Several studies report the anticancer effect of Glycyrrhiza glabra (G. glabra), Glycyrrhiza uralensis (G. uralensis) and their compounds. However, the anticancer effect of Glycyrrhiza cultivar roots are limited. In this study, we compared the anticancer effect of Glycyrrhiza cultivar (Wongam and Shinwongam) extracts with G. glabra and G. uralensis extracts in breast cancer cell lines. Freeze dried Glycyrrhiza root extracts were dissolved in cell culture media at 2 mg/mL and filtered by $0.2{\mu}m$ filter. Glycyrrhiza root extracts were serially diluted at the concentrations of $10{\mu}g/mL$, $100{\mu}g/mL$, $200{\mu}g/mL$, $400{\mu}g/mL$, $800{\mu}g/mL$, $1000{\mu}g/mL$ and $2000{\mu}g/mL$. MCF-7 and MDA-MB-231 breast cancer cells were treated with different concentrations of Glycyrrhiza root extracts and the cell viability was measured using MTT assay. In MCF-7 cells, G. glabra showed no significant difference with Wongam and showed significant difference with Shinwongam at $1000{\mu}g/mL$ (G. glabra 101.2% and Shinwongam 82.68%) and $2000{\mu}g/mL$ (G. glabra 83.07% and Shinwongam 54.05%). G. uralensis showed significant difference with Wongam at $2000{\mu}g/mL$ (G. uralensis 66.48% and Wongam 95.02%) and showed no significant difference with Shinwongam. In MDA-MB-231 cells, G. glabra showed no significant difference with both Wongam and Shinwongam. G. uralensis showed significant difference with Wongam at $2000{\mu}g/mL$ (G. uralensis 72.59% and Wongam 93.47%) and showed no significant difference with Shinwongam. In conclusion, the current study demonstrated that G, glabra and G. uralensis compared with Wongam, and Shinwongam at low concentrations ($10{\mu}g/mL{\sim}800{\mu}g/mL$) display similar cytotoxic potency.

Oxidative stress is common cause of neurodegenerative diseases. The purpose of this study is to investigate the in vitro free radical scavenging activity and protective effect of three Glycyrrhiza species including Glycyrrhiza uralensis, G. glabra, and a new variety of Glycyrrihza (Shinwongam, SW) against hydrogen peroxide-induced oxidative stress in C6 glial cells. In vitro assays, radical scavenging activities of G. uralensis, G. glabra, and SW against 2,2-diphenyl-1-picrylhydrazyl, ·OH, and O2- increased as concentration-dependent manner. In addition, the SW was found to contain the highest polyphenol and flavonoid contents. The treatment of H2O2 to C6 glial cell induced oxidative stress, whereas G. uralensis, G. glabra, and SW significantly increased the cell viability as dose-dependent manner. In particular, SW exerted stronger protective effect on H2O2-induced cytotoxicity, than G. uralensis and G. glabra. Furthermore, reactive oxygen species (ROS) formation was significantly elevated by H2O2 in C6 glial cells. However, treatments of G. uralensis, G. glabra, and SW decreased ROS formation. In addition, SW decreased pro-inflammatory related protein expression levels such as inducible nitric oxide synthase and cyclooxygenase-2, compared to H2O2-treated control group. These results indicated that G. uralensis and G. glavra, especially SW, may be useful for preventing from oxidative stress-induced neuronal damage by regulating inflammatory reaction.

Ultraviolet B (UVB) damages DNA residues in skin keratinocytes. In particular, the formation of cyclobutane pyrimidine dimers (CPD), a pyrimidine residue damage in DNA, is considered a representative indicator of skin photoaging. In this study, we confirmed defensive effect of Glycyrrhiza glabra (G. glabra) extract against UVB induced DNA damage. First of all, we confirmed UVB dependent amount of CPD formation in human keratinocyte cell line. UVB induced CPD was decreased by G. glabra extract by dose dependent manner. In addition, it was confirmed that the expression of mRNA of DNA damage recovery factors was increased by G. glabra extract. Consequently, through this study, it was possible to confirm the DNA protection effect of G. glabra extract in skin keratinocytes.
Objectives : Genetic analysis and taste pattern were performed to identify species between Glycyrrhiza uralensis and G. glabra which are officially listed in Korean Pharmacopoeia IX as origin of Gamcho(g$\={a}$nc$\v{a}$o, licorice root, Glycyrrhizae Radix et Rhizoma). Methods : Genetic analysis showed that identification between two species was done by comparing base sequence of ITS(intergenic transcribed spacer) and trnH-psbA regions from eleven Gamchoes sold in market. There was different taste pattern using by taste sensor in Glycyrrhiza uralensis and G. glabra. Results : Genetic analysis showed that six Gamchoes from China were identified as Glycyrrhiza uralensis and five Gamchoes from Uzbekistan were G. glabra. From the results of taste pattern, sourness and astringency of Glycyrrhiza uralensis from China were significantly higher than G. glabra from Uzbekistan, and aftertaste of astringency, aftertaste of umami, and saltiness of Glycyrrhiza uralensis were signicantly low as compared to G. glabra. There is no significant difference between two species in terms of bitterness, aftertaste of bitterness, and umami. Conclusions : Taken together, Glycyrrhiza uralensis from China and G. glabra from Uzbekistan were identified by taste sensor, and this technic could be applied to establishment of taste pattern marker for identification of different species located in various regions.

This study was investigated to elucidate the growth characteristics and the contents of glycyrrhizin and free sugars of Glycyrrhiza plants, i. e. G. glabra var. glandulifera, G. uralensis, and G. pallidiflora. The growth of G. glabra var. glandulifera and G. pallidiflora was better in plots planted with stolon and stem over 2cm in diameter compared to that of below 1cm and $1{\sim}2cm$. G. glabra var. glandulifera didn't bloom in Suwon but produced stolon, while G. pallidiflora bloomed but did not produce stolon. Total root weight of six-year-old G. glabra var. glandulifera consisted of stolon (73.2%) and primary root (26. 8 %). The contents of glycyrrhizin and free sugars were highest in stolon, followed by tap root and leaf. The glycyrrhizin contents in Glycyrrhiza plants were 7.38% in G. glabra var. glandulifera, 5.29% in G. uralensis and 0. 15% in G. pallidiflora.
The ethanolic extract of some medicinal plants having anti-asthmatic activity such as Solanum xanthocarpum, Curcuma longa, Glycyrrhiza glabra, Piper longum, A. vasica, A. lebbeck, and Tinospora cordifolia was evaluated for antihistaminic and anti-cataleptic activity. The aqueous solution of ethanolic extract of S. xanthocarpum and G. glabra potentiated histamine-induced tracheal chain contractions. Whereas, C. longa, P. longum, and T. cordifolia, and A. lebbeck were without any significant effect on histamine. Only A. vasica inhibited histamine-induced tracheal chain contraction. G. glabra per se produced contraction of the tracheal chain, which was blocked by pretreatment with atropine. Single dose of S. xanthocarpum potentiated clonidine-induced catalepsy but on repeated doses (once in a day for 3 days) inhibited catalepsy. Pretreatment with ethanolic extract of C. longa, P. longum, T. cordifolia inhibited catalepsy whereas G. glabra and A. lebbeck significantly potentiated clonidine-induced catalepsy. None of the extracts inhibited haloperidol-induced catalepsy. Thus the extracts having antihistaminic activity or mast cell stabilizing activity inhibited clonidineinduced catalepsy.

In this work, comparative study on antioxidative activities of extracts from Glycyrrhiza uralensis (G. uralensis) produced in Korea and in China and Glycyrrhiza glabra (G. glabra) produced in Uzbekistan was conducted. Among three origins, 50% ethanol extracts (21.15 ${\mu}g/mL$), ethyl acetate fraction (29.15 ${\mu}g/mL$) and aglycone fraction (3.26 ${\mu}g/mL$) of G. uralensis from Korea showed the higher free radical (1,1-phenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$) than extracts from other origins. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of extracts from three origins on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system were investigated using luminol-dependent chemiluminescence assay 50% ethanol extract (1.00 ${\mu}g/mL$) and ethyl acetate fraction (0.34 ${\mu}g/mL$) of G. uralensis from China showed the most prominent ROS scavenging activity. The protective effects of extract/fractions of G. uralensis and G. glabra extracts on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. 50% ethanol extract and aglycone fraction of G. uralensis and G. glabra extracts from three origins showed cellular protective effects in a concentration dependent manner (5 ~ 50 ${\mu}g/mL$). Aglycone fraction of G. uralensis from Korea (${\tau}_{50}$ = 847.4 min)especially showed cellular protective effects four times higher than that from China (${\tau}_{50}$ = 194.3 min). These results indicate that G. uralensis and G. glabra extracts, which have been used as whitening agent, could be applicable to functional cosmetic ingredient as a natural antioxidant. Judging from the prominent cellular protecitve effects, it is concluded that G. uralensis and G. glabra extracts can protect the skin from $^1O_2$ and various ROS induced by UV.

Licorice species (Glycyrhiza species) are perennial plants belonging to the Leguminosae family. Licorice is world-widely distributed in Asia, Europe, and the Americas. The licorice species, such as Glycyrhiza uralensis (G. uralensis) and G. glabra, have been widely used in traditional oriental medicine. G. uralensis is found in Central Asia to the northeastern part of China and G. glabra is distributed from southern Europe to the northwestern part of China. These licorice species are characterized by having various pharmacological activities, including anti-oxidant, anti-inflammatory, immune improvement, and anti-tumor effects. In this study, we investigated the comparative anti-inflammatory effects of four licorice varieties (G. glabra L., G. uralensis FISCH., Shinwongam, and Wongam) on LPS-induced inflammatory responses in RAW 264.7 macrophage cell line. We evaluated the cytotoxicity of licorices at various concentrations. In addition, the nitric oxide (NO) production was elucidated by the treatment of licorice.
The effects of fermented products from 40 medicinal herbals commonly available in Korea were examined according to concentrations of polyphenolic compound and kojic acid, and the activities of DPPH ($\alpha,\alpha$'-diphenyl-$\beta$-picrylhydrazyl) free radical scavenging and tyrosinase. The polyphenolic compound concentrations were 0.24 by Corydalis turtschaminovill ~ 11.42% (dry matter basis) by Syringa velutina in the extracts and 0.18 by Poria cocos ~ 12.27% by S. velutina in the fermented products. Kojic acid concentrations were 0.02 by Poria cocos Sclerotium ~ 9.67 mM by S. velutina in the extracts and 0.33 by P. cocos ~ 10.32 mM by S. velutina in the fermented products. Syringa velutina contained the highest polyphenolic compound and kojic acid concentrations, which were higher in the fermented product than in the extract. Higher DPPH free radical scavenging activity (>60%) was observed in the extracts of A. sessiliflorum, Citrus nobillis, and Angelica gigas and the fermented product of A. sessiliflorum compared to the other medicinal plants. Higher tyrosinase inhibition activity (>50%) was observed in the extracts of Morus alba, Glycyrrhiza glabra, and Rubus coreanus and the fermented products of G. glabra, Cnidium officinale, and S. velutina. Based on the above results, G. glabra, C. officinale, and S. velutina possessed high tyrosinase-inhibitive activities and kojic acid concentrations, which could be definitely enhanced by the fermentation of Phenillus linteus mycelium.
Colorectal cancer is one of the most common lethal cancer types worldwide. In recent years, widespread and large-scale studies have been done on medicinal plants for anti-cancer effects, including Glycyrrhiza glabra. The aim of this study was to evaluate the effects of an ethanol extract Glycyrrhiza glabra on the expression of HSP90, growth and apoptosis in the HT-29 colon cancer cell line. HT-29 cells were treated with different concentrations of extract (50,100,150, and $200{\mu}g/ml$). For evaluation of cell proliferation and apoptosis, we used MTT assay and flow cytometry technique, respectively. RT-PCR was also carried out to evaluate the expression levels of HSP90 genes. Results showed that Glycyrrhiza glabra inhibited proliferation of the HT-29 cell line at a concentration of $200{\mu}g/ml$ and this was confirmed by the highest rate of cell death as measured by trypan blue and MTT assays. RT-PCR results showed down-regulation of HSP90 gene expression which implied an ability of Glycyrrhiza glabra to induce apoptosis in HT-29 cells and confirmed its anticancer property. Further studies are required to evaluate effects of the extract on other genes and also it is necessary to make an extensive in vivo biological evaluation and subsequently proceed with clinical evaluations.
