Effect of Daehwangmokdanpitang on psoriasis-like skin inflammation (건선 모사 피부 염증모델에서 대황목단피탕(大黃牧丹皮湯)의 효능 연구)

  • Objectives: Psoriasis is a common chronic inflammatory skin disease characterized by keratinocyte hyperproliferation and an excessive inflammatory response. Agents that can attenuate keratinocyte hyperproliferation and excessive inflammatory responses are considered potentially useful for the treatment of psoriasis. Daehwangmokdanpitang (DHMDPT) exhibits a broad range of bioactivities, including anti-proliferative and anti-inflammatory effects. This study aims to evaluate the anti-psoriatic potential of DHMDPT in vitro. Methods: HaCaT keratinocytes were stimulated with a mixture of IL-17A, IL-22, oncostatin M, IL-1α, and TNF-α (M5) to establish an in vitro psoriatic keratinocyte model. Cell viability was measured using the MTT assay. Quantitative real-time PCR (qRT-PCR) was performed to measure the mRNA levels of the hyperproliferative marker gene keratin 6 (KRT6) and inflammatory factors such as IL-6, TNF-α, and IL-23A. Additionally, chemokines including CCL5, CCL2, CCL20, and CXCL1 were measured by qRT-PCR. Results: DHMDPT attenuated M5-induced hyperproliferation, as indicated by a reduction in KRT6 expression in HaCaT keratinocytes. M5 stimulation significantly upregulated the mRNA levels of IL-6, TNF-α, and IL-23A. However, DHMDPT treatment attenuated the upregulation of IL-6 but not TNF-α or IL-23A. Additionally, DHMDPT inhibited the expression of CCL5, CCL2, and CXCL1, but not CCL20. Conclusion: DHMDPT effectively attenuated the M5-induced proliferation and inflammatory response in HaCaT keratinocytes. Therefore, DHMDPT could be an attractive candidate for future development as an anti-psoriatic agent.

The Development of Early Mouse Embryos Depend on $Ca^{2+}$ Concentration (Ca$^{2+}$ 농도에 따른 생쥐 초기배의 발생)

  • Objective: This study was to determine the effect of different concentration of calcium III medium on the preimplantational development of zygotes and early 2 cell embryos. Methods: Female mice of ICR strain ($5{\sim}8$ weeks old) were superovulated and mated with fertile males. Zygotes or early 2-cell embryos were collected by flushing the oviducts $31{\sim}32$ hours after hCG injection. The embryos were cultured in various concentrations of $Ca^{2+}$ in medium or with EDTA, EGTA and $Ni^{2+}$. Result and Conclusion: Treatment of high concentraion of $Ca^{2+}$ (3.42 mM $(2X){\sim}17.l$ mM (10X)) in medium didn't develop well compared to the control. Low concentrations of $Ca^{2+}$ (0.214mM $(1/8X){\sim}0.855$ mM (1/2X) were deterimental to development beyond 2-cell stage. EDTA, $Ca^{2+}$ chelating agent was treated with ranged concentrations of EDTA (0.014 $mM{\sim}0.107$ mM) to medium contaning 1.71 mM $Ca^{2+}$ showed beneficial effect to development to blastocyst compared to the control. EGTA, extracellular $Ca^{2+}$ chelator, was treated with ranged concentrations of EGTA ($0.014{\sim}0.107$ mM) to the medium contaning 1.71 mM $Ca^{2+}$. There is no significant difference with the control. $Ni^{2+}$ (50 ${\mu}M$), T-type $Ca^{2+}$-channel blocker was treated to medium contaning low concentration of $Ca^{2+}$. It overcame 2-cell block significantly. Rate of degenerated embryos decreased and developmental rate to morula and blastocyst increased more than low $Ca^{2+}$ concentration alone. Further studies are needed for the overcoming effect of 2-cell block by $Ni^{2+}$.

Protective Effects of Perilla frutescens Britt var. japonica Extracts from Oxidative Stress in Human HaCaT Keratinocytes (HaCaT 피부각질세포에서 들깻잎 추출물의 산화적 스트레스에 대한 항산화 효과)

  • The aim of this study was to investigate the protective effects of methanolic extract from perilla (Perilla frutescens Britt var. japonica) leaves (PLME) on oxidative injury from hydrogen peroxide ($H_2O_2$) in human HaCaT keratinoctyes. Cells were co-incubated with various concentrations (0~200 ${\mu}g/mL$) of PLME for 24 hr, and then exposed to $H_2O_2$ (500 ${\mu}M$) for 4 hr. $H_2O_2$ significantly decreased cell viability (p<0.05). However, PLME provided protection from $H_2O_2$-induced HaCaT cell oxidation in a dose-dependent manner. To further investigate the protective effects of PLME on $H_2O_2$-induced oxidative stress in HaCaT cells, the cellular levels of lipid peroxidation, and antioxidant enzymes (including superoxide dismutase (SOD), glutathione peroxidase (GSH-px) and catalase (CAT)) were measured. PLME decreased cellular levels of lipid peroxidation, and also increased the activities of antioxidant enzymes. In addition, the antioxidant activities of PLME were also determined by DPPH and hydroxyl (${\cdot}OH$) radical scavenging assay, and major antioxidant compounds of PLME were measured by colorimetric methods. DPPH and ${\cdot}OH$ radical scavenging activities of PLME increased in a dose dependent manner and was similar to the DPPH scavenging activity of ascorbic acid at 50 ${\mu}g/mL$; however PLME activities were stronger than ascorbic acid (50 ${\mu}g/mL$) in the ${\cdot}OH$ scavenging assay. The amounts of antioxidant compounds, including total polyphenolics, total flavonoids, and total ascorbic acid from PLME were $52.2{\pm}1.1$ mg gallic acid (GAE)/g, $33.7{\pm}4.7$ mg rutin (RUE)/g, and $17.0{\pm}0.5$ mg ascorbic acid (AA)/g, respectively. These results suggest that PLME has a strong free radical-scavenging activity and a protective effect against $H_2O_2$-induced oxidative stress in the keratinocytes.

Kojic Acid, a Potential Inhibitor of NF-$textsc{k}$B Activation in Transfectant Human HaCaT and SCC-13 Cells

  • The activation of NF-$\kappa$B induced by kojic Acid, an inhibitor of tyrosinase for biosynthesis of melanin in melanocytes, was investigated in human transfectant HaCaT and SCC-13 cells. These two keratinocyte cell lines transfected with pNF-$\kappa$B-SEAP-NPT plasmid were used to determine the activation of NF-$\kappa$B. Transfectant cells release the secretory alkaline phosphatase (SEAP) as a transcription reporter in response to the NF-$\kappa$B activity and contain the neomycin phosphotransferase (NPT) gene for the dominant selective marker of geneticin resistance. NF-$\kappa$B activation was measured in the SEAP reporter gene assay using a fluorescence detection method. Kojic Acid showed the inhibition of cellular NF-$\kappa$B activity in both human keratinocyte transfectants. It could also downregulate the ultraviolet ray (UVR)-induced activation of NF-$\kappa$B expression in transfectant HaCaT cells. Moreover, the inhibitory activity of kojic Acid in transfectant HaCaT cells was found to be more potent than known antioxidants, e.g., vitamin C and N~acetyl-L-cysteine. These results indicate that kojic Acid is a potential inhibitor of NF-$\kappa$B activation in human keratinocytes, and suggest the hypothesis that NF-$\kappa$B activation may be involved in kojic Acid induced anti-melanogenic effect.

Antioxidant activity of Persimmon Leaves during Growth (감잎의 성장시기별 항산화 효과)

  • Kojongsi persimmon (Diospyros kaki) is the major cultivar of astringent persimmon in southern of Korea. Kojongsi persimmon leaf has been traditionally used for acute and chronic diseases in Oriental countries. The purpose of this study was undertaken to investigate the antioxidative activities of the extract of Kojongsi persimmon leaf during growth. We investigated the antioxidant effects of the persimmon leaf extracts during growth on total polyphenol, total flavonoid, electronic donating ability (DPPH), nitrite (NO) scavenging and superoxide dismutase (SOD)-like activity. The next, we investigated the possible cell protective effects of the persimmon extract treatment against ultraviolet B (UVB)-induced injury in HaCaT keratinocytes. The contents of total polyphenol and flavonoid in leaf extract of Kojongsi persimmon were increased in time-dependent manner. In Jun, DPPH and NO radical scavenging and SOD-like activities in the leaf extract of Kojongsi persimmon was increased to the highest. However, the antioxidant activities in persimmon varieties were not any difference. The cell cytotoxicity by UVB irradation in HaCaT keratinocytes was significantly increased with the compared to the control group. However, the treatment of leaf extract of Kojongsi persimmon in HaCaT keratinocytes was shown to protective effect against UVB-induced cell cytotoxicity. These results suggest that the leaf extract of Kojongsi persimmon has potent antioxidant activity, and protective effect against UVB-induced keratinocyte injury. Thus, these properties may be contributed in the care of acute and chronic diseases.

Effect of Pulsed Electromagnetic Field Stimulation on TNF-α/IFN-γ induced inflammatory response in human skin keratinocytes HaCaT Cell to reduce inflammatory factors (인간 피부각질세포 HaCaT Cell에서 TNF-α/IFN-γ로 유도된 염증 반응에 대한 펄스형 전자기장(PEMF) 자극의 염증 인자 완화 효과)

  • The purpose of this study was to observe cell death in human keratinocytes stimulated against the infectious cytokines TNF-α and IFN-γ, and to observe the expression of Phospho-NF-κB due to phosphorylation of IkB to confirm the mechanism of inhibiting the expression of inflammatory cytokines. As a result of cell viability analysis, differences in PEMF stimulation time were observed little by little after 1 hour, 3 hours, 6 hours, 12 hours, 24 hours, and 48 hours, but there was no statistical significance according to PEMF stimulation time for each time (p>0.05). No significant difference was observed in the total amount of NF-κB present in the cytoplasm and nucleus, but a significant decrease in the expression of phosphorylated NF-κB was observed in the group exposed to PEMF stimulation for 24 hours (*p<0.05). The expression of IL-1β was observed in all inflammation-induced groups, and the concentration of IL-1β compared to α-Tubulin expression was reduced by about 54% in the PEMF-stimulated group for 24 hours compared to the control group (***p<0.001). As a result of the study, it is shown that PEMF stimulation does not negatively affect HaCaT cells from 0 to 48 hours and can inhibit the expression of inflammatory cytokines by inhibiting the pathway of NF-κB.

Inhibitory effect of mushrooms extract on TNF-α/INF-γ induced-cytokine in human keratinocytes, HaCaT (버섯류 추출물의 피부각질세포(HaCaTcell) 내 염증성 사이토카인 억제효과)

  • Mushroom is known for anti-inflammatory and anti-oxidative potential. This study provides evidence that the inhibitory effect of mushroom on the expression of pro-inflammatory cytokines in human keratinocytes, HaCaT cells. To define the underlying mechanisms of action, tumor necrosis factor${\alpha}$/$IFN{\gamma}$-activated human keratinocytes model was used. Mushroom significantly inhibited the expression of cytokines in HaCaT cells. Taken together, the results demonstrate that mushroom inhibited inflammtion, suggesting that mushroom (DW extract: Grifola frondosa Cordyceps militaris), (Ethanol extract: Ganoderma lucidum, Lentinus edodes, Cordyceps militaris, Flammulina velutipes) might be a candidate for the treatment of skin inflammation.

The Experimental Study of Jeondo-san on Antioxidant Effects (전도산(顚倒散)의 항산화 효과에 관한 실험적 연구)

  • Objectives : This study was carried out to investigate the antioxidant effects of Jeondo-san(JDS). Methods : The antioxidant effects of JDS were measured by the scavenging for 2,2-Diphenyl-1-picrylhydrazyl(DPPH) radical, the formation of intracellular glutathione(GSH), the inhibition for reactive oxygen species(ROS). Results : 1. All concentrations of JDS showed antioxidant effect by decreasing the DPPH radicals. 2. All concentrations of JDS did not effect on the formation of intracellular GSH in HaCaT cell. 3. All Concentrations of JDS inhibited the production of ROS in the HaCaT cell stimulated with $H_2O_2$. Conclusion : The present date suggest that JDS has effects on the stage of inflammation.

4-O-Methylhonokiol Protects HaCaT Cells from TGF-β1-Induced Cell Cycle Arrest by Regulating Canonical and Non-Canonical Pathways of TGF-β Signaling

  • 4-O-methylhonokiol, a neolignan compound from Magnolia Officinalis, has been reported to have various biological activities including hair growth promoting effect. However, although transforming growth factor-${\beta}$ (TGF-${\beta}$) signal pathway has an essential role in the regression induction of hair growth, the effect of 4-O-methylhonokiol on the TGF-${\beta}$ signal pathway has not yet been elucidated. We thus examined the effect of 4-O-methylhonokiol on TGF-${\beta}$-induced canonical and noncanonical pathways in HaCaT human keratinocytes. When HaCaT cells were pretreated with 4-O-methylhonokiol, TGF-${\beta}1$-induced G1/G0 phase arrest and TGF-${\beta}1$-induced p21 expression were decreased. Moreover, 4-O-methylhonokiol inhibited nuclear translocation of Smad2/3, Smad4 and Sp1 in TGF-${\beta}1$-induced canonical pathway. We observed that ERK phosphorylation by TGF-${\beta}1$ was significantly attenuated by treatment with 4-O-methylhonokiol. 4-O-methylhonokiol inhibited TGF-${\beta}1$-induced reactive oxygen species (ROS) production and reduced the increase of NADPH oxidase 4 (NOX4) mRNA level in TGF-${\beta}1$-induced noncanonical pathway. These results indicate that 4-O-methylhonokiol could inhibit TGF-${\beta}1$-induced cell cycle arrest through inhibition of canonical and noncanonical pathways in human keratinocyte HaCaT cell and that 4-O-methylhonokiol might have protective action on TGF-${\beta}1$-induced cell cycle arrest.

Antimicrobial and Immunological activities of Vinca minor Extracts (빈카 마이너 추출물의 항균 및 면역활성 연구)

  • Vinca alkaloid compounds including vincamine from Vinca minor L. have been extracted by ethanol and hot water extraction methods. Antimicrobial properties of those extracts have been investigated against dandruff causing microorganism Malassezia furfur, yeast, Gram positive and negative bacteria. Vincamine standard and ethanol extract showed no sign of antimicrobial activity, whereas hot-water extract had the activity against M. furfur. Furthermore hot water extract had antimicrobial activity against Gram positive Bacillus sp. and Gram negative Escherichia coli. Cytotoxic properties of those extracts have also been investigated with HaCaT cell (human keratinocyte), HT-29 cell (human colorectal adenocarcinoma cell) and Raw cell, showing no significant cytotoxic effects. We also measured the ROS using dichlorofluorescein diacetate (DCFDA), a popular fluorescence-based probe for reactive oxygen species detection. The result showed the increasement of reactive oxygen species formation (20%) in HaCaT and HT-29 cell lines.
