The objective of the present study was to assess the antipsychotic and antidiarrhoeal activities of ethanolic extract of roots of Rubia cordifolia in mice and rats. The antipsychotic activity of ethanolic extract of roots of Rubia cordifolia (ERC) was evaluated by observing its effect on amphetamineinduced stereotyped behavior in mice. Effect of ERC was also studied on motor coordination and locomotion in mice. The antidiarrhoeal activity was evaluated using castor oil-induced diarrhoea and excretion of sodium and potassium ions in the intestinal secretion in rats and gastrointestinal transit in mice. The ERC inhibited amphetamine-induced stereotyped behaviour, diminished locomotion and impaired motor coordination. ERC inhibited castor oil-induced diarrhoea, decreased both sodium and potassium excretion in the intestine and decreased gastrointestinal transit. Thus the present study confirms the anti-diarrhoeal activity of Rubia cordifolia. Further studies are necessary to evaluate the potential of Rubia cordifolia as an antipsychotic.

Objectives: The purpose of this study was to investigate the effects of Shengmai-san and Pyungwi-san, a herbal product used in traditional Chinese medicine, on gastrointestinal (GI) motility in mice. Methods: The in vivo effects of Shengmai-san and Pyungwi-san on GI motility were investigated by measuring the intestinal transit rates (ITRs) using Evans blue in normal mice and in mice with experimentally induced GI motility dysfunction (GMD). GMD was induced by injecting acetic acid or streptozotocin intraperitoneally. Results: In normal Institute of Cancer Research mice, ITRs were significantly and dose-dependently increased by Shengmaisan (0.01~1 g/kg) and Pyungwi-san (0.01~1 g/kg). The ITRs of acetic acid induced peritoneal irritation model and streptozotocin-induced diabetic model mice were significantly reduced compared to normal mice, and these reductions were significantly and dose-dependently inhibited by Shengmai-san (0.01~1 g/kg) and Pyungwi-san (0.01~1 g/kg). Conclusions: These results suggest that both Shengmai-san and Pyungwi-san are a good candidate for the development of a prokinetic agent that may prevent or alleviate GMD.
The effect of propionylpromazine, acepromazine maleate, ketamine HCI, xylazine HCI, and pentobarbital sodium as chemical restraint drugs on the transit time of barium sulfate through the stomach and duodenum in 24 healthy cats was investigated. In the present study, propionylpromazine, acepromazine maleate, and ketamine HCI did not reveal significant effect on the gastroduodenal transit time, but xylazine HCI and pentobarbital sodium pro-longed the gastroduodenal transit time markedly compared with control group. Therefore it is concluded that propionylpromazine, acepromazine maleate, and ketamine HCI could be selected for upper gastrointestinal radiographs. but xylazine HCI and pentobarbital sodium should be avoided.

BACKDROUND/OBJECTIVE: Constipation is a condition that can result from intestinal deformation. Because humans have an upright posture, the effects of gravity can cause this shape deformation. Oligosaccharides are common prebiotics and their effects on bowel health are well known. However, studies of the physiological functionality of a product that contains both lactulose and galactooligosaccharides are insufficient. We investigated the constipation reduction effect of a dual-type oligosaccharide, Dual-Oligo, in loperamide-treated rats. MATERIALS/METHODS: Dual-Oligo consists of galactooligosaccharides (15.80%) and lactulose (51.67%). Animals were randomly divided into four groups, the normal group (normal), control group (control), low concentration of Dual-Oligo (LDO) group, and high concentration of Dual-Oligo (HDO) group. After 7 days of oral administration, fecal pellet amount, fecal weight, watercontent of fecal were measured. Blood chemistry, short-chain fatty acid (SCFA), gastrointestinal transit ratio and length and intestinal mucosa were analyzed. RESULTS: Dual-Oligo increased the fecal weight, and water content of feces in rats with loperamide-induced constipation. Gastrointestinal transit ratio and length and area of intestinal mucosa significantly increased after treatment with Dual-Oligoin loperamide-induced rats. A high concentration of Dual-Oligo tended to produce more acetic acid than that observed for the control group, and Dual-Oligo affected the production of total SCFA. Bifidobacteria concentration of cecal contents in the high-concentration oligosaccharide (HDO) and low-concentration oligosaccharide (LDO) groups was similar to the result of the normal group. CONCLUSIONS: These results showed that Dual-Oligo is a functional material that is derived from a natural food product and is effective in ameliorating constipation.
Our recent study has revealed that in vivo intestinal transit rate (ITR) in normal mice was significantly increased by the administration of an aqueous extract of dried Gentiana scabra rhizomes and roots (GS-W) in a dose-dependent manner. Following on from our previous study, the effect of GS-W on ITR was measured in mice with experimentally induced gastrointestinal motility dysfunctions (GMDs) in the present study. GS-W showed no significant acute toxicity even at an oral dose of 5 g/kg to mice. ITR was significantly retarded in the GMD mice compared with that in normal mice, and this retardation was significantly recovered by the oral administration of GS-W in a dose-dependent manner. Furthermore, the ITR value of GS-W at a dose of 1 g/kg appeared to be higher than that of cisapride, which was predominantly prescribed for human patients with various GMDs in the late 1900s but was withdrawn from the market in 2000 due to its fatal side effects. The current results suggest that GS-W is a potential substitute for cisapride to prevent or alleviate human GMDs.
For comparison with conventional barium-gastrointestinal study, modified method using barium and methylcellulose was performed in 16 normal dogs (4-8 kg) divided into two groups. The group 1 received 8ml/kg of 40% w/v barium suspension only, and group 2 had taken 15 ml/kg of 0.6% w/v methylcellulose after administration 4 ml/kg of 40% w/v barium suspension by feeding tube. Between two techniques, the modified method with 4ml of 40% w/v barium suspension and 15 ml of 0.6% w/v methylcellulose showed much better image quality ["excellent" result in 7 of the 8 subjects (88%)] and shorter transit time (107 minutes) toe the cecum. In addition, the best image quality was obtained at 20 and 40 minutes after admin-istration of contrast agent. Ardipusilloside-I is a natural triterpenoid saponin, which was isolated from Ardisia pusilla A. DC. The aim of the study was to evaluate the stimulation of ardipusilloside-I on gastrointestinal motility in vitro and in vivo. The experiment of smooth muscle contraction directly monitored the contractions of the isolated jejunal segment (IJS) in different contractile states, and the effects of ardipusilloside-I on myosin were measured in the presence of $Ca^{2+}$-calmodulin using the activities of 20 kDa myosin light chain ($MLC_{20}$) phosphorylation and myosin $Mg^{2+}$-ATPase. The effects of ardipusilloside-I on gastro emptying and intestinal transit in constipation-predominant rats were observed, and the MLCK expression in jejuna of constipated rats was determined by western blot. The results showed that, ardipusilloside-I increased the contractility of IJS in a dose-dependent manner and reversed the low contractile state (LCS) of IJS induced by low $Ca^{2+}$, adrenaline, and atropine respectively. There were synergistic effects on contractivity of IJS between ardipusilloside-I and ACh, high $Ca^{2+}$, and histamine, respectively. Ardipusilloside-I could stimulate the phosphorylation of $MLC_{20}$ and $Mg^{2+}$-ATPase activities of $Ca^{2+}$- dependent phosphorylated myosin. Ardipusilloside-I also stimulated the gastric emptying and intestinal transit in normal and constipated rats in vivo, respectively, and increased the MLCK expression in the jejuna of constipation-predominant rats. Briefly, the findings demonstrated that ardipusilloside-I could effectively excite gastrointestinal motility in vitro and in vivo.
The purpose of this study was to elucidate effects of fructooligosaccharide on gastrointestinal tract and blood lipids of rats when this was supplied as purchased condition or oligosaccharide containing sponge cake. Male Sprague-Dawley rats were assigned to one of 3 treatments 1) control diet 2) 7.5% fructooligosaccharide containing diet (FOS diet) 3) lyophilized sponge cake powder containing diet (FOS-C diet). The sponge cake was made with fractooligosaccharide which replaced 40% of its surose, and the final concentration of fructooligosaccharide in FOS-C diet was 7.5%. Cecal and fecal water contents, amount of cecal content, and cecal wall weight were higher from fructooligosaccharide consumption, whereas total gut transit time was longer in rats consuming fructooligosaccharide compared with those fed control diet. Cecal and fecal pH were lower in FOS and FOS-C groups than in control group. Total cecal SCFA pools were higher from ingesting fructooligosaccharide containing diets compared with control diet. Serum triglyceride levels were lower in rats fed FOS and FOS-C diet than those fed control diet, while serum cholesterol levels were unaffected by treatment. Therefore the effects of fructooligosaccharide in sponge cake on serum lipids and gastrointestinal tract were similar to those of intact fructooligosaccharide. Also, adding 7.5% of FOS accompanied diarrhea symptom which suggests some precaution are needed when using FOS.
Probiotics are live microorganisms that, when administered in adequate amounts, confer health benefits to the host. This study was conducted for the isolation of potential lactic acid bacteria (LAB) with probiotic properties from goat milk and yogurt. Several tests were conducted in vitro using the standard procedures for evaluating the inhibitory spectra of LAB against pathogenic bacteria; tolerance to NaCl, bile salt, and phenol; hemolytic, milk coagulation, and bile salt hydrolase activities; gastrointestinal transit tolerance; adhesion properties; and antibiotic susceptibility. Among 40 LAB strains screened according to culture characteristics, five isolates exhibited antagonistic properties. Three were identified as Pediococcus acidilactici, and two were identified as Enterococcus faecium, exploiting 16S rRNA gene sequencing. All the isolates succeeded in the gastrointestinal transit tolerance assay and successively colonized mucosal epithelial cells. Based on the results of these in vitro assays, both P. acidilactici and E. faecium can be considered as potential probiotic candidates.

BIOVITA 3 bacterial species (BIOVITA 3), a probiotic blend powder containing Clostridium butyricum IDCC 1301, Weizmannia coagulans IDCC 1201, and Bacillus subtilis IDCC 1101, has been used as a food ingredient for gut health. However, its efficacy in improving constipation has not been reported. Therefore, we aimed to investigate the functional effects of oral administration of BIOVITA 3 as well as its component strains alone (at 1.0×109 CFU/day) in Sprague-Dawley (SD) rats with loperamide-induced constipation. The study included fecal analysis, gastrointestinal transit ratio, histopathological analysis, short chain fatty acids (SCFAs), and metagenome analysis. As results, the BIOVITA 3 group showed significant improvements in fecal number, water content, gastrointestinal transit ratio, and thickening of the mucosal layer. In the SCFAs analysis, all probiotic-treated groups showed an increase in total SCFAs compared to the loperamide-constipated group. Changes in microbial abundance and the diversity index of three groups (normal, constipated, and BIOVITA 3) were also defined. Of these, the BIOVITA 3 showed a significant improvement in loperamide-constipated SD-rats. This study suggests the possibility that BIOVITA 3 can be applied as an ingredient in functional foods to relieve constipation.
