청소년 및 고령층을 위한 잡곡밥의 혼합비율 설정 및 영양성, 기능성 평가 (Establishment of Mixing Ratio of Multigrain Rice for Adolescent and Aged People and Its Nutritional and Functional Estimation)

  • 본 연구는 청소년과 고령층을 대상으로 기호성이 높은 잡곡밥 혼합비율을 설정하고, 영양성과 기능성을 평가하였다. 청소년과 고령층을 대상으로 잡곡 혼합비율에 따른 잡곡밥의 기호도를 측정한 결과, 청소년층을 위한 혼합잡곡(MGA)은 40% 잡곡 첨가군을, 고령층을 위한 혼합잡곡(MGAP)은 소화흡수율과 기호도를 고려하여 30% 잡곡 첨가군을 선정하였다. 또한 청소년 및 고령층의 전체 잡곡 중 특화잡곡 혼합 비율은 전체적 기호도가 가장 높은 10%와 20%로 각각 선정하였다. 특화잡곡 간의 비율은 전체적인 기호도가 가장 높은 혼합비율로 선정하였으며, 그 결과 MGA는 녹두와 백태의 혼합비율을 2:1로, MGAP는 율무와 메밀의 혼합비율은 1:1로 결정하였다. 혼합작곡의 영양성 및 기능성을 평가한 결과, MGA에서 칼로리 및 조단백질, 조지방 함량이 가장 높게 나타났으며, 필수아미노산 함량 또한 다양하게 존재하였고, 무기질의 함량도 백미(PR) 및 MGAP에 비해 다량 함유되어 있음을 확인하였다. 반면 기능성과 관련된 총 폴리페놀 및 플라보노이드 함량은 MGAP에서 가장 높았으며, hydroxyl 라디칼 소거능을 제외한 나머지 라디칼 소거능 또한 MGAP에서 가장 높게 나타났다.

사람 골육종 세포 Saos2에서 아미노산 수송계 L의 발현 및 기능적 특성 (EXPRESSION AND FUNCTIONAL CHARACTERIZATION OF AMINO ACID TRANSPORT SYSTEM L IN SAOS2 HUMAN OSTEOGENIC SARCOMA CELLS)

  • Amino acids are required for protein synthesis and energy sources in all living cells. The amino acid transport system L is a major nutrient transport system that is responsible for $Na^+$-independent transport of neutral amino acids including several essential amino acids. In malignant tumors, the L-type amino acid transporter 1 (LAT1), the first isoform of system L, is highly expressed to support tumor cell growth. In the present study, the expression and functional characterization of amino acid transport system L were, therefore, investigated in Saos2 human osteogenic sarcoma cells. RT-PCR and western blot analyses have revealed that the Saos2 cells expressed the LAT1 and the L-type amino acid transporter 2 (LAT2), the second isoform of system L, together with their associating protein heavy chain of 4F2 antigen (4F2hc) in the plasma membrane, but the expression of LAT2 was very weak. The uptakes of [${14}^C$]L-leucine by Saos2 cells were $Na^+$-independent and were completely inhibited by the system L selective inhibitor, 2-aminobicyclo-(2,2,1)-heptane-2-carboxylic acid (BCH). The affinity of [${14}^C$]L-leucine uptake and the inhibition profiles of [${14}^C$]L-leucine uptake by various amino acids in the Saos2 cells were comparable with those for the LAT1 expressed in Xenopus oocytes. The majority of [${14}^C$]L-leucine uptake is, therefore, mediated by LAT1 in the Saos2 cells. These results suggest that the transports of neutral amino acids including several essential amino acids into Saos2 human osteogenic sarcoma cells are for the most part mediated by LAT1. Therefore, the Saos2 human osteogenic sarcoma cells are excellent tools for examine the properties of LAT1. Moreover, the specific inhibition of LAT1 in tumor cells might be a new rationale for anti-tumor therapy.

대장균 세포 내 다양한 외부 스트레스에 대한 DPS 단백질의 생리적 기능 (Physiological Function of a DNA-Binding Protein from Starved Cells in Combating Diverse External Stresses in Escherichia coli)

  • 대장균에서 DNA 결합 단백질로 확인된 DNA-binding Protein from Staved cells (DSP)는 DNA를 보호하는 중요한 기능을 한다는 것을 보여주었다. 이 연구의 목표는 야생형 대장균과 dps 유전자 결손 대장균(${\Delta}dps$ E.coli)의 특성 비교를 통해 여러 종류의 스트레스에 대해 대장균에서 DPS의 기능적 역할을 설명하는 것이다. 다양한 스트레스 상태에서 자외선 흡광도계(UV-spectrophotometer)를 이용하여 야생형 대장균과 dps 유전자 결손 대장균의 세포성장을 측정하였으며, 각각의 대장균 세포 성장 속도를 비교함으로써 우리는 대장균에 존재하는 DPS 단백질의 기능적 역할을 확인하였다. 야생형 대장균에 비해 dps 유전자 결손 대장균은 영양분 결핍, 산성화, 열충격, 다양한 활성산소종 스트레스들에 민감한 현상을 나타내었으며, 이것은 DPS가 다양한 극단적인 스트레스에 중요한 기능을 한다는 것을 제안하였다. 결론적으로 대장균의 DPS는 다양한 환경적인 스트레스로부터 DNA와 강하게 결합하여 유지함으로써 세포를 보호하고 세포성장에 결정적인 기능을 한다는 것을 증명하였다.

동결건조 누에분말의 일반 성분 및 영양 성분 (Proximate and nutritional compositions of freeze-dried silkworm powder as edible insect resources)

  • 혈당 조절에 도움을 줄 수 있는 건강기능식품 기능성 원료로 인증을 받은 동결건조 누에분말의 일반 성분 및 영양 성분에 대하여 보고된 바를 고찰하여 다음과 같은 결론을 얻을 수 있었다. 1. 동결건조 누에분말의 일반 성분은 조단백질 57.7%, 조지방 9.5%, 조섬유 5.4%, 회분 9.3%, 수분 4.7% 내외였다. 식육성 재료나 식용 곤충에 비하여 단백질 성분이 높고 지방 성분은 낮았다. 2. 동결건조 누에분말을 이루고 있는 주요 아미노산은 glutamic acid와 aspartic acid로 아미노산의 조성은 누에의 성장 단계에 따른 차이가 있었다. 3. 동결건조 누에분말은 포화지방산과 불포화지방산이 약 3:7의 비율로 있으며, linolenic acid 34.7%, oleic acid 24.4%, palmitic acid 20.2%, stearic acid 9.2%, linoleic acid 7.7%가 많이 포함되어 있다. 4. 동결건조 누에분말에 많이 포함되어 있는 5대 무기물은 칼륨, 인, 칼슘, 마그네숨, 황 등이 있으며, 그 외에도 구리, 망간, 아연, 철 등의 무기물을 가지고 있다.

Characterization of intrinsic molecular structure spectral profiles of feedstocks and co-products from canola bio-oil processing: impacted by source origin

  • Objective: Feed molecular structures can affect its availability to gastrointestinal enzymes which impact its digestibility and absorption. The molecular spectroscopy-attenuated total reflectance Fourier transform infrared vibrational spectroscopy (ATR-FTIR) is an advanced technique that measures the absorbance of chemical functional groups on the infrared region so that we can identify and quantify molecules and functional groups in a feed. The program aimed to reveal the association of intrinsic molecular structure with nutrient supply to animals from canola feedstocks and co-products from bio-oil processing. The objective of this study was to characterize special intrinsic carbohydrate and protein-related molecular structure spectral profiles of feedstock and co-products (meal and pellets) from bio-oil processing from two source origins: Canada (CA) and China (CH). Methods: The samples of feedstock and co-products were obtained from five different companies in each country arranged by the Canola Council of Canada (CCC). The molecular structure spectral features were analyzed using advanced vibrational molecular spectroscopy-ATR-FTIR. The spectral features that accessed included: i) protein-related spectral features (Amide I, Amide II, α-helix, β-sheet, and their spectral intensity ratios), ii) carbohydrate-related spectral features (TC1, TC2, TC3, TC4, CEC, STC1, STC2, STC3, STC4, TC, and their spectral intensity ratios). Results: The results showed that significant differences were observed on all vibrationally spectral features related to total carbohydrates, structural carbohydrates, and cellulosic compounds (p<0.05), except spectral features of TC2 and STC1 (p>0.05) of co-products, where CH meals presented higher peaks of these structures than CA. Similarly, it was for the carbohydrate-related molecular structure of canola seeds where the difference between CA and CH occurred except for STC3 height, CEC and STC areas (p>0.05). The protein-related molecular structures were similar for the canola seeds from both countries. However, CH meals presented higher peaks of amide I, α-helix, and β-sheet heights, α-helix:β-sheet ratio, total amide and amide I areas (p<0.05). Conclusion: The principal component analysis was able to explain over 90% of the variabilities in the carbohydrate and protein structures although it was not able to separate the samples from the two countries, indicating feedstock and coproducts interrelationship between CH and CA.

  • 코로나19 바이러스의 대유행으로 바이러스와 같은 외부 병원균으로부터 우리의 몸을 보호하는 면역 기능 개선 건강기능식품의 시장은 점차 증가하고 있다. 우리는 본 연구에서 높은 조단백, 조지방, 식이섬유 함량을 나타내는 고영양식품인 청국장이 면역 강화 기능을 나타냄을 밝혀내고자 하였다. 청국장 열수 추출물은 RAW 264.7 세포에서 세포독성을 나타내지 않으며, 대식세포의 nitric oxide, reactive oxygen species 및 interleukin (IL)-6, IL-1β, tumor necrosis factor-α 사이토카인의 생산량을 증가시켰다. 또한, 청국장 열수 추출물은 RAW 264.7 세포에서 inducible nitric oxide synthase 및 cyclooxygenase-2의 발현을 유의적으로 증가시켰다. 청국장 열수 추출물은 RAW 264.7 세포에서 I kappa B kinase α/β와 I kappa B (IκB)α의 인산화 및 IκBα의 degradation을 증가시켰으며, Nuclear factor-kappa B p65의 인산화를 증가시켜 p65의 세포질에서 핵으로의 이동을 촉진하였다. 이러한 연구 결과는 청국장 추출물이 선천성 면역 반응을 강화하는데 유망한 건강기능식품 소재로 활용될 수 있음을 시사한다.

조명 및 카메라 이동속도가 토양 영상에 미치는 영향 (Effect of light illumination and camera moving speed on soil image quality)

  • Soil texture has an important influence on agriculture such as crop selection, movement of nutrient and water, soil electrical conductivity, and crop growth. Conventionally, soil texture has been determined in the laboratory using pipette and hydrometer methods requiring significant amount of time, labor, and cost. Recently, in-situ soil texture classification systems using optical diffuse reflectometry or mechanical resistance have been reported, especially for precision agriculture that needs more data than conventional agriculture. This paper is a part of overall research to develop an in-situ soil texture classification system using image processing. Issues investigated in this study were effects of sensor travel speed and light source and intensity on image quality. When travel speed of image sensor increased from 0 to 10 mm/s, travel distance and number of pixel were increased to 3.30 mm and 9.4, respectively. This travel distances were not negligible even at a speed of 2 mm/s (i.e., 0.66 mm and 1.4), and image degradation was significant. Tests for effects of illumination intensity showed that 7 to 11 Lux seemed a good condition minimizing shade and reflection. When soil water content increased, illumination intensity should be greater to compensate decrease in brightness. Results of the paper would be useful for construction, test, and application of the sensor.

한약자원을 이용한 약선차 조성물이 전북 일부지역에 거주하는 여대생의 혈액 성상에 미친 영향 (Effect of Yak-Sun Tea Prescription from Oriental Medicinal Herbs for Blood Metabolic Factors of University Women in Jeonbuk Area)

  • This research evaluated how the composition of Yak-sun(oriental diet therapy), taken as a nutritional supplement with daily meals, can effect the medical condition of those suffering from modern diseases like obesity and hyperlipideia. We produced Yak-sun tea, determined its nutrient composition and physical characteristics and evaluated how this tea effects affects the active oxygen concentration inside the body by clinical practices. The study results indicated that this tea significantly decrease active oxygen concentration. We think that scientific and objective evaluation was done on the components of Yak-sun tea prescription. We concluded that we could apply the components not only in the form of tea, but also in other food forms. The information we received from this conclusion will provide basic information for the application of oriental medicinal resources into other foods and will also be a steppingstone in the field of functional food research for the development of medicinal herbs which is already an important research field world-wide.

동결건조 낫토를 첨가한 영양바의 품질특성 (Quality Characteristics of Nutritional Bar Supplemented with Freeze-Dried Natto)

  • This study evaluated the color, DPPH radical activity, and sensory characteristics of nutritional bars supplemented with varying concentrations (0, 2.5, 5, and 7.5%) of freeze-dried Natto. An inconsistent pattern was observed for the L value, whereas both a and b values increased significantly up to 5% addition of freeze-dried Natto. The DPPH free radical scavenging activity of nutritional bars supplemented with 0, 2.5, 5, and 7.5% freeze-dried Natto were determined to be 59.54, 59.70, 44.85, and 55.07%, respectively. The appearance, color, taste, texture, and overall preference of nutritional bars supplemented with freeze-dried Natto showed significant differences between the samples. The overall preference of the nutrient bar was highest at 6.08 points, determined for the 2.5% Natto supplemented bar. The quality characteristics of the 2.5% added samples exhibited significantly higher values as compared to the controls, thereby indicating the potential of development in health-functional cereal bars.

Association between nutrient intakes and prevalence of depressive disorder in Korean adults: 2014 Korean National Health and Nutrition Examination Survey

  • Objective: Dietary nutrients may play a significant role in depressive disorders. However, sufficient evidences in epidemiological studies are limited. We investigated the cross-sectional association between dietary nutrients and the prevalence of depressive disorder in Korean adults using representative Korean data. Methods: Participants were 2,938 adults aged 19 ~ 64 years from the Korea National Health and Nutrition Examination Survey (KNHANES) conducted in 2014. Dietary intakes were assessed using 24-h recall method. Depressive disorder was assessed using Patients Health Questionnaire-9 (PHQ-9, self-depression test) as applied in 2014 KNHANES only. We defined depressive disorder as having a PHQ-9 score of ${\geq}10$, which was characterized as moderate depression and more. A multivariate logistic regression analysis was performed to estimate the adjusted odd ratios (ORs) and 95% confidence interval (CIs) of depressive disorder. Results: Among the 2,938 subjects, 170 were identified as having depressive disorder. The multivariate-adjusted regression analysis demonstrated that the risk of depression was significantly associated with riboflavin (OR = 0.44, 95% CI: 0.24-0.85, p for trend = 0.018), thiamin (OR = 0.48, 95% CI: 0.23-0.99, p for trend = 0.045), and vitamin C (OR = 0.57, 95% CI: 0.34-0.95, p for trend = 0.025) in the highest versus lowest tertiles of intake. Conclusion: The high consumption of riboflavin, thiamin, and vitamin C was associated with the low prevalence of depressive disorder in Korean adults.
