Glutamate Decarboxylase (GAD)를 이용한 Gamma-Amino Butyric Acid (GABA) 생산 및 최적화 (Effects and Optimization of Gamma-Amino Butyric Acid (GABA) Production Process using Glutamate Decarboxylase (GAD))

  • pH controlled batch reactor and bubble column reactors have been developed in this research. They were used to produce high concentration of GABA and to determine optimal pH for GABA production. Glutamate decarboxylase (GAD) was isolated from recombinant E. coli and used for GABA production from monosodium glutamate (MSG). pH control was inevitable because the pH increased with MSG consumption. GAD showed highest activity at acidic conditions at pH 5.5 but the optimal pH for GABA production was pH 6.0. When 1.5 mole of MSG was used as reactant, the 1.05 mole of GABA was produced after 10 hrs batch reaction. Using bubble column reactors, 80 % of MSG was converted to GABA for 6 hrs reaction and 1.2 mole of GABA was produced.

  • Novel poly(aspartic acid) derivatives conjugated with $\gamma$-amino butyric acid, GABA, moieties, and their amphiphilic analogs were synthesized and characterized. The chemical structures of these polymers were confirmed by FT-IR and $^1H$ NMR spectroscopy. Their physicochemical properties in aqueous media were characterized by electrophonetic light scattering spectrophotometry (ELS), acid-base titration, and UV-spectroscopy. In addition, the in vitro cell activity of the GABA-conjugated polymer was examined. These results indicated that GABA-conjugated poly(aspartic acid) derivatives showed cell-growth activity and nanoparticle formation of a suitable size within aqueous media. These polymers have potential application in the cosmetic and pharmaceutical fields.

Characteristics of Red Pepper Paste by Using Germinated Barley with Increased γ-Amino Butyric Acid

  • Germinated barley, instead of glutinous rice, was used to make health-enhancing fermented red pepper paste. The proximate components of commercial glutinous rice red pepper paste (CGRPP) and germinated barley red pepper paste (GBRPP) were analyzed during fermentation. The sensory characteristics and ${\gamma}$-amino butyric acid (GABA) contents of CGRPP and GBRPP were evaluated. The contents of ${\beta}$-glucan and GABA showed the highest value after 48 hrs of germination. During the fermentation, the contents of GABA in GBRPP increased up to 28 days and then decreased. During sensory evaluation, the consumer liked the GBRPP more than CGRPP. The GABA contents were increased during fermentation and GABA contents of GBRPP were twice as much as that of CGRPP. These results suggest that the GBRPP can have consumer acceptance for its health benefits and taste and can therefore become commercialized.

Production of GABA (gamma amino butyric acid) by Lactic Acid Bacteria

  • Gamma-amino butyric acid (GABA) is a kind of pharmacological and biological component and its application is wide and useful in Korea specially, becoming aging society in the near feature. GABA is request special dose for the purposed biological effect but the production of concentrated GABA is very difficult due to low concentration of glutamic acid existed in the fermentation broth. To increase GABA concentrate using fermentation technology, high content of glutamic acid is required. For this reason, various strains which have the glutamic acid decarboxylase (GAD) and can convert glutamic acid to GABA, were isolated from various fermented foods. Most of GABA producing strains are lactic acid bacteria isolated from kimchi, especially added monosodium glutamate (MSG) as a taste enhancer. Optimizing the formulation of culture media and the culture condition, GABA conversion yield and amounts were increased. Finally GABA concentration of fermentation broth in batch or fed batch fermentation reached 660 mM or 1000 mM, respectively. Furthermore formulation of culture media for GABA production developed commercially. Many studies about GABA-rich product have been continued, so GABA-rich kimchi, cheese, yogurt, black raspberry juice and tomato juices has been also developed. In Korea many biological effects of GABA are evaluated recently and GABA will be expected to be used in multipurpose.

  • 혈압저하작용, 이뇨기능 등의 다양한 생리활성을 나타내는 $\gamma$-amino butyric acid(GABA)를 고농도로 생산하는 유산균인 Lactobacillus sakei B2-16을 이용하여 GABA의 산업적 생산 배지를 연구하였다. L. sakei B2-16의 최적 상용 배지는 Lactobacilli MRS 배지였으며, Lactobacilli MRS 배지에 1% mono sodium glutamate(MSG)를 첨가하고, L. sakei B2-16을 배양했을때 MSG의 99.3% 는 GABA로 전환되었다. MRS 배지를 기본으로 최적 배지조성을 검토한 결과, 탄소원으로 4% sucrose와 질소원으로 1% yeast extract를 첨가하였을 때 균체 증식과 GABA 생산량이 가장 우수하였다. 산업적 배지를 확립하기 위하여 미배아를 온수 추출하여 얻은 추출액 배지에 L. sakei B2-16을 배양한 결과, 7%의 MSG를 100% GABA로 전환시켰으며, 미배아 추출액을 이용한 배지는 산업적 생산용 배지로의 응용이 기대된다.

The crystal and molecular structure of $\gamma$-hydroxy-.$\beta$-amino butyric acid

  • The crystal structure or $\gamma$-hydroxy-$\betha$-aminobutyric acid was determined by MULTAN system with X-ray intensity data on a diffractometer and refined by the least-squares method to an R-value 0.034 for 711 reflections. The crystals were orthorhombic, space group $P2_{1}2_{1}2_{1}$, Z = 4, with a = 10.220, b = 8.257 and c = 6.556$\AA$. The molecule takes the zwitterionic form and skeletal conformation is trans-transform. The molecules are held together by intra-and intermolecular NH-O and OH--O hydrogen bonds.

김치로부터 GABA를 생산하는 커드 형성 젖산균의 분리 및 동정 (Isolation and identification of soycurd forming lactic acid bacteria which produce GABA from kimchi)

  • 비단백질 구성 아미노산인 gamma amino butyric acid(GABA)는 뇌에서 억제성 신경전달물질로 작용하며 혈압 저하, 신경 안정, 이뇨 작용 등에 효과가 있는 것으로 알려져 있다. 김치로부터 두유에서 커드를 형성하는 40개 균주를 분리하였다. GABA 생산력이 높은 균주를 선별하기 위하여 분리된 균주들을 두유에 접종하여 $37^{\circ}C$에서 48시간동안 발효하였다. 분리된 균주들 가운데 발효두부에서 가장 높은 GABA 를 생산하는 383 균주는 API 분석과 16S rDNA sequence를 통하여L. sakei로 동정되었으며 L. sakei 383으로 명명하였다. L. sakei 383은 MRS 배지에서 $35^{\circ}C$, 24시간 배양할 때 가장 높은 생육을 나타내었으며 두유에 균을 접종하여 $35^{\circ}C$에서 18시간 발효 했을 때를 GABA 생산을 위한 최적 배양 조건으로 정하였으며 최대 GABA 함량은 8.65 mg/100 g이었다.

  • 본 연구는 막걸리로부터 ${\gamma}$-amino butyric acid (GABA) 생성 유산균을 분리 및 동정하고 최적 GABA 생산조건을 확립하는데 그 목적이 있다. 막걸리로부터 64균주의 유산균은 MRS 배지에서 성장된 집락의 색과 모양의 특성에 따라 분리하였다. 분리균주의 GABA 생산은 1% MSG가 첨가된 MRS 액체배지에서 배양하여 TLC와 HPLC 방법에 의해 평가되었다. B-134 균주는 GABA생성을 위한 우수균주로 선발하였다. 16S rRNA 유전자 및 glutamate decarboxylase B (gadB) 유전자의 염기서열분석을 통하여, B-134 균주는 Lactobacillus plantarum subsp. plantarum B-134 균주로 명명하였다. GABA 생성을 위한 온도, pH, NaCl 및 MSG 농도를 달리하여 최적배양조건을 조사하였다. 그 결과 B-134 균주의 최적배양 조건은 온도 $37^{\circ}C$, pH 5.7, NaCl 농도 0% (w/v), 그리고 MSG 농도 3% (w/v)로 결정되었으며 본 조건에서 48시간 배양시 25 mM의 GABA를 생산하였다. 이러한 결과로부터 B-134 균주는 GABA함유 건강기능식품개발을 위한 유용한 균주로 판단된다.

굴, Crassostrea gigas 부착기 유생의 부착에 미치는 신경전달물질종의 영향 (The Effect of Neuroactive Compounds on Settlement of Pacific Oyster, Crassostrea gigas Pediveliger Larvae)

  • 굴 부착기 유생의 부착 유인 효과를 조사하기 위하여 6종류(serotonin, ${\gamma}$-amino butyric acid, L-3,4-dihydroxyphenylalanine, norepinephrine, epinephrine, methyl bromide)의 신경전달물질을 채묘기질에 직접 처리하여 굴 부착기 유생의 부착 유인 효과를 조사하였다. 조사결과 평균 채묘율는 각각 $29.1{\pm}2.2%$, $29.1{\pm}2.2%$, $19.2{\pm}2.1%$, $11.0{\pm}1.2%$, $15.2{\pm}0.9%$$42.7{\pm}2.7%$로 MB 처리구에서 가장 높은 결과를 보였고, norepinephrine 처리구에서 유의적으로 가장 낮은 결과를 보였다 (P < 0.05). 채묘기질 $cm^2$당 부착밀도와 부착빈도는 ${\gamma}$-amino butyric acid와 methyl bromide 처리구에서 각각 $1.97{\pm}1.42$$2.37{\pm}1.86$ 마리/$cm^2$로 가장 높은 부착밀도를 보였다 (P < 0.05). 본 실험결과 굴 인공종묘배양장 등에서 부착기 유생의 부착요율을 높이는데, ${\gamma}$-amino butyric acid와 methyl bromide를 이용하면 효과적일 것으로 보인다.
