Objective: Daweizi (DWZ) is a famous indigenous pig breed in China and characterized by tender meat and high fat percentage. However, the expression profiles and functions of transcripts in DWZ pigs is still in infancy. The object of this study was to depict the transcript profiles in DWZ pigs and screen the potential pathway influence adipogenesis and fat deposition, Methods: Histological analysis of backfat tissue was firstly performed between DWZ and lean-type Yorkshire pigs, and then RNA sequencing technology was utilized to explore miRNAs, lncRNAs and mRNAs profiles in backfat tissue. 18 differentially expressed (DE) transcripts were randomly selected for quantitative real-time polymerase chain reaction (QPCR) to validate the reliability of the sequencing results. Finally, gene ontology (GO) and Kyoto encyclopedia of genes and genomes (KEGG) enrichment analysis were conducted to investigate the potential pathways influence adipocyte differentiation, adipogenesis and lipid metabolism, and a schematic model was further proposed. Results: A total of 1,625 differentially expressed transcripts were identified in DWZ pigs, including 27 upregulated and 45 downregulated miRNAs, 64 upregulated and 119 down-regulated lncRNA, 814 upregulated and 556 downregulated mRNAs. QPCR analysis exhibited strong consistency with the sequencing data. GO and KEGG analysis elucidated that the differentially expressed transcripts were mainly associated with cell growth and death, signal transduction, peroxisome proliferator-activated receptors (PPAR), AMP-activated protein kinase (AMPK), PI3K-Akt, adipocytokine and foxo signaling pathways, all of which are strongly involved in cell development, lipid metabolism and adipogenesis. Further analysis indicated that the BGIR9823_87926/miR-194a-5p/AQP7 network may be effective in the process of adipocyte differentiation or adipogenesis. Conclusion: Our study provides comprehensive insights into the regulatory network of backfat deposition and lipid metabolism in pigs from the point of view of miRNAs, lncRNAs and mRNAs.
본 연구에서는 진피 소수성 추출물(CECU)의 U87MG 교모세포종 암줄기세포에 대한 항암 활성을 확인하였다. 그 결과, CECU는 25-200 ㎍/mL의 농도 범위에서 U87MG 교모세포종 암줄기세포의 증식, 종양구체 형성과 이동능력을 유의적으로 저해하였다. 특히, CECU는 G0/G1기에서 세포주기 정지와 세포사멸을 유도하여 교모세포종 암줄기세포의 증식을 억제할 수 있었다. 게다가, 교모세포종 암줄기세포에 대한 CECU의 항암 활성은 CD133, Oct4, Nanog, Integrin α6, ALDH1A1과 같은 줄기세포능 조절인자들의 발현과 STAT3 신호전달경로를 저해함으로써 유도된 것임을 확인하였다. 마지막으로, CAM assay를 통해 CECU가 U87MG 교모세포종 암줄기세포의 in vivo 종양 형성을 효과적으로 억제함을 입증하였다. 따라서, 본 연구는 진피 소수성 추출물이 주요 stemness marker들의 발현과 핵심 stemness 조절 신호전달경로를 억제함으로써 U87MG 교모세포종 암줄기세포에 대한 항암 활성을 나타냄을 입증하여, 교모세포종의 예방 및 치료를 위한 천연물 소재로서의 활용 가능성을 새롭게 제시하였다.
The anti-oxidant enzyme heme oxygenase-1 (HO-1) is known to exert anti-inflammatory effects. From a library of pyrazolo[3,4-d]pyrimidines, we identified a novel compound KKC080096 that upregulated HO-1 at the mRNA and protein levels in microglial BV-2 cells. KKC080096 exhibited anti-inflammatory effects via suppressing nitric oxide, interleukin1β (IL-1β), and iNOS production in lipopolysaccharide (LPS)-challenged cells. It inhibited the phosphorylation of IKK and MAP kinases (p38, JNK, ERK), which trigger inflammatory signaling, and whose activities are inhibited by HO-1. Further, KKC080096 upregulated anti-inflammatory marker (Arg1, YM1, CD206, IL-10, transforming growth factor-β [TGF-β]) expression. In 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridinetreated mice, KKC080096 lowered microglial activation, protected the nigral dopaminergic neurons, and nigral damage-associated motor deficits. Next, we elucidated the mechanisms by which KKC080096 upregulated HO-1. KKC080096 induced the phosphorylation of AMPK and its known upstream kinases LKB1 and CaMKKbeta, and pharmacological inhibition of AMPK activity reduced the effects of KKC080096 on HO-1 expression and LPS-induced NO generation, suggesting that KKC080096-induced HO-1 upregulation involves LKB1/AMPK and CaMKKbeta/AMPK pathway activation. Further, KKC080096 caused an increase in cellular Nrf2 level, bound to Keap1 (Nrf2 inhibitor protein) with high affinity, and blocked Keap1-Nrf2 interaction. This Nrf2 activation resulted in concurrent induction of HO-1 and other Nrf2-targeted antioxidant enzymes in BV-2 and in dopaminergic CATH.a cells. These results indicate that KKC080096 is a potential therapeutic for oxidative stress-and inflammation-related neurodegenerative disorders such as Parkinson's disease.

Background: Highly pathogenic avian influenza virus (HPAIV) is considered a global threat to both human health and the poultry industry. MicroRNAs (miRNA) can modulate the immune system by affecting gene expression patterns in HPAIV-infected chickens. Objectives: To gain further insights into the role of miRNAs in immune responses against H5N1 infection, as well as the development of strategies for breeding disease-resistant chickens, we characterized miRNA expression patterns in tracheal tissues from H5N1-infected Ri chickens. Methods: miRNAs expression was analyzed from two H5N1-infected Ri chicken lines using small RNA sequencing. The target genes of differentially expressed (DE) miRNAs were predicted using miRDB. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analysis were then conducted. Furthermore, using quantitative real-time polymerase chain reaction, we validated the expression levels of DE miRNAs (miR-22-3p, miR-146b-3p, miR27b-3p, miR-128-3p, miR-2188-5p, miR-451, miR-205a, miR-203a, miR-21-3p, and miR-200a3p) from all comparisons and their immune-related target genes. Results: A total of 53 miRNAs were significantly expressed in the infection samples of the resistant compared to the susceptible line. Network analyses between the DE miRNAs and target genes revealed that DE miRNAs may regulate the expression of target genes involved in the transforming growth factor-beta, mitogen-activated protein kinase, and Toll-like receptor signaling pathways, all of which are related to influenza A virus progression. Conclusions: Collectively, our results provided novel insights into the miRNA expression patterns of tracheal tissues from H5N1-infected Ri chickens. More importantly, our findings offer insights into the relationship between miRNA and immune-related target genes and the role of miRNA in HPAIV infections in chickens.

The Hippo kinase cascade functions as a central hub that relays input from the "outside world" of the cell and translates it into specific cellular responses by regulating the activity of Yes-associated protein 1 (YAP1). How Hippo translates input from the extracellular signals into specific intracellular responses remains unclear. Here, we show that transforming growth factor β (TGFβ)-activated TAK1 activates LATS1/2, which then phosphorylates YAP1. Phosphorylated YAP1 (p-YAP1) associates with RUNX3, but not with TEAD4, to form a TGFβ-stimulated restriction (R)-point-associated complex which activates target chromatin loci in the nucleus. Soon after, p-YAP1 is exported to the cytoplasm. Attenuation of TGFβ signaling results in re-localization of unphosphorylated YAP1 to the nucleus, where it forms a YAP1/TEAD4/SMAD3/AP1/p300 complex. The TGFβ-stimulated spatiotemporal dynamics of YAP1 are abrogated in many cancer cells. These results identify a new pathway that integrates TGFβ signals and the Hippo pathway (TGFβ→TAK1→LATS1/2→YAP1 cascade) with a novel dynamic nuclear role for p-YAP1.
Cryptococcus neoformans causes life-threatening meningoencephalitis in humans, but the treatment of cryptococcosis remains challenging. To develop novel therapeutic targets and approaches, signaling cascades controlling pathogenicity of C. neoformans have been extensively studied but the underlying biological regulatory circuits remain elusive, particularly due to the presence of an evolutionarily divergent set of transcription factors (TFs) in this basidiomycetous fungus. In this study, we constructed a high-quality of 322 signature-tagged gene deletion strains for 155 putative TF genes, which were previously predicted using the DNA-binding domain TF database (http://www.transcriptionfactor.org/). We tested in vivo and in vitro phenotypic traits under 32 distinct growth conditions using 322 TF gene deletion strains. At least one phenotypic trait was exhibited by 145 out of 155 TF mutants (93%) and approximately 85% of the TFs (132/155) have been functionally characterized for the first time in this study. Through high-coverage phenome analysis, we discovered myriad novel TFs that play critical roles in growth, differentiation, virulence-factor (melanin, capsule, and urease) formation, stress responses, antifungal drug resistance, and virulence. Large-scale virulence and infectivity assays in insect (Galleria mellonella) and mouse host models identified 34 novel TFs that are critical for pathogenicity. The genotypic and phenotypic data for each TF are available in the C. neoformans TF phenome database (http://tf.cryptococcus.org). In conclusion, our phenome-based functional analysis of the C. neoformans TF mutant library provides key insights into transcriptional networks of basidiomycetous fungi and ubiquitous human fungal pathogens.
진피 상층부 모세혈관은 젊은 사람에 비해 노인에서 그 수와 크기가 감소되어 있어 피부에서 모세혈관이 차지하는 비율이 나이에 따라 급격히 낮아진다. 연교는 물푸레나무과에 속하는 개나리 열매를 건조한 것으로 주로 염증성 또는 항균성 질환에 오랫동안 사용되어져온 약재로서 지금까지 피부 혈관신생과 관련된 효능은 보고되지 않았다. 따라서 본 연구에서는 연교 추출물이 혈관신생과 관련된 인자들에 미치는 영향을 피부 각질형성 세포주를 이용하여 조사하고자 하였다. 우리는 먼저 연교 추출물이 혈관신생과 관련된 인자들의 발현에 어떤 영향을 주는지 알아보고자 각질형성세포에 연교 추출물을 처리하고 혈관신생과 관련된 55개 단백질의 발현을 분석하였다. 발현 변화를 보인 인자들 중 혈관내피세포 성장인자(VEGF, vascular endothelial growth factor)는 강력한 혈관신생 촉진인자로서 연교 추출물에 의해 유의하게 발현이 증가되었다. 따라서 연교 추출물이 VEGF 생성에 미치는 영향에 대해 자세히 알아보고자 연교 추출물을 농도별로 세포에 처리하고 단백질 발현과 mRNA 발현 변화를 조사한 결과, 연교 추출물은 VEGF의 유전자 수준뿐만 아니라 단백질 수준의 발현을 2배이상 농도 의존적으로 증가시켰다. 다음으로 연교 추출물에 의한 VEGF 발현 증가에 관여하는 신호전달 기전을 밝히고자 MAPK 활성을 살펴본 결과, 연교 추출물을 세포에 처리하면 5 min 내 p38 MAPK의 활성이 관찰되었으며, 특이적 억제제 전처리를 통해 p38 MAPK 활성을 억제하면 연교 추출물을 처리하더라도 VEGF의 유전자 및 단백질 발현이 완전히 억제됨을 확인하였다. 이러한 결과로부터 연교는 피부 각질형성세포에 작용하여 p38 MAPK 활성을 통해 VEGF 생성을 유도함을 알 수 있었고, 피부에서 노화에 따른 표피아래 모세혈관 손상을 개선하는데 도움을 줄 수 있는 후보 물질로서 연교의 새로운 효능을 제안한다.
본 연구에서는 랫트를 이용한 제2형 당뇨 동물모델로 같은 혈당조절 효과가 나타나는지 검토하고 이러한 효과가 당화 혈색소를 포함한 최종당화산물(advanced glycation end products, AGEs)과 어떤 상관관계가 있는지 또한 단백질과 당화를 촉진해 당화혈색소 생성의 원인 중 하나인 산화적 스트레스와 관련된 기전을 규명하고자 하였다. 기존의 db/db 마우스에서 실험한 결과와 마찬가지로 랫트를 이용한 제2형 당뇨모델에서도 가시오가피 추출물의 섭취는 혈당을 강하시키고 homeostasis model assessment(Homa-IR)를 감소시켜 인슐린 저항성 개선에 도움을 주는 것으로 확인되었다. 특히 혈중 당화혈색소량의 감소가 두드러졌는데 이는 산화적 스트레스 감소로 인한 지질과산화물 생성의 억제가 중요한 원인으로 생각되며 이와 관련된 혈중 사이토카인 IL-$1{\beta}$와 TNF-${\alpha}$의 농도도 감소한 것으로 나타났다. 당화혈색소는 산화적 스트레스에 의해 최종당화산물로 전환이 되어 인슐린 저항성 세포의 protein kinase C(PKC)를 활성화하여 transforming growth factor(TGF)-${\beta}$를 생성하는데 가시오가피 추출물의 섭취는 최종당화산물의 농도, PKC 그리고 TGF-${\beta}$ 모두를 억제하는 것으로 확인되었으며, 이것은 가시오가피 추출물 성분이 PKC와 TGF-${\beta}$에 직접 작용하기보다는 신호전달체계의 상위에 존재하는 최종당화산물을 억제하여 나타난 결과로 생각한다. 향후 연구에서는 가시오가피 추출물을 분획화하여 어떤 성분에 의하여 당화혈색소와 최종당화산물 생성을 억제하는지에 대한 구체적인 실험이 이루어져야 할 것으로 여겨진다.
본 연구에서는 부처꽃 에탄올 추출물(ELM)에 대한 항암효능을 알아보기 위하여 인체백혈병 U937 세포의 증식에 미치는 영향과 이와 연관된 apoptosis 유발 여부와 함께 그에 따른 분자생물학적 기전에 대해서 조사하였다. 먼저 ELM 처리에 따른 증식 억제 정도를 조사한 결과, ELM 처리 농도 의존적으로 생존율 및 증식억제 현상이 나타났으며, 핵의 형태 변화, DNA 단편화 및 apoptosis 유발에 관하여 조사한 결과 역시 ELM 처리 농도 의존적으로 증가됨을 확인할 수 있었다. ELM 처리에 따른 U937 세포에서의 apoptosis 유발에 있어서 미토콘드리아 막의 기능 손상이 관여하는 지를 확인하기 위하여 MMP의 변화 정도를 확인한 결과, ELM 처리 농도 증가에 따라 MMP의 소실이 증가하는 것을 관찰할 수 있었다. 이러한 MMP의 소실에 가 관여하는 지를 확인하기 위하여 사멸수용체(DR4, 5, Fas) 및 사멸수용체에 결합하는 리간드(FasL, TRAIL)의 발현 변화를 확인한 결과, DR4 및 DR5의 발현이 증가하는 것으로 관찰되었다. 또한 내인적 경로에 관여하는 Bcl-2 family 유전자들의 발현변화를 확인한 결과, Bcl-2 발현 감소 및 Bax의 발현 증가의 변화를 보였으며, Bid 단백질의 발현감소가 나타났으므로 상대적으로 tBid의 생성이 증가되었음을 추측할 수 있었다. 한편apoptosis 유발에 직접적으로 관여하는 것으로 알려진 caspase-3, -8 및 -9의 발현에 미치는 ELM의 영향에 대해서 조사하였다. 결과에서 알 수 있듯이 ELM은 death receptor에 의하여 활성화 되는 것으로 알려진 caspase-8 및 세포질로 방출된 cytochrome c에 의하여 활성화 되는 것으로 알려진 caspase-9의 활성화를 유발하였으며, caspase cascade에 의하여 apoptosis에 직접적으로 관여하는 caspase-3의 발현도 증가시키는 것으로 나타났다. 또한 활성화된 caspase-3에 의하여 분해가 일어나는 기질 단백질인 PARP의 경우 ELM 처리에 의하여 모두 단편화가 유발되는 것으로 나타났다. 이상의 결과를 종합해 보면 인체 백혈병 U937 세포에 ELM을 처리하였을 경우에 유발되는 apoptosis는 외인적 경로인 DR4 및 DR5의 발현 증가를 통한 caspase-8의 활성화와 이로 인한 Bid 단백질의 단편화와 함께 내인적 경로의 미토콘드리아 기능 손실에 의하여 caspase-9 및 -3의 활성화 유발과 기질단백질들의 분해가 중요한 역할을 하는 것으로 생각되며, IAP family의 발현 감소로 인하여 caspase의 활성이 억제되지 못하는 것도 apoptosis 유도에 어느 정도 관여했을 것으로 생각된다. 따라서 ELM 처리에 의하여 유발되는 apoptosis는 외인적 경로 및 내인적 경로를 모두 경유하는 multiple apoptotic pathway에 의하여 조절되며, 이때 caspases가 중요한 역할을 한다는 것을 알 수 있었다.
배경 : 골관절염은 단순 노화로 인한 질병이 아니라 연골대사의 이상으로 인한 기계적 그리고 생화학적 불안정성이 나타나는 질환이다. Transforming growth factor-${\beta}$와 같이 연골세포의 기능을 향상시키는 촉진인자가 있는 반면 Interleukin(IL)-1이나 Tumor necrosis factor-${\alpha}$는 염증반응을 증가시킨다. 이중 IL-1은 골관절염의 병인에서 가장 중요한 염증 유발 인자로 알려져 있으며 이에 대한 자료도 축적되고 있다. 이 논문의 목적은 IL-$1{\beta}$에 대한 human chondrosarcoma cell (SW1353)의 유전자 발현 양상을 파악하여 골관절염 병인의 이해를 증대시키는데 있다. 재료 및 방법 : Chondrosarcoma cell line (SW 1353)은 연골세포의 IL-$1{\beta}$를 통한 세포노화에 대한 유전자 조절을 실험실에서 연구하는데 유용한 것으로 알려져 있다. 골관절염에서 IL-1에 의한 전체적인 유전자 발현의 변화를 연구하고 분석하기 위해 SW1353을 각각 1시간, 6시간, 24시간동안 IL-1에 노출시킨후 각각 총 RNA를 정제하였다. 유전자 발현의 변화는 17k human cDNA microarray로 분석하였고 semiquantitative RT-PCR로 확인하였다. 결과 : Metallothioneins, matrix metalloproteinases, extracellular proteins, antioxidant protein, cytoskeletal proteins, cell cycle regulatory proteins, 세포성장과 세포 자멸사에 대한 단백질, signal protein, transcriptional factor를 포함한 1,200개 유전자에서 2배 이상의 변화가 관찰되었다. 이러한 변화는 초기 관절염에서 보이는 병리생리학적 변화와 상관관계가 있는 것으로 생각된다. 결론 : cDNA microarray 분석으로 유전자 발현의 의미있는 변화를 관찰하였으며 골관절염 발병기전에서 분자생물학적 변화에 대한 인식과 치료 목표를 정립하는데 대한 새로운 자료로서 가치가 있을 것으로 생각된다.

ID의 비밀번호 관리소홀, 부정사용에 의하여 발생하는 모든 결과에 대한 책임은 회원에게 있습니다.

제 4 장 서비스의 이용
제 12 조 (서비스 이용 시간)
① 서비스 이용은 당 사이트의 업무상 또는 기술상 특별한 지장이 없는 한 연중무휴, 1일 24시간 운영을
원칙으로 합니다. 단, 당 사이트는 시스템 정기점검, 증설 및 교체를 위해 당 사이트가 정한 날이나 시간에
서비스를 일시 중단할 수 있으며, 예정되어 있는 작업으로 인한 서비스 일시중단은 당 사이트 홈페이지를
통해 사전에 공지합니다.
② 당 사이트는 서비스를 특정범위로 분할하여 각 범위별로 이용가능시간을 별도로 지정할 수 있습니다. 다만
이 경우 그 내용을 공지합니다.
제 13 조 (홈페이지 저작권)
① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
