungal properties of various herbal medicines belonging to the Ranunculaceae family and to find an extraction method effective in inhibiting fungal growth. When antifungal activity was measured in a liquid medium with extracts obtained by either hot water extraction or organic solvent extraction of the herbal medicines Clematis apiifolia, Coptis chinensis, and Pusatilla chinensis, effective results were obtained from the chloroform extract. In addition, fungal growth inhibition experiments were performed on unicellular fungi, Candida albicans, Candida tropicalis, and Candida lusitaniae, and on filamentous fungi, such as Pythium ultimum, Aspergillus fumigatus, and Fusarium oxysporum, using disk diffusion experiments on solid media. It was confirmed that P. chinensis extract has excellent antifungal properties against Candida spp. and C. apiifolia extract against filamentous mold. Finally, GC-MS analysis was performed to explore the useful antifungal substances present in the extract. As a result of the study, thurbergenone from C. apiifolia and 16-hydroxycleroda-3, 13(14)-dien-15, 16-olide (16-HCDO) from C. chinensis were confirmed as antifungal candidates. In conclusion, it was confirmed that C. apiifolia, C. chinensis, and P. chinensis have antifungal activity against various fungi, and in GC-MS analysis, all herbal medicines were confirmed to have different antifungal candidates. These results indicate that the Ranunculaceae family has evolved in several directions for fungal resistance traits.
The extraction of polyphenol and flavonoid from citrus peel was performed by the ethanol, sugar, hot water (80$^{\circ}C$), and subcritical water extraction methods. The maximum yields of total polyphenolic compounds (27.25${\pm}$1.33 mg QE/g DCP, QE and DCP indicate quercetin equivalent and dried citrus peel, respectively) and flavonoids (7.31${\pm}$0.41 mg QE/g DCP) were obtained by subcritical water extraction (SWE) with operating conditions of 190$^{\circ}C$, 1300 psi, and 10 min. The yields by SWE were over 7.2-, and 8.5-fold higher than those of total polyphenols (3.79${\pm}$0.73 mg QE/g DCP) and flavonoids (0.86${\pm}$0.27 mg QE/g DCP) obtained using the ethanol extraction, which showed the highest extraction efficiency among tested conventional methods, respectively. Antioxidant activities of extracts obtained by different methods showed no significant differences. However, the relative antioxidant yield per 1 g dried citrus peel by SWE (190$^{\circ}C$, 10 min) was over 9.5-fold higher than that by the ethanol extraction.
