Composition of the insect diet in feces of yellow-throated marten, Martes flavigula, in Jirisan National Park, South Korea

  • The composition of the Martes flavigula diet has been extensively studied, but little is known about its insect component. This study characterized the insect diet and insect nest materials in the feces of M. flavigula. A total of 952 fecal samples were collected in Jirisan National Park from January 2009 to November 2011, and 1379 species or taxonomic groups were identified. M. flavigula fed on insects (8.7%) and bee wax (5.0%), which comprised 2.9% and 4.4% of the dry weight of M. flavigula feces, respectively. A total of 12 insect species belonging to 8 families in 3 orders were identified. The most frequently found insects were Hymenoptera (frequency of occurrence, FO, 89.1), including Vespa simillima simillima (FO, 37.3) and Vespula koreensis koreensis (FO, 20.0). The Vespidae constituted the majority of the insect diet in autumn when the diversity of marten's prey was strongly reduced, probably because the numbers of social insects were sufficient for M. flavigula. In addition, the data suggest that M. flavigula attacks the nests of social wasps in late autumn when males do not have a venomous sting and new queens are less aggressive than workers. Bee wax appeared in all seasons and the highest rate was from spring to early summer. However, the remains of honeybees were not found in feces; thus, M. flavigula presumably eats honey but not honeybees.

Honokiol as an Effective Antimicrobial Compound against Causative Agent of American foulbrood, Paenibacillus larvae

  • Recently, number of honeybees (Apis mellifera) has visibly decreased because they are vulnerable to some diseases like American foulbrood disease. American foulbrood disease, which is caused by Paenibacillus larvae, is emerged as great cause of decrease in number of honeybees. After antibiotic-resistant strain emerged, it is now more difficult to treat those pathogens successfully. Researches on finding alternative antibacterial compound are ongoing. In this study, we examined the antibacterial effect of honokiol on P. larvae. Honokiol showed great antibacterial effect with minimum inhibitory concentration of 12.5 ㎍/mL and minimum bactericidal concentration of 50 ㎍/mL. An agar diffusion test also confirmed the anti-Paenibacillus larvae activity of honokiol with an inhibitory zone of 9±0.5 mm. Since honokiol is known to interact membrane of some bacteria, we measured 260 nm absorbing particles, which could be induced by leakage of cells, and confirmed that the leakage of P. larvae occurred in dose-dependent manners. However, result of crystal violet assay suggested that honokiol has only mild anti-biofilm formation effect on P. larvae, which means honokiol controls the bacteria by inducing the bursting of membrane. Finally, an additive effect of honokiol with tetracycline and terramycin was found using a checkerboard assay with a fractional inhibitory concentration index value of 0.5.

Anti-Parasitic Activity of Lespedeza cuneata Extract on Causative Agent of Nosemosis Type C, Nosema ceranae

  • Although honeybees (Apis mellifera) are crucial for maintenance of the ecosystem, population of honeybee has been steadily decreasing due to diseases including nosemosis. Nosemosis is a disease caused by Nosema ceranae and is now considered as a major threat to honeybees. N. ceranae is a microsporidian that stays in form of spore even before the infection, which makes it harder to control than other pathogens. People are now aware of this parasite, however, cure and preventive candidates for nosemosis are hardly found until today. In this study, in vitro experiment of Lespedeza cuneata treatment to prevent nosemosis were done using Trichoplusia ni cell line, BTI-TN5B1-4. Normal T. ni cells exhibited round shape without abnormal size. On the other hand, when N. ceranae were treated, cells deteriorated and some cells abnormally enlarged due to N. ceranae infection. Interestingly, treatment of T. ni cells with L. cuneate extract protected abnormal cell shape induced by N. ceranae infection to normal shape. Some N. ceranae spores were observed outside of the cells. Effective concentration range for N. ceranae control were experimented. Lowest concentration which can control nosemosis were 50 ㎍/mL. When the concentration of L. cuneata extract was exceeded 200 ㎍/mL, cytotoxicity started to show up.

Bacillus thuringiensis 유래 Vip3Aa 단백질 순수분리 및 꿀벌 (Apis mellifera)에 대한 위해성평가 (Purification and risk assessment of Bacillus thuringiensis Vip3Aa protein against Apis mellifera)

  • 본 연구는 LMO 유전산물의 위해성평가를 위해 바실러스로부터 증폭된 Vip3Aa 유전자를 이용하여 대장균에서 단백질 순수분리 하였으며, MALDI-TOP 분석법을 통해 기존의 알려진 살충성 Vip3Aa 단백질과 동등한 단백질임을 증명하였다. 순수 분리한 Vip3Aa 단백질을 이용하여 꿀벌 과독성 급성섭식독성평가를 수행하였다. 그 결과 무처리군, Hepes buffer, Vip3Aa 단백질 처리군 모두 치사 및 일반 중독증상을 보이는 개체는 발견되지 않았다. 이 결과를 통해 Vip3Aa 단백질은 꿀벌에 위해성을 나타내지 않는다는 결론을 얻을 수 있었다. 본 연구 결과는 향후 국내 LMO 유전자 산물 위해성평가에 유용하게 활용될 것이라 사료된다.

천연항생제로서 봉독의 기원 및 지표성분 설정 (Origin and effective ingredient standards of honeybee venom as natural antibiotic ingredients)

  • This research was performed in order to investigate the origin, standard compound, and structural and physical properties of honeybee venom which used as natural antibiotic ingredients to animal. We compared the nucleotide sequence of mitochondrial cytochrome c oxidase subunit 1 gene (COI) of honeybees were collected from Gangwon, Gyeonggi, Chungnam, Gyeongbuk, Gyeongnam province and Suwon. As major constituent of honeybee venom, melittin was assayed by liquid chromatography. X-ray, differential scanning calorimetry (DSC) and fourier transform infrared spectroscopy (FT-IR) were utilized to examine the structural and physical properties of honeybee venom. Based on the 627bp sequence of COI, Apis mellifera ligustica was determinated honeybees collected from all six regions. Melittin content varied from 50.7 to 68.6 and averaged 59.8%. According to XRD analysis, honeybee venom showed regular crystal structure peaks at $2{\Theta}=8.5^{\circ}$ and $21.5^{\circ}$. DSC showed that the maximum degration temperature of powder was around $230^{\circ}C$. Through FT-IR analysis, we could identify cross-linking by the presence of peptide peak at 1,500~1,600 $cm^{-1}$. In conclusion, the origin of honeybee venom was Apis mellifera ligustica and effective ingredient standards was melittin content varied from 50.7 to 68.6 as natural antibiotic ingredients.

Propolis Inhibits Neurite Outgrowth in Differentiating SH-SY5Y Human Neuroblastoma Cells

  • Propolis is a multicomponent, active, complex resinous substance collected by honeybees from a variety of plant sources. We have studied the effect of propolis on neurite outgrowth of SH-SY5Y human neuroblastoma cells induced to differentiate by all-trans-retinoic acid (RA). Propolis, at a concentration of $3{\mu}g/mL$, had no significant effect on the viability of differentiating SH-SY5Y cells. However, the neurite outgrowth of the differentiating SH-SY5Y cells treated with propolis ($0.3{\sim}3{\mu}g/mL$) for 48 hr was significantly inhibited in a dose-dependent manner. Treatment of RA-stimulated differentiating SH-SY5Y cells with 0.3 to $3{\mu}g/mL$ propolis resulted in decreased level of transglutaminase and 43-kDa growth-associated protein (GAP-43) in a dose-dependent manner. The results indicate that propolis is able to inhibit neurite outgrowth of differentiating SH-SY5Y cells.

  • 우리나라에 분포하고 있는 재래꿀벌(Apis cerana)의 일벌을 대상으로 춘계 11지역 및 하계 12지역 개체군을 선발하였으며, 총 47개 정량형질에 대해 지역별로 판별분석을 실시 하였고, 또한 양 계절간 변이도 분석하였다. 그 결과, 지역별 비교에서는 춘계 91.67%, 하계 88.44%의 평균분리율을 보였으며, 양계절간 비교에서는 97.58%로 나타났다. 반면에 양계절을 통합한 비교에서는 70.16%로 낮았다.

  • 우리나라에 분포하고 있는 재래꿀벌(Apis cerana)의 일벌(worker)을 대상으로 춘계 15지역 및 하계 16지역 개체군을 선택하였으며, 총 47개 정량형질에 대해 계절 및 개체군별로 판별분석(discriminant analysis)을 실시하였다. 그 결과, 각 계절별 및 개체군별 분리도는 모든 비교방법에서 매우 뚜렷하였다. 특히, 전체 47형질 중 앞다리 경절 길이(FTL)가 분리 기여도가 가장 큰 형질로 나타났다.

생봉독을 이용한 세균성 설사 자돈의 치료효과 (The Therapeutic Effect of Piglets with Bacterial Diarrhea by Natural Honeybee (Apis mellifera) Venom.)

  • This study was designed to examine the therapeutic effect of Italian honeybee (Apis mellifera) venom in piglets with bacterial diarrhea. Preweaning piglets were assigned to treated and nontreated control groups. In treated group, 47 piglets were given natural honeybee venom once a day for 3 consecutive days. The natural honeybees were stung acupoints of GV-1 (Jiao-Chao, at the indentation between the base of tail and the anus) and ST-25 (Hai-men, about 1 cm lateral to the umbilicus). In control group, 44 piglets were intramuscularly injected with a standard dosage of colistin sulfate (300,000 IU/kg of body weight) and antid-iarrheal drug (berberine, 2 ml/kg) once a day for 3 consecutive days. At post-treatment, 90.9% of control piglets and 93.6 % of piglets in treated group recovered from bacterial diarrhea. Bee acupuncture therapy did not show in piglets without any side effects such as allergy, intoxication, hemorrhage, or infection. It might be concluded that honeybee venom therapy was effective in controlling of piglets with bacterial diarrhea.

  • Governments and industry have a growing interest in the harmonization of environmental test methods and risk assessment procedures. OECD are currently producing a set of harmonised test guidelines for studying the environmental fate and ecological effects of pesticides. FAO has published an environmental risk assessment procedure. This procedure, which is similar to those used in US and Europe, is based on calculating the ratio of the toxicity of a pesticide to indicator organisms to their level of exposure to the pesticide. The exposure depends on both the concentration of the pesticide and its bioavailability. Ratios which indicate a pesticide will not produce a harmful effect have been established using ecological field studies. Examples are presented for assessing the risk to aquatic ecosystems, earthworms and honeybees. Long-term field studies(up to 20 years) have also shown that pesticides can be used indefinitely without harming soil fertility. Herbicides can be used to avoid the ecologically damaging effects of using soil cultivations excessively for weed control.
