Effect of Charge Carrier Lipid on Skin Penetration, Retention, and Hair Growth of Topically Applied Finasteride-Containing Liposomes

  • The aim of this study was to investigate the effect of charge carrier lipid on the skin penetration, retention, and hair growth of topically applied finasteride-containing liposomes. Finasteride-containing liposomes were prepared by traditional thin film hydration method using Phospholipon$^{(R)}$ 85 G and cholesterol with or without charge carrier lipid (1,2 dimyristoyl-sn-glycero-3-phosphate or 1,2-dioleoyl-trimethylammonium-propane for anionic and cationic charge, respectively). Freshly prepared finasteride-containing liposome suspension was applied on the hairless mouse skin, and skin penetration and retention were measured using Keshary-Chien diffusion cell. Non-liposomal formulation (ethanol 10% solution containing 0.5 mg/ml of FNS) was also used as a control. The amount of finasteride in the diffusion cell and mouse skin was measured by HPLC. The hair growth was evaluated using depilated male C57BL/6N mice. Mean particle size of all finasteride-containing liposomes was less than a micron, and polydispersity index revealed size homogeneity. Skin penetration and retention studies showed that significantly less amount of finasteride was penetrated when applied as anionic liposome while more amount of the drug was retained. Specifically, in liposome prepared with 10% anionic charge carrier lipid, penetration was 12.99 ${\mu}g/cm^2$ while retention was 79.23 ${\mu}g/cm^2$ after 24 h of application. In hair growth study, finasteride-containing anionic liposomes showed moderate efficacy, but the efficacy was not found when applied as cationic liposomes. In conclusion, topical application of finasteride using anionic liposome formulation appears to be useful option for the treatment of androgenetic alopecia to avoid systemic side effects of the drug.

참도박의 Wnt 경로 활성화를 통한 모발성장 효과 (Hair-growth Promoting Effect of Grateloupia elliptica Via the Activation of Wnt Pathway)

  • Grateloupia elliptica has been reported to have the proliferation effect of dermal papilla cells (DPCs), which play important roles in the regulation of hair cycle. In the present study, we examined in vitro and in vivo hair growth-promoting effect of Grateloupia elliptica. When isolated rat vibrissa follicles were treated with extract of G. elliptica, the hair-fiber lengths of the vibrissa follicles significantly increased. Furthermore, the G. elliptica extract accelerated the telogen-angen transition in C57BL/6 mice. To investigate the molecular mechanisms of the G. elliptica extract on the proliferation of DPCs, we examined the activation of $wnt/{\beta}$-catenin signaling which is known to regulate hair follicle development, differentiation and hair growth. The G. elliptica extract activated $wnt/{\beta}$-catenin signaling via the increase of ${\beta}$-catenin and phospho-$GSK3{\beta}$. In addition, the G. elliptica extract increased the level of cyclin E and CDK2, while the level of $p27^{kip1}$ was decreased. These results suggest that the the G. elliptica extract may induce hair growth by proliferation of DPCs via cell-cycle progression and the activation of $Wnt/{\beta}$-catenin signaling.

금잔화, 상황, 어성초 에탄올 추출물의 세포독성 평가 및 3T3-L1 세포에 대한 육모 효과 (Cell Viability and Hair Growth Effect on 3T3-L1 Cells of Ethanol Extract from Calendula officinalis L. Flower, Phellinus linteus Fruit Body and Houttuynia cordata Thunb. Whole Plant)

  • Background: Hair loss related syndromes are increasing due to environmental pollution and stress. Hair care products are mainly prepared by mixing chemicals and natural extracts, such as those obtained from medicinal plants. The purpose of this study was to investigate the effects of 70% ethanol extracts from the flowers of Calendula officinalis, fruit body of Phellinus linteus, and the whole plant of Houttuynia cordata on the growth of CCD-986 cells, hair follicle dermal papilla cells (HFDPC), and 3T3-L1 cells. Methods and Results: All sample extracts at all concentrations, except for that from P. linteus fruit body at $500{\mu}g/m{\ell}$, were cytotoxic to CCD-986 cells. However, none of the sample extracts were cytotoxic to HFDPC. The lipid differentiation of 3T3-L1 cells regulates hair regeneration via secretion of platelet derived growth factor. The 70% ethanol extract of H. cordata whole plant promoted hair growth. Adipogenesis rate significantly increased in a treatment concentration-dependent manner. Conclusions: These results suggest that 70% ethanol extracts of C. officinalis flower, P. linteus fruit body and H. cordata could be used for the development of hair care products.

Flavonoid Silibinin Increases Hair-Inductive Property Via Akt and Wnt/β-Catenin Signaling Activation in 3-Dimensional-Spheroid Cultured Human Dermal Papilla Cells

  • Hair loss, also known as alopecia, is a common dermatological condition of psychosocial significance; development of therapeutic candidates for the treatment of this condition is, hence, important. Silibinin, a secondary metabolite from Silybum marianum, is an effective antioxidant that also prevents various cutaneous problems. In this study, we have investigated the effect of silibinin on hair induction using three-dimensional (3D) cultured, human dermal papilla (DP) spheroids. Silibinin was found to significantly increase viability through AKT serine/threonine kinase (AKT) activation in 3D DP spheroids. This was correlated with an increase in the diameter of the 3D DP spheroids. The activation of the wingless and INT-1 (Wnt)/${\beta}$-catenin signaling pathway, which is associated with hair growth induction in the DP, was evaluated using the T cell-specific transcription factor and lymphoid enhancer-binding factor (TCF/LEF) transcription factor reporter assay; results indicated significantly increased luciferase activity. In addition, we were able to demonstrate increased expression of the target genes, WNT5a and LEF1, using quantitative real-time PCR assay. Lastly, significantly elevated expression of signature genes associated with hair induction was demonstrated in the 3D DP spheroids treated with silibinin. These results suggest that silibinin promotes proliferation and hair induction through the AKT and Wnt/${\beta}$-catenin signaling pathways in 3D DP spheroids. Silibinin can be a potential candidate to promote hair proliferation.

케라틴 펩타이드에 의한 모발 및 두피 특성 변화 연구 (Study on Changes of Hair and Scalp Characteristics by Keratin Peptides)

  • 본 연구에서는 Fervidobacterium islandicum AW-1를 이용하여 케라틴 펩타이드를 생산하고, 두피 및 모발에 관련된 인자를 확인하여, 화장품 원료로서 케라틴 펩타이드의 가능성을 알아보았다. 모유두 세포주에 케라틴 펩타이드를 농도에 따라 세포 독성 및 증식을 살펴본 결과, 세포 독성은 나타나지 않았고, 세포증식에도 효과가 없었다. 케라틴 펩타이드를 함유한 헤어 샴푸, 헤어 에센스를 제조하여 인체 첩포 시험을 거쳐 무자극을 확인하였다. 30 명의 건강한 성인 피험자를 대상으로 4 주 동안 샴푸 및 에센스를 2 그룹으로 나누어 사용한 결과, 육안평가에서 윤기, 탈모, 두피 트러블, 푸석거림에서 통계적으로 유의한 긍정적 결과를 나타내었다. 두피 수분량은 샴푸와 에센스 사용 전 대비 2, 4 주 후에 통계적으로 유의한 증가를 나타내었다. 두피에 대한 경피수분손실양은 에센스를 사용한 그룹에서 사용 전 대비 4주 후에 통계적으로 유의한 감소를 나타내었다. 두피 피지량 역시, 에센스를 사용한 그룹에서 사용 전 대비 4 주 후에 통계적으로 유의한 감소를 나타내었다. 정상 모발, 손상 모발을 이용한 헤어 샴푸와 에센스의 사용 전후의 빗질에 의한 마찰력을 측정한 결과, 사용 전 대비 모두 통계적으로 유의한 변화를 나타내었다. 정상 모발의 경우, 사용 전보다 빗질의 마찰력이 증가하였고, 손상 모발의 경우 사용 전보다 빗질의 마찰력이 감소하였다. 이를 통해 케라틴 펩타이드는 모발 및 두피 관련한 제품에 사용할 수 있는 원료로 확인하였다.

측백엽(側柏葉) 추출물의 발모효과(發毛效果)에 관한 실험적 연구 (Influence of Extracts of Biotae Orientalis on the Efficiency of Hair Growth in Hair Removed C57BL/6N Mice)

  • Biotae Orientalis has been widely used for treatment of relaxion of smooth muscle, gastrointestinal hemorrhage and alopecia in Oriental Medicine. This experiment examined the effect of an extracts, obtained from the acetone and MeOH extracts of dried or fresh Biotae Orientalis, on hair growing activity of the C57BL/6N mice after topical application to skin. First, We examined on hair growth activity of extracts of Biotae Orientalis compare to control and 1% minoxidil groups. Second, We investigated on the number of hair follicle and mast cells after topical application of extracts of the Biotae Orientalis to skin for 16 day. Third, We investigated immunoreactive density of vascular endothelial growth factor(VEGF), protein kinase C-${\alpha}$(PKC-${\alpha}$) and stem(mast) cell factor(SCF) in skin of C57BL/6N mice by immunohistochemical methods. I fourth investigated changes of subpopulation of splenocytes and thymocytes in C57BL/6N mice for 16day using laser flow cytometry. The results were as follows : Hair growing effect of acetone and MeOH extracts of dried and fresh Biotae Orientalis was observed in 70%, 90% and 60% in hair removed skin area in 16 day respectively. Immunoreactive density of VEGF and PKC-${\alpha}$ in skin of experimental groups was weakly stained compare to control group in 10 day. Immunoreactive density of stem cell factor in skin of experimental group was heavily stained compare to control group in 10 day. Splenic TH/TC Iymphocytes of lived MeOH extracts group significantly increased compare to control group. TH cells in thymic T lymphocytes were increased compare to control group. These experiment suggest that acetone and MeOH extracts of Biotae Orientalis may be used for topical treatment of alopecia areata.

황칠나무 발효 추출물의 육모효과 (The Effect of Fermented Extracts of Korean Dendropanax Morbifera Levéille on Hair Growth)

  • 이전 연구에서 우리는 Lactobacillus plantarum ilchiwhangchil 1785와 Lactobacillus plantarum ilchiwhangchil 2020 균주로 발효시킨 황칠나무 잎 가지 추출물의 효과적인 항균 활성을 확인하였다. 본 연구에서는 황칠나무 잎 가지 추출물을 L. plantarum ilchiwhangchil 1785와 L. plantarum ilchiwhangchil 2020 균주로 발효시킨 발효액이 인간모유두세포에 미치는 육모효과를 알아보았다. 황칠나무 잎 가지 발효 추출물의 농도가 1, $10{\mu}g/ml$ 범위에서 인간모유두세포는 세포사멸 증가 없이 유의적으로 세포 성장이 농도의존적으로 증가하였다. 또한 황칠나무 잎 가지 발효물로 처리한 세포가 대조군보다 세포성장이 활발함을 이중염색법으로 확인하였다. 더욱이 황칠나무 잎 가지 발효 추출물 처리한 세포는 처리하지 않은 세포에 비해 각각 18.84%, 23.31% 세포 이동성이 증가하였다. 고성능액체크로마토그래피(HPLC)를 이용하여 육모효과의 활성 물질을 분석하였다. 모발성장효과가 알려진 ${\beta}$-sitosterol 함량이 황칠나무 잎 가지 추출물에 비해 황칠나무 잎 가지 발효 과정에서 약 10배 증가되는 것을 확인하였다. 결과를 종합하면, 우리는 본 연구에서 황칠나무 잎 가지 추출물 발효액이 모발 성장을 촉진시킴을 확인하였다.

A novel tetrapeptide for the treatment of hair loss identified in ginseng berry: in silico characterization and molecular docking with TGF-β2

  • Hair loss causes psychological stress due to its effect on appearance. Therefore, the global market for hair loss treatment products is rapidly growing. The present study demonstrated that ginseng berry-derived and sequence-modified peptides promoted the proliferation rate of dermal papilla (DP) cells and keratinocytes, in addition to having antioxidant properties. Moreover, the potential role of these ginseng berry peptides as TGF-β2 antagonists was confirmed through in silico computer docking. In addition to promoting the growth of ,the ginseng berry-derived peptides also promoted the proliferation of keratinocytes experimental Particularly, an unmodified ginseng berry-derived peptide (GB-1) and two peptides with sequence modifications (GB-2 and GB-3) decreased ROS generation and exhibited a protective effect on damaged HaCaT keratinocytes. Computer-aided peptide discovery was conducted to identify the potential interactions of important proteins with transforming growth factor-beta 2 (TGF-β2), a key protein that plays a crucial role in the human hair growth cycle. Our results demonstrated that MAGH, an amino acid sequence present in herbal supplements and plant-based natural compounds, can inhibit TGF-β2.

miR-133a-3p and miR-145-5p co-promote goat hair follicle stem cell differentiation by regulating NANOG and SOX9 expression

  • Objective: Hair follicle stem cells (HFSCs) differentiation is a critical physiological progress in skin hair follicle (HF) formation. Goat HFSCs differentiation is one of the essential processes of superior-quality brush hair (SQBH) synthesis. However, knowledge regarding the functions and roles of miR-133a-3p and miR-145-5p in differentiated goat HFSCs is limited. Methods: To examine the significance of chi-miR-133a-3p and chi-miR-145-5p in differentiated HFSCs, overexpression and knockdown experiments of miR-133a-3p and miR-145-5p (Mimics and Inhibitors) separately or combined were performed. NANOG, SOX9, and stem cell differentiated markers (β-catenin, C-myc, Keratin 6 [KRT6]) expression levels were detected and analyzed by using real-time quantitative polymerase chain reaction, western blotting, and immunofluorescence assays in differentiated goat HFSCs. Results: miR-133a-3p and miR-145-5p inhibit NANOG (a gene recognized in keeping and maintaining the totipotency of embryonic stem cells) expression and promote SOX9 (an important stem cell transcription factor) expression in differentiated stem cells. Functional studies showed that miR-133a-3p and miR-145-5p individually or together overexpression can facilitate goat HFSCs differentiation, whereas suppressing miR-133a-3p and miR-145-5p or both inhibiting can inhibit goat HFSCs differentiation. Conclusion: These findings could more completely explain the modulatory function of miR-133a-3p and miR-145-5p in goat HFSCs growth, which also provide more understandings for further investigating goat hair follicle development.

Effect of the hedgehog signaling pathway on hair formation-related cells

  • Alopecia has emerged as one of the biggest interests in modern society. Many studies have focused on the treatment of alopecia, such as transplantation of hair follicles or inhibition of the androgen pathway. Hair growth is achieved through proper proliferation of the components such as keratinocytes and dermal papilla cells (DPCs), movement, and interaction between the two cells. The present study examined the effect of the hedgehog (Hh) signaling pathway, which is an important and fundamental signal in the cell, on the morphology and the viability of human keratinocytes and DPCs. Upregulation of Hh signaling caused a morphological change and an increase in epithelium-mesenchymal transition-related gene expression but reduced the viability of keratinocytes, while the alteration of Hh signaling did not cause any change in DPCs. The results show the possibility that the regulation of Hh signaling can be applied for the treatment of alopecia.
