Hepatoprotective Lignan Compound from Schizandrae Fructus on Tacrine-induced Cytotoxicity in Hep G2 Cells

  • In the course of search for hepatoprotective agents from oriental traditional medicines, one compound was isolated from the MeOH extract of the fruits of Schizandra chinensis, and its structure was determined as gomisin J(1). The evaluation for hepatoprotective activity of isolated compound 1 on drug-induced cytotoxicity was conducted, and compound 1 showed protective effect with an EC$_{50}$value of 86.0 ${\pm}$ 5.3 ${\mu}$M against tacrine-induced cytotoxicity in Hep G2 cells. One of the well-known hepatoprotective agents, silybin, used as a positive control, and the hepatoprotective effect of compound 1 is similar with that of silybin (EC$_{50}$ value = 90.5 ${\pm}$ 9.8 ${\mu}$M).

Modification of C2,3,23,28 Functional Groups on Asiatic Acid and Evaluation of Hepatoprotective Effects

  • For the development of novel hepatoprotective agents, C2, C3, C23 and C28 functional groups on asiatic acid were modified, and the prepared compounds were evaluated for their hepatoprotective effects. Among the prepared compounds, 9, 13 and 16 showed significant hepatoprotective activities against CCl4- and galactosamine (GaIN)-induced hepatotoxicity. Especially, compound 9 showed the most significant hepatoprotective effects against GaIN-induced hepatotoxicity (66.4% protection at 50 μM) and moderate hepatoprotective activities against CCl4-induced hepatotoxicity (20.7% protection at 50 μM).

오수목과탕(吳茱木瓜湯)의 자유(自由)라디칼 소거활성(演去活性) 및 肝(간) 세포(細胞) 보호효과(保護效果) (Free Radical Scavenging and Hepatoprotective Effects of Osumokkya-tang)

  • In order to find the new hepatoprotective agents preliminary screening of twelve prescriptions which have been used for the treatment of hepatic disease in Oriental traditional medicine has been carried out. Both DPPH free radical scavenging and hepatoprotective effect on tacrine-induced cytotoxicity in Hep G2 cells are performed. Osumokkya-tang which is one of the prescription belongs to interior-warming properties showed the DPPH free radical scavenging effect as well as hepatoprotective activity significantly. It is also suggested that its hepatoprotective effect is derived from Chaenomelis Fructus.

  • Hepatoprotective activities of 26 traditional Korean herbal prescriptions described in Donghibogam were evaluated in vivo and in vitro screening system. Twenty one prescriptions appeared to be active. Six prescriptions among them showed highly significant hepatoprotective activities; Shihogyegy-Tang(柴胡桂枝湯), Soshiho-Tang(小柴胡湯), Shiho plus Younggolmoryo-Tang(柴胡加龍骨牡蠣湯), Yinjinho-Tang(茵蔯蒿湯), Yinsam-Tang(人蔘湯) and Shashim-Tang(瀉心湯). Shihogyegy-Tang was most active. They reduced the release of transaminases from dissociated hepatocytes by thioacetamide, and serum transaminases and alkaline phosphatase activities of rat with carbontetrachloride-induced hepatitis. They inhibited significantly lipid peroxidation and cellular fatty change of liver by carbontetrachloride.

  • For the development of novel hepatoprotective agents, C11, C28, C2,3,23 or C2,23,28 functional groups on asiatic acid were modified, and their hepatoprotective effects were evaluated. Most of the prepared compounds displayed potent hepatoprotective activities against CCl4- and galactosamine (GaIN)-induced hepatotoxicity. Especially, compounds 16 and 20 showed the most significant hepatoprotective effects against GaIN-induced hepatotoxicity (54.2% and 46.4% protection at 50 mM, respectively).

Hepatoprotective Constituents of the Edible Brown Alga Ecklonia stolonifera on Tacrine-induced Cytotoxicity in Hep G2 Cells

  • In this study, ethanolic extracts from 18 seaweed variants were assessed for hepatoprotective activity against tacrine-induced cytotoxicity in Hep G2 cells. Only one of these, Ecklonia stolonifera Okamura (Laminariaceae), a member of the brown algae, exhibited promising hepatoprotective activity. Bioassay-guided fractionation of the active ethyl acetate (EtOAc) soluble fraction obtained from the ethanolic extract of E. stolonifera, resulted in the isolation of several phlorotannins [phloroglucinol (1), eckstolonol (2), eckol (3), phlorofucofuroeckol A (4), and dieckol (5)]. Compounds 2 and 4 were determined to protect Hep G2 cells against the cytotoxic effects of tacrine, with $EC_{50}$ values of 62.0 and 79.2 $\mu$g/mL, respectively. Silybin, a well characterized hepatoprotective agent, was used as a positive control, and exhibited an $EC_{50}$ value of 50.0 $\mu$g/mL. It has been suggested that the phlorotannins derived from marine brown algae might prove useful sources in the development of novel hepatoprotective agents.

  • Solanum lyratum(Solanaceae) has been used as a traditional analgesic, antipyretic and hepatoprotective agents in Korea. In this study, we investigated the hepatoprotective effect of ethylacetate extract of Solanum lyratum (SL) on the dimethylnitrosamine (DMN)-induced liver damage in rats. Oral administration of SL (150, 300 mg/kg daily for 4 weeks) into the DMN-treated rats remarkably prevented the elevation of serum alanine transaminase, aspartate transaminase and alkaline phosphatase levels. SL also increased serum protein level and reduced the hepatic level of malondialdehyde in DMN-treated rats. Furthermore, DMN-induced elevation of hydroxyproline content was reduced by the treatment of SL. In conclusion, these results demonstrated that SL exhibited in vivo hepatoprotective effect against DMN-induced liver injury, and suggest that SL may be useful in the prevention of liver damage.

In vitro Free Radical Scavenging and Hepatoprotective Compound from Sanguisorbae Radix

  • In the course of searching for hepatoprotective agents from natural products, four compounds were isolated from the MeOH extract of Sanguisorbae Radix, as guided by their DPPH free radical scavenging activity. The structures were determined as 4,5-dimethoxy-3-hydroxybenzoic acid methyl ester (1), (+)-gallocatechin (2), methyl $6-O-galloyl-{\beta}-D-glucopyranoside$ (3), and pomolic acid $3-O-[{\alpha}-L-arabinopyranoside]-28-O-[{\beta}-D-glucopyranosyl]$ ester (ziyu-glycoside I) (4). Compounds 2 and 3 showed significant DPPH free radical scavenging effects, exhibiting $IC_{50}$ values of 11.4 and $13.0\;{\mu}M$, respectively. L-Ascorbic acid was used as a positive control and exhibited the $IC_{50}$ value of $50.3\;{\mu}M$. In evaluation of the hepatoprotective activity of the isolated compounds on drug-induced cytotoxicity, compound 2 showed the significant hepatoprotective effect with the $EC_{50}$ value of $91.84\;{\pm}\;11.0\;{\mu}M$ on tacrine-induced cytotoxicity in Hep G2 cells, while silybin, a positive control, exhibited $EC_{50}$ value of $122.4\;{\pm}\;12.5\;{\mu}M$.

순무의 d-galactosamine 유발 간장해 보호효과 (Hepatoprotective Effects of Brassica rapa (Turnip) on d-Galactosamine Induced Liver Injured Rats)

  • Brassica rapa L. (Turnip) which is one of the specialized crops in Ganghwa island, has been used for diuretic, digestive, and curative for jaundice, etc. In this study, the anti oxidative effects and hepatoprotective effects of turnip in vitro and in vivo were investigated in order to evaluate the possibility as hepatoprotective agents. Ethanol extract of turnip potently showed the scavenging effect on DPPH and inhibitory effect on lipid peroxidation. Oral administration of turnip extract to dgalactosamine-induced experimental liver injured rats was significantly reduced the serum AST, ALT and LDH enzyme activities. And the decrease of catalase and SOD activities in liver microsolmal cytosol was significantly improved by the treatment of turnip. Based on these findings, it is presumed that ethanol extract of turnip may have the hepatoprotective effect on d-galactosamine-induced hepatotoxicity rat.
