Germinability of Resting Cysts Associated with Occurrence of Toxic Dinoflagellate Alexandrium Species (유독 와편모조류 Alexandrium속의 출현에 미치는 휴면포자의 발아율)

  • To study the causes of occurring toxic dinoflagellate Alexandriwn species, an experiment was undertaken in Jinhae Bay shellfish harvesting areas. The water and sediment samples were collected to record the abundance of Alexandriwn species, and to study the distribution and the germinability of those benthic cysts from September 1993 to July 1994. Alexandrium species were began to appear at all the sample stations after January, and reached maximum concentration (530 cells/l) at Taekok station (Chilcheon-do) in March 1994. Alexandrium cysts were also found at every station surveyed, of which several sites showed the higher concentration of 700-800 $cysts/cm^3$ at the upper sediment profile (0-4cm), but the concentrations were wide range of 100-800 $cysts/cm^3$. The results of each sampling season showed a great difference in the cyst germination experiments, were potentially high in cold season; $72.5\%$ (Jan.), $68\%$ (Apr.),$44\%$(Jul.), and $9\%$ (Oct.). These results suggested that germination of Alexandrium resting cysts in 15 m depth of coastal waters in Jinhae Bay would be controlled by a seasonal endogenous clock instead of the general environmental factors like temperature. Therefore, it is possible that Alexandrium species could be abundant by the germination of resting cysts in cold season, and contribute to the regional paralytic shellfish poisoning (PSP) toxification.

Conditions for Solid Matrix Priming of Carrot Seeds and Physiological Changes in the Seed During the Treatment (당근 종자의 Solid Matrix Priming 적정 조건구명과 처리중 종자의 생리적 변화)

  • To improve germinability of carrot seeds of 'Inari', 'Mansan', 'Mussang', and 'Hongsim' cultivars, various levels of solid matrix priming (SMP) and the physiological changes of seeds associated with SMP were examined. As the ratio of water in a seed-Micro Cel E-water mix and priming duration increased, seed germinability enhanced. However, there was radicle protrusion during SMP in some cultivars, indicating the existence of cultivar different variations of the optimum conditions. The optimum ratio for a seed-Micro Cel E-water mix was 5 : 3 : 10.5. In this mix, the optimum durations for the treatment were 3 days for 'Mansan' and 5 days for other cultivars. SMP at $20^{\circ}C$ to $25^{\circ}C$ resulted in a higher germination percentage and a shorter $T_{50}$. Seeds absorbed water quickly within an hour and the percentage of moisture was stable at 43 to 49 after 24 hours of the treatment. Conductivity of the mix declined for the first 6 hours to $100{\mu}S{\cdot}cm^{-1}$ with little changes thereafter. On the other hand, the pH of the mixed increased slightly during the early phase, and then returned to the initial level as the treatment proceeded.

Seed Dormancy of Jerusalem Artichoke (Helianthus tuberosus L.) and Seed Treatment for Germination Induction (돼지감자 종자의 휴면성과 발아유도를 위한 종자처리)

  • Jerusalem artichoke (Helianthus tuberosus L.) as a biomass potential crop has several distinct advantages such as vigorous growth on poor land and high yield of carbohydrate. In this crop, seed dormancy has hampered the efforts of seed-propagration and to use them in breeding programs for improving jerusalem artichoke. Several seed treatments were tested to determine their effectiveness in overcoming the seed dormancy found in five collected varietes of jerusalem artichoke. The first results showed that the seed fertilities of five collected varieties ranged from 2.4% to 14.7% and the number of seed produced by one plant ranged from 88 to 1058. Germinability of seeds stored for 3 months at room temperature after harvest was almost 0% and it was not improved by addition to the treatments of temperature, light and GA3, while germinability of seeds stored for 27 months at room temperature after harvest increased to 47.5% in germination rate. But the removal and pin-pricking of seedcoat were very effective in breaking the seed dormancy, giving germination of 96.8% and 82.3%, respectively. These results showed that the seed dormancy of jerusalem artichoke was induced by the seedcoat. Besides the treatment of seedcoat removal and seedcoat-pinpricking, the treatment of low and wet stratification was also effective in breaking the seed dormancy of jerusalem artichoke. Whole dormant seeds incubated for 70 days in low and wet condition germinated over 85%.

Effect of Priming and Seed Pellet Technique for Improved Germination and Growth in Fraxinus rhynchophylla and Alnus sibirica (프라이밍 및 종자펠렛 제조를 통한 물푸레나무와 물오리나무의 발아율 향상 및 생장증대 효과)

  • This study was carried out to select new pelleting binder and material for seeds from Fraxinus rhynchophylla Hance and Alnus sibirica Fisch. ex Turcz. The optimum treatments of the various concentrations and species of priming agents to improve seed germination of both woody medicinal plants were also estimated. Germinability was increased when the seeds of Fraxinus rhynchophylla Hance was soaked in -1.0 MPa of PEG6000 solution at $15^{\circ}C$ for 4 days significantly, the optimum treatment for improving germination of Alnus sibirica Fisch. ex Turcz was observed when the tested seeds was soaked in 100 mM of KCl at $15^{\circ}C$ for 4 days. The influence of physical and chemical properties of pelleting solid materials, the mixture of gypsum, diatomaceous earth, dalma ceramic and vermicuolite (6:1:1:1 ratio) were found as the best pelleting materials for Fraxinus rhynchophylla Hance and Alnus sibirica Fisch. ex Turcz. seeds. To satisfy the requirements of absorption and compatibility for multi-layer seed pelleting, SGPA (Starch-grafted cross-linked polyacrylates) hydrogel was prepared using starch, acrylonitrile, ceric ammonium nitrate, nitric acid, methyl alcohol and potassium hydroxide. The resulting SGPA hydrogel showed high water absorption but not plant compatibility. It suggested that seed pelleting using pelleting materials and SGPA hydrogel (multi-layer coating) after priming agent treatment is to increase germinability and seedling growth and it can reduced irrigation labours and can also save seed.

Response of Germination Rate and Ascorbate Peroxidase Activity to Cryopreservation of Perilla (Perilla frutescens) Seeds with Variable Initial Viabilities (들깨 종자 활력 수준별 초저온 동결보존 후 발아율 및 Ascorbate Peroxidase 활성 변화)

  • BACKGROUND: Seed of perilla (Perilla frutescens var. japonica Hara) is short-lived in conventional storage conditions. For long-term conservation of plant species, cryopreservation is the method currently available. This study was performed to find out reliable methods for a long-term storage of seeds of perilla as a genetic resource. METHODS AND RESULTS: Using seeds of 9 perilla cultivars, the effects of desiccation, aging, and cryopreservation on seed germinability and ascorbate peroxidase activity in the seeds were investigated. Initial germinability of the seeds was various, and dry seeds of all cultivars survived cryopreservation without loss of viability. The highest germination was achieved at 4-5% moisture content, and stimulatory effect of cryogenic temperature on the seed germination was observed in some cultivars. Accelerated aging of perilla seeds led to reduction in germination and ascorbate peroxidase activity, and the susceptibility of seeds to aging was different among the tested cultivars. No significant difference in germination was observed for the aged seeds of control and liquid nitrogen exposed. CONCLUSION: The results of this study suggest that cryopreservation at 4-5% moisture content would be a suitable method for long-term conservation of perilla seeds without detrimental effects on germination.

An Optimum Harvest Time for Chinese Milk Vetch (Astragalus sinicus L.) Seed Production (자운영 종자생산을 위한 적정 수확시기 구명)

  • To determine an optimum harvest time for chinese milk vetch (CMV) seed production, the seeds were harvested at 4 times, according to 25, 30, 35, and 40 day after flowering (DAF), in Miryang, southern part of Korea. CMV plants were manually harvested at each time and seed threshing was done by rice threshing machine. Seed yield, 1,000-seed weight, germinability, and hard coat ratio were investigated. Seed yield was the highest, 53.9 kg/300 kg by dry weight (DW) of CMV plant, at 35 DAF. 1,000-seed weight increased according to seed harvest time from 25 DAF to 40 DAF when it was 3.10 g. The germination ratios of seeds harvested at 4 times were not significantly different when the seeds stored until August 1. In case of long period of CMV seeds stored, the seeds harvested later showed higher germination rate. On the other hand, because the hard coat ratio causing germination inhibition was declined with an increase of storage period, it was higher in the seeds harvested later. There was no difference among the seeds harvested at 4 times at October 1. In conclusion, it was presumed that an optimum harvest time for CMV seed production should be at 35 DAF considering seed yield, weight and germinability.

Study on the Seed Germination Characteristics of Trapa japonica FEROV. (마름(Trapa japonica FEROV.) 종자의 발아특성에 관한 연구)

  • In this study, germination characteristics and effect of some chemicals on germination of Trapa japonica $F_{EROV}$ seeds were investigated to obtain a fundamental data for controlling them. When the seeds were stored at low temperature for at least 3 months, their dormancy was completely released with a germinability of above 90%. However, those stored under the room or high temperature condition were never germinated. Germination was best at constant temperature of around $25^{\circ}C$ or at alternate temperature of 25/$15^{\circ}C$ (12 hr-day/12 hr-night), and was nat dependent on light. They could germinate though planted at the depth of 15 cm, but their initial growth were: suppressed with increase of planting depth, especially above 10 cm depth. Germinability was lost when the fresh weight was decreased more than 30%, indicating that the seed is sensitive to dehydration. The freezing at $-20^{\circ}C$ for 1 hr didn't affect germination, but 5 hr treatment completely killed them. Their germination was inhibited by 100% at 100 ppm ABA and slightly inhibited above 25 ppm of butachlor, whereas pyrazosulfuron-ethylnt is likely to control T. japonica physically and herbicide treatment to inhibit germination may not be recommendatory. and bensulfuron-methyl didn't inhibit the germination at the concentration of 100 ppm. Taken together, dehydration treatme is likely to control T. japonica physically and herbicide treatment to inhibit germination may not be recommendatory.

Effect of Sucrose, Calcium and Boron Added in the Medium on Pollen Germination of Peach(Prunus persica SIEB) (Sucrose, 칼슘, 붕소 첨가가 복숭아 화분 발아에 미치는 영향)

  • This study was carried out to identify the effect of sucrose, calcium, and boron on the increase of pollen germination frequency in peach $(Prunus\;persica\;_{SIEB})$ under various temperature levels. The highest pollen germination was obtained at $20^{\circ}C$. Pollen germination was completed in 3.5 hrs. after planting of pollen on the basal medium which was composed of 1% agar and 10% sucrose, and conditioned pH 5.5. The optimum concentrations of sucrose and $H_3BO_3$ at $25^{\circ}C$ were 10% and $100\;mg\;{\cdot}\;L^{-1}$, respectively. However, $CaCl_2$ inhibited pollen germination in peach. Eight peach cultivars showed significantly different germinabilities at $10^{\circ}C$ on the basal medium. 'Nagasawa-Hakuho' showed the highest germinability of 26.1%, and 'Baekmijosaeng' showed the lowest germinability of 0.1%. $H_3BO_3$ strongly promoted pollen germination frequency when added to the germination medium at $10^{\circ}C$. Optimum concentration was $100\;mg\;{\cdot}\;L^{-1}$.

Seed Quality, Germinability and Initial Growth of Pterocarpus erinaceus (African Rosewood). How Important are Mother Tree Size, Source and Timing of Fruit Harvest?

  • Pterocarpus erinaceus is a multipurpose tree species indigenous to semi-arid and Guinean-savanna woodlands of Africa. Natural regeneration on the savanna is being hampered by higher fire frequencies and other land use changes. Simultaneously, demand for timber from the species on the international market is on the rise, raising conservation concerns. To ensure sustainability, good quality seeds, sources of which have not received much research focus, are needed for afforestation. This study investigated how seed quality, germinability and initial seedling growth of P. erinaceus might be influenced by land-use type, mother tree size as well as source and timing of fruit harvest, using both correlational and experimental approaches. The results showed that up to 94.6% of all harvested fruits contained seeds, with no differences found between fruits harvested from cultivated and non-cultivated lands. Percentage of (sound) unblemished seeds was found to be higher for fruits harvested early March (47.3%) than fruits from late April (39.5%). Percentage sound seeds was 41.4% for dispersed fruits (i.e. detached fruits picked from under mother trees) which was not found to differ from undispersed fruits (i.e. fruits harvested while still attached to mother trees) at 45.5%. Also, the influence of fruit harvest time was not found to be different for dispersed and undispersed fruits. Correlations between seed set (proportion of fruits containing seeds) and mother tree size (both tree height and DBH) were found to be very low and non-significant for both dispersed and undispersed fruits. Across mother trees, mean emergence percent was 79.7%, and mean seedling height at three weeks following emergence was 5.32 cm. Both emergence percentage and seedling height were not found to differ among mother trees, but seeds from dispersed fruits had a higher emergence percent (85%) than seeds from undispersed fruits (74%). Implications of findings are discussed.

Establishment of Days after Anthesis(DAA) and Fruit After-ripening Period(FAP) for High-Quality Seed Production of Watermelon (우량 수박종자 생산을 위한 적정 성숙일수 및 후숙일수 확립)

  • This study was conducted to investigate days after anthesis (DAA) and fruit after-ripening period (FAP) for seed-harvesting of high quality watermelon seeds. Fruit weight and number of seed per fruit increased according to DAA, while those did not significant about FAP. Ratio of cotyledon at whole seed was higher about 2 to 4% compared to seed coat irrespective of DAA and FAP. Germinability of watermelon was not a significant effect by DAA, however, it had differences by FAP. Percent of germination (PB) was below 50%, when 30 days maturated fruits after anthesis was omitted ripening, while PB was increased to 92% by ripening. In addition, seeds at DAA 40 and FAP 20 were higher general seedling vigors (hypocotyl length, diameter etc.) in BP test. Results indicated that considering seed productivity, it had maximized seed viability at DAA 40 and FAP 20.
