Optimal Extract Methods of Antioxidant Compounds from Coat of Grape Dreg (포도부산물인 과피로부터 항산화 물질 최적 추출방법 확립)

  • Optimal extraction method for antioxidant compounds from coat of grape dreg was established. Extracts were prepared with ethanol solutions containing phosphoric, formic, acetic, HCl, TFA, and citric acids. Antioxidant compounds of grape coat were determined by HPLC analysis and evaluated for antioxidizing effects using in vitro model system. Peonidin-3-glucoside content was the highest in 0.1% HCl-added ethanol extract. The extract prepared from pure ethanol without organic acids showed the highest content of cyanidin-3-glucoside among samples tested. Resveratrol and quercetin contents, the most important antioxidants, were highest in 0,1% HCl-added extract. Electron-donating ability was high in 0.1% acetic acid-added extract. SOD-like activities were 95.08% and 94.39% in 0.1% formic and 0.1% citric acid extracts, respectively. Inhibitory effects on peroxidation of egg yolk lecithin were observed in phosphoric (60.51%), citric (59,27%), and formic acid (56,77%) extracts. Hydrogen radical-scavenging activity was 59.47% in 0,1% HCl extract. Results suggest addition of 0.1% HCl in ethanol solution affords the highest yield in antioxidant compounds and antioxidant activities.

Influence of Acid and Salt Content on the Ethanol Production from Laminaria japonica (산 농도 및 염 농도가 다시마 에탄올 발효에 미치는 영향)

  • In the study, the effect of acid and salt concentrations during the production of bio-ethanol from various brwon-algae raw materials was investigated. Especially, the possibility of the conversion of various polysaccarides contained in Laminaria japonica was studied. Bio-ethanol was produced by Saccharomyces cerevisiae KCCM1129 strains in Laminaria japonica. The maximum bio-ethanol production of 2.09 g/L using heat-treatment of Laminaria japonica was achieved. The optimum concentration for reducing sugar conversion by Laminaria japonica was found to be 3.95 g/L at the HCl concentration of 0.1 N. But bio-ethanol production was higher than the case without the non-acid pretreatment. Among the various polysaccharides, only mannitol produced maximum 3.09 g/L bio-ethanol. In case of laminaran, the ethanol was produced only at 0.15 g/L only in 0.1 N HCl pretreatment medium and cell growth was higher than other pretreatment.

Protective Effect of Cotoneaster mongolicus Pojark. Extract in HCl/ethanol-induced Gastritis Mice (Cotoneaster mongolicus Pojark. 추출물의 HCl/ethanol로 유발된 위염 mice에 대한 보호효과)

  • Cotoneaster mongolicus Pojark. (CM), in the family Rosaceae is an endemic plant to the Mongolian region (its name: Moнroл чapraй). In Mongolia, Cotoneaster species as a crude drug is mainly used for inflammatory diseases, diarrhea, and stomach indigestion. In this study, we evaluated the gastro-protective activity underlying mechanism of CM. For in vivo experiments, mice were divided into 5 groups; normal mice (Normal), gastritis mice (Control), gastritis mice treated with sucralfate 10 mg/kg (SC), gastritis mice treated with CM 100 mg/kg (CML), gastritis mice treated with CM 200 mg/kg (CMH). Gastritis was provoked by HCl/ethanol (60% ethanol in 150 mM HCl). After oral administration of each drug, HCl/ethanol was orally administered 90 mins later to induce gastritis. CM alleviated the damage to the gastric mucosa caused. As a result of confirming the expression of protein in gastric tissue through western blot, CM significantly reduced the expression of NF-κB activated due to gastritis. Also, it significantly modulated the Nrf2-Keap1 pathway. These results indicate that CM not only inhibits the nuclear metastasis of NF-𝛋B but also modulates the Nrf2-Keap1 pathway to relieve inflammation of the gastric mucosa.

Immobilization of Yeast Pichia stipitis for Ethanol Production (에탄올 생산을 위한 효모 Pichia stipitis의 고정화)

  • In this study, DEAE-cotton [derivatized by 2-(diethylamino)ethyl chloride hydrochloride (DEAE HCl)] was prepared as a carrier for immobilized $Pichia$ $stipitis$ for ethanol production. When cotton was derivatized with 0.5 M DEAE HCl, the yeast cell suspension was adsorbed at 100% of the initial cell $OD_{600}$. The adsorbed yeast cells were estimated to be 101.8 mg-dry cells/g-DEAE-cotton. In particular, when a flask culture using the immobilized yeast cells was conducted in a glucose and xylose-containing medium, the yeast cells on the DEAE-cotton gradually produced ethanol, according to glucose and xylose consumption; the ethanol yield was approximately 0.33 g-ethanol/g-monosaccharide. Because DEAE-cotton was successfully used as a carrier for ethanol production from a glucose and xylose-containing medium, we expect that this bioethanol production process may be used for the bioethanol production process from the hydrolysate of lignocellulosic biomass. All the results of DEAE-cotton were compared with those of DEAE-cellulose as a carrier for immobilization.

Preparation of Corncob Grits as a Carrier for Immobilizing Yeast Cells for Ethanol Production

  • In this study, DEAE-corncobs [delignified corncob grits derivatized with 2-(diethylamino)ethyl chloride hydrochloride ($DEAE{\cdot}HCl$)] were prepared as a carrier to immobilize yeast (Saccharomyces cerevisiae) for ethanol production. The immobilized yeast cell reactor produced ethanol under optimized $DEAE{\cdot}HCl$ derivatization and adsorption conditions between yeast cells and the DEAE-corncobs. When delignified corncob grit (3.0 g) was derivatized with 0.5M $DEAE{\cdot}HCl$, the yeast cell suspension ($OD_{600}$ = 3.0) was adsorbed at >90% of the initial cell $OD_{600}$. This amount of adsorbed yeast cells was estimated to be 5.36 mg-dry cells/g-DEAE corncobs. The $Q_{max}$ (the maximum cell adsorption by the carrier) of the DEAE-corncobs was estimated to be 25.1 (mg/g), based on a Languir model biosorption isotherm experiment. When we conducted a batch culture with medium recycling using the immobilized yeast cells, the yeast cells on DEAE-corncobs produced ethanol gradually, according to glucose consumption, without cells detaching from the DEAE-corncobs. We observed under electron microscopy that the yeast cells grew on the surface and in the holes of the DEAE-corncobs. In a future study, DEAE-corncobs and the immobilized yeast cell reactor system will contribute to bioethanol production from biomass hydrolysates.

Protective effects of red ginseng according to steaming time on HCl/ethanol-induced acute gastritis (염산/에탄올로 유도된 급성 위염 동물모델에서 증숙시간에 따른 홍삼의 보호 효과)

  • The objective of the present study was to evaluate the protective effect of red ginseng (RG) according to steaming time on 150 mM HCl/60 % ethanol induced gastric ulcer models in mice. The sample was divided into 3 groups-G (dried ginseng), RG 4 (steamed 4 h and dried ginseng), RG 6 (steamed 6 h and dried ginseng), and determined through in vitro experiments, such as 1,1-diphenyl-2-picrylhydrazyl and 2,2'-azinobis-3-ethyl-benzothiazoline-6-sulfonic acid radical scavenging activity, HPLC analysis, total polyphenol, and flavonoid contents. In vitro experiment results were depended on steaming hours. Based on the results, we chose two samples (G, RG 6) and conducted in vivo experiments. Mice were divided into 5 groups: Nor (normal group), Con (acute gastritis mice treated with distilled water), G (gastris induced by HCl/Ethanol treated with 100 mg/kg G), RG 6 (gastris induced by HCl/ethanol treated with 100 mg/kg RG 6), and SC (gastris induced by HCl/Ethanol treated with 10 mg/kg sucralfate). In our results revealed that RG 6 suppressed elevated reactive oxygen species, and inflammatory related makers, such as cyclooxygenase-2, inducible nitric oxide synthase, tumor necrosis factor alpha, and interleukin-1 beta. In addition, gastric lesion area was improved. These results suggest that RG 6 protects the stomach by attenuating oxidative stress and inflammatory response under gastric ulcer conditions. Therefore, RG 6 should be a potential therapeutic agent for the treatment of acute gastric ulcer.

Protective Effects of a Lycium chinense Ethanol Extract through Anti-oxidative Stress on Acute gastric lesion mice (급성 위염 유발 마우스 동물 모델에서 구기자(枸杞子) 에탄올 추출물의 위점막 손상 보호 효과)

  • Objectives : Gastric lesions affect many people around the world and their development are results of the imbalance between destructive and protective factors in the gastric mucosa. Lycium chinense has been widely used as a traditional Korean medicine, it was recently reported that they have potent anti-inflammatory effects in chronic hepatitis models. Therefore, this study aimed to investigate the anti-inflammatory activity of Lycium chinense extract (LCE) on HCl-Ethanol induced gastric lesion mice.Methods : The ICR mice were divided randomly into five groups of six animals each. Group A was normal mice, and group B was treated orally with 0.5 ml 150 mM HCl-60% Ethanol. Mice in group C and D were pre-treatment of LCE (100 mg/kg and 200 mg/kg bodyweight, p.o before HCl/ethanol treatment) and group E was orally administered sucralfate (10 mg/kg).Results : 150mM HCl/60% ethanol-induced gastric mucosal injury mice were ameliorated mucosal damage upon histological evaluation by treatment of LCE. Pre-treatment of LCE attenuated reactive oxidative species (ROS) and produces peroxynitrite (ONOO-) in stomach tissues. As results of stomach protein analyses, LCE effectively reduce inflammatory-related factors such as cyclooxygenase-2 (COX-2), tumor necrosis factor alpha (TNF-α), inducible nitric oxide synthase (iNOS) and interleukin-6 (IL-6) in gastric lesion mice. In addition, nuclear factor kappa B (NF-κB) and inhibitor of phosphorylation of nuclear factor kappa B (p-IκB) were down-regulated in LCE-administrated gastric lesion mice.Conclusions : Our discovery supports that the therapeutic activity of LCE ameliorate the development of gastric lesion via suppressing the oxidative stress and gastric partial inflammation induced by 150 mM HCl/60% ethanol.

The protective effect of Citrus unshiu Peel water extract through PI3K/Akt/NF-κB signaling pathway in mice with HCl/ethanol-induced acute gastritis (HCl/ethanol로 유발한 급성 위염 마우스에서 PI3K/Akt/NF-κB 신호전달경로를 통한 진피 열수 추출물의 보호 효과)

  • This study aimed to verify the effect of Citrus unshiu peel water extract (CUP) on a mouse model of acute gastritis (AG) induced by HCl/ethanol. Several studies have found that CUP has anti-inflammatory effects. The AG model was induced by oral administration of 150 mM HCl/60% ethanol (550 µL) to all groups except the control group. Also, for drug treatment, sucralfate (10 mg/kg) and CUP (100 or 200 mg/kg) were orally administered for 90 minutes before induction. The effect of CUP treatment was confirmed by gross gastric mucosal damage measurement, and the levels of Glutamic Oxaloacetic Transaminase (GOT), Glutamic Pyruvic Transaminase (GPT), and myeloperoxidase were reduced as well as the levels of oxidative stress biomarkers and their related proteins. In addition, the levels of inflammatory proteins, mediators, and cytokines were significantly downregulated byPI3K/Akt signaling. Taken together, these results show that CUP treatment alleviates AG by regulating PI3K/Akt signaling.

Study on Antigastritic and Anti Helicobacter Pylori Effects from Water Extract of Ulmus Davidiana var. Japonica Nakai (유근피(楡根皮) 수추출물의 Helicobacter pylori에 대한 항균효과)

  • Present study was carried out for development of a new herb product with gastroprotective effect. Natural herbs mentioned that have gastrointenstial protective property in Donggeuibogam and reports were evaluated anti-bacterial activity against Helicobacter pylori, then Ulmus davidiana var. japonica Nakai (YGP) were selected. The material used for the test were water extract of Ulmus davidiana var. japonica Nakai. They were tested individually on HCl ethanol-induced gastric lesion in rats showed the most significant effectiveness, respectively. Then YGP was made, and tested on HCl ethanol, indomethacin-induced gastric lesion, shay ulcer and gastric secretion. Conclusions : YGP showed significant effect on HCl ethanol, indomethacin-induced gastric lesion, and Shay ulcer. It can be regarded that the antigastritic and anti-ulcerative effects of YGP is originated from reduction of total acid output identified by gastric secretion test.

  • The ethanol extract of Sida acuta Burm. (ALSA) whole plant was studied for its anti-ulcer activity against aspirin plus pylorous ligation induced gastric ulcer, HCl-ethanol induced ulcer, and water immersion stress induced ulcer (WISIU) in rats. We found that ALSA at a dose of 300 mg/kg, (orally) markedly decrease the incidence of ulcers in the first two models. ALSA showed reduction in gastric volume, free acidity, and ulcer index (53.69%). It has not reduced the total acidity significantly and no significant change in pH. It also showed 55.14% gastro protective activity, wheres standard drug sucralfate showed 94.85%. WISIU showed protection index 24.4%, whereas standard drug omeprazole (OMEZ) showed protection index 100%.
