Analysis of Amino Acids, Vitamins and Minerals of Fruiting Body of Fomitopsis pinicola (소나무 잔나비버섯의 아미노산, 비타민 및 무기물 분석)

  • The nutritional composition of Fomitopsis pinicola (F. pinicola) fruiting body has been analyzed for medicinal and edible uses. The contents of crude fibers, carbohydrates, crude protein, moisture, crude fats and ashes were 43.3%, 26.3%, 12.8%, 12.6%, 3.3% and 1.7%, respectively. Eighteen amino acids were found in F. pinicola. Among total amino acids, glutamate content was the highest (457 mg/100 g dry mushroom) and arginine, glycine, valine, aspartate and isoleucine were followed. Concerning free amino acids, glutamine, arginine, trytophan, and glutamate were dominant. The vitamin E content was the highest (276 mg/100 g dry mushroom), then vitamin H and vitamin B, were followed. The mineral contents were as follows: K 165.06 mg, P77.57 mg, Mg 46.11 mg, Fe 21.56 mg, and Ca 16.90 mg based on 100 g dry mushroom.

Identifying N sources that affect N uptake and assimilation in Vanda hybrid using 15N tracers

  • Vanda is an aerial tropical orchid native to Thailand and nitrogen (N) fertilizer is mainly used to promote its growth and quality. However, little is known about the characteristics of N absorption and assimilation in Vanda. The objective of this study was to determine the appropriate source of N for Vanda cultivation. In this experiment, shoots and roots of Vanda 'Ratchaburi Fuchs-Katsura' were sprayed weekly with 100 ml of $^{15}N$ tracer solution (1) 10 mM of $^{15}NO_3{^-}$, (2) 5 mM of $^{15}NO_3{^-}$ plus 5 mM of $NH_4{^+}$, (3) 5 mM of $NO_3{^-}$ plus 5 mM of $^{15}NH_4{^+}$ and (4) 10 mM of $^{15}NH_4{^+}$. The results indicated that plants fed with a combined N fertilizer gave the highest of $^{15}N$ use efficiency ( $^{15}NUE$) of about 21.8%, 30 days after the first feeding (DAF), compared with those fed sole sources of $^{15}NO_3{^-}$ (21.0%) and $^{15}NH_4{^+}$ (16.6%). However, a sole nitrate fertilizer or combination fertilizer did not significantly affect the total N and labelled N content. Alanine was a major amino acid found in leaves and roots at 7 DAF, whereas glutamine was mainly found in stems. At 30 DAF, tyrosine and alanine became major components in the leaves, and glutamine decreased in stems when plants were fed with a single $^{15}NH_4{^+}$ source.

T-Cell Death-Associated Gene 51 Is a Novel Negative Regulator of PPARγ That Inhibits PPARγ-RXRα Heterodimer Formation in Adipogenesis

  • The nuclear receptor peroxisome proliferator-activated receptor γ (PPARγ) is the master transcriptional regulator in adipogenesis. PPARγ forms a heterodimer with another nuclear receptor, retinoid X receptor (RXR), to form an active transcriptional complex, and their transcriptional activity is tightly regulated by the association with either coactivators or corepressors. In this study, we identified T-cell death-associated gene 51 (TDAG51) as a novel corepressor of PPARγ-mediated transcriptional regulation. We showed that TDAG51 expression is abundantly maintained in the early stage of adipogenic differentiation. Forced expression of TDAG51 inhibited adipocyte differentiation in 3T3-L1 cells. We found that TDAG51 physically interacts with PPARγ in a ligand-independent manner. In deletion mutant analyses, large portions of the TDAG51 domains, including the pleckstrin homology-like, glutamine repeat and proline-glutamine repeat domains but not the proline-histidine repeat domain, are involved in the interaction with the region between residues 140 and 506, including the DNA binding domain, hinge, ligand binding domain and activation function-2 domain, in PPARγ. The heterodimer formation of PPARγ-RXRα was competitively inhibited in a ligand-independent manner by TDAG51 binding to PPARγ. Thus, our data suggest that TDAG51, which could determine adipogenic cell fate, acts as a novel negative regulator of PPARγ by blocking RXRα recruitment to the PPARγ-RXRα heterodimer complex in adipogenesis.

Chemosensitizing effect and mechanism of imperatorin on the anti-tumor activity of doxorubicin in tumor cells and transplantation tumor model

  • Multidrug resistance of tumors has been a severe obstacle to the success of cancer chemotherapy. The study wants to investigate the reversal effects of imperatorin (IMP) on doxorubicin (DOX) resistance in K562/DOX leukemia cells, A2780/Taxol cells and in NOD/SCID mice, to explore the possible molecular mechanisms. K562/DOX and A2780/Taxol cells were treated with various concentrations of DOX and Taol with or without different concentrations of IMP, respectively. K562/DOX xenograft model was used to assess anti-tumor effect of IMP combined with DOX. MTT assay, Rhodamine 123 efflux assay, RT-PCR, and Western blot analysis were determined in vivo and in vitro. Results showed that IMP significantly enhanced the cytotoxicity of DOX and Taxol toward corresponding resistance cells. In vivo results illustrated both the tumor volume and tumor weight were significantly decreased after 2-week treatment with IMP combined with DOX compared to the DOX alone group. Western blotting and RT-PCR analyses indicated that IMP downregulated the expression of P-gp in K562/DOX xenograft tumors in NOD/SCID mice. We also evaluated glycolysis and glutamine metabolism in K562/DOX cells by measuring glucose consumption and lactate production. The results revealed that IMP could significantly reduce the glucose consumption and lactate production of K562/DOX cells. Furthermore, IMP could also remarkably repress the glutamine consumption, α-KG and ATP production of K562/DOX cells. Thus, IMP may sensitize K562/DOX cells to DOX and enhance the antitumor effect of DOX in K562/DOX xenograft tumors in NOD/SCID mice. IMP may be an adjuvant therapy to mitigate the multidrug resistance in leukemia chemotherapy.

Studies on the Chemical Composition of Grape Juices in Korea (한국산(韓國産) 포도의 과즙성분(果汁成分)에 관(關)한 연구(硏究))

  • The chemical composition of the of grape juices produced in Korea in 1969 were analyzed and the amino acids and sugars in these juices were detected by means of paper partition chromatography. The results are summarized as follows: 1. Less sugar and amino-N and more acid were contained in Korean grape juices than those of foreign's, and tannin content was not so different as foreign's. 2. More sugar, amino-N and tannin and less acid were contained in the juices of Campbell Early obtained in Anyang district than in Taejon and Pohang districts. 3. Twenty-one amino acids detected in grape juices were distributed as following frequency. Aspartic acid, serine, glycine, asparagine, lysine, arginine, threonine, alanine ${\gamma}-amino$ butyric acid, valine, leucine, proline (in 11 varieties), glutamine, tyrosine(10), cystine(9), glutamic acid, Hydroxyproline(8), isoleucine(4), phenylalanine, unknown(3). 4. Alanine was mostly abundant in all varieties and ${\gamma}-amino$ butyric acid was next and the decreasing order were arginine, valine, leucine, proline, glutamine, threonine, etc. in the amount. 5. Number of amino acid detected in grape juices of each varieties were 20 sorts in Delaware(Anyang), 19 in Black Hamburg, and Schuyler, 18 in Campbell Early(three districts), Delaware(seedless, Taejon) and Alden, 17 in Niagara, 16 in Muscat Hamburg, and 15 in Golden Muscat. 6. Number of essential amino acids contained in Delaware(Anyang) and Black Hamburg were 6 sorts and in Campbell Early (Anyang), Niagara and Muscat Hamburg were 5 and in others 4. 7. The same number of amino acid were detected in the juices of Campbell Early obtained from three districts, but hydroxyproline was detected in that of Anyang only, while isoleucine was appeared in those of Taejon and Pohang. 8. Glucose, fructose were detected in all grape juices.

Analysis of Nectar Characteristics of Idesia polycarpa (유망 밀원수종 이나무의 화밀 특성 분석)

  • We assessed the nectar source potential of a prospective honey plant, Idesia polycarpa, by analyzing nectar volume, free sugar content, and free amino acid content. Idesia polycarpa is a dioecious tree; the males bloom approximately four days earlier than females, and the blooming period is approximately 17 days-from March 14th to March 30th. Upon investigating the patterns of nectar secretion, it was found that male flowers peak on the third day of blooming at 5.0 ± 2.5 μL, and female flowers peak on the second day of blooming, at 1.1 ± 0.4 μL. There was a significant difference between males and females in the total nectar volume (9.7 ± 2.9 μL for males and 1.7 ± 0.5 μL for females) and the dried nectar volume (2.2 ± 0.6 μL for males, 0.8 ± 0.3 μL for females) during the blooming period. The free sugar content of floral nectar was 54.6 ± 15.4 ㎍/μL for males and 20.5 ± 4.9 ㎍/μL for females, and the sugar content per flower was higher in males (170.7 ± 15.4 ㎍) than in females (24.9 ± 5.5 ㎍). Our analysis of the amino acid content showed that 20.4 ± 3.9 mg/L (comprised of 19 amino acids) is produced in male flowers and 3.2 ± 0.1 mg/L (11 amino acids) in female flowers. In the male flower, the main amino acid was glutamine, followed by asparagine and proline, whereas in the female nectar, asparagine was the main amino acid, followed by glutamic acid and glutamine. Idesia polycarpa blooms after the blooming period of a major honey plant, Robinia pseudoacacia, and its nectar volume and nectar characteristics, such as free sugar content and amino acid content, make it a viable honey plant.

  • A high cell density culture system for the anchorage dependent CHO cells was developed based on the combination of in removal of ammonium ion and microcarrier culture system, and semi-fed-batch feeding of glucose and glutamine was employed to the developed culture system. The glass bead was selected as an optimum microcarrier in terms of cell growth. An ammonium ion selective zeolite, Phillipsite-Gismondine, was packed in a dialysis menium ion. The semi-fed-batch operation was employer to the novel culture system for the high density cell culture, and the results showed the cell growth was improved by 32% and tPA productivity by 250%.

  • Some biological activities of Beauveria bassiana were studied to elucidate pharmacological function of B. bassiana-infected larva of the silkworm. The mycelium consisted mainly of carbohydrate (65.8%), followed by protein (15.9%) and fat (8.3%). Glucose (68.8%), mannose (7.1%), and galactose (6.1%) were major components in carbohydrates. Ten amino acids including glutamine, threonine, valine, aspartic acid, alanine, leucine, serine, glycine, arginine, and isoleucine were found in protein as major amino acids. Various extracts were prepared from the freeze-dried mycelium of B. bassiana by systemic extraction and their biological activities were investigated. Among tested fractions, the hot-water extract (HW) contributed significantly to the anti-coagulant activity, anti-complementary activity, and stimulation of intestinal immune system. The methanol extract (ME) increased acetylcholinesterase (AChE) inhibition activity and reactive oxygen species (ROS) scavenging activity.

  • 1) Rabbit follicular oocytes from preovulatory follicles were cultured for 12 hr in vitro and fertilized in vivo by transferring the oocytes to the first foster-mother. 2) Two youngs were bron from transferred embryos from the first foster-mother to the second foster-mother. This demonstrates that in vitro cultured follicular oocytes are normal and they can develop into normal young born when transferred to the foster-mother. 3) A simple chemically defined culture medium, salt sol. with glutamine (2mM), which was developed by Bae and Foote(1975) proves fully good enough for rabbit follicular oocyte culture. We call this B-F medium. 4) Twelve hours culture in vitro of the rabbit follicular oocyte may be a proper culture time for further development.

Influence of Amino Acid and Vitamin Addition on the Growth and Metabolism of a Hybridoma in Batch Culture (하이브리도마의 회분식배양에서 아미노산과 비타민의 첨가에 따른 세포성장과 대사의 변화)

  • The effects of various step-fortifications of the initial medium with amino acids, glucose, and vitamines on the growth and metabolism of a hybridoma cell line in batch cultures were quantified. Comparisons between the metabolic rates of the various cultivations were made for the exponential growth phase. Fortification of the basal medium resulted in higher cell densities through a prolonged growth phase, but the maximum specific growth rate was not affected. The uptake rate of glutamine increased with the addition of amino acids but did not change upon the addition of glucose or vitamines. The specific glucose consumption decreased slightly with the addition of amino acids but increased production of lactate and {{{{ { NH}`_{4 } ^{ +} }}}}. A reciprocal relationship between the yields of {{{{ { NH}`_{4 } ^{+ } }}}} and lactate indicated a joint regulation of glycolysis and glutaminolysis.
