Emerging role of Hippo pathway in the regulation of hematopoiesis

  • In various organisms, the Hippo signaling pathway has been identified as a master regulator of organ size determination and tissue homeostasis. The Hippo signaling coordinates embryonic development, tissue regeneration and differentiation, through regulating cell proliferation and survival. The YAP and TAZ (YAP/TAZ) act as core transducers of the Hippo pathway, and they are tightly and exquisitely regulated in response to various intrinsic and extrinsic stimuli. Abnormal regulation or genetic variation of the Hippo pathway causes a wide range of human diseases, including cancer. Recent studies have revealed that Hippo signaling plays a pivotal role in the immune system and cancer immunity. Due to pathophysiological importance, the emerging role of Hippo signaling in blood cell differentiation, known as hematopoiesis, is receiving much attention. A number of elegant studies using a genetically engineered mouse (GEM) model have shed light on the mechanistic and physiological insights into the Hippo pathway in the regulation of hematopoiesis. Here, we briefly review the function of Hippo signaling in the regulation of hematopoiesis and immune cell differentiation.

Drosophila blood as a model system for stress sensing mechanisms

  • The Drosophila lymph gland is the hematopoietic organ in which stem-like progenitors proliferate and give rise to myeloid-type blood cells. Mechanisms involved in Drosophila hematopoiesis are well established and known to be conserved in the vertebrate system. Recent studies in Drosophila lymph gland have provided novel insights into how external and internal stresses integrate into blood progenitor maintenance mechanisms and the control of blood cell fate decision. In this review, I will introduce a developmental overview of the Drosophila hematopoietic system, and recent understandings of how the system uses developmental signals not only for hematopoiesis but also as sensors for stress and environmental changes to elicit necessary blood responses. 단삼사물탕이 조혈작용에 미치는 영향 (The Effect of DanSamSaMul-Tang on Hematopoiesis)

  • Objectives : Korean traditional medicinal herbs and some formulas have been used to improve the function of the hematopoietic system. The present research is to examine the effect of DanSamSaMul- Tang(DSSMT) as a biological response modifier related to hemato-potentiating function using bone marrow cell as a testing system. Methods : Mice with leukopenia and thrombocytopenia induced by CTX were treated with DSSMT Extracts. Then we observed cytokines mRNA expression level, the number of CFU-GEMM and BFU-E, and the expression of TPO and SCF in bone marrow cells of mice. Results : In the DSSMT group, the gene expressions of hematopoietic cytokines were increased significantly and the number of CFU-GEMM and BFU-E of this group was significantly increased compared to the group treated only with EPO and IL-3. The expressions of TPO and SCF in the group treated with DSSMT were higher than those of the control group. Conclusions : We showed that DSSMT has a good effect on the hematopoietic system. This could also be used as important research data for Korean oriental medicine about hemato-potentiating.

Characterization and function of human Ly-6/uPAR molecules

  • Human Ly-6/uPAR molecules are a superfamily composed of two subfamilies; one is the membrane bound proteins with a GPI-anchor and the other are secreted proteins without the GPI-anchor. Ly-6/uPAR molecules have remarkable amino acid homology through a distinctive 8-10 cysteine-rich domain that is associated predominantly with O-linked glycans. These molecules are encoded by multiple tightly linked genes located on Chr. 8q23, and have a conserved genomic organization. Ly-6/uPAR molecules have an interesting expression pattern during hematopoiesis and on specific tumors indicating that Ly-6/uPAR molecules are associated with development of the immune system and carcinogenesis. Thus, Ly-6/uPAR molecules are useful antigens for diagnostic and therapeutic targets. This review summarizes our understanding of human Ly-6/uPAR molecules with regard to molecular structure as well as what is known about their function in normal and malignant tissues and suggest Ly-6/uPAR molecules as target antigens for cancer immunotherapy.

외기요법(外氣療法)과 영연온담탕(靈連溫膽湯) 및 적연온담탕(赤連溫膽湯)이 복수암(腹水癌) 생쥐의 조혈계통(造血系統)에 미치는 영향(影響) (Effects of Emitted-Qi Therapy(外氣療法), Youngyeonondamtang(靈連溫膽湯) and Jeokyeonondamtang(赤連溫膽湯) on Hematopoiesis System in Mice bearing Ascites Cancer)

  • In order to study the effects of emitted-qi therapy, Youngyeonondamtang and Jeokyeonondamtang on the mice bearing ascites cancer caused by Sarcoma 180, the author divided the mice into normal group, control group, EQ group, YO group, JO group, CTX+QE group, CTX+YO group, CTX+JO group, and treated with emitted -qi therapy and administrated Youngyeonondamtang, Jeokyeononondaintang and Cyclophosphamide (CTX). After collecting blood from the mice, measured the values of Prothrombin Time(PT), Partial Thromboplastin Time(PTT), Fibrinogen, White Blood Cell(WBC) and Platelet. The results were as follows, 1. PT was not showed any significant change in ever)r group, being compared with the control group. 2. H increased significantly in YO group, being compared with the control group. 3. Fibrinogen decreased significantly in EQ group, YO group, CTX+EQ group, CTX+YO group, CTX+JO group, being compared with the control group. 4. WBC increased significantly in CTX+EQ group, CTX+YO group, CTX+JO group, being compared with the CTX group. 5. Platelet decreased significantly in every group, being compared with the control group. These results suggest that emitted-qi therapy, Youngyeonondamtang and Jeokyeonondamtang had the effects that could heal the hematopoiesis system in the mice bearing ascites cancer, and especially had the better effects in the case that hematopoiesis system has been impaired by CTX.

화학요법제 5-flurouracil의 조혈 및 면역독성 억제를 위한 조성물 개발 연구 (Inhibitive Effect of KH against Toxicity Induced by 5-Flurouracil(FU))

  • The objective on this study was to investigate effect of KH, which was composed of 9 kinds of oriental herbs tonifing the blood, against toxicity of 5-flurouracil(FU) through animal study. Reduction of WBC and platelet after treating S-FU was significantly recovered by KH. KH also rehabilitated immune gene expression of interleukin-2(IL2) and $Interferon-{\gamma}\;(IFN-{\gamma})$ by RT-PCR. In addition, in situ hybridization of spleen showed that KH treatment increased mRNA expression of IL2. In addition to the immune action, antitumor activity on 5-FU was not affected by KH treatment. In conclusion, our study demonstrated that KH alleviated damage of hematopoiesis system induced by f-FU without toss of antitumor activity.

The increasing hematopoietic effect of the combined treatment of Korean Red Ginseng and Colla corii asini on cyclophosphamide-induced immunosuppression in mice

  • Background: Hematopoiesis is the production of blood cells from hematopoietic stem cells (HSCs) that reside in the bone marrow. Cyclophosphamide (CTX) is a chemotherapy drug that suppresses the immune system. Korean Red Ginseng (KRG) and Colla corii asini (CCA) have been traditionally used for boosting the immune system. Methods: HSCs in the bone marrow, and immune cell subtype in splenocytes, PBMCs, and thymocytes were investigated. Serum levels of hematopoietic-related markers were analyzed using ELISA. Protein expression in spleen tissue was analyzed using western blot analysis. Hematoxylin & eosin staining in the femurs of mice were also conducted. Results: The combination of KRG and CCA with a ratio of 3:2 increased HSCs, CD3 and CD8+ T cells in the circulation, and CD3 T cells in the spleen. A ratio of 2:3 (KRG:CCA) increased the thymic regulatory T cells and recovered the CD3 T cells in the spleen and circulation while recovering proteins in the JAK-STAT pathway in the spleen. Overall, blood cell population and differentiating factors vital for cell differentiation were also significantly recovered by all combinations especially in ratios of 3:2 and 2:3. Conclusion: A ratio of 3:2 (KRG:CCA) is the most ideal combination as it recovered the HSC population in the bone marrow of mice.

  • NF-${\kappa}B$/Rel transcription factor family participates in diverse biological processes including embryo development, hematopoiesis, immune regulation, as well as neuronal functions. In this review, the NF-${\kappa}B$/Rel signal transduction pathways and their important roles in the regulation of immune system will be discussed. NF-${\kappa}B$/Rel members execute distinct functions in multiple immune cell types via the regulation of target genes essential for cell proliferation, survival, effector functions, cell trafficking and communication, as well as the formation of lymphoid architecture. Consequently, proper activation of NF-${\kappa}B$/Rel during immune responses to allergens, auto-antigens, allo-antigens, and pathogenic infection is crucial for the integrity of host innate and adaptive immunity.

  • Global attractivity and oscillatory behavior of the following nonlinear impulsive parabolic differential equation which is a general form of many population models $$\array{\{{{\frac {{\partial}u(t,x)}{{\partial}t}=\Delta}u(t,x)-{\delta}u(t,x)+f(u(t-\tau,x)),\;t{\neq}t_k,\\u(t^+_k,x)-u(t_k,x)=g_k(u(t_k,x)),\;k{\in}I_\infty,}\;\;\;\;\;\;\;\;(*)$$ are considered. Some new sufficient conditions for global attractivity and oscillation of the solutions of (*) with Neumann boundary condition are established. These results no only are true but also improve and complement existing results for (*) without diffusion or impulses. Moreover, when these results are applied to the Nicholson's blowflies model and the model of Hematopoiesis, some new results are obtained.

  • 비정상적인 척추 질환 및 기능은 단순한 근·골격계 문제 외에도 항상성을 방해하고 직·간접적인 생리적 부작용을 일으킬 수 있다. 특히 척추를 통해 나오는 신경이 면역체계를 조절하는 기관에 적절한 신호를 전달하지 못하면 면역 기능의 일부 또는 전부에 문제가 생겨 더 많은 질병에 노출될 수 있다. 이 연구는 기본적인 해부학 및 생리학적 지식에 초점을 맞추고 척추 기능이 잠재적으로 면역 기능을 유지하거나 개선하는 데 도움이 될 수 있는 잠재적 메커니즘을 고찰하고자 한다. 이를 위해 조혈, 스트레스, 호흡, 척추와 신경의 관계, 면역체계와 관련된 척추의 역할을 살펴보고 이러한 역할이 면역기능에 영향을 미칠 수 있음을 확인한다.
