Probe DNAs consisting of mercaptohexyl moiety at their 5-end were spotted on the gold electrode using micropipette or DNA arrayer utilizing the affinity between gold and sulfur. Cyclic voltammetry in 5mM ferricyanide/ferrocyanide solution at 100 ㎷/s confirmed the immobilization of probe DNA on the gold electrodes. When several DNAs were detected electrochemically, there was a difference between target DNA and control DNA in the anodic peak current values. It was derived from specific binding of Hoechst 33258 to the double stranded DNA due to hybridization of target DNA. It suggested that this DNA chip could recognize the sequence specific genes. It suggested that multichannel electrochemical DNA microarray is useful to develop a portable device for clinical gene diagnostic System.

We report the passivation characteristics of dodecanethiol (DDT) molecules on gold electrodes of field effect transistor bio-sensors. The leakage current between the electrolyte and the electrode can be as small as 3pA when 7 mM of DDT passivation is performed. The DDT layer is also shown to resist the attack of acid up to 120 mins.

For the purpose of developing a direct label-free electrochemical detection system, we have systematically investigated the electrochemical signatures of each step in the preparation procedure, from a bare gold electrode to the hybridization of label-free complementary DNA, for the streptavidin-modified electrode. For the purpose of this investigation, we obtained the following pertinent data; cyclic voltammogram measurements, electrochemical impedance spectra and square wave voltammogram measurements, in $Fe(CN)_6^{3-}/Fe(CN)_6^{4-}$ solution (which was utilized as the electron transfer redox mediator). The oligonucleotide molecules on the streptavidin-modified electrodes exhibited intrinsic redox activity in the ferrocyanide-mediated electrochemical measurements. Furthermore, the investigation of electrochemical electron transfer, according to the sequence of oligonucleotide molecules, was also undertaken. This work demonstrates that direct label-free oligonucleotide electrical recognition, based on biofunctional streptavidin-modified gold electrodes, could lead to the development of a new biosensor protocol for the expansion of rapid, cost-effective detection systems.
A label-free DNA detection method was developed for a simple electrochemical DNA sensor with a short assay time. Self-assembled monolayers of peptide nucleic acid were used as a probe on gold electrodes. The formation of the self-assembled monolayers on the gold electrodes was successfully checked by means of cyclic voltammetry. The target DNA, hybridized with peptide nucleic acid, can be detected by the anodic peak current of ferrocene dendrimers, which interact electrostatically with the target DNA. This anodic peak current was measured by square wave voltammetry at 0.3 V to decrease the detection limit on the order of the nanomolar concentrations. As a result, the label-free electrochemical DNA sensor can detect the target DNA in concentrations ranging from 1 nM to $1\;{\mu}M$ with a detection limit of 1 nM.
In this paper, we fabricated and evaluated polydimethylsiloxane(PDMS)-based flexible and implantable micro electrodes. The electrode patterning was carried out with the photolithography and chemical etching process after e-beam evaporation of 100 ATi and 1000 A Au. The PDMS substrate was treated by oxygen plasma using reactive ion etching(RIE) system to improve the adhesiveness of PDMS and metal layers. The minimum line width of fabricated micro electrode was 20 $\mu$m. After finished patterning, we did packaging with PDMS and then brought up the electrode's part about 40 $\mu$m with gold electroplating. The Hank's balanced salt solution(HBSS) test was carried out for 6 month for endurance of fabricated micro electrode. We carried out in-vivo test for the evaluation of biocompatibility by implanting electrodes under the ICR mouse skin for 42 days.
Structually organized mono- and multilayers were developed on gold for the catalytic and affinity biosensing using hyper-branched dendrimers. For the catalytic biosensing interface, a new approach to construct a multilayered enzyme film on the electrode surface was developed. The film was prepared by layer-by-layer depositions of dendrimers and periodate-oxidized glucose oxidase. The voltammograms obtained from the GOx/dendrimer multilayered electrodes revealed that bioelectrocatalytic response is directly correlated to the number of deposited bilayers. From the analysis of voltammetric and ellipsometric signals, the coverage of active enzyme per layer during the layering steps was estimated, demonstrating the spatially-ordered multilayer formation. As an extension of the study, dendrimers having various degrees of ferrocenyl modification were prepared and used. The resulting electrodes were electrochemically characterized, and the density of ferrocenyl groups, active enzyme coverage, and sensitivity were estimated. For the affinity-sensing surrface, a biosensor system based on avidin-biotin interaction was developed. As the building block of affinity monolayer, G4 dendrimer having partial ferrocenyl-tethered surface groups was prepared and used. And the biotinylated and electroactive dendritic monolayer was used for the affinity-sensing surface interacting with avidin. Electrochemical characterization of the resulting biosensor was conducted using free enzyme in electrolyte in terms of degree of surface coverage with avidin and subsequent surface shielding.
전극과 용액 사이 계면에서 일어나는 전기화학 반응 현상을 보다 정확하게 이해하기 위하여 전기화학 반응과정을 분광학적 방법으로 실시간으로 모니터링 할 수 있는 전극으로도 작동할 뿐만 아니라 표면증강라만산란(SERS) 활성도 강한 금 마이크로쉘을 제조하였다. 기존에 보고된 금 마이크로쉘에서 핵으로 사용한 폴리스티렌의 경우 균일성이 떨어지고 유기용매에 약하며 독성이 있다. 이에 본 연구에서는 폴리스티렌 보다 균일한 구조를 가지고, 유기 용매에서도 사용 가능하며 무독한 실리카 비드를 이용하여 금 마이크로쉘을 만들고 높은 SERS 신호를 낼 수 있도록 최적화시켰다. $2{\mu}m$ 실리카 비드 표면에 서로 다른 양의 3-aminopropyl triethoxysilane (APTES)를 반응시켜 얻은 금 마이크로쉘에서 SERS 신호가 가장 월등히 증폭되는 조건을 비교한 결과 1% (v/v) APTES 조건에서 SERS 신호의 증폭이 가장 컸다. 표면증강라만산란 스펙트럼 및 전계방출형 주사전자현미경(FE-SEM) 이미지를 통해 금 마이크로쉘을 분석하였다.
Excessive presence of heavy metals in environment affects plants and fruits grown in the contaminated area. Rapid on-site monitoring of heavy metals can provide useful information for efficiently characterizing heavy metal-contaminated sites and for minimizing the exposure of the contaminated food crops to humans. This study reports on the evaluation of gold and glassy carbon (GC) electrodes with mercury or bismuth as a coating material for simultaneous determination of cadmium (Cd) and lead (Pb) in 0.1 M $HNO_3$ solution by anodic stripping voltammetry (ASV). The use of a square-wave voltammetric potential between a working electrode and a reference electrode caused Cd and Pb ions deposited on the electrode surface to be oxidized, thereby generating electric currents at different potentials. The mercury-coated gold electrode was not sensitive enough to detect the usable range of Cd concentrations (1 to 100 ppb). The GC electrodes with mercury or bismuth displayed well-defined, sharp and separate current peaks for Cd and Pb ions when the square-wave voltammetric potentials were applied. The peak currents measured with both mercury- and bismuth- coated GC electrodes were linearly proportional to Cd and Pb concentrations in the range of 1 to 200 ppb in 0.1 M $HNO_3$ with strong linear relationships between concentration and peak current ($R^2$ > 0.95), indicating that both of Cd and Pb ions could be quantitatively measured.
Si Nanowire (NW) field effect transistors (FETs) were fabricated on hard Si and flexible polyimide (PI) substrates, and their electrical characteristics were compared. Si NWs used as channels were synthesized by electroless etching method at low temperature, and these NWs were refined using a centrifugation method to get the NWs to have an optimal diameter and length for FETs. The gate insulator was poly(4-vinylphenol) (PVP), prepared using a spin-coating method on the PI substrate. Gold was used as electrodes whose gap was 8 ${\mu}m$. These gold electrodes were deposited using a thermal evaporator. Current-voltage (I-V) characteristics of the device were measured using a semiconductor analyzer, HP-4145B. The electrical properties of the device were characterized through hole mobility, $I_{on}/I_{off}$ ratio and threshold voltage. The results showed that the electrical properties of the TFTs on PVP were similar to those of TFTs on $SiO_2$. The bending durability of SiNWs TFTs on PI substrate was also studied with increasing bending times. The results showed that the electrical properties were maintained until the sample was folded about 500 times. But, after more than 1000 bending tests, drain current showed a rapid decrease due to the defects caused by the roughness of the surface of the Si NWs and mismatches of the Si NWs with electrodes.
