Process Development for Production of Functional material from Citrus Madurensis(Citrofortunella microcarpa) Using Innovative Extraction by Focused Ultrasound(INEFU). (깔라몬딘오렌지로부터 집속초음파추출을 이용한 기능성 물질 생산 공정 최적화)

  • 집속초음파 추줄법(Innovative Extraction by Focused Ultrasound, INEFU)은 기존의 추출법에 비교하여 식물세포벽으로부터 기능성 성분을 낮은 온도에서 높은 추출 효율을 얻기 위한 추출공정으로 적용하기 위하여 연구되고 있다. 본 연구에서는 깔라몬딘오렌지(깔라만시, Citrus Madurensis, Citrofortunella microcarpa)에서 Vitamin C와 Polyphenol의 기능성 성분 추출을 위해 INEFU를 적용하여 추출 효율에 미치는 요인을 평가하였다. 추출 변수의 최적화를 위해 추출 온도 및 추출 시간을 요인으로 하여 순차적인 최적화를 진행하였다. 동일한 추출조건에서 저온추출(CEM)과 INEFU를 비교하였을 때 INEFU에서 Vitamin C는 고순도로 26배 증가하여 추출되는 것을 확인하였다. 또한 Polyphenol는 저온추출(CEM)에서는 polyphenol이 극소량 추출되었으나, INEFU에서는 Feruric acid가 10.3배 증가하여 추출되는 것으로 확인하였다. 본 연구를 통해 INEFU가 기존의 추출 공정에 비해 기능성 성분 생산에 보다 효과적임을 확인하였다.

  • Nanoparticles are substances that are smaller in size and smaller than cells that make up the skin. Therefore, they are very suitable as mediators for transmitting drugs or genes across cell membranes, and also deliver specific ingredients into the skin.In this study, nanoparticles were extracted from mugwort and particles of around 100 nm were obtained through dynamic light scattering (DLS), and the results of concentration-dependent enhancement of cell viability in fibroblasts were obtained through MTT assay. In addition, it was confirmed that the COL1A1 mRNA expression level was increased and the IL-6 mRNA expression level was decreased through the quantitative real-time PCR analysis method. Moreover, as these nanoparticles were confirmed to be stable, they can be applied not only to cell experiments but also to cosmetic formulations. While the demand for plant-derived ingredients continues to increase, excluding chemical ingredients from the recent cosmetics industry trend, there is a limitation in that there are few research results suggesting the application field of plant-derived nanoparticles. Therefore, in order to overcome the limitations of the cosmetic industry at the present time, the results obtained in this study present nanoparticles derived from Artemisia princeps (NDAP) as a highly functional cosmetic material.

Antioxidant, Antimicrobial and Anti-inflammatory of Mixed Medicinal Herb Extract (한약재 복합 추출물의 항산화, 항균 및 항염 효과)

  • Objectives : This paper aimed to verify the applicability of mixed extract ofAngelica gigasNakai,Cnidium officinaleMakino,Paeoniala ctifloraPall,Rechmannia glutinosaLibosch,Scutellaria baicalensisGeorgi, which were prescribed for improving inflammation in Donguibogam, as the materials for beauty food and functional medicinal herb cosmetics by manufacturing such mixed extract and evaluating the biological activity of the extract.Methods : The mixed medicinal herb water extract(MMW) and ethanol extract(MME) were freeze-dried to be used as the specimen. We performed electron donating ability, lipid acidification inhibitory activity, anti-inflammatory activity against skin flora, MTT assay, NO inhibitory activity and the protein expression inhibitory activity of iNOS and COX-2.Results : For anti-oxidation experimentation, the electron donating abilities of MMW and MME were above 60.0% and 90.0% at 500 μg/ml, respectively. In the inhibition rate of lipid peroxidation, MMW and MME showed 43.1% and 52.1% at 1,000 μg/ml, respectively. As a result of antimicrobial activity, both the MMW and MME showed significant clear zones forPropionibacterium acnesat 4 mg/disc, but did not indicated the clearzones forStaphylococcus aureus, Escherichia coliandStaphylococcus epidermidis. Anti-inflammatory activity by NO assay showed LPS-induced NO was significantly inhibited in a concentration-dependent manner. Also, the expression of iNOS and COX-2 proteins were significantly inhibited following treatment with MMW and MME of 50 μg/ml.Conclusions : Accordingly, it can be concluded that mixed medicinal herb extract has the potential to beused as a functional food and cosmetic material.

A Study on Whitening and Anti-inflammatory Effects of Eriobotrya Japonica Leaf Extracts with Different Extraction Methods (추출 방법에 따른 비파엽 추출물의 미백 및 항염활성에 관한 연구)

  • In this study, we prepared Eriobotrya japonica leaf extracts by several extraction processes and then evaluated their biological activities for their potential application as a new raw material of functional cosmetic. Their whitening effects were measured by tyrosinase inhibitory activities, and anti-inflammatory effects were determined by inhibitory activities of nitric oxide (NO) production. Among the several extracts obtained from E. japonica leaf, supercritical fluid extract showed tyrosinase inhibitory activities at the concentration of 10%. Inhibitory activity on NO production effect related to anti-inflammatory efficacy was in the order: supercritical fluid extract > ethanol extract > hot water extract. According to the results of MTT assay, cell cytotoxicity was not observed at all concentrations except for a 5% concentration of the 70% ethanol extract. For whitening effects, 30% ethanol and 70% ethanol extract showed mushroom tyrosinase inhibitory activity at the concentration of 5%. These results indicated that E. japonica leaf extracts could have the functional effects when they are added as ingredients in cosmetics.

Subjective Skin Improvement and Safety of the Spicules-containing Cosmetics (스피큘을 함유한 화장품의 피부안전성 및 주관적 피부개선감 연구)

  • This study has purposes : evaluating how safe the skin is when applied with the Spicules-containing cosmetic, and whether Diamond-spicules is permeated into the skin and showed the efficacy of the active ingredients well. For these aims, women in their 30s and 60s were surveyed for skin irritation and changes after applying cosmetics to their skin for four weeks. As a result, Spicules-containing cosmetics were safer on the skin than general functional cosmetics, and in particular, in the case of cosmetics containing Diamond-spicules, statistically significant results (p<.001) were obtained in improvement of anti-wrinkle, firmness, moisturizing, whitening, alleviation of inflammation, and sensitivity. Thus, It has been proved Diamond-spicules containing functional active ingredients, not only as transdermal delivery system, but also can be used as multifunctional cosmetic material.

Study on Effect of Human Hair by Deer Antler Fermented Keratin Peptide (녹용 발효 케라틴 펩타이드에 의한 인체 모발 연구)

  • In this study, keratin peptides were produced through high-temperature anaerobic fermentation of keratin, a protein contained in deer antler, with Fervidobacterium islandicum AW-1, and factors related to human hair, confirming the possibility of keratin peptides as cosmetic ingredients. As a result of the cytotoxicity and proliferation of deer antler fermented keratin peptide according to the concentration in the dermal papilla cell line, cytotoxicity was not observed and the cell proliferation effect was shown. For human dermal papilla cells, statistically significant increasing in growth factors according to the deer antler fermented keratin peptide was determined possiblity of effects on hair growth. Cosmetic products containing deer antler fermented keratin peptides were manufactured and skin safety and anti hair loss efficacy clinical tests were conducted. As a result, after 12 weeks of use, the total number of hairs statistically significant increased compared to before using the product and the difference in total number of hairs compared to the control group was found. In conclusion, we suggest that the possibility of fermented deer antler keratin peptide as a cosmeceutical ingredient as well as a health functional food material was confirmed.

Antioxidant Activity and Whitening activity of Psidium guajava leaf extract (구아바 잎 추출물의 항산화 및 미백 활성 효과)

  • The purpose of this study was to investigate the applicability of the Psidium guajava leaf extract as a whitening functional cosmetic material. We measured DPPH radical scavenging activity, intracellular ROS, cytotoxicity in B16F10 melanoma cells and cytoprotective effect on ultraviolet A, in vitro tyrosinase inhibitory effect and melanin biosynthesis inhibitory effect. The antioxidative effect was confirmed through high DPPH radical scavenging activity and intracellular ROS activity inhibition measurement of the Psidium guajava leaf extract. The survival rate of B16F10 melanoma cells was more than 98% at all concentrations, and the cytoprotective effect from ultraviolet ray A was found to increase in a concentration-dependent manner. In addition, in vitro tyrosinase activity inhibitory effect of 10% and melanin biosynthesis inhibitory effect of 20% were observed. Through less toxicity for B16F10 melanoma cell, high antioxidant activity, inhibition of tyrosinase activity and melanin biosynthesis inhibitory effect, we confirmed the possibility of developing the Psidium guajava leaf extract as a whitening functional cosmetic material with a safe and excellent whitening effect.

Anti-wrinkle efficacy of isoquercitrin isolated from Nymphoides indica (어리연꽃에서 분리된 Isoquercitrin의 주름개선 효능 연구)

  • In this study, we evaluated the possibility of wrinkle-reducing functional cosmetic material of isoquercitrin (IQC) isolated from Nymphoides indica (NP) by measuring the efficacy of ROS inhibition, procollagen synthesis induction, and matrix metalloproteinase (MMP)-1 inhibition. As a result, ROS tended to be inhibited in a concentration dependent manner, and the procollagen protein synthesis rate was increased by 70% ($5{\mu}g/mL$) and the MMP-1 protein was decreased by 41% ($5{\mu}g/mL$) compared with the control. This was suggested the ability of IQC to induce the procollagen synthesis and to reduce the ROS and MMP-1 production. Therefore, it is considered that IQC isolated from NP has enough value to be a functional cosmetic material.

Analysis on Antioxidant Activity and Agronomic Characteristics of Extract from Smilacis Chinae Radix

  • The Smilacis chinae Radix refers to the root of Smilax chinae L distributed in mountain and filed of Korea, and it is a vine shrub in the Lilaceae family, called Berchemia berchemiaefolia, and is referred to as Smilacis chinae Radix in it's a natural medicine name. Antibacterial, inflammatory, and antioxidant activity were studied in Smilacis chinae Radix. In this study, biological activities such as antioxidant (DPPH, ABTs, TPC), cytotoxicity, wrinkle improvement, and whitening improvement to increase the utilization value of Smilacis chinae Radix and identify the botanical value. Therefore, we tried to explore the applicability of Smilacis chinae Radix as a functional cosmetic material. Smilacis chinae Radix (SCR) was dried and extracted with ethanol. In order to measure the biological activity of the SCR, antioxidant activity, inhibition activities of collagenase, tyrosinase and cell viability were measured. The DPPH (1,1-diphenyl-2-picryl hydrazyl) radical scavenging activity in the extract with a concentration of 400㎍/mL is 91.22% ± 0.41%%. ABTs (2,2'-azinobis-3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity in the extract with a concentration of 400㎍/mL is 99.60% ± 0.03%. Total polyphenol contents (TPC) are 0.203 ± 0.05 mg GAE/mg Ext when SCR was lmg/mL. And the Cell viability for HaCaT derived human keratinocyte and Raw264.7, a mouse-derived macrophage was determined using the MTT assay. When cell was treated with 100㎍/mL of SCR, HaCaT cell showed cell viability of 78.09 ± 0.1% and Raw264.7 cell showed cell viability of 91.88 ± 0.42%. From the above results, we have shown the possibility that the CSR have antioxidant ability, inhibition activity of collagenase and tyrosinase and cell safety ability which can be useful in a functional cosmetic material.

Anti-inflammation and Anti-aging Effect of Lactic Acid Bacteria Fermented Lycium chinense Extracts (젖산균 발효 구기자(Lycium chinense ) 추출물의 항염증 및 항노화 효능 연구)

  • In this study, fermented Lycium chinense (L. chinense) extract was used to analyze the efficacy of nitric oxide (NO) and anti-aging inhibition to verify its usefulness as a functional cosmetic material. L. chinense was extracted with hydrothermal and 70% ethanol as a solvent, fermented, concentrated, and freeze-dried to prepare a sample, followed by MTT assay, NO inhibition assay, western blot assay, and UPLC analysis.As a result of the analysis of NO inhibitory efficacy, fermented L. chinense water extract (FLW) and fermented L. chinense 70% ethanol extract (FLE) showed excellent NO inhibitory efficacy of 47.96% and 56.71%, respectively. As a result of measuring the expression patterns of the wrinkle-related proteins MMP-1, and TRPV-1 through Western blot, both FLW and FLE confirmed the inhibitory efficacy of MMP-1 and TRPV-1. Based on the results of the experiment, the fermented L. chinense extract is expected to have a high application value as a functional cosmetic material related anti-inflammatory, and anti-aging.
