고온내성 연료용 알코올 효모균주 Saccharomyces cerevisiae KNU5377에서 HSF1 유전자의 변이주 구축 (Construction of hsf1 Knockout-mutant of a Thermotolerant Yeast Strain Saccharomyces cerevisiae KNU5377)

  • 출아효모인 Sacharomyces cerevisiae S288C균주를 이용한 효모의 게놈이 완성된 후 S. cerevisiae는 다양한 연구 모델로 이용되어져 왔다. 현재까지 효모를 이용한 기능 유전체학 측면에서의 연구는 laboratory strainin인 S288C 균주 또는 그 유래의 균주들이다. 그러나 자연에서 분리된 효모 또는 산업적으로 이용되어지고 있는 S. cerevisiae의 유전학 측면에서의 연구는 낮은 포자형성률 및 형질전환률, 그리고 S288C 균주와의 게놈상의 상이성 때문에 거의 이루어지지 않고 있다. 여기서 우리 연구진은 자연에서 분리된 Saccharomyces cerevisiae KNU5377 균주를 이용하여 random spore analysis를 통해 MATa 및 $MAT{\alpha}$ 타입의 각각의 haploid cell을 분리 후 이미 보고된 KanMX module를 가지고 round PCR기법에 의한 short flanking homology 기법을 이용하여 전사조절인자인 HSF1 유전자가 치환된 변이주를 구축할 수 있었다. 덧붙여, 모든 유전자에 이 기법을 적용할 수는 없다는 것을 확인하였다. 앞으로 이 변이주를 통해 기능 유전체학적인 측면에서 이 유전자의 스트레스와의 관련성을 연구하고자 한다.

조기경보시스템 검증을 위한 무인기상관측망 실황자료 표출 시스템 (A System Displaying Real-time Meteorological Data Obtained from the Automated Observation Network for Verifying the Early Warning System for Agrometeorological Hazard)

  • 농촌진흥청 농업기상재해 조기경보시스템은 기상청으로부터 제공되는 기상정보를 활용하여 농장 단위로 상세 추정하고, 추정된 상세 기상정보를 바탕으로 작물의 생육 추정 및 생육이 진행됨에 따라 발생할 수 있는 기상 재해를 예측하여 사용자에게 미리 전달한다. 이들 예측 정보를 검증하기 위한 무인기상관측망을 연구 지역 내에 구축하였으며, 관측망으로부터 수집되는 기상 실황 자료의 실시간 웹 표출 시스템을 구축하였다. 기상관측장비로부터 수집되는 기상요소로는 기온, 습도, 일사량, 강우량, 토양수분, 일조시간, 풍속, 풍향 등이며, 1분단위로 수집 및 10분 간격으로 서버로 전송된다. 자료 표출 시스템은 기상관측장비로 부터 수집되는 1분 단위의 기상자료를 DB로 구축하는 1단계, 수집된 기상자료를 10분, 1시간, 1일 단위로 통계 분석하는 2단계, 수집 및 분석한 기상자료를 웹으로 표출하는 3단계로 구성된다. DB에 수집된 기상자료는 웹 페이지를 통해, 전체 지점 또는 1개 지점의 1분단위, 10분단위, 1시간 단위, 1일 단위로 조회할 수 있으며, CSV 포맷으로 다운로드 할 수 있다. 자료 표출 시스템 접속 URL은 http://aws.agmet.kr 이다.

난임부부의 한방의료기관 이용 실태 및 한방난임치료 지원사업 수요에 관한 설문조사연구 (A Survey Study on Use Condition of Korean Medical Institution and Demand of Korean Medical Treatment Project of Infertility in Infertile Couple)

  • Objectives: The purpose of this study is to investigate use condition of Korean medical institution and demand of Korean Medical Treatment Project of Infertility in infertile couple. Methods: Questionnaires were distributed to all members of the incorporated association AGAYA(http://www.agaya.org/) via e-mail and to infertile women who visited to Korean medicine OB & GY outpatient clinic of university Korean medicine hospitals of the country and an infertility specialized Korean medicine hospital. One hundred twenty four of completed questionnaires were analyzed. Results: 1. Korean medical institution utilization rate of infertile couple was 89.6% and was similar with western medical institution utilization rate of 87.1%. 2. 96.8% of infertile couple answered that Korean Medical Treatment Project of Infertility is necessary. And 90.3% of infertile couple answered that herbal medicines should be supported first of all. 3. 90.3% of infertile couple answered that they will participate if Korean Medical Treatment Project of Infertility go ahead. Conclusions: It was confirmed that most of the infertile couple have been received Korean medical treatment for infertility, and think that Korean Medical Treatment Project of Infertility is necessary.

초등학생 대상 영양교육 웹사이트의 활용효과 (Evaluation of a Nutrition Education Website for Children)

  • This study was carried out to evaluate the usefulness and the effectiveness of the nutrition education website 'ifood(http://ifood.or.kr)', for children. A total of 262 children (5th and 6th graders) participated in the self-education program using the website which consisted of 4 sessions every two weeks. In each session, they were asked to answer some questions on the comprehensiveness of each topic while navigating the website. Nutrition knowledge (20 items), dietary attitudes (7 items), and dietary behaviors (10 items) were evaluated using questionnaires before and after the education. After the education, the usefulness and readability of the website was also evaluated. Most (97.2%) of the responders reported that the education using this website was helpful, 97.3% thought that the contents were reliable, 90.1% responded that the contents were easy to understand, and 88.8% answered that they would revisit 'ifood'. The menus they liked best were 'Interesting games' for 35.8%, 'How can I cook?' for 17.8%, 'How can I become slim' for 16.4%. This website might be more attractive by providing a 'greater variety of contents in detail', 'more music, motion pictures and images', 'more interesting games'. After the education, there was a significant increase in nutrition knowledge scores from 10.9 to 13.1. Although the total scores of dietary attitudes and dietary behaviors did not improve, there was a significant increase in each of some questions. These results showed that 'ifood', a nutritional education website, was desirable in reliability, aesthetics and enjoyability, and was effective in improving nutrition knowledge and some of dietary attitudes and behaviors in children.

중의학 변증과 양방 검사의 상관관계 연구 현황 -CNKI를 이용하여- (A Review Study in the Correlation between Pattern Identification of Traditional Chinese Medicine and Western Medicine Examination -Research on CNKI-)

  • Objectives : The purpose of this study is to investigate the correlation between pattern identification of traditional Chinese medicine (TCM) and western medicine, examined by a systematic research of Chinese medicine papers. Methods : We searched for the papers regarding pattern identification of TCM published from 1994 to 2012 in CNKI (China National Knowledge Infrastructure http://www.cnki.net) at April, 2012. Results : A total of 30 studies were finally included; 18 studies of them were related to stroke (cerebral infarction) and there were 12 studies regarding other diseases, such as hypertension, chronic colonitis, vascular dementia, mild cognitive impairment and etc. All 30 studies were analyzed and classified by diseases, differentiation of syndromes, numbers of subjects, the instrument of pattern identification, items of western medicine examination and statistical results. Conclusions : According to our study, there are some correlations between pattern identification of TCM and various items of western medicine examination. The result suggests a possibility of using the western medicine examination data for pattern identification of TCM.

한반도 특산식물 꼬리말발도리의 분포와 생육환경특성 (Geographical distribution range and growth environmental characteristics of Deutzia paniculata Nakai, a Korean Endemic Plant)

  • The current status of geographical distribution range of Deutzia paniculata, one of the korean endemic plants, was investigated based on the species distribution information. The information of the geographical range used for analysis includes all the published references to the distribution of D. paniculata, herbarium specimens in Korea National Arboretum (KH) including its online database: http://www.nature.go.kr, and field research. Although, D. paniculata was firstly recorded in Wonsan, Hamgyeongnam-do, North Korea, this research revelaved that D. paniculata is mainly distributed in Gyeongsangbuk-do and Gyeongsangnam-do, southern regions of the Korean Peninsula. According to the distribution map, this species was estimated on distribution edge of in Anyang region of Gyeonggi-do, Mt. Naejang of Jeollabuk-do and Mt. Taebaek of Gwangwon-do on the distribution edge. However, it was made use of intensive field survey to identify the natural population of the species in these regions. D. paniculata habitat was mainly distributed between 290 meters and 491 meters in altitude, but it was also found ranged from 936 to 959 meters in Gun-wi, Gyeongsangbuk-do, South Korea. The distribution slope was 5° to 35° and the north, N, NE and NW in the aspect. The flora of vascular plants in D. paniculata habitats was listed in 137 taxa: 54 families, 103 genera, 120 species, 2 subspecies, 12 varieties and 3 forms while Korean endemic plants were found in three taxa, Stewartia koreana, Weigela subsessilis and D. Paniculata. Based on a collection of non-biological environment and biological environment data, D. paniculata population were discussed to make strategy and basic research methods for sustainable preservation.

하천 일부 구간에서의 수변림 조성에 따른 수질 개선 효과 모의를 위한 SWAT-REMM ArcView GIS 시스템의 개선 및 평가 (Enhancement of the SWAT-REMM System for Simulation of Water Quality Improvement with Riparian Buffer System)

  • 유역내 비점오염 부하량의 증가로 인하여 그동안 많은 수질 개선 노력이 진행되어 왔다. 그 중 하나의 대안으로 수변림 조성이 거론되고 있으나, 현재 수변림 조성에 따른 수질 개선 효과 평가 방법의 부재로 유역내 어느 곳에 어떠한 식생을 조성해야 할지 정확한 판단을 하기 어려운 실정이다. 이러한 문제를 해결하기 위하여 SWAT-REMM Prototype 버전이 개발되었으나, 다른 유역에 적용하기에는 여러 가지 문제점을 나타내고 있다. 또한 SWAT-REMM Prototype 버전은 소하천내 모든 유역내 수변림을 조성한다는 가정하에 수변림 조성에 따른 수질개선효과를 모의 분석하는데 활용될 수 있다. 그러나 국내 상당부분의 하천에서는 여름철 집중강우로 인한 수해 발생으로 많은 구간이 석축과 같은 정비사업이 진행되어 왔다. 이러한 구간에 대해서 수질 개선을 목적으로 수변림을 조성한다는 것은 현실적으로 불가능하다. 따라서 SWAT-REMM 시스템을 이용하여 유역 전체 모든 하천이 아닌, 하천 일부구간에 대해서 수변구역을 조성에 따른 효과 분석을 수행할 수 있는 기능에 대한 많은 요구가 있어 왔다. 이에 본 연구에서는 SWAT-REMM Prototype 버전의 여러 가지 기능을 수정 보완하였으며, 도암댐 유역내 모든 하천에 대해서 수변림 조성에 따른 수질 개선 효과를 모의 분석하였다. 수변림 조성은 1차 하천 (지천)에 대해서는 하천 양안 1m 정도의 초생대를 조성한다고 가정하였고, 최고 차수 하천인 도암댐 유입 송천에 대해서는 하천 양안 10m 의 수변림을 조성한다고 가정하여 수질 개선효과를 분석한 결과 상당량의 유사가 하천으로 유입되기 전 유역내 설치된 수변림에 의해서 저감되는 것으로 모의 분석되었다. 본 연구에서는 분석한 수변림 조성 시나리오는 현실적으로 실행하기 어려운 점이 있기 때문에 수변림 조성 적지 분석후, 본 연구에서 개발된 하천 구간 수변림 조성 시나리오 모듈을 이용한다면, 보다 현실적인 수변림 조성에 따른 유사저감효과를 평가할 수 있을 것으로 기대된다.(SWAT-REMM ArcView GIS: http://www.EnvSys.co.kr/$\sim$swat-remm)

  • Recent advances in high-throughput genotyping technologies have enabled us to conduct a genome-wide association study (GWAS) on a large cohort. However, analyzing millions of single nucleotide polymorphisms (SNPs) is still a difficult task for researchers conducting a GWAS. Several difficulties such as compatibilities and dependencies are often encountered by researchers using analytical tools, during the installation of software. This is a huge obstacle to any research institute without computing facilities and specialists. Therefore, a proper research environment is an urgent need for researchers working on GWAS. We developed BioSMACK to provide a research environment for GWAS that requires no configuration and is easy to use. BioSMACK is based on the Ubuntu Live CD that offers a complete Linux-based operating system environment without installation. Moreover, we provide users with a GWAS manual consisting of a series of guidelines for GWAS and useful examples. BioSMACK is freely available at http://ksnp.cdc.go.kr/biosmack.

Detection of Quantitative Trait Loci Affecting Fat Deposition Traits in Pigs

  • Quantitative trait loci (QTL) associated with fat deposition traits in pigs are important gene positions in a chromosome that influence meat quality of pork. For QTL study, a three generation resource population was constructed from a cross between Korean native boars and Landrace sows. A total of 240 F2 animals from intercross of F1 were produced. 80 microsatellite markers covering chromosomes 1 to 10 were selected to genotype the resource population. Intervals between adjacent markers were approximately 19 cM. Linkage analysis was performed using CRIMAP software version 2.4 with a FIXED option to obtain the map distances. For QTL analysis, the public web-based software, QTL express (http://www.qtl.cap.ed.ac.uk) was used. Two significant and two suggestive QTL were identified on SSC 6, 7, and 8 as affecting body fat and IMF traits. For QTL affecting IMF, the most significant association was detected between marker sw71 and sw1881 on SSC 6, and a suggestive QTL was identified between sw268 and sw205 on SSC8. These QTL accounted for 26.58% and 12.31% of the phenotypic variance, respectively. A significant QTL affecting IMF was detected at position 105 cM between markers sw71 and sw1881 on SSC 6.

수술실의 간호오류 및 과오 예방을 위한 E-learning 실무교육 프로그램의 개발 및 평가 (Development of an E-learning Education Program for Preventing Nursing Errors and Adverse Events of Operating Room Nurses)

  • Purpose: This study was to develop, implement, and evaluate an e-learning education program for improving practical knowledge and preventing nursing errors and adverse events of nurses working in the operating room (OR). Method: The e-learning program was developed and evaluated according to the following processes: 1) preparation phase 2) implementation phase 3) evaluation phase. In evaluation phase, the effectiveness was analyzed based on the Kirkpatrick's model. Results: The e-learning program consisted of OR basic nursing skills and techniques and nursing activities' manual based on the categories of nursing errors: surgical operation preparation, nursing skills and techniques, environment management, patient safety and comfort, and patient monitoring. The program was provided through on-line, http://cafe.daum.net/pnuhorn, for 4 weeks. The mean score(percent) of participants' satisfaction was $21.24{\pm}1.71$(68.2%). Their total knowledge level was significantly improved(Z=-3.00, p=.003) and specifically in the category of environment management(Z=-3.77, p<.001) and patient monitoring(Z=-2.46, p=.014). The occurrence of nursing errors or adverse events was a little decreased, but not statistically significant(Z=-3.10, p=.756). Conclusion: E-learning for nurses is one way of effective and efficient teaching-learning strategies. For better e-learning, it is important to develop the vital content of the education and objective measures for detecting nursing errors and adverse events.
