한국(韓國) 자생차엽(自生茶葉)의 향기성분(香氣成分) (Volatile Components of Korean Native Tea Leaves (Camellia sinensis O. Kuntze))

  • 12개 지역의 한국자생차 향기성분을 기체크로마토그래프 및 질량분광계를 이용하여 분석한 결과는 다음과 같다. 다엽중의 향기성분으로 70~80개 성분을 GC를 이용해 분리할수 있었고, 그중 52개 성분은 GC/MSD로 동정하였는데, 이들을 분류하면 alcohol류 19종, aldehyde류 5종, hydrocarbon류 2종, ketone류 6종, ester류 4종, lactone류 3종, acid류 2종, phenol류 3종, pyrazine류 4종, 그리고 질소화합물 4종이였다. 이중 geraniol, linalool oxide, 1-hexanol, ehtanol등이 주요 향기 성분이었다.

GC-ECD와 GC-MSD를 이용한 유니코나졸 잔류실태조사 (Monitoring of Uniconazole by Using GC-ECD and GC-MSD)

  • 본 연구에서는 2012년 1월부터 8월까지 농산물 중 엽채류에 대하여 GC-ECD 및 GC-MSD를 이용하여 uniconazole의 잔류실태조사를 실시하였다. 겨자채에 표준용액을 0.05, 0.5 mg/kg 농도로 하여 회수율을 측정한 결과 98.9%에서 109.4%까지 양호한 결과를 나타내었다. 2012년 1월부터 8월까지 서울지역에 유통되고 있는 농산물 중 엽채류 3,632건에 대하여 모니터링을 실시한 결과 14건에서 uniconazole이 검출되었다. Uniconazole의 검출양은 0.098 mg/kg에서 2.2 mg/kg로 나타났다. 본 연구를 통하여 동시분석법으로 보다 효율적인 검사가 가능할 것으로 기대된다.

삼채잎 향기 성분 분석과 항산화 및 항염 효과 연구 (Analysis of Aroma Components by Part of Allium Hookeri and Research on Antioxidant and Anti-inflammatory)

  • 삼채는 Allium 속 식물로써, 항산화, 항염 및 항균 등에 대한 선행연구가 실시되어왔지만, 정유 추출로써의 연구는 미비한 실정이다. 그에 본 연구는 삼채 잎, 뿌리, 통 부위에 대한 정유 추출에 따른 GC-MSD, 삼채잎의 세포독성, 항산화, 항염을 확인하여 화장품 소재로서의 가능성을 확인하였다. 삼채잎, 뿌리, 통부위의 정유추출 수율은 0.01, 0.02, 0.01%로 확인되었고, GC-MSD 향기성분을 분석한 결과 삼채잎 정유의 주요 성분은 Diallyl trisulfide(34.02%)와 Methyl allyl trisulfide(25.14%)으로 나타났다. 세포독성이 확인되지 않은 10%의 농도에서 NO 생성 39.69%억제, DPPH 라디칼 소거활성 88.26%를 보였다. 이를 통하여 삼채 잎 정유는 화장품 및 식품 산업분야에서 항산화 및 항염 효능이 있는 원료로서 유용하게 활용 가능성을 제시한다.

GC/MSD를 사용한 원유 내 잔류농약의 분석법 연구 (Research for the Analytical Method of Various Pesticides in Raw Milk by Gas Chromatography-Mass Spectrometry)

  • The aim of this study was to optimize a simple, fast, and economical analysis procedure for the determination of 16 different pesticides in raw milk via GC/MSD. Analyses were performed via gas chromatography with electron impact mass spectrometric detection in the selected ion monitoring mode (GC/MSD-SIM) using Pentachloronitrobenzene as the internal standard. The modified sample preparation methodology was based on the Pesticide Analytical Manual (PAM) of the FDA concerning fat extraction, ACN-ether partitioning, and clean-up of the Sep-Pak florisil cartridge. The modified methodology for the determination of the 16 pesticides was validated. The range of LOQs of the 16 pesticides was likely three times lower than their Maximum Residence Levels (MRLs). The recoveries of most of the pesticides were acceptable at the fortification levels of 0.5 and 1.0 ${\mu}g/mL$ and their RSD (%) level was less than 20%. None of the 16 pesticides were detected in the selected raw milk samples.

수삼과 홍삼농축액 중 유기인계와 유기염소계 농약의 잔류 (Residue of Organophosphorus and Organochlorine Pesticides in Fresh Ginseng and Red Ginseng Extract)

  • 본 연구에서는 경북의 풍기와 상주에 있는 인삼재배포장에서 직접 시료를 채취하고, 이를 이용한 홍삼농축액제품을 제조하였다. 수삼과 홍삼농축액 중 유기인계와 유기염소계 농약을 GC-NPD와 GC-ECD로 각각 정량분석 하였으며 GC-MSD로 정성분석을 실시하여, 식품의약품안전청이 2007년 1월1일부터 시행한 농약잔류허용기준에 따라 안전성을 평가하였다. 풍기 수삼에서 유기인계 농약은diazinon을 비롯한pyrimethanil, tolclofos-methyl, metalaxyl, diethofencarb, parathion, cyprodinil, tolylfluanid와 kresoxim-methyl이 모두 불검출 되었다. 그러나 상주 수삼에서는 tolclofos- methyl이 평균 $0.054{\pm}0.008\;mg/kg$ 검출되어 수삼의 농약잔류허용기준 0.3mg/kg의 $18{\pm}2%$ 수준이었다. 풍기와 상주 수삼에서 유기염소계 농약은 BHC, DDT의 이성질체와 aldrin, azoxystrobin, captan, cypermethrin, deltamethrin, dieldrin, difenoconazole, endosulfan-sulfate, endrin, fenhexamid, quintozene, ${\alpha}-endosulfan$, ${\beta}$-endosulfan도 검출 되지 않았다. 한편 풍기와 영주의 인삼으로 제조한 홍삼농축액 중에도 유기인계와 유기염소계 농약은 모두 검출 되지 않았다.

GC/MS를 이용한 잎담배 중 알칼로이드 함량 분석 (Determination of Nicotine and Other Minor Alkaloids in Tobacco Leaves by GC/MS)

  • To obtain the optimum condition for analysis of 10 alkaloids in tobacco leaves, such as nicotine, nornicotine, anatabine, anabasine, myosmine, cotinine, 2,3'-dipyridyl, $\beta-nicotyrine,\;\beta-nornicotyrine\;and\;\beta-formylnornicotin$, 5 types of extraction method were investigated by GC-FID and GC/MS. The optimum condition of alkaloid extraction was achieved by using methanol:dichloromethane(1:3, v/v) after NaOH treatment. The use of mass selective detector (MSD) provided unambiguous nicotine related alkaloid analysis. Alkaloids in various tobacco leaves were extracted with the optimum extraction condition and quantified by GC/MS/SIM mode. Compared with concentrations of alkaloids among the various tobacco leaves, the concentration of alkaloids was generally in the order burley > flue-cured > oriental tobacco. In flue-cured tobacco leaves, the order of concentration of alkaloids was nicotine > anatabine > nornicotine > $\beta-nicotyrine\;>\;\beta-formylnornicotine\; >\;myosmine\;>\;2,3'-dipyridyl\;>\;cotinine\;>\;anabasine\;>\;\beta-nornicotyrine$. However, in the case of burley and oriental tobacco leaves, the concentration of nornicotine was higher than that of anatabine.

원각사지 십층석탑 오염물의 유기산 분석 (Analysis of the Organic Acid Contaminants on the surface of TEN-STORIED STONE PAGODA ON THE SITE OF WON-GAKSA)

  • TEN-STORIED STONE PAGODA ON THE SITE OFWON-GAKSA(Temple) which is one of three marble pagodas in South Korea, were dated from the thirteenth year of the reign of King Sejo(1467). On the roof and surface of each the stories, there were large amount of the contaminants such as pigeon′s excretions, dust and environmental elements for a long time. The pH of contaminants is not acid, but is 7.2, neutral. To find the species of organic acidscontained in the contaminants and the degree of damaging for a marble pagoda, we analyzed the contaminants using GC-MSD method by the following procedures. Organic acids were extracted by saponifying whole contaminants. After Saponification, the organic acids were mathylated to increase their volatility upon subsequent GC-MSD analysis. The mathyl esters of the organic acids are extracted from the acidified aqueous solution. And the organic extracts were washed with adilute base solution. The washed extract were analyzed by GC(Hewlett Packard 5890)with a nonpolar capillary column(Crosslinked 5% Ph Me Silicone, $50×0.2㎜×0.33\mum$film thickness, USA) and Mass Spectrometric Detector(Hewlett Packard 5970B).As the result, it was found that 12 organic acids were the main compound in pagoda′contaminants, and the amount of organic acid were negligible.

한약재중의 잔류농약 분석 - GC에 의한 18종 잔류농약의 분석 - (Determination of residual pesticides in crude drugs - Gas chromatographic Analysis of 18 pesticides -)

  • This study was carried out to determine the 11 organochlorine, 7 organophosphorus residual pesticides in 251 crude drugs. These residual pesticides in herbal drugs were extracted with acetonitrile and the extracts were cleaned up via LC-florisil solid phase extraction column. The prepared samples were assayed for pesticide residues using GC-ECD, NPD with capillary column and identified by GC-MSD. Recoveries were $63.9{\sim}111.5%$ in the organochlorine pesticides and $69.8{\sim}92.4%$ in the organophosphorus pesticides, and detection limits were $0.001{\sim}0.65\;ppm$ in the organochlorine pesticides and $0.0009{\sim}0.0074\;ppm$ in the organophosphorus pesticides. Pesticide residues were detected in 9 cases.

Lipoxygenase 결핍 콩과 그 가공품의 휘발성 성분 분석 (Studies on Volatile Compounds in Lipoxygenase Deficient-soybean and Its Products)

  • Lipoxygenase(LOX) in soybeans is responsible for beany flavors which limit the wide utilization of soybeans to foods. This study was conducted to analyze beany flavor compounds of the normal Hwagkeumkong and LOX-deficient soybean cultivars, Jinpumkong which lacks L-2, L-3, and Jinpumkong 2 which lacks all L-1, L-2, L-3. Using the combination of dynamic headspace sampling and gas chromatography-mass selective detector(DHS-GC-MSD) for analyzing volatile compounds, hexanal and hexanol were identified in whole soy flour of all three soybena cultivars. Hwangkeumkong had more volatile compounds than Jinpumkong and Jinpumkong 2 in defatted soy flour. Hexanal and acetic acid were identified in soy milk of all three soybean cultivars but Hwangkeumkong had more volatile compounds than Jinpumkong 2. From the analysis with a static headspace sampling(SHS) and GC-MSD the major compounds were hexanal, acetic acid, 1-hexanol, and 1-octen-3-ol. The content of acetic acid was similar among three cultivars. But contents of hexanal and pentanal in Jinpumkong 2 were less than that of Jinpumkong and Hwangkeumkong. Using GC-FID, Jinpumkong 2 had less contents of hexanal and pentanol than Hwangkeumkong in whole soy flour and defatted soy flour. In this study, LOX-deficient soybean cultivars showed less hexanal, pentanol and other compounds than the normal Hwangkeumkong. However quite amount of beany flavor compounds were identified in Jinpumkong and Jinpumkong 2. So further studies are required to characterize LOX isozymes, to understand the mechanisms of beany flavors production, and to develop some other methods for removing beany flavor.

목제 주방용품 및 위생용품에 잔류하는 유해물질에 관한 연구 (Study on the Hazard Compounds from Wooden Kitchenwares and Sanitary Produce)

  • 시중 유통되고 있는 50종의 목제 주방기구와 위생용품을 각각 benzene과 n-heptane에 용출시켜 GC-MSD를 이용하여 분석한 결과, naphthalene과 diethyl phthalate는 모든 조건에서 검출되지 않았으나, biphenyl의 경우 전 제품에서 미량이 검출되었다. Biphenyl은 자작나무 수입 원목과 국산 원목에서도 각각 0.25 ppm, 0.34 ppm 함유되어 있는 것으로 확인되었다. 또한 biphenyl의 검출량은 목제를 가공하는 과정 중에 감소하며, 그 검출량은 과일의 잔류 허용치 (110 ppm)에 비해 극히 미량인 것으로 판단된다. 따라서 biphenyl의 유입경로는 제품의 제조 및 생산과정에서 첨가되었다기 보다는 제제 자체에 잔존한다고 추정되며, 이러한 biphenyl의 잔존량은 인체에 영향을 미칠 수 있는 농도는 아닌 것으로 판단된다.
