trans-(3R,5S)-Atorvastatin Ca의 합성 및 Rat에서 고지방식이로 유도된 고지혈증 치료효과 (Synthesis of trans-(3R,5S)-Atorvastatin Ca and Curative Effect on Hyperlipidemia Induced by a High-Fat Diet in Rats)

  • 고지혈증 치료제인 cis-(3R,5R)-atorvastatin Ca (1)은 4개의 입체이성질체가 있으며, 각각의 이성질체들을 선택적으로 제조한다는 것은 매우 어려운 일이다. 본 연구에서는 입체이성질체 중의 하나인 trans-(3R,5S)-atorvastatin Ca (7)을 초산에서 3,5-diketo atorvastatin ester (3)를 $Me_4NHB(OAc)_3$을 사용하여 환원시켜 cis-(3R,5R) (4)과 trans-(3R,5S)-atorvastatin ester (5)를 각각 1.5%와 98.5%의 비율로 입체선택적으로 제조할 수 있었다. 또한, 고지혈증 치료제인 cis-(3R,5R)-atorvastatin Ca (1)과 그의 입체이성질체인 trans-(3R,5S)-atorvastatin Ca (7)을 쥐에서 고지방식이에 의해 유발된 고지혈증의 치료효과에 대하여 알아보았고, 이러한 연구의 수행을 위해 2010년 1월에 실험을 실시하였다. 그 결과, 화합물 1과 7의 total cholesterol (TC), high-density lipoprotein-cholesterol (HDL-c), low-density lipoprotein-cholesterol (LDL-c)과 triglyceride (TG)는 각각 $93.0{\pm}0.5$, $43.5{\pm}0.8$, $40.4{\pm}1.4$, $45.6{\pm}0.9\;mg/d{\ell}$$110.0{\pm}0.7$, $33.3{\pm}0.6$, $65.8{\pm}1.9$, $54.8{\pm}1.2\;mg/d{\ell}$를 atherogenic index (AI)와 cardiac risk factor (CRF)는 $1.14{\pm}0.05$ $2.14{\pm}0.05$$2.31{\pm}0.06$, $3.31{\pm}0.06$을 나타냈으며 aspartate aminotransferase (AST)와 alanine aminotransferase (ALT)는 $51.9{\pm}4.6$, $16.0{\pm}2.1\;IU/{\ell}$$75.8{\pm}4.4$, $35.1{\pm}9.7\;IU/{\ell}$로 두 화합물 모두 고지혈증에 대한 치료효과를 나타내었으나, 화합물 1이 우수한 치료효과가 있는 반면 화합물 7은 치료효과가 낮은 것을 확인하였다. 이러한 결과들로 입체선택적 이성질체의 합성에 대한 새로운 방법을 제시하고, 향후 시판되고 있는 의약품들의 입체이성질체에 대한 임상적 활용이 가능할 것으로 사료된다.

고지방 식이 유도 비만 마우스에서 고올레산 땅콩 섭취의 혈장 지질 개선 효과 (Effect of Monounsaturated Fatty Acid-enriched Peanut Consumption on Serum Lipid in High Fat Diet-induced Mice)

  • This study was conducted to evaluate the effect of high-oleate and normal-oleate peanut consumption on adipose mass and serum lipids in obese-induced C57BL/6J mice. After four weeks of the high-fat diet, mice were randomly divided into six groups: normal control (NC) diet, high-fat control (HFC) diet, high-oleate peanut-seed (HOPS) diet, normal-oleate peanut-seed (NOPS) diet, high-oleate peanut-oil (HOPO) diet, and olive-oil (OO) diet. After four weeks, all four experimental diet groups showed significantly lower body weight and epididymal fat weight than HFC group. In four experimental diet groups, serum triglycerides (TG), total cholesterol (TC) and low-density lipoprotein cholesterol (LDL-C) were significantly lower, and high-density lipoprotein cholesterol (HDL-C) was significantly higher than HFC group. TG was significantly decreased in HOPS group (92.1±1.2 mg/dL) than NOPS group (101.7±5.3 mg/dL, p<0.05). Similarly, LDL-C was significantly lower in HOPS group (66.1±2.8 mg/dL) than NOPS (76.9±1.5 mg/dL, p<0.05), on the other hand, HDL-C indicated a significant elevation in HOPS (50.5±2.1 mg/dL) than NOPS group (45.2±1.6 mg/dL, p<0.05). This result suggests that the consumption of high-oleate peanut has a favorable effect on the plasma lipid profile.

쌀 배아와 흑미 미강 색소 첨가 배아젤리가 고콜레스테를 식이 흰쥐의 지질대사와 항산화효소 활성에 미치는 영향 (Effects of Rice Embryo and Embryo Jelly with Black Rice Bran Pigment on Lipid Metabolism and Antioxidant Enzyme Activity in High Cholesterol-Fed Rats)

  • 본 연구는 쌀 배아를 활용한 만성 대사성질환 예방용 제품 및 건강기능성 식품 개발의 일환으로써 고콜레스테롤 식이를 급여하여 고지혈증을 유발시킨 흰쥐에서 15%의 배아와 25%의 흑미 미강색소 배아젤리의 첨가가 혈장과 간 조직의 지질 대사와 항산화효소의 환성에 미치는 영향을 살펴보았다. 실험 식이를 6주 간 급여한 결과, 배아와 흑미 미강색소 배아젤리 첨가는 실험동물의 식이섭취에 영향을 미치지 않았다. 배아군과 흑미 미강색소 배아젤리군은 고콜레스테롤 급여 대조군에 비해 혈장의 총 콜레스테롤과 LDL-콜레스테롤 및 간의 중성지방과 총 콜레스테롤 농도를 감소시키고, HDL-콜레스테롤 농도와 HDL-C/TC 비는 증가시켰으며, 동맥경화지수는 감소시켜 체내 지질대사의 개선 효과가 있었다. 혈장 GOT와 GPT 수치는 배아와 흑미 미강색소 배아젤리를 첨가하였을 때 감소하여 고콜레스테롤혈증 상태에서 간 기능 보호에 긍정적인 효과가 있었다. 또한 배아와 흑미 미강색소 배아젤리는 고콜레스테롤 급여로 인해 증가된 혈장과 간 내의 지질과산화를 억제시키는 효과가 있었다. 반면, 항산화 효소인 간 조직의 SOD와 CAT활성은 배아와 배아젤리 첨가에 따라 증가하였다. 이상의 결과로 볼 때 배아와 흑미 미강색소 배아젤리는 고콜레스테롤 식이 흰쥐의 간 조직에서의 항산화 활성을 강화시키고 산화적 손상을 억제시키는 자용이 있으며 현장과 간조직의 지질대사를 개선하여 심혈관계 질환을 예방 및 감소시킬 수 있을 것으로 사료된다.

일부 대학생들의 타우린 섭취가 생화학적 및 혈액학적 검사에 미치는 영향 (The Effect of Taurine Intake among Korean College Students: Serum Biochemistry and Blood Hematology)

  • 타우린은 심혈관 질환을 예방하고 간 기능 개선 및 당뇨병 및 혈소판 기능을 향상시키는 여러 작용을 보고하고 있다. 하지만, 타우린이 인체에 미치는 영향에 대하여 우리나라에서 연구된 결과가 많지 않다. 이에 기본 용량을 복용 후 혈당질환과 간질환, 지질 질환에 대한 영향을 알아보고자 하였다. 대상자는 타우린 복용군 15명과 대조군 15명을 대상으로 시행하였다. 타우린은 기본 용량인 1,000 mg을 식후에 2주 동안 복용 후 변화를 확인하였다. 대상자 모두 기숙사에서 제공하는 식사 외에 약이나 기타 음식을 절제하도록 하였다. 그 결과 타우린 복용군에서 간 기능 검사인 GGT는 섭취 전 $23.53{\pm}25.73IU/L$, 섭취 후 $15.15{\pm}4.91IU/L$로 감소하였다(P=0.186). 지질 대사인 TG는 섭취 전 $100.4{\pm}28.33mg/dL$, 섭취 후 $80.22{\pm}17.08mg/dL$로 유의한 결과를 보였다(P<0.05). T-cho, LDL-C이 감소를 보였고, HDL-C이 약간 증가를 보였다. 결과적으로 간 기능과 지질대사 개선에 도움을 주는 것으로 나타났다. 혈액학적 검사에서는 segmented neutrophil 백분율이 감소하고, lymphocyte 백분율이 증가를 보였다. 결과적으로 면역학적 기능과 관계가 있으리라 사료된다.

석류·참다래 혼합추출물의 체지방 감소에 대한 유효성 및 안전성을 평가하기 위한 8주, 무작위 배정, 이중눈가림, 위약-대조 인체적용시험 (A 8-Week, Randomized, Double-Blind, Placebo-Controlled Human Trial to Evaluate the Efficacy and Safety of Punica granatum L.·Actinidia chinensis Planch. Mixed Extract on Body Fat)

  • Objectives: This study was conducted to evaluate efficacy and safety of Punica granatum L. Actinidia chinensis Planch. mixed extract on body fat. Methods: Thirty women were recruited ($25kg/m^2{\leq}body$ mass index [BMI]${\leq}29.9kg/m^2$) ($19yr{\leq}age{\leq}38yr$) and randomized to receive Punica granatum L. Actinidia chinensis Planch. mixed extract or placebo for 8 weeks. During the test, they visited four times including screening. Body fat mass, body fat rate, anthropometric dimensions at waist and hip, waist-hip ratio, total cholesterol, triglyceride, low density lipoprotein-cholesterol (LDL-C), and high density lipoprotein-cholesterol (HDL-C) were measured at screening and 8 weeks after screening. Adverse reaction were also checked each visit. Results: There were no significant differences in all index. But when divide experimental group by BMI index, age, and compliance, less than BMI $27kg/m^2$ group decreased body weight, body fat mass and more than BMI $27kg/m^2$ group also decreased body weight, body fat mass and waist circumference. Less than age 30 years group showed decline of waist circumference, fat free rate and T-cholesterol and more than age 30 years group showed decline of fat free mass. Group with compliance more than 95% was decreased BMI, fat free mass, and hip circumference. Conclusions: There was no significant efficacy as compared with placebo group, but as divide experimental group according to BMI index, age, compliance, mixed extract might be effective to reduce obesity index.

손목굴증후군 환자의 신경생리학적 소견과 혈청 지질 수치와의 상관관계 (Correlation between Serum Lipid Level and Neurophysiological Findings in Patients with Carpal Tunnel Syndrome)

  • Background: Carpal tunnel syndrome (CTS) is the most common entrapment neuropathy caused by compression of the median nerve beneath the transverse carpal ligament. CTS can be correctly diagnosed by the patients' description of symptoms and electrophysiological tests that measure nerve conduction through the wrist. Many previous studies reported various risk factors of CTS, such as obesity, diabetes mellitus, thyroid disease and trauma. Obesity is associated with both hyperlipidemia and CTS. This study focused on the relationship between severity of CTS and serum lipid level. Methods: One hundred fourteen patients with CTS and 74 controls were divided into four groups according to the severity; normal, mild, moderate and severe. And then serum total cholesterol (TC), triglyceride (TG), low-density lipoprotein (LDL-C) and high-density lipoprotein (HDL-C) were measured in each group. Results: There was a positive correlation between TG and CTS severity (p<0.001). But TC, LDL-C and HDL-C were not correlated with CTS severity. Conclusions: These results suggest that high serum TG may act as an aggravating factor of CTS.

타이치 운동이 중년여성의 대사증후군 위험인자 및 건강관련 삶의 질에 미치는 영향 (The Effects of the Tai Chi Exercise on Metabolic Syndrome and Health-related Quality of Life in Middle-aged Women)

  • Purpose: The purpose of this study was to test the effects of the Tai Chi exercise on metabolic syndrome and health-related quality of life in middle-aged women. Methods: A quasi-experimental design was used. Subjects were sixty middle-aged women with metabolic syndrome. All of the subjects were met the criteria of the National Cholesterol Education Program-Adult Treatment Panel III (NCEP-ATP III). The subjects were divided into the experiment group (n=33) trained the Tai Chi for 12 weeks and the control group (n=27). Metabolic syndrome risk factors including blood pressure, waist circumference, glucose, triglyceride (TG), and high density lipoprotein cholesterol (HDL-C) were measured before and after the 12-week period. Euro Quality of Life Questionnaire 5-Dimensional Classification (EQ-5D) was used to evaluate the health-related quality of life. Results: The experiment group showed significant decreases in diastolic blood pressure, waist circumference, glucose, and TG; and increase in HDL-C compared to the control group. For the health-related quality of life evaluation, the experiment group showed significant improvement more than the control group. Conclusion: The Tai Chi exercise may be effective intervention in preventing cardiovascular disease caused by metabolic syndrome in middle-aged women.

The effects of physical training on antioxidative status under exercise-induced oxidative stress

  • This study investigated the effect of physical training and oxidative stress on the anti oxidative activity and on plasma lipid profile. Forty eight rats were given either a physical training or no training for 4 weeks and were then subdivided into 3 groups: before-exercise (BE); during-exercise (DE); after-exercise (AE). The antioxidative activity was evaluated with the activities of catalase in plasma and superoxide dismutase (SOD), the ratio of reduced glutathione/ oxidized glutathione (GSH/GSSG) and the level of malondialdehyde (MDA) in liver. The plasma concentrations of triglyceride (TG), total cholesterol (TC), high-density lipoprotein-cholesterol (HDL-C)) were also compared. Compared to those of non-training group. catalase activities of training group were lower before exercise but higher during and after exercise. SOD activities were higher regardless of exercise. GSH/GSSG ratio was higher before exercise but was not significantly different during exercise and even lower after exercise. There were no differences between non-training group and training group in MDA levels regardless of exercise. Compared to those of non-training group, atherosclerotic index of training group was lower after exercise and there were no significant differences before and during exercise. There were no differences between non-training group and training group in HDL-C regardless of exercise. These results suggest that moderate physical training can activate antioxidant defenses and decrease the atherosclerotic index and this beneficial effect is evident under exercise-induced oxidative stress.

비타민 C 보충이 인슐린 비의존형 당뇨병 환자의 혈당 및 혈청지질에 미치는 영향 (Effect of Vitamin C Supplementation on Blood Sugar and Serum Lipid in NIDDM Patients)

  • The purpose of this study is to determine the effect of oral vitamin C supplements on blood sugar and serum lipid level(total cholesterol, triglyceride, high density lipoprotein, low density lipoprotein) in non-insulin independent diabetes mellitus. The study design was a non equivalent control group pre-test post-test design. Data for the study were collected from June 24 to August 31, 2001. The ninty-five research subject were assigned to experimental group(51) and control group(44). Vitamin C(3g/day) was given to 51 subjects for 4 weeks. Following a 12h overnight fasting, blood sample was obstaind at baseline and at the end of 4week - supplementation. Blood samples were taken for plasma vitamin C concentration, fasting blood sugar, HbA1c and serum lipid level. The pre-equivalent test was used by Chi-squre, t-test and two group's pre and post experimental differences were analyzed by t-tset to compare with each other. The results of this study were as follows ; 1. The difference between the two groups in plasma vitamin C concentration was significant(t=-12.950, p=.000). 2. The difference between the two groups in fasting blood sugar was significant(t=5.293, p=.000). 3. The difference between the two groups in HbA1c was not significant(t=1.758, p=.082). 4. The difference between the two groups in total cholesterol, HDL, LDL was significant(t=3.786, p=.000 ; t=-5.515, p=.000 ; t= 4.169, p = .000). These results suggest that megadose vitamin C supplementation be effective in lowering fasting blood sugar, serum lipids and increasing plasma vitamin C. Thus dietary measures to increase plasma vitamin C may be on important health strategy for reducing the compliance of diabetic patients.

잣 성분의 혈중 콜레스테롤 저하효과 (Cholesterol-Lowering Effect of Pine Nut in Plasma of Rats)

  • 잣의 혈중 콜레스테롤 저하효과를 동물실험을 통하여 확인하기 위하여 기본식이(정상군), 기본식이에 콜레스테롤을 첨가한 식이(대조군), 그리고 기본식이에 콜레스테롤과 여러 농도의 잣을 함유한 실험식이(실험군)를 각 군별로 섭취하게 한 결과 잣의 농도에 따른 식이섭취량과 체중변화량의 유의적인 차이는 크게 나타나지 않았다. 식이효율은 저농도로 잣을 섭취한 경우에는 대조군과 차이를 나타내지 않았지만 10% 이상의 고농도의 잣을 섭취한 경우에는 식이효율이 높았다. 본 연구에 사용한 농노범위(1-20%)에서는 잣 섭취에 의해 혈중 콜레스테롤 함량과 동맥경화지수가 감소하는 효과가 나타나지 않았으며 이는 잣에 함유된 다량의 지방이 섭취됨으로써 잣이 지니는 콜레스테롤 저하효과가 감쇄하였기 때문으로 사료된다. 또한 잣 성분이 HMG-CoA reductase와 cholesterol esterase의 저해활성을 지니는지 확인하기 위하여 hexane과 물을 사용하여 잣 성분을 추출한 후 각 추출물에 대한 HMG-CoA reductase와 cholesterol esterase의 저해활성을 측정한 결과 잣의 hexane 추출물은 66%의 HMG-CoA reductase 저해활성을 지니고 있었으며 물 추출물은 저해활성을 나타내지 않았다. 한편 잣의 hexane 추출물과 물 추출물은 cholesterol esterase에 대한 저해활성은 지니고 있지 않았다. 결론적으로 혈중 콜레스테롤의 함량 저하효과는 가공하지 않은 잣의 섭취에 의해서는 기대하기 어려우므로, 향후 잣의 hexane 추출물 중의 유용성분이 혈중 콜레스테롤 함량 저하효과를 지니는지에 대한 동물실험을 수행할 가치가 있다고 판단된다.
