Antioxidant Properties of Lentinula edodes after Sawdust Bag Cultivation with Different Oak Substrates (참나무 수종별 톱밥재배에 따른 표고의 항산화 특성)

  • We evaluated the antioxidant properties of Lentinula edodes upon sawdust bag cultivation with 5 oak substrates: Quercus acutissima, Q. mongolica, Q. serrata, Q. aliena, and Q. variabilis. We found that the optimal extraction conditions were 70% (v/v) methanol shaken at 150 rpm at $25^{\circ}C$ and 150 rpm for 24 h. The methanolic extracts from L. edodes contained high phenolic and flavonoid contents, they also exhibited stronger antioxidant activities. The total phenolic contents and total flavonoid contents of the mushroom extracts ranged from 2.37 to 3.12 milligrams of gallic acid equivalents per gram of dried mushroom (mg GAE/g) and 0.48 to 0.48 milligrams of quercetin equivalents per gram of dried mushroom (mg QE/g), respectively. In addition, the mushroom extracts exhibited 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity (15.8% to 89.4%) at 2 to 10 mg/mL, ferric reducing antioxidant power (0.153 to 0.425) at 5 to 20 mg/mL, and reducing power (0.078 to 0.359) at 5 to 20 mg/mL, respectively. Q. aliena more effectively increased total phenolic contents, total flavonoid contents, and antioxidant activities than the other oak substrates.

Bioactive Compounds and Antioxidant Activity of Jeju Camellia Mistletoe (Korthalsella japonica Engl.) (제주 동백나무 겨우살이의 용매별 기능성 성분 및 항산화 작용)

  • Mistletoes are hemi-parasitic plant growing on different host tree and shrubs. They are traditionally used in folkloric medicine for the treatment of diarrhea, cough, diabetes, hypertension, cancer and skin infection. The purpose of this study was to determine the contents of phenolics and antioxidant activity of 70% ethanol, 100% methanol and hot water extracts of Jeju camellia mistletoe (Korthalsella japonica Engl.). Ethanol was most effective in extracting total phenols (7,427 mg gallic acid equivalent (GAE)/100 g) and flavonoid (1,777 mg rutin equivalent (RE)/100 g). The free radical scavenging activity, 1,1-diphenyl-2-picryl hydrazyl (DPPH) (EC50 = 7.8 mg/ml) and hydrogen peroxide (H2O2) (EC50 = 1.4 mg/ml), and the capacity for chelating metal ions (EC50 = 8.0 mg/ml) and reducing power (EC50 = 14.9 mg/ml) of the samples also higher in ethanolic extracts. The strong correlation (r2 = −0.996~−0.881) between antioxidant capacities and the phenolic contents implied that phenolic compounds are a major contributor to the antioxidant activity of the ethanolic extracts of Jeju camellia mistletoe. As conclusions, Jeju camellia mistletoe contains bioactive substances with a potential for reducing the physiological as well as oxidative stress and this could explain the suggested cancer preventive effect of these plants as well as their protective role on other major diseases.

Effect of Inonotus obliquus Extract on the Expression MMPs and HAS-2 (차가버섯 추출물의 MMPs 발현 저해 및 HAS-2 발현 증가효과)

  • In order to investigate the possibility of Inonotus obliquus extract as an active ingredient for wrinkle-care cosmetics, we prepared 70 % ethanolic extract of Inonotus obliquus and measured its DPPH radical scavenging activity and elastase inhibitory activity. We also evaluated the effect of Inonotus obliquus extract on expression of MMPs and HAS-2 in fibroblast and HaCaT cell, respectively. Inonotus Obliquus extract showed DPPH radical scavenging activity and elastase inhibitory activity in a dose-dependant manner ($SC_{50}$ = 91 ${\mu}g$/mL, $IC_{50}$=124 ${\mu}g$/mL). Inonotus obliquus extract inhibited the expression of MMP-1 mRNA 50 ~ 79 % at concentrations of 5 ~ 25 ${\mu}g$/mL, and reduced the expression of MMP-2 around 20 % at a concentration of 10 ${\mu}g$/mL. And it also reduced the expression of MMP-9 54 ~ 70 % in tconcentrations of 5 to 25 ${\mu}g$/mL. Furthermore, Inonotus obliquus extract increased HAS-2 mRNA expression in a dose-dependent manner in concentrations of 5 to 25 ${\mu}g$/mL without cytotoxicity. These results suggested that Inonotus Obliquus extract could be useful as an active ingredient for wrinkle-care cosmetics.

Antioxidant and antidiabetic Activity of Jehotang according to Different Cooking Methods (제조방법에 따른 제호탕의 항산화 및 항당뇨 활성)

  • Jehotang is a cold traditional Korean drink made with honey and several ingredients used in traditional Korean medicine. The ingredients include Fructus mume, Fructus amomi, Fructus tsaoko, Santalum album and honey. In this study, Jehotang and its ingredients were determined through the analysis of antioxidant activity, total phenolic content, ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibitory activity. In addition, quality characteristics of Jehotang made by a traditional recipe(Yeonmil) and a modern recipe(double boiling and boiling) were also compared in terms of pH, color and sugar content. Total phenolic content of extract from Fructus ammomi was found to be 120.45 mg, and Yeonmil recipe was discovered 152.66 mg equivalent of gallic acid per g of extract. DPPH free radical scavenging activity were Feuctus amomi(93.13%) and Yeonmil recipe(56.44%). The Fructus amomi extract showed the highest ${\alpha}$-glucosidase inhibitory activity(89.51%) at the concentration of $100{\mu}g/mL$. ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibitory activity of boiled Jehotang were 52.38% and 72.52%, respectively. These results suggest that extract of Fructus amomi has an antioxidant activity and antidiabetic effects. Yeonmil recipe is useful for antioxidant effects more than the others. Also, the double boiling recipe has an excellent antidiabetic effect.

Antioxidant Properties of Water Extracts from Lentinula edodes Cultivars Grown on Oak Log (원목재배 표고버섯 물추출물의 항산화 특성)

  • We investigated the antioxidant activities of nine Lentinula edodes varieties cultivated on oak log. The total polyphenol content (TPC), total flavonoid content (TFC), and antioxidant properties of L. edodes (varieties: Dasanhyang, Chunbaegko, Poongnyunko, Soohyangko, Baekwhahyang, Chunjang 1ho, Chunjang 2ho, Sanlim 5ho, and Sanlim 7ho) were evaluated using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging assay and the ferric reducing antioxidant power (FRAP) assay. The TPC and TFC of mushroom extracts ranged from 5.10 to 10.88 mg gallic acid equivalent (GAE)/g and 0.76 to 5.78 mg quercetin equivalent (QE)/g, respectively. In addition, mushroom extracts exhibited DPPH radical scavenging activity of 24.22~90.41%, 2.0 mg/mL and FRAP of 0.056~0.231, 2.0 mg/mL. The Chunbaegko variety showed the highest TPC (10.88 mg GAE/g), TFC (5.78 mg QE/g), DPPH activity (38.19~90.11%, 0.2~2.0 mg/mL), and FRAP (0.075~0.231, 0.2~2.0 mg/mL).

Antioxidant and antimicrobial activities of muscadine grape extracts (머스커다인 포도 추출물의 항산화 및 항균 활성)

  • The objective of this research was to investigate the antioxidant and antimicrobial activities of muscadine grape extracts. Three different cultivars of muscadine grapes including Higgings, Jumbo, and Noble were selected. The skin/pulp and seed parts of three selected muscadine grape cultivars were used for extraction. The total phenolic contents of muscadine grape extracts were expressed as gallic acid equivalents (GAE). The antioxidant activity of muscadine grape extracts were determined by scavenging activity of diphenylpicrylhydrazyl (DPPH) radical and expressed as effective concentration ($EC_{50}$), which represented the concentration of the extract exhibiting 50% DPPH radical scavenging. The antimicrobial activity against E. coli K12 was determined and expressed as the minimum inhibition concentration (MIC). The seed extracts exhibited greater total phenolic contents than the skin/pulp extracts, ranging from 231.24 to 294.81 mg/mL GAE. The seed extracts exhibited greater antioxidant activities than the skin/pulp extracts ($EC_{50}$ of Higgins seed extract=0.026 mg/mL). However, the skin/pulp extracts exhibited greater antimicrobial activities than the seed extracts, exhibiting the minimum inhibition concentration (MIC) in Higgins skin/pulp extract (MIC=4.0 mg/mL). This research indicated that the seed part and skin/pulp parts of the muscadine grapes possessed antioxidant activity and antimicrobial activity, respectively. Therefore, it was concluded that muscadine grapes possess the potential to be utilized as functional foods or nutraceuticals.

Comparison of the Flavonoid and Urushiol Content in Different Parts of Rhus verniciflua Stokes Grown in Wonju and Okcheon (원주산과 옥천산 참옻나무의 부위별 flavonoid 및 urushiol 함량 비교)

  • Fustin comprised >98 and 73.0-86.7% of the total flavonoid content in the bark, and the stems and lignum of Rhus verniciflua, respectively. The butein, fisetin, and sulfuretin content varied between 0.31-2.17, 0.27-3.32, and 0.15-0.80 mg/g on a dry weight basis, respectively, in different parts of Rhus verniciflua. The urushiol content was 5.09-6.29, 55.05-56.30, and 0.38-0.39 mg/100 g on a dry weight basis in the stems, bark, and lignum, respectively. This showed that the bark of the tree had the highest urushiol content. C15:3 (pentadecatrienyl catechol), C15:1 (pentadecenyl catechol), and C15:2 (pentadecadienyl catechol) comprised 63, 33-35, and 2-3% of urushiol congeners in the tree bark, respectively.

Comparison of Antioxidant Activities of Pileus and Stipe from White Beech Mushrooms (Hypsizygus marmoreus) (부위별 흰색 느티만가닥버섯 추출물의 항산화 활성)

  • Hypsizygus marmoreus (white cultivar), also called white beech mushrooms, are edible mushrooms commercially cultivated in Korea and Japan. This study was carried out to evaluate the antioxidant properties of H. marmoreus. H. marmoreus fruit bodies were divided into pileus and stipe. The pileus and stipe were extracted into water and 80% ethanol and their antioxidant activities were analyzed. The total polyphenol content was highest in the water extract (pileus 1137.39±0.38 mg of GAE (gallic acid equivalents)/100 g, stipe 700.86±0.06 mg of GAE/100 g) compared to the ethanol extract (pileus 923.47±0.18 mg of GAE/ 100 g, stipe 324.05±0.03 mg of GAE/100 g). Ethanol extracts from pileus showed better scavenging ability on DPPH (47.32±0.23% at 10 mg/ml) and ABTS (57.33±0.10% at 10 mg/ml) than the stipe and water extract groups. Water extract from pileus were more effective in reducing power and ORAC (oxygen radical absorbance capacity) value than stipe and ethanol extract. The toxicity of water and ethanol extracts was investigated using WST-1 (Water Soluble Tetrazolium salt) assay on the mouse macrophage cell line RAW 264.7. Overall, total polyphenol content and antioxidant activities of extracts from H. marmoreus increased in a dose dependent manner while pileus was showed better total polyphenol content and antioxidant activities than stipe.

Antioxidant Activity and Alpha-Glucosidase Inhibitory Activity of Stings of Gleditsia sinensis Extracts (조각자 추출물의 항산화 활성 및 alpha-glucosidase 저해 활성)

  • This study was performed to investigate the physiological activities of stings of Gleditsia sinensis extracts. Antioxidant activity was evaluated by measuring total phenolic contents (TPC), comet assay, and 2.2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity (RSA). Anti-diabetic activity was measured by inhibition activities on $\alpha$-glucosidase. Stings of Gleditsia sinensis extracts were prepared by extracting them with methanol and ethanol. The methanolic extracts showed the highest phenol content (1.12 g/100 g gallic acid equivalents). The $\alpha$-glucosidase inhibitory activity of methanol extracts were 17.9% higher, and that of ethanol extracts were 10.3% higher at a concentration of 1 mg/ml. These results indicate that stings of Gleditsia sinensis might be potential candidates as antioxidant and anti-diabetic agents.

Antioxidant Compounds and Antioxidant Activities of Methanolic Extracts from Milling Fractions of Oat (귀리 도정 부위별 메탄올 추출물의 항산화 성분 및 항산화 활성)

  • Oats (Avena sativa L.) are well known for having high contents of bioactive compounds and health benefits. The objective of this study was to evaluate the antioxidant compounds and antioxidant activities of methanolic extracts from milling fractions and whole grains of oat. The contents of total polyphenolics and flavonoids were analyzed by spectrophotometric methods. ABTS radical and DPPH radical scavenging activities and reducing power were used to compare relative antioxidant activities of milling fractions from oat. Total polyphenolics and flavonoids were much more abundant in oat bran extract than in extracts from whole grain and endosperm. In addition, high levels of ABTS radical (42.34 mg Trolox equivalent antioxidant capacity (TEAC)/100 g sample) and DPPH radical (24.18 mg TEAC/100 g sample) scavenging activities and reducing power ($A_{700}=0.76$) were detected in oat bran. The results of this study indicate that oat bran has significantly higher antioxidant activities and appears to have significant health benefits.
