Agrobacterum tumefaciens-mediated transformation (ATMT) is becoming an effective system as an insertional mutagenesis tool in filamentous fungi. We developed and optimized ATMT for two Colletotrichum species, C. falcatum and C. acutatum, which are the causal agents of sugarcane red rot and pepper anthracnose, respectively. A. tumefaciens strain SK1044, carrying a hygromycin phosphotransferase gene (hph) and a green fluorescent protein (GFP) gene, was used to transform the conidia of these two Colletotrichum species. Transformation efficiency was correlated with co-cultivation time and bacterial cell concentration and was higher in C. falcatum than in C. acutatum. Southern blot analysis indicated that about 65% of the transformants had a single copy of the T-DNA in both C. falcatum and C. acutatum and that T-DNA integrated randomly in both fungal genomes. T-DNA insertions were identified in transformants through thermal asymmetrical interlaced PCR (TAIL-PCR) followed by sequencing. Our results suggested that ATMT can be used as a molecular tool to identify and characterize pathogenicity-related genes in these two economically important Colletotrichum species.

The feasibility of using bleaching treatments for removing fungal stain was evaluated on heavily stained raiadta pine sapwood. Sodium chlorite and sodium hypochlorite appeared to destain fungal discoloration by providing proper treatment conditions (chemical concentration, treatment temperature, and treatment time), while hydrogen peroxide did not remove fungal stain under the bleaching regimes evaluated. The addition of sodium silicate and sodium hydroxide in the hydrogen peroxide solution as a buffer could remove fungal discoloration completely; however, the color of wood surface turned faint green after bleaching, thereby reducing the lightness of bleached samples. The results suggest that hydrogen peroxide bleaching could be a feasible method for removing fungal discoloration of stained radiata pine sapwood, although further research is needed to solve the problem of color change after bleaching. Also, further tests under field conditions are recommended.

The Ulmus davidiana have been used in traditional oriental medicine as remedies for inflammation, ulcers, cancers, bacterial infections and scabies. In this study, the anti-fungal, anti-oxidant, and anti-inflammatory activitiesof a supercritical extract of U. davidiana were investigated in vitro. To explore the anti-oxidant and anti-inflammatory characteristics of the supercritical extract, ABTS radical scavenging activity, and the inhibition of nitric oxide production in LPS-stimulated RAW 264.7 cells were examined, respectively. In addition, the anti-fungal activities of the extract were assessed. The results showed a concentration-dependent increase in ABTS radical scavenging activity. Cells stimulated with LPS produced more nitric oxide than normal control cells; however, cells treated with the supercritical fluid extract decreased this production in a concentration-dependent manner. Finally, the supercritical fluid extracts showed significant anti-fungal activity. These results suggest that extracts of the U. davidiana might be used to develop potent anti-fungal, anti-oxidant, and anti-inflammatory agents, and may be useful as ingredients for related new functional cosmetic materials.
To see effect of marine rock powder and fungal culture supernatant, we analyzed the biodegradation rates of harmful marine dinoflagellate, Heterosigma akashiwo and Prorocentrum minimum for developing the effective control methodology of algal bloom. Relatively low removal rates were observed in the treatment of marine rock powder or buffer solution alone. However, the lysis of H. akashiwo and P. minimum was enhanced in the combined treatments of marine rock powder with fungal supernatant. The effective concentration and exposure time of fungal supernatant for the lysis of H. akashiwo and P. minimum were 5 ml/l and 30 minutes, respectively. These results suggest that the fungal supernatant may be a biocontrol agent for the control of algal blooms in seawater.
Fluctuation of fungal zoospores on agar strips were observed in the rumen of sheep fed three different levels of dietary concentrate, timothy hay: concentrate = 3:0 (AF diet), timothy hay: concentrate = 2:1 (MC diet), timothy hay : concentrate = 1:2 (HC diet) respectively. The number of zoospores on the strip was drastically decreased after morning feed with AF diet. The number was the highest at 0 h ($1.34{\times}10^2/cm^2$), then declined to $2.0{\times}10^3/cm^2$ at 9 h after feeding. In the rumen of animals fed MC diet, the number of zoospores decreased with time after feeding, although the decrement was slower than that with AF diet. During 0-3 h after feeding, number of zoospores was $1.6{\times}10^4/cm^2$. Although the number slightly decreased at 6 and 9 h, relatively high levels were maintained. It seems that the inducers for zoospore-release were maintained at relatively high concentration throughout incubation period. The fluctuation pattern of number of germinated zoospores was different in the rumen of animals fed HC diet from those of AF and MC diets. The number of zoospores was constantly maintained at lower level ($1.0{\times}10^3/cm^2$) than the other diets. For MC diet, continuous high number of germinated zoospores may be due to the continuous release of zoospores by hemes in timothy hay and concentrate feed, and by unknown mechanisms. Unlike AF diet which promoted relatively rapid decline of zoosporogenesis, supplementation of concentrate feed to the timothy hay did not promote such rapid decline of zoosporogenesis. It was suggested that release of inducers for zoosporogenesis from concentrate feed persisted longer time than from timothy hay. HC diet promoted the lowest zoospore production, suggested the lowest fungal population size in this experiment. These results show that an appropriate amount of concentrate may support fungal growth and stimulate zoosporogenesis in the rumen.

Objectives: This study was performed in order to determine airborne fungi levels in homes and find related factors that may affect airborne fungi concentration. Methods: Fifty homes were study subjects for measuring airborne fungi. For sampling airborne fungi, the impaction method on agar plates was used and samples were counted as colony forming units per cubic meter of air ($CFU/m^3$). In addition, information regarding housing characteristics and atopic disease in each home were collected via questionnaire. Results: The geometric means (GM) of airborne fungi concentrations in fifty living rooms and bedrooms were 68.03 and 62.93 $CFU/m^3$, respectively. The GM of airborne fungi concentration in atopy homes was 78.42 $CFU/m^3$. This was higher than non-atopy homes' 54.34 $CFU/m^3$ (p-value=0.051). In the results of the multiple regression analysis, outdoor airborne fungal concentration proved a strong effective factor on indoor airborne fungal concentration. Also, construction year, floor area of house, indoor smoking and frequency of ventilation were factors that showed a significant association with indoor airborne fungi concentration. Conclusions: The results of this study show that some housing and living characteristics may affect the development and increase of airborne fungi. In addition, exposure to airborne fungi may be a risk factor for the prevalence of childhood atopic diseases.
Several challenges arise in DNA extraction and gene amplification for airborne fungal metagenome analysis from a particulate matter (PM) samples. In this study, various conditions were tested to optimize the DNA extraction method from PM samples and polymerase chain reaction (PCR) conditions with primer set and annealing temperature. As a result of comparative evaluation of DNA extraction under various conditions, chemical cell lysis using buffer and proteinase K for 20 minutes and bead beating treatment were followed by using a commercial DNA extraction kit to efficiently extract DNA from the PM filter samples. To optimize the PCR conditions, PCR was performed using 10 primer sets for amplifying the ITS2 gene region. The concentration of the PCR amplicon was relatively high when the annealing temperature was 58℃ with the ITS3tagmix3/ITS4 primer set. Even under these conditions, when the concentration of the PCR product was low, nested PCR was performed using the primary PCR amplicon as the template DNA to amplify the ITS2 gene at a satisfactory concentration. Using the methods optimized in this study, DNA extraction and PCR were performed on 15 filter samples that collected PM2.5 in Seoul, and the ITS2 gene was successfully amplified in all samples. The optimized methods can be used for research on analyzing and interpreting the fungal metagenome of atmospheric PM samples.
The patterns of fungal growth and fiber digestion under the microscope, and tile productions of fibrolytic enzyme were studied in an in vitro culture with Neocallimastix frontalis SA when either filter paper or rice straw was provided as sole energy source. Under the microscopic observation, active zoospores attachment, sporangium development and complex rhizoidal system were founded on the surface and at the edge of filter paper. After 7 days of incubation, a reduced fiber mass, a decreased fiber cohesion and a weakened fiber structure by fungal digestion were clearly observed. Similar fungal development was observed with rice straw, but fungal growth and digestion took place mostly on the damaged and exposed portion of rice straw. Although there were some differences in absolute concentration and pattern, the concentration of both cellulase and xylanase increased with incubation time with the higher activity being obtained with filter paper. Their differences were large especially after 48 and 96hr of incubation(P< 0.05). The filter paper was more good inducer of cellulolytic and xylanolytic enzymes compared with complex substrate, rice straw. These findings suggest that the filter paper is the better energy source for N frontalis than the complex substrate, and structural disintegration by physical process is able to help rumen fungal growth on the lignified roughage although anaerobic rumen fungi have mechanical and enzymatic functions for fiber digestion.
Objectives: Exposure to mold is strongly associated with adverse health effects (development or exacerbation of allergic diseases). We reviewed the health effects of mold exposure and explored to determine the annual distribution of indoor mold in facilities with susceptible populations. Methods: The health effects of mold exposure were mainly summarized by reviewing related papers and WHO research reports. We selected 10 facilities, including daycare centers, postpartum care centers, medical institutions, and elderly care facilities within the Seoul Metropolitan. Mold sampling was performed once every week or once every quarter from February 2016 to 2017. In addition, fungal species analyses was performed, and distribution status by month and facility was analyzed in the same manner as concentration. Results: Adverse health effects attributed to fungal exposure are largely divided into allergic symptoms, toxic effects, and infectious effects. Monthly mean concentrations of mold indoors and outdoors was 368.8 CFU/㎥ (geometric mean 213.4 CFU/㎥) and 496.0 CFU/㎥ (327.9 CFU/㎥), respectively. The indoor concentration has begun to increase in February, peaked in July, declined in August, increased again until October, and then decreased in November. About 36 genera of indoor fungal species were found in each facility. Cladosporium sp., Penicillium sp., Fusarium sp., Aspergillus sp., Alternaria sp., and Arthrinium sp. were observed as the dominant species. Conclusions: Our findings showed that the overall level of indoor mold was below the 500 CFU/㎥ level recommended by the Ministry of Environment. The development of DNA-based assessment and expanding facilities to be monitored for mold would be necessary for preventive aspects.

For reduction of side effects by anti-fungal agents, a less toxic anti-fungal substance or a synergistic substance with a new mechanism is needed. The anti-yeast substance (AYS) originated from Rahnella aquatilis strain AY2000 is like to be a heterogeneous protein. The AYS inhibited the growth of Candida albicans in culture broth, and AYS-treated cells were arrested in each phase during cell cycle. Among AYS-treated cells, the population of the cells belonging to sub-G1 phase was not increased during cell cycle. Therefore, AYS has rather yeaststatic than yeastcidal effect to C. albicans. Moreover, with combination of itraconazole or fluconazole, AYS had a synergistic anti-yeast activity against Saccharomyces cerevisiae based on the analysis of fractional inhibitory concentration index.
