Antioxidative Components and Anti-Oralmicrobial Effect of Bamboo (Phyllostachys nigra var. henonis Stapf) Leaves (죽엽(솜대)의 항산화 성분 및 구강세균에 대한 항균 효과)

  • The objective of this study was to investigate the antioxidative components and anti-oralmicrobial effect of bamboo (Phyllostachys nigra var. henonis Stapf) leaves. The moisture, crude protein, crude fat, crude ash, and carbohydrate contents were 6.30%, 5.10%, 1.73%, 10.61%, and 76.26%, respectively. Vitamin C content was higher than Vitamin A and E contents. Among organic acids, citric acid content was the most abundant organic acid, followed by succinic acid, acetic acid, malic acid, and formic acid. Total polyphenol and flavonoid contents were 21.66 mg/g and 42.78 mg/g, respectively. Minimum inhibitory concentrations (MICs) of extracts of bamboo leaves for Streptococcus mutans and Streptococcus sobrinus were determined to be 0.04% and 0.16%, respectively. MICs of extracts of bamboo leaves for Porphyromonas gingivalis and Prevotella intermedia were determined to be 0.02%. Extract of bamboo leaves had strong antimicrobial activity against S. mutans, S. sobrinus, P. gingivalis, and P. intermedia at a concentration of 0.32%. At this concentration, extract of bamboo leaves inhibited growth of these pathogenic bacteria up to 60 h. The results of the present study demonstrate the antimicrobial effects of bamboo leaves ethanol extract against oral pathogenic bacteria, suggesting that bamboo leaves could be an effective natural agent for oral hygiene.

Effects of Rapamycin on Cell Apoptosis in MCF-7 Human Breast Cancer Cells

  • Background: Rapamycin is an effective anti-angiogenic drug. However, the mode of its action remains unclear. Therefore, in this study, we aimed to elucidate the antitumor mechanism of rapamycin, hypothetically via apoptotic promotion, using MCF-7 breast cancer cells. Materials and Methods: MCF-7 cells were plated at a density of $1{\times}10^5$ cells/well in 6-well plates. After 24h, cells were treated with a series of concentrations of rapamycin while only adding DMEM medium with PEG for the control regiment and grown at $37^{\circ}C$, 5% $CO_2$ and 95% air for 72h. Trypan blue was used to determine the cell viability and proliferation. Untreated and rapamycin-treated MCF-7 cells were also examined for morphological changes with an inverted-phase contrast microscope. Alteration in cell morphology was ascertained, along with a stage in the cell cycle and proliferation. In addition, cytotoxicity testing was performed using normal mouse breast mammary pads. Results: Our results clearly showed that rapamycin exhibited inhibitory activity on MCF-7 cell lines. The $IC_{50}$ value of rapamycin on the MCF-7 cells was determined as $0.4{\mu}g/ml$ (p<0.05). Direct observation by inverted microscopy demonstrated that the MCF-7 cells treated with rapamycin showed characteristic features of apoptosis including cell shrinkage, vascularization and autophagy. Cells underwent early apoptosis up to 24% after 72h. Analysis of the cell cycle showed an increase in the G0G1 phase cell population and a corresponding decrease in the S and G2M phase populations, from 81.5% to 91.3% and 17.3% to 7.9%, respectively. Conclusions: This study demonstrated that rapamycin may potentially act as an anti-cancer agent via the inhibition of growth with some morphological changes of the MCF-7 cancer cells, arrest cell cycle progression at G0/G1 phase and induction of apoptosis in late stage of apoptosis. Further studies are needed to further characterize the mode of action of rapamycin as an anti-cancer agent.

Pharmacological Effects of Proteoglycans Extracted from Fruiting Bodies of Fomitella fraxinea (장수버섯 자실체의 열탕추출액으로부터 분리한 단백다당체의 약리적 효과)

  • Anti-complementary assay for immuno-stimulating polysaccharide screening, human tumor colony-forming assay for discovering anti-tumor drugs, and toxic assay against mouse were performed to examine pharmacological activities of polysaccharides extracted from fruiting bodies of Jang-soo mushroom (Fomitella fraxinea). Hot water $(100^{\circ}C,\;FF-I)$, 1% ammonium oxalate solution $(80^{\circ}C,\;FF-II)$, and 5% sodium hydroxide solution $(80^{\circ}C,\;FF-III)$ were used for extraction of three polysaccharides from fruiting bodies of it. Anti-complementary activity of FF-I was more effective than the others. FF-I was further fractionated into three groups of polysaccharide by DEAE-Sephadex A25 column chromatography (FF-NP, FF-AP1, and FF-AP2). FF-AP1 showed not only the highest anti-complementary activity but also the growth-inhibitory activity against Snu-I (human stomach cancer cell) among 9 kinds of human tumor cell lines. But FF-AP2 exhibited its activity against Hep-2(larynx cancer) and KB(mouth epidermal cancer) cell lines at $500\;{\mu}g/ml$ although its anti-complementary activity was lower than it of FF-AP1. When FF-I was orally administrated to mice with dosage of 5000 mg/kg, no remarkable changes were observed in viewpoint of tissue-pathology.

Accelerated Germination of Orchardgrass(Dactylis glomerata) Seed Under Water Stress by Seed Pre-treatments (수분 장애시 종자 전처리가 오차드그라스의 발아에 미치는 영향)

  • It is important to improve the germination of grass seeds because they are poor germination under water stress. This experiment was conducted to investigate the response of seeds pre-treated individually with Polyethylene glycol 6000 (PEG) and growth regulators(GA$_3$, Kinetin, NAA) on the germination characteristics when two levels of osmotic potential (0, -5 bar) were put to seeds of Orchardgrass. In the untreated seeds, the total germination rate was decreased under water stress, and the mean germination time was delayed. Priming of Orchardgrass seeds using PEG or GA$_3$ resulted in improved germination under water stress(-5 bar), whereas the opposite was true of kinetin or NAA. The response of seeds primed in solutions of either -15 bar PEG or 100 ppm GA$_3$ was particulary marked compared with the untreated seeds. It is suggested that PEG and GA$_3$ may have positive regulatory effects in triggering the system for water stress alleviation. But the inhibitory effect of the water stress was not completely removed by allowing the seeds to pre-treat in solution of PEG or GA$_3$.

Immunomodulation by Bioprocessed Polysaccharides from Lentinus edodes Mycelia Cultures with Rice Bran in the Salmonella Gallinarum-infected Chicken Macrophages (Salmonella Gallinarum 감염닭의 대식세포에서 표고버섯 균사체 발효 미강생물전환소재에 의한 면역조절효과)

  • In this study, we investigated the effect of bioprocessed polysaccharides (BPPs) from liquid culture of Lentinus edodes fungal mycelia containing rice bran (BPP-RB) on a chicken-derived macrophage cell line, HD-11, when infected with Salmonella Gallinarum, an etiological agent of fowl typhoid. Experimental results demonstrated water extract of BPP-RB did not show growth inhibitory effects on S. Gallinarum 277. Protein expression profiles were also not altered by its treatment. Nonetheless, it could (i) enhance phagocytic activity of HD-11 cells, (ii) activate transcriptional expression of Th1-type cytokines such as tumor necrosis factor-${\alpha}$ and interleukin $(IL)-1{\beta}$, iNOS, as well as an immunosuppressive cytokine IL-10, and (iii) negatively regulate Th2-type cytokines such as IL-4 and IL-6. Together results suggest that BPP-RB may be applicable for preventing fowl typhoid or other Salmonella infections in poultry farms as a potential feed additive.

Characterization of Probiotic and Functional Properties of Lactobacillus curvatus ML17, a Mukeunji Starter (묵은지 스타터 Lactobacillus curvatus ML17의 생균제적 특성 및 기능성 조사)

  • This study investigated the probiotic and functional characteristics of Mukeunji starter, Lactobacillus curvatus ML17, isolated from Mukeunji. Lb. curvatus ML17 was confirmed as a safe microorganism due to its non-hemolytic activity and non-production of harmful ${\beta}$-glucuronidase and ${\beta}$-glucosidase. Tolerance to artificial gastric and bile juice of Lb. curvatus ML17 was investigated. After incubation in artificial gastric and bile juice, the number of surviving cells was $1.38{\times}10^8$ CFU/mL. According to the results of adhesion assay, this strain also exhibited good adhesion to Caco-2 cells. Lb. curvatus ML17 showed good antimicrobial activity against food borne pathogens, especially Micrococcus luteus, Bacillus cereus, Salmonella enterica subsp. enterica, and Pseudomonas aeruginosa. Cell-free extract of Lb. curvatus ML17 exhibited high levels of DPPH scavenging capacity and inhibitory effects on growth of AGS human gastric adenocarcinoma cells and HT-29 human colon carcinoma cells. These results suggest that Lb. curvatus ML17 has potential for application in functional foods.

EFFECTS OF AMYLASE ON THE DEMINERALIZATION IN HYDROXYAPATITE (Amylase가 Hydroxyapatite 탈회에 미치는 영향)

  • Salivary proteins which are produced in the saliary acinar cells have been known to be involved in the Calcium and phosphate metabolism. The acquired pellicle resulting from such metabolism is considered as a secondary defence membrane against tooth caries. In this respect, some proteins included in saliva probably play an important role in the prevention of demineralization in enamel. On the other hand, fluoride has long been known to prevent the demineralization of enamel by the inhibition of the growth of Streptococcus mutans(S. mutans) and by the chemical reaction with calcium and phosphate, Therefore, I have examined the roles of amylase and albumin in the demineralization of enamel and compared these preteins with fluoride in terms of anticariogenic effect. 1. The demineralization caused by S. mutans occurred slowly and progressively for the first 60 min, then the rate of demineralization was accelerated afterwards. 2. pH decreased continuously during the entire period of each experiment. 3. The demineralization was significantly inhibited by the preteatment of amylase and fluoride but albumin had little effect on it. 4. An addition of 0.1 mM lactic acid (final concentration 0.1 ${\mu}M$) caused a rapid increase in calcium concentration reaching a maximum within 10 min. 5. pH decreased rapidly by the addition of 0.1 mM lactic acid and reached a minimum within a few seconds followed by an increase in pH. pH reaced a plateu with 10 min. 6. Fluoride, amylase and albumin played little role in the 0.1 mM lactic acid-induced demineralization. 7. A slow infusion of 0.1 M lactic acid at a rate of 5 ${\mu}l/min$ caused a slower increase in calcium concentration compared with the bolus addition of lactic acid. 8. Fluoride had an inhibitory effect on the calcium release caused by slow infusion of lactic acid while amylase and albumin had no effect on it. These results suggest that fluoride inhibits demineralization by protecting the HA from the acid attack whereas amylase has a direct effect on S. mutans to prevent demineralization.

  • 1. Kum Nam Poong which is highly susceptible to bacterial leaf blight was used as the host plant throughout this experiment. Xanthomonas oryzae 6526 was inoculated on the top of upper leaves by single needle inoculation method. After 14 days, the enlarged spots were examined in the experimental pots. Each of 3 levels of nitrogen, phosphate and potassium pots were arranged at random with three replications. 2. The amounts of nitrogen fertilizer applied and the lesion development of bacterial leaf blight were positively correlated regardless of application of phosphate and potassium fertilizers. 3. The effect of phosphate fertilizer on the lesion development was not significantly different from standard level. The lesion development was stimulated when the amount of phosphate fertilizer was increased as twice as standard level. 4. The inhibitory effect of potassium fertilizer on leaf blight was maximum by applying standard level. The stimulative effect of potassium fertilizer on the lesion development, however, was noticed. when potassium fertilizer applied was increased as twice as the standard level. 5. The heading date and spike number of rice plant were significantly correlated with the lesion development, and such phenomena were depended on the amount of nitrogen fertilizer applied regardless of the other fertilizers applied in this experiment.

Antioxidant and Whitening Activity of Essential Oils (Essential Oils의 항산화 활성 및 미백작용)

  • This study is performed to investigate the effects of citrus essential oils on melanin production in B16 melanoma cells and reactive oxygen species (ROS) generation in RBL 2H3 cells. Five kinds of citrus essential oil (bergamot, grapefruit, lemmon, mandarin, petigrain) did not have any influence on DPPH radical scavenger activity, cell growth and cytotoxicity in B16 melanoma cells. In purified tyrosinase assay, both mandarin and petigrain essential oils dose-dependently inhibited its activity, but bergamot did not. In $1{\mu}M\;{\alpha}-MSH-stimulated$ B16 melanoma cells, all of 5 citrus essential oils inhibited melanin production in $\underline{a}$ dose dependent manner. On the other hand, four kinds of citrus essential oil dose-dependently increased ROS generation in RBL 2H3 mast cells, but mandarin did not. From the above results, it is possible that citrus essential oils nay be developed to be anti-melanogenic agent on the basis of their inhibitory effect on MSH-induced melanin production. Hut we can not rule out the possibility of the induction of allergy and inflammation since citrus essential oils caused ROS generation in RBL 2H3 mast cells.

Isolation and identification of soycurd forming lactic acid bacteria which produce GABA from kimchi (김치로부터 GABA를 생산하는 커드 형성 젖산균의 분리 및 동정)

  • Gamma amino butyric acid (GABA), known as a non-protein amino acid and major inhibitory neurotransmitter in the brain, has several functional properties such as neurotransmission, induction of hypotension, tranquilizer, and diuretic effects. The purpose of this study was to isolate and identify lactic acid bacteria, producing high GABA in fermented soy curd. Thirty-two strains of tofu-forming lactic acid bacteria were isolated from kimchi which a traditional Korean food fermented with many kind of microorganism. Among 32 strains, four strains (strain No. 10, 104, 214, 249) formed firm soycurd. In order to select lactic acid bacteria having high GABA producing potential, the isolated strains were cultured in the soymilk and fermented for 48 hr at $37^{\circ}C$. A strain No. 383, which showed highest GABA contents in fermented soycurd, was identified as L. sakei by 16S rDNA sequencing and API analysis, and named as L. sakei 383. L. sakei 383 showed optimal growth up to 24 hr at $35^{\circ}C$ in MRS broth. The optimal time and temperature for GABA production were 18 hr and $35^{\circ}C$ in soymilk. In the optimal condition time and temperature, GABA content of fermented soycurd by L. sakei 383 was 8.65 mg/100 g.
