The antioxidant activities of water, ethanol, methanol, and chloroform extracts of Ulmus davidiana were evaluated using various antioxidant assays: DPPH (1,1-diphenyl-2-pricrylhydrazyl) free radical scavenging; hydroxyl radical scavenging; lipid peroxidation scavenging; and reducing power assays. All extracts, except the chloroform extract, demonstrated strong antioxidant activity in all assays. The chloroform extract had the highest hydroxyl radical scavenging activity, and its activity was equivalent to $\alpha$-tocopherol at a concentration of 0.5 mg/mL. Additionally, the antidiabetic activity of their extracts was also evaluated using a rat intestinal $\alpha$-glucosidase inhibition assay. Among all extracts investigated, the methanol extract had the highest $\alpha$-glucosidase inhibitory activity, although its activity was less than acarbose at $0.5\;{\mu}g/mL$. This result suggested that U. davidiana extracts may have antidiabetic activity. Total phenolic compounds and flavonoids were also measured. Phenolic compounds such as tannic acid, p-coumatric acid, and kaempferol were detected by high-performance liquid chromatography (HPLC). These results suggest that U. davidiana extracts may be useful as a potential source of antioxidant and antidiabetic materials.
Various parameters such as solvent selection, concentration, solid/liquid ratio, soaking time, temperature, stationary, shaking conditions, and repeated extractions were investigated in order to determine the optimum extraction conditions of ${\beta}$-glucosidase from bagasse fermented by mixed culture of Aspergillus niger NRC 7A and Aspergillus oryzae NRRL 447. Among various solvents tested, non ionic detergents gave the best results than the inorganic or organic salt solutions and distilled water. The optimum conditions for extraction of ${\beta}$-glucosidase were 30 min soaking time at $40^{\circ}C$ under shaking condition at 150 rpm, with solid/liquid ratio 1:15 (w/v), which yielded $2882.74{\pm}95.52U/g$ fermented culture (g fc) of enzyme activity. With repeated washes under the above optimum conditions, the results showed that enzyme extracted in the $1^{st}$ and $2^{nd}$ washes represents about 90% of the total activity.
The aqueous extract from the fruit body of Lentinus edodes was evaluated for inhibitory activities against $\alpha$-glucosidase isolated from Spargue-Dawley male rats. A aqueous extract of Lentinus edodes exhibited 13.8% inhibitory activity on using 2 mM p-nitrophenyl $\alpha$-D-glucopyranoside as a substrate ($IC_{50}$ 75.3 mg/ml). The aqueous extract of Lentinus edodes inhibition type on $\alpha$-glucosidase was determined to be competitive inhibition. When it was oral administered to increase of blood glucose levels after STZ-induced in a dose dependent dietary. These results suggest that aqueous extract of Lentinus edodes effect a metabolism of intestine, and thereby reducing the increase of blood glucose after STZ-induced.
The effects of water soluble materials in paper mill sludge on cellulase and $\beta$-glucosidase activities were studied while the optimization of enzyme system for hydrolysis of the paper mill sludge for production of glucose was made. Water soluble materials in the paper mill sludge showed stimulatory effect on carboxymethyl cellulose (CMC) activity, inhibitory effect on filter paper (FP) activity, and no effect on avicelase and $\beta$-glucosidase activities. CMC and ${\beta}$-glucosidase activities at 5 and 10, 5 or 10 and 10, and 10 and 10 U/ml were optimal for hydrolysis of 5, 10, and 20% of the paper mill sludge, respectively.
In this study ethanol extracts of aerial part of Geranium thunbergii Sieb. et Zucc. was investigated for their ability to inhibit a-glucosidase, and thus was fractionated using two organic solvents, including dichloromethane, ethyl acetate. The ethyl acetate-soluble fraction, which manifested potent enzyme inhibitory properties, was then followed by tracking down the active compound by combining HPLC micro-fractionation to an enzyme assay in 96-well plate. The ${\alpha}$-glucosidase inhibitory activity profile showed that two peaks exhibited potent inhibitory activity, and then the structural analyses of the two peaks were carried out by HPLC-UV-MS. The main ${\alpha}$-glucosidase inhibitory compounds in the ethyl acetate-soluble fractions of ethanol extracts of Geranium thunbergii Sieb. et Zucc. were tentatively identified as geraniin and kaempferol-7-rhamnoside.

Electron donating ability (EDA), reducing power, total phenolic contents, total flavonoid contents, antimicrobial activity, and ${\alpha}$-glucosidase inhibitory effects of twenty-two sorghum varieties were examined using ethanol extracts (70%, v/v). The electron donationg abilities (EDA) of Mesusu and Bulgeunjangmoksusu (94% at 10 ${\mu}g$/mL) indicated higher antioxidant activity compared to those of other varieties and standard antioxidants such as BHT (13%) and BHA (74%). The reducing power of Mesusu ($OD_{700}$=0.71) suggested higher antioxidant activity, which was dependent on sample concentration. Bitjarususu showed the highest content of total phenolics (22.9 mg GAE/g); however, extracts from Heuinsusu exhibited the lowest content of total phenolics (16.4 mg GAE/g). Jangmoksusu showed the highest total flavonolic contents (3.5 mg QE/g), and Sigyeongsusu and Chal (GS) susu displayed the most antibacterial activity (MIC=8 ${\mu}g$/mL) against Escherichia coli. Extracts of Bulgeunjangmoksusu, Moktaksusu, and Ginjangmoksusu showed the highest ${\alpha}$-glucosidase inhibitory effect (98%) at the concentration of 5 ${\mu}g$/mL.
This study was performed to investigate the biological activities of Hericium erinaceus fruiting body, including antioxidative, fibrinolytic, thrombin inhibitory and ${\alpha}$-glucosidase inhibitory activities. The water extract of H. erinaceus was fractionated into hexane, chloroform, ethyl acetate, butanol and water fractions, and each of these fractions was individually assayed. The water extract showed high antioxidative activity at 87.78%. The water fraction showed the highest extraction yield at 35.68% (w/w). In the fraction activity tests, butanol and water fractions showed strong antioxidative activities at 87.91% and 92.27%, respectively. Assays for fibrinolytic activity indicated that only the ethyl acetate fraction has significant efficacy at 2.96 plasmin units/mL. The 10-fold dilution of hexane fraction showed high thrombin inhibitory activity, and ${\alpha}$-glucosidase inhibitory activity at 77.67% and 90.53%, respectively. In conclusion, solvent fractions of H. erinaceus can be used as materials for the development of biofunctional foods to prevent cardiovascular diseases.

The purpose of this study was to measure total phenolic compounds as a measure of antioxidant activity as well as ${\alpha}$-amylase inhibitory and ${\alpha}$-glucosidase inhibitory activities as a measure of anti-diabetic efficacy in methanol extracts from 23 kinds of medicinal plants. Extracts of three medicinal plant species showing high total polyphenol contents were selected (Euonymus alatus stem, Taxus cuspidata fruit, and Eucommia ulmoides leaf). Extracts of six medicinal plant species showing over 60% DPPH radical scavenging activity were also selected [Eucommia ulmoides barks (80.10%), Lycium chinense roots (64.25%), Euonymus alatus stem (73.59%), Lespedeza cuneata (78.20%), Taxus cuspidata fruits (70.52%), and Tilia taquetii leaf and stem (67.81%)]. Regarding ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibitory activities acarbose showing approximately 80% inhibitory activity was selected as a control group, and six species (Eucommia ulmoides heartwood, Eucommia ulmoides bark, Euonymus alatus stem, Dioscorea batatas, Coix lachryma-jobi, and Phaseolus radiatus) showed greater than 80% ${\alpha}$-glucosidase inhibitory activity. Extracts of nine medicinal plant. species showing over 80% ${\alpha}$-amylase inhibitory activity (Pueraria thunbergiana root, Eucommia ulmoides bark, Eucommia ulmoides leaf, Lycium chinense fruits, Euonymus alatus leaf and stem, Euonymus alatus stem, Sasa borealis whole, Dioscorea batatas leaf and stem, and Tilia taquetii leaf and stem). Based on these results, medicinal plants showing high antioxidant and antidiabetic activities can be used as fundamental products in developing new medicines, as well as functional foods to prevent adult disease.

This study was carried out to investigate the total polyphenol, total flavonoid content, antioxidant activity, angiotensin I-converting enzyme (ACE) inhibitory activity, $\alpha$-glucosidase inhibitory activity, and antiproliferation activity of the citron seed. The citron seed were separated to hull and embryo, and extracted with n-hexane and 70% ethanol. Antioxidant activity of ethanol extract was higher than that of n-hexane extract. IC50 value for DPPH radical scavenging activity of ethanol extract of hull (CSE1) and embryo (CSE2) were 3.18 and 8.43 mg/mL, and those of total antioxidant activity were 19.96 and 11.28 mg AA eq/g, respectively. ACE inhibitory activity and $\alpha$-glucosidase inhibitory activity on CSE1 showed the highest values of 31.61 and 45.17%, respectively. Antiproliferation effects on the MCF7, HepG2, H460, HCT-116, and PC3 cell line showed the highest values of 14.09, 19.12, 12.29, 9.78, and 9.12% in extract concentration of 5 mg/mL, respectively. These results suggested that citron seed can be used for development of functional food material which have biological activities.

To establish the anti-diabetic(α-glucosidase inhibitory) activity of D. lotus leaf extract, isolate and identify the constituents responsible for the activity. The methanolic extract of leaves was partitioned between water, n-butanol and ethyl acetate. Bio-assay guided fractionation, based on inhibition of ;${\alpha}$-glucosidase, allowed isolation and identification of the active components. Liquid chromatography/mass spectrometry(LC/MS), 1 H-NMR and 13 C-NMR spectra analyses demonstrated that the active compound was myricetin-3-O-;${\alpha}$-L-rhamnoside(1). Compound 1 demonstrated a strong inhibition on the α-glucosidase, in vitro and ;${\alpha}$-glucosidase inhibitory value was calculated as 98.08%, when that of a reference drug, acarbose was estimated as 83.03%. The present study indicates compound 1 could be considered as an ;${\alpha}$-glucosidase inhibitor and developed as an important antidiabetes agent for type II diabetes therapy.
