반강성 특성이 반영된 전정 유모세포 모델의 활동전위 생성에 관한 연구 (A Study on the Action Potential Generations of the Vestibular Hair Cell Model with Negative Stiffness Feature)

  • 본 논문에서는 섬모 번들 특성 모델과 통합 전정 유모세포 모델을 제안한다. 기존 전정기관 모델에 관한 연구는 외력이 없는 상태에서 전정 유모세포의 섬모 번들이 가지는 특성이나 시냅스에서 발생하는 활동전위만을 대상으로 진행되었으며 섬모번들의 고유 특성을 고려한 전정신경의 활동전위에 대한 연구는 이루어지지 않았다. 제안된 통합 전정 유모세포 모델은 외력을 반영하였고 서로 다른 규칙성을 가진 유모세포에 대해 각각의 섬모번들의 반강성 특성을 고려하였고 이를 기존의 외력없는 모델과 비교하였다. 그 결과 외부 자극의 변화에 따른 주파수 변화가 큰 불규칙적 신경섬유와 중간규칙적 신경섬유에서는 반강성 구간의 감소를 보였으나 그렇지 않은 규칙적 신경섬유에서는 기존의 반강성 특성과 유사한 특성을 보여주었다. 또한 제안된 전정 유모세포 모델을 통해 11개의 주파수 대역에서의 모델링 데이터와 기존의 동물 실험 데이터가 거의 일치함을 보여 주었다. 제안된 섬모 번들 특성 모델이 적절히 모델링되었음이 확인된다.

아젤라인산 및 비타민 B6의 육모효과 검증에 관한 연구 (A Study for Verification of Hair Growth Effect of Azelaic Acid and Vitamin B6)

  • Purpose: Interest in the augmentation of hair growth for functional and aesthetic purpose has increased dramatically in recent years. Many hair growth products have been released, but most of these have not been proven scientifically. This study aims to measure the hair growth effect of azelaic acid and vitamin $B_6$, which have been known as hair growth materials, in animal models. Methods: Six weeks old C57BL/6 mice were used in this study and hair of mice were removed by topical treatment. The mice were divided into five experimental groups according to the testing material such as saline (negative control), propylene glycol(vehicle control), azelaic acid, vitamin B6 and azelaic acid plus vitamin B6 in combination. Hair growth was documented photographically and histologically, and then analysed by the high quality hair analysis program system. The quantity of endocrine factors, IGF-I and TGF-${\beta}1$ in the skin of mice was measured by PCR analysis. Results: The topical treatment of azelaic acid and vitamin B6 in combination for 2 weeks to dorsal skin accelerated hair regrowth more than other groups. The azelaic acid and vitamin $B_6$-combined treatment also promoted hair follicle elongation and thickness compared to the others. Histologic studies showed increased number of basal cells in azelaic acid and vitamin $B_6$-combined treatment. Furthermore, the azelaic acid and vitamin $B_6$-combined group significantly increased the expression of IGF-I but decreased the expression of TGF-${\beta}1$ in the skin of mice compared to other groups. Conclusion: These results suggest that azelaic acid and vitamin $B_6$, when used together, have an additive effect and might be used as hair growth materials.

황기(黃芪) 추출물의 외용 도포가 자발성 원형탈모 생쥐에 미치는 영향 (Effects of Topical application of Astragalus membranaceus in Spontaneous Alopecia Mice Model)

  • Objectives: Astragalus membranaceus has been reported to inhibit immune responses, but its effect on hair loss is not clear. In this study, the effect of A. membranaceus extract (AM) on hair regrowth in C57BL/6 mice with natural hair loss in the telogen phase was investigated. Methods: Mice with natural hair loss were topically treated with 1% AM on the dorsal skin for 2 weeks. Dorsal skin samples were stained with hematoxylin and eosin and probed with an anti-mouse CD8a IgG. The mRNA expression levels of tumor necrosis factor $(TNF)-{\alpha}$, interferon $(IFN)-{\gamma}$ and interleukin (IL)-4 were measured by reverse transcription polymerase chain reaction and quantitative real-time polymerase chain reaction. Results: AM treatment induced hair regrowth in hair loss mice, while control mice suffered continued hair loss. Tapering hair shafts and broken hair follicles were decreased as well as CD8+ T lymphocyte infiltration. In addition, the expressions of $TNF-{\alpha}$, $IFN-{\gamma}$ and IL-4 were reduced by AM treatment. Also, AM treatment significantly increased the KGF expressions in Hs68 fibroblast cells. Conclusion: These results suggest that topical application of A. membranaceus may be an alternative therapy for hair loss.

현미 주정 추출물의 항산화 활성 및 melanin 합성 촉진 효과 (Ethanolic Extract of Oryza sativa Displays Antioxidative Activity and Promotes Melanin Synthesis)

  • 모발의 검정색은 노화됨에 따라 백색으로 변화된다. 이러한 현상은 모낭내 tyrosinase 활성의 감소와 $H_2O_2$에 의한 축적에 의하여 기인된다. 따라서 본 연구의 목적은 현미주정추출물(OREE)를 이용하여 백발화의 원인인 과산화 수소에 대한 항산화 활성과 멜라닌 생성 촉진 효과를 조사한 것이다. 본 연구에서 OREE은 낮은 DPPH radical 소거능과 환원력을 보여주었다. 그러나 B16F1 세포내에서 $H_2O_2$ 소거에 대해서는 높은 항산화효과를 나타내었다. 그리고 OREE는 in vitro에서 DOPA 산화 활성은 나타나지 않았지만 $64{\mu}g/ml$에서 tyrosinase 활성을 증가시켰다. MTT assay에서 OREE는 $32{\mu}g/ml$ 이상의 농도에서 세포독성을 나타내었다. 또한 OREE는 $8{\mu}g/ml$ 이상의 농도에서 멜라닌 합성은 농도에 비례하여 증가하였고, $H_2O_2$로 멜라닌 생성을 저하시킨 세포에서도 멜라닌 합성을 증가시켰다. 멜라닌 합성에 대한 OREE의 효과를 확인하기 위하여 Western blot 분석이 수행되었다. $H_2O_2$로 멜라닌 생성을 저하시킨 세포에서 멜라닌 합성 기전 관여하는 tyrosine hydroxylase와 tyrosinase-related protein-2 (TRP-2) 발현은 OREE의 존재 하에서 증가하였다. 이상의 발견들은 OREE가 멜라닌 합성을 촉진시킬 수 있어 이와 관련된 모발화장품의 개발에 적용시킬 수 있다는 것을 시사하고 있다.

Establishment and characterization of an immortalized human dermal papilla cell line

  • Establishment of immortalized human dermal papilla cells (DPCs) retaining the characteristics of DPCs would be a great help for hair researchers. We recently established a simian virus 40T (SV40T)-transformed human DP cell line (SV40TDPC). However, the cell line senesced around passage 25 and ceased proliferation. In this study, we introduced the human telomerase reverse transcriptase (hTERT) gene into SV40T-DPC and established an immortalized human DP cell line. The cell line, SV40T-hTERT-DPC, did not induce tumors when inoculated into nude mice. SV40T-hTERT-DPC maintained morphology of early passage DPCs, expressed markers of DPCs, and retained responses to Wnt/${\beta}$-catenin and bone morphogenic protein (BMP) signaling pathways known to be required for hair-inducing activity of DPCs. The data strongly suggest that SV40T-hTERT-DPC retains many characteristics of human DPCs in vivo without malignant transformation.

자외선 조사에 따른 모발의 물리화학적 변화에 관한 연구 (Study on the Physicochemical Change of Human Hair Shaft Following Radiation with Ultraviolet)

  • 자외선 B를 사람의 두피에서 채취한 모발에 6시간, 12시간, 24시간. 48시간 동안 각각 조사한 후 모발의 미세구조적인 변화 과정과 모발 단백질의 분해에 의한 원소 성분의 함량 변화를 전자현미경 및 에너지분산분광분석기를 이용하여 분석하였다. 자외선에 6시간 조사된 모발에서 모발 표면은 중첩된 큐티클층이 분리되어 있거나 부러져있는 상태로 나타났다. 큐티클세포의 내큐티클에는 직경이 30nm에서부터 700nm크기의 공포를 형성하고 있었다. 자외선에 12시간 조사된 모발에서 피질은 치밀하게 배열되어 있는 각화세포의 세포막이 파괴되어 공포가 형성되었는데 공포는 세포막을 따가서 일렬로 배열되어 있었다. 또한, 멜라닌 과립과 접촉하고 있는 거대원섬유에서 공포가 형성되어 멜라닌 과립과 분리되는 것을 확인할 수 있었다. 이 시기에도 멜라닌 과립의 모양이나 크기의 변화는 나타나지 않았다. 자외선에 24시간 조사된 모발에서 큐티클세포들은 절단된 상태로 관찰되었고 세포질의 내큐티클에 형성된 공포들은 인접해 형성된 공포들과 융합되었다. 자외선에 48시간 조사된 모발은 표면이 거칠게 나타났고, 큐티클의 안쪽 층에 있는 세포에서부터 형성된 공로로 인해 위쪽에 있는 큐티클 세포는 들뜨면서 조각나 떨어져 나갔다. 자외선 조사를 받아 손상된 모발의 원소성분을 에너지 분산 분광분석기로 분석한 결과 정상모발 보다 산소성분의 함량이 높게 나타났으나 황의 성분은 감소되었다.

참가죽그물바탕말 1,9-Dihydroxycrenulide 및 Epiloliolide의 양모 효능 (The Effect of 1,9-Dihydroxycrenulide and Epiloliolide from Dictyota coriacea on the Hair Growth)

  • This study was conducted to evaluate the effects of Dictyota coriacea extract and its active components such as 1,9-dihydroxycrenulide and epiloliolide on the hair growth. Treatment with D. coriacea extract and the hexane and EtOAc fractions of D. coriacea extract significantly increased the proliferation of dermal papilla cells (DPCs), a central regulator of the hair cycle. Especially, 1,9-dihydroxycrenulide and epiloliolide from D. coriacea extract, caused an increase in the DPC proliferation. When isolated rat vibrissa follicles were treated with 1,9-dihydroxycrenulide or epiloliolide for 21 d, the hair-fiber lengths for the vibrissa follicles increased. When examined the activity of 5α-reductase, which converts testosterone to dihydrotestosterone (DHT), a main cause of androgenetic alopecia, the several solvent fractions of D. coriacea extract significantly decreased the 5α-reductase activity while 1,9-dihydroxycrenulide and epiloliolide scarcely inhibited 5α-reductase activity. In addition, we found that the D. coriacea extract and several solvent fractions of D. coriacea extract could not act as a KATP channel opener, which could be a contributory factor in the effect on hair growth. These results suggest that D. coriacea extract and 1,9-dihydroxycrenulide and epiloliolide, principals of D. coriacea, have the potential to treat alopecia via the proliferation of DPCs.

Effect of the hedgehog signaling pathway on hair formation-related cells

  • Alopecia has emerged as one of the biggest interests in modern society. Many studies have focused on the treatment of alopecia, such as transplantation of hair follicles or inhibition of the androgen pathway. Hair growth is achieved through proper proliferation of the components such as keratinocytes and dermal papilla cells (DPCs), movement, and interaction between the two cells. The present study examined the effect of the hedgehog (Hh) signaling pathway, which is an important and fundamental signal in the cell, on the morphology and the viability of human keratinocytes and DPCs. Upregulation of Hh signaling caused a morphological change and an increase in epithelium-mesenchymal transition-related gene expression but reduced the viability of keratinocytes, while the alteration of Hh signaling did not cause any change in DPCs. The results show the possibility that the regulation of Hh signaling can be applied for the treatment of alopecia.

Effects of Natural Extracts on the Radiation-induced Pink Mutations in Tradescantia Stamen Hair Cells

  • The effect of a water-soluble extract from natural materials on radiation-induced mutations was studied by means of TSH assay in Tradescantia 4430 stamen hair cells. Inflorescence cuttings, with or without pretreatments of natural extracts for 3 hours, were exposed to 1 Gy of gamma ray. Comparisons were made on the basis of pooled data during the peak interval between the mean pink mutation frequencies of the experimental groups. Pretreatments of FB or FB-I resulted in about two-fold increases of the pink mutation frequencies, compared to those of the control group. Synergism between certain fractions and radiation was a possible cause of the increased DNA damage. FB and FB-I had a radiosensitizing effects on the pink mutations in Tradescantia 4430 stamen hair cells (p<0.001). On the other hand, the extract PP in a proper concentration significantly reduced the pink mutation frequencies (p<0.05). The result means that PP has a protective effect on the radiation-induced cell damage.

Wnt/β-catenin signaling activator restores hair regeneration suppressed by diabetes mellitus

  • Diabetes mellitus is one of the most prevalent diseases in modern society. Many complicationssuch as hepatic cirrhosis, neuropathy, cardiac infarction, and so on are associated with diabetes. Although a relationship between diabetes and hair loss has been recently reported, the treatment of diabetic hair loss by Wnt/β-catenin activators has not been achieved yet. In this study, we found that the depilation-induced anagen phase was delayed in both db/db mice and high-fat diet (HFD) and streptozotocin (STZ)-induced diabetic mice. In diabetic mice, both hair regrowth and wound-induced hair follicle neogenesis (WIHN) were reduced because of suppression of Wnt/β-catenin signaling and decreased proliferation of hair follicle cells. We identified that KY19382, a small molecule that activates Wnt/β-catenin signaling, restored the capabilities of regrowth and WIHN in diabetic mice. The Wnt/β-catenin signaling activator also increased the length of the human hair follicle which was decreased under high glucose culture conditions. Overall, the diabetic condition reduced both hair regrowth and regeneration with suppression of the Wnt/β-catenin signaling pathway. Consequently, the usage of Wnt/β-catenin signaling activators could be a potential strategy to treat diabetes-induced alopecia patients.
