The galactolipid M-5, which showed anti-inflammatory activity is glycoglycerolipid isolated from the Okinawa marine sponge Phyllospongia foliascens. Glycolipids have been synthesized by various methods, especially it were generally known that synthetic method of M-5 analogue and synthetic method of various glycolipids by glycosidation after synthesis of glycerolipid part. The others, it was not suggested that synthetic method via glycosylglycerol obtained by degradation from diglycoside. This study was carried out to investigate the synthesis of galactosylglycerol from melibiose as M-5 intermediate. Synthesis of galactosylglycerol was accomplished by selective protection of hydroxy group of sugar and diol cleavage by Pb(OAc)$_4$. As a result, galactosylglycerol was synthesized by 8 steps pathway and their structures were elucidated by analysis instrument.

For a studyon the production of ${\gamma}$-linolenic acid(GLA) by fungi 3 fungal strains were isolated from soil. Their cell growth lipid content and fatty acid composition were compared in shake flask culture. Among these fungi the fungus designated as FA-007 has high lipid content(21.1%) and GLA content(15.6% of total fatty acids) The fungal strain FA-007 was tentatively identified as Mucor sp. on the basis of morphological characteristics, Fungal oil produced by this fungus was composed of 75.2% neutral lipid 5.3% glycolipid and 19.5% phospholipid. Although the GLA content in phospholipid was higher than it in neutral lipid the GLA content in neurtal lipid was high as 15.5%.
To investigate the possible roles of LAMMER kinase homologue, Lkh1, in Schizosaccharomyces pombe, whole proteins were extracted from wild type and lkh1-deletion mutant cells and subjected to polyacrylamide gel electrophoresis. Differentially expressed proteins were identified by tandem mass spectrometry (MS/MS) and were compared with a protein database. In whole-cell extracts, 10 proteins were up-regulated and 9 proteins were down-regulated in the mutant. In extracellular preparations, 6 proteins were up-regulated in the lkh1+ null mutant and 4 proteins successfully identified: glycolipid anchored surface precursor, $\beta$-glucosidase (Psu1), cell surface protein, glucan 1,3-$\beta$-glucosidase (Bgl2), and exo-1,3 $\beta$-glucanase (Exg1). These results suggest that Lkh1 is involved in regulating cell wall assembly.

Further investigation of Lycium chinense fruits gave a mixture of (6'-O-palmitoyl)- and (6'-Ostearoyl)-$\beta$-sitosterol-3-O-$\beta$-D-glucopyranoside (1) and two glycolipids, 1-O-(9Z, 12Z, 15Z-octadecatrienoyl)-2-O-(9Z, 12Z, 15Z-octadecatrienoyl)-3-O-$\beta$-D-galactopyranosyl glycerol (2) and 1-O-(9Z, 12Z-octadecadienoyl)-2-O-(9Z, 12Z, 15Z-octadecatrienoyl)-3-O-$\beta$-D-galactopyranosyl glycerol (3). These compounds were newly isolated as constituents of L. chinense.
Purpose : Infection with Shiga-like toxin (SLT)-producing Escherichia coli, an emerging human pathogen found particularly in young children under 5 years of age, causes a spectrum of illnesses with high morbidity and mortality, ranging from diarrhea to hemorrhagic colitis and hemolytic uremic syndrome. Host mediators play an important role in the pathogenesis of SLT-I toxicity. The experiments described here were designed to investigate the effect of SLT-I on TNF-${\alpha}$ production and to understand the effect of TNF-${\alpha}$ on GB3 expression. We also further examine the relationship between the Gb3 level and the differential susceptibility of cells to the cytotoxic action of SLT-I. Methods : The effect of purified SLT-1 from E. coli O157 : H7 (ATCC 43890) on tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) production in Raw264.7 cells was investigated. Many mediators regulate endothelial cell membrane expression of the glycolipid globotriaosyleramide (Gb3), which serves as the toxin receptor, suggesting that the host response to the toxin or other bacterial products may contribute to pathogenesis by regulating target cell sensitivity to the toxins. Therefore, the relationships between Gb3 expression and cytotoxicity against SLT-I on three types of cells were evaluated. Results : Detectable levels of TNF-${\alpha}$ were produced as early as six hours after induction and continued to increase during 48 hours by SLT-I. It was also found that Vero cells and dendritic cells (DC2.4 cells) expressed high levels of Gb3, 83% and 68%, respectively, and that Raw264.7 cells had a low level of Gb3 (29%) and appeared refractory to cytotoxicity against SLT-I. Vero cells and DC2.4 cells expressing high levels of Gb3 were highly susceptible to SLT-I. Furthermore, macrophages showed a resistance to SLT-I cytotoxicity, despite the fact that Gb3 expression was enhanced. Conclusion : These results strongly suggest that the expression of Gb3 is necessary but not sufficient to confer sensitivity of macrophages to SLT-I and further underpin the important role of SLT-I and its Gb3 receptors in the pathogenesis of E. coli O157 infection.

To evaluate the quality of lipids and their fatty acid compositions in diploid and triploid carps(Cyprinus carpio), we analyzed the compositions of neutral lipid, phospholipid and glycolipid, and fatty acid contents of each lipids in these carp muscles. Total lipid contents of diploid carps, nonspawning season and spawning season, and triploid carp were $2.5\~3.0\%,\;1.4\~1.9\%\;and\;2.4\~3.1\%$, respectively. Regardless of belly and dorsal muscles, diploid carp of nonspawning season and triploid one were higher in neutral lipid, but lower in glycolipid and phospholipid than those of diploid one of spawning season. The neutral lipid and phospholipid were mainly consisted of triglyceride($66.9\~86.1\%$), and phosphatidyl-choline($55.6\~62.9\%$), respectively. In fatty acid compositions of total lipid, triploid carp showed higher content in monoenes such as 16: 1, 18: 1, but lower in polyenes such as 22:6 than those of diploid carp. The major fatty acids in total lipid were 18:1, 16:9, 18:2, 18:3 and 16:1 in order. The fatty acid compositions of neutral lipids showed similar trend to those of total lipid, and the major fatty acids in phospholipid were 16:0, 18:1, 18:2 and 22:6, These fatty acid compositions of neutral lipid, glycolipid and phospholipid were slightly difference between dorsal and belly muscles in all the carps.
Barley koji was made in order to investigate the lipid contents of barely koji during preparation. Diethyl ether extracts and 85% methanol extracts were extracted and purified. The lipid components were classified. The individual neutral lipids, glycolipids and phospholipids were fractionted, quantified and fatty acid compositions of the three lipids were determined. Total lipid contents of diethyl ether and 85% methanol extract of barley koji increased during preparation. Neutral lipid, glycolipid and phospholipid contents in diethyl ether extract increased, however, neutral lipid, glycolipid and phospholipid contents in 85% methanol extract decreased during koji preparation. TG content of the neutral lipid in diethyl ether extract decreased. Conversely, DG, FS, FFA and ES contents increased. But TG, DG and FS contents of the neutral lipid in 85% methanol extract decreased. LPC, (PC+PS), PI, PG and PE contents of the phospholipid on diethyl ether extract increased. But LPC, (PC+PS), PE and PI contents in 85% methanol extract decreased during koji preparation. Palmitic acid, linoleic acid and linolenic acid of neutral lipid in diethyl ether extract decreased, however, palmitic, stearic, oleic, linoleic acid in 85% methanol extract decreased. Palmitic acid, stearic acid, oleic acid and linoleic acid of glycolipid in diethyl ether extract increased, but in 85% methanol extract they decreased except oleic acid. Palmitic, stearic, oleic, linoeic and linolenic acid of phospholipid in diethyl ether extract increased during koji preparation. On the other hand palmitic, oleic and linoleic acid in 85% methanol extract decreased but stearic and linoleic acid increased.
