Electrophoretic Variations in Protein and Some Enzymes during Stratification of Panax ginseng Seeds (인삼종자의 개갑처리과정에 있어서 단백질 및 몇가지 효소의 전기영동특성변이)

  • The electrophoretic variations in protein and some enzymes during stratification of Panax ginsseng seeds were checked in relation to the differentiation and development of immature embryo, Seed-protein on PAGE showed eight major bands during the period of investigation, Four bands of Rf : 0.12, 0.24, 0.66 and 0,72 remained in dark stained condition, whereas the remainders (Rf : 0.40, 0.48, 0.56 and 0,76) became lighter in the staining condition as the treatment period was prolonged, In embryo, which was separately sampled from 75 days after the treatement, the only band of Rf 0.76 was not observed, In peroxidase, the number of bands was gradually increased and came to four bands at 45 days after treatment, and their staining color became gradually darker until the time of final samling. Also, the embryo had the same patterns as those in the intact seed. Esterase showed four bands during the period of investigation. As the treatment period was prolonged, the degree of staining condition came to darker in the band of Rf 0.36, but lighter in the band of Rf 0.56. The band of Rf 0.20 was not detected in the intact seed at 90 days after the treatment, but clearly observed in embryo. Glutamate dehydrogenase had only two bands: one (Rf : 0.16) was detected only at 60 days after the treatment and the other (Rf: 0.14) was always observed throughout the period, and showing simple pattern in the electrophoretic variation. Glutamate-oxaloacetate transaminase showed the only band of Rf 0.36 during the period of investigation and even in embryo, and showing the simplest pattern, indeed.

Protection of spontaneous and glutamate-induced neuronal damages by Soeumin Sibjeundaibo-tang and Soyangin Sibimijihwang-tang in cultured mice cerebrocortical cells

  • Soeumin Sibjeundaibo-tang (SJDBT) and Soyangin Sibimijihwang-tang (SMJHT) have been used traditionally to improve the systemic blood circulation and biological energy production in the patients with circulatory and neuronal diseases. The object of this study is to determine the protective effects of SJDBT and SMJHT extracts on the spontaneous and glutamate-induced neuronal damages in cultured cells derived from mice cerebral cortex. At 14 days after beginning the cultures, the activity of lactate dehydrogenase released into the culture media was significantly decreased by treatment of cerebroneuronal cells with SJDBT and SMJHT (0.1 mg/ml) for 7 days. By comparison with the normal cells, cerebroneuronal morphology was dramatically changed by treatment of glutamate (1 mM) for 12 hrs, and this was conspicuously recovered by pretreatment of cerebroneural cells with SJDBT and SMJHT (0.1-1.0 mg/ml) for 2 days. Moreover, glutamated-induced DNA fragmentation was also protected by pretreatment of cerebroneuronal cells with those extracts. These results suggest that naturally occurring and glutamate-induced degeneration of cultured cerebrocortical cells may be related, in part, to the process of apoptotic cell death. The pharmacological properties of SJDBT and SMJHT extracts to improve cerebroneuronal degeneration may be considered as one of useful medicines that can prevent cerebrocortical impairments resulted from age-dependent and excitotoxicity-induced neuronal degeneration in human brain.

Morphological and Biochemical Characteristics in the Korean Landraces of Mulberry Tree, Morus (한국 재래뽕의 생태 및 품종간 특이성에 관한 연구)

  • This study was conducted to characterize mulberry varieties by the analysis of morphological characters as well as biochemical and molecular biological markers. As for the budding stage Geumsang, Chosang i and Yeongbyeonchuwoo were early, but Dangsang 6, Hwangchuwoo were late. The lowest varietiy in rate of death atop was Dangsang 8(0.0%). Suncheonppong was the highest leaf yields in spring and autumn rearing season. In biochemical isozyme analysis, peroxidase gave good zymogram patterns in isoelectric focusing electrophoresis. There were high variations in RAPD analysis among the mulberry trees. From the obtained peroxidase and RAPD variations, cluster phylogenetic analysis was carried out using NT-SYS PC program. There were no clear grouping patterns between native varieties and leading varieties. The highest similarity was observed between Suwonsang 1 and Suwonsang 2 at about 90% similarity level.

  • Enzyme electrodes for amperometric measurement of ammonia was prepared by immobilization of L-glutamate dehydrogenase on an Immobilon-AV Affinity membrane and attachment to a glassy carbon electrode. Reduced nicotinamide adenine dinucleotide (NADH) was used as the electroactive species. The electrochemical oxidation of NADH was monitored at +1.0 volt vs. Ag/AgCl. Response was linear from $4.0\;{\times}\;10^{-5}\;to\;4.0\;{\times}\;10^{-4}$ M. The detection limit was 2.0 ${\times}\;10^{-6}$ M. Response time, the optimum pH and life time of enzyme immobilized membrane were 2 min, pH 7.3∼7.6 (Dulbecco's buffer solution) and about 25 days respectively. When the enzyme electrode was applied to the $NH_4^+$ determination with amperometric method, other physiological materials had no interference.

Isozyme Variants in Genus Pinus by Simulated Acidic Rain (인공 산성비가 소나무속의 동위효소 양상에 미치는 영향)

  • The effect of acid rain on plant isozyme response was studied with the treatment of simulated acid rain to Pinus densiflora, P. nigida and P. koraiensis for 9 months. The isozyme pattern of $\alpha$-Esterase($\alpha$-Est), Peroxidase(POD) and Glutamate dehydrogenase(GDH) were observed in the control (pH 5.6) and simulated acid rain (pH 3.5) treatment. No changes of isozyme pattern in $\alpha$-Est was observed in P. densiflora after 9 month treatment of simulated acid rain, but, two new isozymes were activated in P. nigida in the same treatment. In P. koraiensis, two new isozyme were activated but five isozymes were not activated. P. densiflora did not show any difference in POD and GDH after the treatment of simulated acid rain. P. nigida showed activation of eight and two isozymes in POD and GDH, respectively. P. koraiensis showed inactivation of 4 isozymes in POD but showed no changes In GDH.

Effect of Medicinal Plant Extracts on Alcohol Metabolism in Rat Liver

  • The experiment was conducted to evaluate the effects of medicinal plants on ethanol-metabolism. Sprague Dawley rats divided into 6 groups (n=8), fed with 10% ethanol and diets supplemented with each 1% of four plant extracts, ${\alpha}-tocopherol$ (as positive control) and fiber (as negative control) for 4 weeks. Group supplemented with plant extract of Ulmus davidiana showed the most high value (322 nM NADH/min/mg protein) in alcohol dehydrogenase (ADH) activity among the experimented groups $(144{\sim}312\;nM\;NADH/min/mg\;protein)$ at p<0.05. Groups fed with Lagerstroemia indica and Zelkova serrata extract-supplemented diets indicated high activity in aldehyde dehydrogenase (ALDH, 16.7 & 12.3 M NADH/min/mg protein), which were comparatively lower than 20.1 M NADH/min/mg protein of ${\alpha}-tocopherol$ fed group. All of the groups fed with plant extracts indicated very low GPT activities $(13.9{\sim}17.3\;IU/l)$ compared to those (146.1 & 128.6 IU/l) fed with ${\alpha}-tocopherol$ and fiber at p<0.05. From these results, it is suggested that Lagerstroemia indica have a potent ethanol-metabolizing activity.

Protective Effect of MeOH Extract of Evodia officinalis on Cyanide-induced Neurotoxicity in Cultured Neuroblastoma Cells (오수유 MeOH 추출물이 Cyanide에 의한 신경세포의 보호효과에 미치는 영향)

  • We reported that neurotoxicity may contribute to cyanide-induced neuronal injury. Cyanide stimulates the release of glutamate which can activate glutamate receptors to propagate excitotoxic processes. We examined the role of plant extracts in mediating the cyanide-induced cytotoxicity and report here that the cytotoxicity assessed in SK- N-SH cell cultures by measuring lactate dehydrogenase (LDH) in the culture media was significantly blocked by Evodia officinalis MeOH extract (OMU). Also, when OMU was treated in NaCN level cultures, the neurite outgrowth was regenerated as much as in the treatment of NaCN only. These results indicate that OMU treatment were not only protected the neurons against NaCN-induced damage but also regenerated the neurite outgrowth of neuroblastoma cells.

Anti-ecotoxicological Glycoprotein Isolated from Ulmus davidiana Nakai Inhibits Fecal Malodor and Promotes Feed Efficiency in Mice (환경 독성을 억제하는 느릅 당단백질이 마우스의 분뇨 악취저감 및 사료 효율에 미치는 영향)

  • Ulmus davidiana Nakai (UDN) has been traditionally used as a herbal medicine in Korea. In the present study, we investigated the anti-ecotoxic potential of a 116 kDa glycoprotein isolated from UDN (UDN glycoprot ein) in regulating fecal malodor and feed efficiency in mice. We found that UDN glycoprotein (200 μg/ml) has an inhibitory effect on the cell death induced by an ecotoxicological endocrine disrupting chemical, bisphenol A, in colon epithelial HT-29 cells. UDN glycoprotein did not show significant differences regarding the weight of ecotoxicity-related organs (liver, heart, kidneys, and spleen) and the levels of serum glutamate oxaloacetate transaminase, glutamate pyruvate transaminase, and lactate dehydrogenase in mice for 2 weeks, compared to the control. Additionally, UDN glycoprotein reduced the levels of hydrogen sulfide and ammonia as markers of fecal malodor in mice. Interestingly, UDN glycoprotein can improve the mouse feed efficiency. In conclusion, our data indicate that anti-ecotoxicological UDN glycoprotein has the ability to increase the feed efficiency and reduce the fecal malodor by maintaining the viability of colonic epithelial cells in mice.

Effects of Electroacupuncture on Activity of GOT, GPT, LDH and Functional Recovery in the Motor Injury Rats by the 6-hydroxydopamine (6-hydroxydopamine에 의한 운동손상 흰 쥐에서 전침이 GOT, GPT, LDH 활성도 및 기능회복에 미치는 영향)

  • Purpose : This study was investigated the effect of electroacupuncture stimulation on the change of blood biochemical components in the rat spinal cord injury(SCI) damaged by the 6-hydroxydopamine. Methods : SCI model rats were damaged in L1-L2 injected with 6-hydroxydopamine. The thirty Sprague-Dawley adult male rats were randomly divided into normal group, control group and electroacupuncture group. Experimental groups were applied as electroacupuncture(Es-160, ITO, Japan) for 15minutes during the low frequency(2 Hz) stimulation to zusanli. The enzyme concentration levels analysis of the hematological changes were measured of Glutamate Oxaloacetate Transaminase(GOT), Glutamate Pyruvate Transaminase(GPT), Lactate dehydrogenase(LDH) and motor function recovery change was evaluated by the rota-rod test. Results : This study were as follow : The concentration of GOT, LDH in experimental group was lower than control group(p<.05). The experimental group showed increase of motor function recovery more in compared to control group(p<.05). Conclusion : The results of this study showed that electroacupuncture to zusanli point have an effect on functional recovery after the 6-hydroxydopamine induced SCI in rats.

Dioscorea batatas Decne Glycoprotein Prevents Ecotoxicological Effects of Bisphenol A in Gastrointestinal Epithelial Cells and Improves Fecal Malodor and Feed Efficiency in Mice (환경호르몬 비스페놀 A가 유도한 위장관 세포독성 제어효과를 가진 마 당단백질이 마우스의 식이 효율 및 악취저감에 미치는 영향)

  • As a herbal supplement, Dioscorea batatas Decne (DBD) presents potent antioxidant activity and diverse health benefits. In the present study, functions of a 30 kDa glycoprotein isolated from DBD (hereafter, DBD glycoprotein) in the regulation of feed efficiency and fecal malodor in mice were explored. DBD glycoprotein produced protective effect against cytotoxicity induced by the ecotoxicological endocrine-disrupting substance bisphenol A in gastrointestinal epithelial HT-29 cells. To investigate its potential roles in the regulation of feed efficiency and fecal malodor, mice were administered an oral injection of DBD glycoprotein for 2 weeks. Compared with the control values, the weight of internal organs (liver, heart, kidney, and spleen) and levels of glutamate pyruvate transaminase, glutamate oxaloacetate transaminase, and lactic dehydrogenase were not significantly changed during DBD glycoprotein administration for 2 weeks. Interestingly, DBD glycoprotein improved feed efficiency and reduced hydrogen sulfide concentration without altering the ammonia level in mouse feces. Collectively, these results indicate that DBD glycoprotein is a functional agent that exerts gastrointestinal protective effects against ecotoxicological substances, improves feed efficiency, and reduces fecal malodor.
