The Anti-allergic Effects of Dictamni Radicis Cortex(白鮮皮) on the PMA plus A23187-stimulated RBL-2H3 Cells (PBL-2H3 세포에서 백선피의 항알레르기 효과)

  • RBL-2H3 cell line을 이용하여 백선피의 항알레르기 효과를 알아보기 위하여 PMA와 A23187로 자극한 후 RBL-2H3 내의 과립에 Histamine와 같이 존재하는 ${\beta}-hexosaminidase$의 유출량의 억제 정도 및 합성된 후 분비되는 $TNF-{\alpha}$, IL-4의 합성조절 정도를 살펴본 결과, 백선피는 세포생존율에는 영향을 미치지 않고 백선피의 농도가 증가함에 따라 ${\beta}-hexosaminidase$의 세포 위로의 유출을 억제하였으며, 또한 알레르기 반응과 밀접한 관련이 있는 사이토 카인 중에 하나인 $TNF-{\alpha}$, IL-4의 mRNA 수준에서의 발현의 조절을 통한 합성도 감소하였다. 이 결과로 보아 백선피는 알레르기와 관련된 질환에 응용이 가능할 것으로 사료된다.

Anti-Allergic Effects of Nodakenin in IgE/Ag-Induced Type I Hypersensitivity (노다케닌의 항 알러지 효과)

  • Mast cells are major effector cells associated with allergic responses. They are activated through the release of histamine, arachidonic acid, and proinflammatory cytokines. We investigated the effect of nodakenin, derived from the roots of Angelica gigas Nakai, on mast cell degranulation and on an allergic response in an animal model. We also investigated the effect of nodakenin on expression of multiple cytokines. Nodakenin suppressed the release of ${\beta}$-hexosaminidase, a marker of degranulation, as well as the expression of interleukin IL-4 and TNF-${\alpha}$ mRNA. Nodakenin inhibited the passive cutaneous anaphylaxis (PCA) reaction in ICR mice in a dose-dependent manner. These results suggest that nodakenin can inhibit mast cell degranulation through the inhibition of IL-4 and TNF-${\alpha}$ mRNA expression, and that nodakenin may potentially serve as an anti-allergic agent.

Inhibitory Effect of Artemisiae Asiaticae Herba on Degranulation, Production of Cytokine, and $Fc{\varepsilon}RI$ Expression in RBL-2H3 Cells (애엽(艾葉)의 RBL-2H3 비만세포 탈과립, cytokine 생산 및 $Fc{\varepsilon}RI$ 유전자 발현에 미치는 영향)

  • Artemisiae Asiaticae Herba (AAH) has been used to remedy of symptoms such as bleeding, dysmenorrhea, eczema and itchy skin in Oriental Medicine. In this study, we investigated the protective effect of AAH on allergic response. The effect of AAH was analyzed by ELISA and RT-PCR in RBL-2H3 cells. We investigated cell viability, ${\beta}$-hexosaminidase and histamine as markers of degranulation, production of IL-4 and TNF-${\alpha}$, and gene expression of HDC2, cytokines and $Fc{\varepsilon}RI$ ${\alpha}{\beta}{\gamma}$ subunit. We found that AAH suppressed ${\beta}$-hexosaminidase and histamine release, the production of IL-4 and TNF-${\alpha}$ in RBL-2H3 by the anti-DNP IgE plus DNP-HSA stimulation. AAH also significantly decreased cytokine mRNA expressions, such as IL-$1{\beta}$, IL-2, IL-3, IL-4, IL-5, IL-6, IL-12, IL-13, TNF-${\alpha}$, and GM-CSF, and increased cytokine mRNA expressions of IL-10 in RBL-2H3. In addition, AAH suppressed mRNA expression of $Fc{\varepsilon}RI$ ${\alpha}{\beta}{\gamma}$ subunit on cell surface. Our results indicate that AAH protects against allergic response and exerts an anti-inflammatory effect through the inhibition of degranulation and production of cytokines and expression of $Fc{\varepsilon}RI$ ${\alpha}{\beta}{\gamma}$ subunit.

The Comparative Study on the Difference of Anti-allergic Effects Based on Different Form of Seunggal-tang (승갈탕(升葛湯)의 제형변화에 따른 항알레르기 효과의 비교 연구)

  • Objectives Seunggal-tang is one of the prescriptions of oriental herbal medicine, which has been applied to several allergic diseases such as atopic dermatitis. This study was planned to compare differences of anti-allergic effects based on different form of Seunggal-tang by manufacturing differently. Methods Two types herb medicine products were used; aqueous extract (SG-T, Seunggal-Tang) and powder (SG-S, Seunggal-San) which were made from the same mixed formula of Seunggal-tang. To investigate in vitro anti-allergic activities, rat basophilic leukemia (RBL-2H3) cells were treated with SG-T and SG-S for 1 hour, and then stimulated with the phorbol 12-myristate 13-acetate (PMA) plus calcium ionophore A23187. We examined the release of beta-hexosaminidase, as a marker of degranulation, and the releases of tumor necrosis factor (TNF)-alpha and interleukin (IL)-4, as proinflammatory cytokines. Results SG-T and SG-S didn't have effects on cell viabilities in concentrations under 2㎎/㎖. In additionto that, SG-T more inhibited releasing ${\beta}$-hexosaminidase, TNF-${\alpha}$ and IL-4 than SG-S. Conclusions These results indicate that SG-T is more effective against mast cell-mediated allergic reactions than SG-S.

Anti-allergic Effect of Graviola Leaf Extract in Human Mast Cells (인간 비만세포에서 그라비올라 잎 추출물의 항알러지 효과)

  • This study is aimed at determining whether graviola (Annona muricata) beneficially influences immunoglobulin (Ig) E-mediated allergic reactions in human mast cells (LUVA cells). To examine the anti-allergic effect of graviola leaf extract (GLE), we treated antigen/IgE sensitized mast cells with extracts of various concentrations. In this study, the results of the in vitro model of IgE-mediated mast cell degranulation showed that GLE significantly inhibited the release of histamine, β-hexosaminidase, TNF-α, IL-4 and IL-6 in LUVA cells. Pretreatment with GLE suppressed the phosphorylation of antigen-induced Lyn and Syk, thus suppressing the downstream MAPKs pathways. The above results indicate GLE could suppress mast cell activation and allergic responses. Accordingly, it can be supported that GLE has the potential to be used as functional cosmetic material.

  • Objectives : Recently, research has been actively conducted on the efficacy of complexes based on oriental medicine prescriptions for improving immune activity and allergies. In this study, In this study, we aimed to examine the effect of Angelica gigas Nakai and Corni fructus extract (AC), medicinal herbs, among candidate drugs derived through preliminary experiments with various components of oriental medicine prescriptions for allergies, on allergies in RBL-2H3 cells. Methods : We evaluated the effect of the ethanol extract of Ulmus on the allergic inflammatory response in anti-DNP-IgE activated DNP-HSA in RBL-2H3 cells. Cell toxicity was determined by WST-1 assay and the markers of degranulation such as beta-hexosaminidase, histamine, TNF-α and IL-6 production of inflammatory mediators and FcεRI-mediated expression. Results : The results showed that treatment with AC extract (20, 40 and 80㎍/㎖) noncytotoxic levels and significantly inhibited the release of β-hexosaminidase, histamine and the production of TNF-α and IL-6 in RBL-2H3 by the antigen stimulation. Conclusions : These results indicate that AC extract exhibits anti-allergic activity through inhibition of degranulation and inhibition of inflammatory mediators and cytokine release. These findings suggest that AC extract may have potential as a prophylactic and therapeutic agent for the treatment of various allergic diseases.

Inhibitory Effects of Anthocyanins Isolated from Black Soybean (Glycine max L.) Seed Coat on Degranulation and Cytokine Generation in RBL-2H3 Cells (검정콩 껍질 유래 안토시아닌의 RBL-2H3 세포에서 탈과립화와 사이토카인 생성 저해 효과)

  • Anthocyanins belong to a group of flavonoid compounds and are well known for their various health beneficial effects, which include antioxidative activities. Among them, the major anthocyanins isolated from seed coat of black soybean (Glycine max L.) were previously characterized as glycosides containing glucopyranose. Asthma is an allergic disease that is strongly associated with various immune cells, including basophils and mast cells. Eosinophils, basophils, and mast cells play important roles in allergic asthma through the release of inflammatory mediators such as asthma-specific T-helper 2 (Th2) cytokines and subsequent amplification of asthma symptoms via degranulation. Rat basophilic leukemia RBL-2H3 cells are the most common in vitro models for evaluating allergic reactions. In this study, we examined the effects of anthocyanin from seed coat of black soybean on antigen-stimulated degranulation and Th2 cytokine production in RBL-2H3 cells. Cell degranulation was evaluated by measuring the release of ${\beta}$-hexosaminidase. ${\beta}$-Hexosaminidase release and Th2 cytokine production in RBL-2H3 cells was much higher upon stimulation with IgE-antigen complex than those in untreated control cells. Anthocyanins significantly suppressed IgE-antigen complex-induced degranulation of RBL-2H3 cells and inhibited IgE-antigen complex-mediated interleukin (IL)-4, IL-13, and tumor necrosis factor ${\alpha}$ (TNF-${\alpha}$) production in RBL-2H3 cells. These findings suggest that anthocyanins from seed coat of black soybean effectively inhibit allergic reactions and may have beneficial effects against allergic asthma.

Comparative Analysis of Anti-oxidative, Anti-inflammatory, Anti-allergy, and Whitening Effects of Different Solvent Extracts from Zizania latifolia (고장초 추출 용매의 에탄올 함량에 따른 항산화, 항염증, 항알러지, 미백 활성 비교 분석)

  • This study was performed to evaluate the anti-oxidative, anti-inflammatory, anti-allergy, and whitening effects of Zizania latifolia ethanol extracts prepared from 5 different ethanol concentrations (10, 30, 50, 70, and 90%). As the ethanol concentration in the extraction solvent was increased, the radical scavenging activities also increased. The inhibitory activity of Z. latifolia ethanol extracts on nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells tended to increase as the content of ethanol increased. The highest inhibitory activity was obtained with 70% ethanol extract. The antiallergy effects of Z. latifolia ethanol extracts were tested by measuring the release of ${\beta}-hexosaminidase$ in IgE-sensitized RBL-2H3 cells. The suppressive effect of Z. latifolia ethanol extracts increased in a dose-dependent manner as the proportion of ethanol increased, except for the 10% ethanol extract. Furthermore, the inhibitory effects of Z. latifolia ethanol extracts against melanin production in ${\alpha}-melanocyte$ stimulated hormone (MSH)-stimulated B16F0 cells increased as the ethanol ratio increased, and 70 and 90% ethanol extracts showed similar inhibitory activities to arbutin, a positive control, at $250{\mu}m$. The present study confirmed the efficacy of Z. latifolia ethanol extracts in various areas, demonstrating antioxidative, anti-inflammation, antiallergy, skin protective, and skin whitening effects, with no cytotoxicity. It could be used as a raw material in functional foods, as well as in cosmetics.

Effect of Hwangryunhaedoktang on Contact Hypersensitivity and Passive Cutaneous Hypersensitivity in Mice

  • During the screening program to discover antiatopic agents from herbal formulas, we investigated the inhibitory effect of Hwangryunhaedoktang (HT) on passive cutaneous anaphylaxis and oxazolone-induced mouse ear dermatitis. HT significantly inhibited PCA reaction in mice at doses of 50 and 200 mg/kg with inhibitory activity of 31 and 53%, respectively. HT at concentration of 0.05 and 0.1% inhibited ear swelling by 23 and 46% at 16 days in oxazolone-induced mouse ear dermatitis. Both HT with anti without human intestinal microflora showed potent inhibitory activity on the ${\beta}$-hexosaminidase release induced by IgE. Based on these findings, HT may be a usable agent for skin disorder contact dermatitis.

  • We studied the effect of methanol extract of Carpopeltis affinis (CA) on atopic allergic reaction. CA dose-dependently inhibited interleukin (IL)-8 and tumor necrosis factor (TNF)-$\alpha$ secretion from the PMA- plus A23187- stimulated HMC-1. CA also dose-dependently inhibited the histamine and $\beta$-hexosaminidase release from mast cells. CA had no cytotoxic effect. (omitted)
