The Changes in the Dietary Pattern and Health and Nutritional status of Korean During the last one Century (지난 일세기 동안의 한국인 식습관의 변화와 보건영양상태의 추이 분석)

  • The changes in the dietary pattern of Koreans during the last one century and its consequences are summarized as follows; 1. Until the beginning of 20th century, Koreans used variety of cereals, vegetables and fruits for their staple food, but the variety has been largely reduced by the agricultural development and urbanized life style. 2. The well balanced traditional dietary pattern of Korean had been deeply deteriorated by the food shortage during the Japanese occupation and Korean war. 3. The deteriorated nutritional condition of Korean was not remedied by the restoration of traditional dietary pattern, but attempted to overcome it by the adoption of Western food habit. 4. The people were trained to eat milk and flour-meals during the starvation of Korean war, and it was continued after Korean war through the animal husbandry promotion policy. 5. The importation of food and feed cereals has been increased rapidly during the economic growth in 1970's and the food self-sufficiency droped below 50%. 6. In 1970's, the food supply pattern of Korean was restored to the level of early 1900, but the consumption of lipid increased extraordinarily. 7. The overconsumption of animal food and lipid continues in 1980's, and it coincides with the rapid increase in the occurence of food related degenerative deseases. 8. The establishment of Korean dietary goal which is based on the traditional dietary pattern is needed.

The Roles of Dietary Polyphenols in Brain Neuromodulation (뇌 신경조절에서의 식이 폴리페놀 화합물의 역할)

  • Over recent years, it has become evident that the central nervous system bidirectionally interacts with the gastrointestinal tract along the gut-brain axis. A series of preclinical studies indicate that the gut microbiota can modulate central nervous system function through a multitude of physiological functions. Polyphenols are ubiquitous plant chemicals included in foods such as fruits, vegetables, tea, coffee and wine, and their consumption is directly responsible for beneficial health effects due to antioxidant, anti-inflammatory, antimicrobial, immunomodulatory, anticancer, vasodilating, and prebiotic-like effects. There is increasing evidence that dietary polyphenol can contribute to beneficial effects in neuronal protection acting against oxidative stress and inflammatory injury as well as in cognitive functions. In this paper, we overview the neuroprotective role of dietary polyphenols especially focusing on the neuroinflammation and neurovascular function by interaction with the gut microbiome. Polyphenol metabolites could directly act as neurotransmitters crossing the blood-brain barrier and modulating the cerebrovascular system or indirectly modulating gut microbiota. In addition, evidence suggests that dietary polyphenols are effective in preventing and managing neurological disorders, such as age-related cognitive decline and neurodegeneration, through a multitude of physiological functions. Dietary polyphenols are increasingly envisaged as a potential nutraceuticals in the prevention and treatment of neurological disorders, because they possess the ability to reduce neuroinflammation, to improve memory and cognitive function and to modulate the gut microbiota.

Inhibitory Effect of Grapefruit Seed Extract Mixture on the Physiological Function of Botrytis cinerea (식물성 항균소재 처리가 Botrytis cinerea의 생리기능에 미치는 저해효과)

  • Botanical antimicrobial agent-grapefruit seed extract mixture (BAAG) have an unknown compounds which exhibit the antibiotic activities aganist microorganisms including bacteria and fungi. We have examined the effects of BAAG on the physiological function of Botrytis cinerea which was isolated from necrotic lesions of decayed fruits and vegetables such as cucumbers, grapes, tomatoes, and red peppers during storage. In the results of enzymatic activities related to the energetic metabolism there was no inhibitory effect of BAAG on the activities of several enzymes in vitro including glucose 6-phosphate dehydrogenase and malate dehydrogenase, while there was inhibitory effect of BAAG on the activities of hexokinase and succinate dehydrogenase. O-nitrophenyl-$\beta$-D-galactopyranoside(ONPG), the artificial substrate of $\beta$-galactosidase was hydrolyzed in the presence of BAAG, indicating that the membrane was pertubated by the BAAG. From the results we suggested that the antibiotic activity of BAAG is due to the change of membrane permeability of the cell. BAAG was fractionated and purified by silica gel and sephadex column chromatography. Among active fractions two peaks were identified as naringin and limonin when they were analyzed by by NMR and Fast atomic bombardment.

Application of ATP Bioluminescence Assay for Measurement of Microbial Contamination in Fresh-cut Produce Processing Lines (신선편이 농식품 생산라인의 환경미생물 오염도 측정을 위한 ATP 검사법의 이용)

  • With the rapid growth of fresh-cut produce market, the South Korean fresh-cut industry is facing the challenge of ensuring food safety. As the estimation of the microbial numbers in fresh-cut produce processing lines (tools, and equipment) using the conventional microbiological techniques takes days, so there is a need for faster and easier monitoring methods. This study was conducted to investigate the use of ATP bioluminescence assay to measure the degree of microbial contamination from three actual fresh-cut processing lines. The samples collected from frech-cut vegetables, and fresh-cut fruits processing plants were tested for the estimation of the bacterial number, using the ATP bioluminescence and microbiological methods. The result of former was transferred to log RLU/100 $cm^2$, and that of the latter was transferred to log CFU/100 $cm^2$. A positive linear correlation between the ATP bioluminescence assay value and aerobic-plate count was found for fresh-cut processing lines, with a correlation coefficient of 0.8772 (n=50). The results of this study indicate that ATP bioluminescence assay can be used to monitor microbial contamination in fresh-cut produce processing plants, and can help improve the hygiene therein.

β-Sitosterol Induced Growth Inhibition is Associated with Up-regulation of Cdk Inhibitor p21WAF1/CIP1 in Human Colon Cancer Cells (β-Sitosterol에 의한 인체 대장암 HCT116 세포의 증식억제에 관한 연구)

  • $\beta$-Sitosterol is the major phytosterol in higher plants, including fruits and vegetables. The molecule has been shown to have the potential for prevention and therapy for human cancer. We investigated the effects of $\beta$-sitosterol on the cell proliferation of HCT116 human colon cancer cells in order to understand its anti-proliferative mechanism. $\beta$-Sitosterol treatment resulted in the inhibition of cell proliferation in a concentration-dependent manner. The anti-proliferative effect of HCT116 cells by $\beta$-sitosterol was associated with formation of apoptotic bodies and degradation of $\beta$-catenin protein. In addition, $\beta$-sitosterol-treatment induced a marked accumulation of tumor suppressor p53 and a concomitant induction of cyclin-dependent kinase (Cdk) inhibitor p21 without alteration in the levels of cyclins and Cdks. Taken together, these findings provide important new insights into the possible molecular mechanisms of the anti-cancer activity of $\beta$-sitosterol.

Isolation and Identification of Bacteriocin-Producing Lactic Acid Bacteria (유용 박테리오신을 생산하는 유산균의 분리와 동정)

  • Lactic acid bacteria are microorganisms that are closely associated with human and/or animal environments, and are categorized as generally recognized as safe (GRAS) organisms due to their ubiquitous appearance in foods and their contribution to the healthy microflora of mucosal surfaces. This study was performed to isolate and identify lactic acid bacteria with antagonistic effects against food-borne pathogens. A total of 3,000 acid-producing bacteria were isolated from infant feces, cattle feces, goat feces, dog feces, pig feces, vaginal tracts, vegetables, fruits, Kimchi, Jeotgal, fermented sausages, raw milk, cheese, yogurt, Cheonggukjang, Meju, and Makgeolli cultured on MRS agar with 0.05% bromocresol purple. For the isolation of bacteriocin-producing bacteria, the diameter of the clear zone was measured on MRS agar plates. Twenty-six isolates exhibited strong antibacterial activity against indicator strains such as Listeria monocytogenes, Escherichia coli O157:H7, and Salmonella enterica serovar Enteritidis. Lactic acid bacteria were identified as Enterococcus faecalis, Enterococcus faecium, Enterococcus hirae, Lactobacillus acidophilus, Lactobacillus amylovorus, Lactobacillus curvatus, Lactobacillus plantarum, and Pediococcus acidilactici by 16S rDNA gene sequence analysis. The results of this study suggest that the isolates could be used as potential probiotic starters for functional food applications.

The Effects of Perceived Stress on Dietary Habits and Oral Health Behaviors in Korean Adolescents (우리나라 청소년의 스트레스 인지수준이 식습관 및 구강건강행태에 미치는 영향)

  • The purpose of this study was to analyze the effects of perceived stress on dietary habits and oral health behaviors in Korean adolescents. Data were from the Korea Youth Risk Behavior Web-based Survey (KYRBS) in 2011. A total of 74,186 adolescents were surveyed using the self-administered questionnaire. Logistic regression analyses were performed to elucidate the effects of perceived stress on dietary habits and oral health behaviors. Statistical analyses were conducted, and p<0.05 were considered significant. We found that adolescents with higher stress were more likely to increase the risk of adverse dietary habits (low consumption of fruits, vegetables and milk, high consumption of carbonated soft drinks, fast food and cookie) compared to those with lower stress. Regarding the relationship between perceived stress and oral health behaviors, adolescents with higher stress were less likely to practice oral health behaviors such as brushing teeth after snack consumption and brushing teeth before sleep compared to those with lower stress. This result indicates that adolescents' perceived stress might play a significant role in the negative dietary habits and oral health behavior.

Multiresidue Analysis of 240 Pesticides in Apple and Lettuce by QuEChERS Sample Preparation and HPLC-MS/MS Analysis (QuEChERS 전처리법과 HPLC-MS/MS 기기분석을 이용한 사과와 상추 중 240종 농약의 동시분석)

  • The study tested QuEChERS (quick, easy, cheap, effective, rugged and safe) sample preparation and HPLC-MS/ MS analysis for measurement of pesticide residues in fruit and vegetable. 240 kinds of pesticides spiked at three levels of 90, 45, 9 ng/g in lettuce and apple. For QuEChERS sample preparation, graphitized carbon black (GCB) was used for only lettuce in dispersive-SPE as absorbent. Matrix-matched standard calibration was used for quantitative analysis of HPLC-MS/MS. 218 pesticides (91%) in apple and 207 pesticides (86%) in lettuce showed recoveries in the range of 70~120% with $RSD{\leq}20%$. The lowest calibrated level (LCL) were 4.5 ng/g for 192 pesticides, 9 ng/g for 42 pesticides, 45 ng/g for 3 pesticides and 3 pesticides were not detected at all concentration levels. The results showed that the QuEChERS sample preparation and HPLC-MS/MS analysis can be applied to multi-residue analysis of pesticides in vegetables and fruits.

Changes of Nutritional Compounds and Texture Characteristics of Peaches (Prunus persica L. Batsch) during Post-irradiation Storage at Different Temperature (감마선 조사와 저장온도에 따른 복숭아의 품질특성 변화)

  • The gamma irradiation process was performed to prolong a shelf-life of peaches and the effects of a gamma irradiation on the nutritional, physiochemical and sensory characteristics of peaches were evaluated during a post-irradiation storage at $4^{\circ}C$ and $25^{\circ}C$. Nutritional, physiochemical and sensory characteristics of peaches were stable at 1 kGy of an irradiation, a recommended dose for fruits and vegetables by CODEX, and the viable cell counts of contaminated microorganisms were reduced by 2 decimal reduction, at this dose. After a 1-week storage at ambient condition, the microbiological quality of the 1 kGy irradiated peach was stable, while all the non-irradiated peaches were deteriorated Furthermore, cold storage enhanced the shelf-life of the gamma irradiated peach and the nutritional and physiochemical characteristics of the peaches were comparatively stable up to 6 weeks. Right after gamma irradiation, the sensory evaluation results were not different in any of the samples, and the sensory quality of the irradiated peaches was adequate for a 4-week storage at $4^{\circ}C$. Our results suggested that gamma irradiation at 1 kGy can be used to enhance the shelf-life of peach without a significant loss in the quality attributes, especially upon cold storage after radiation treatment.

Protective Effects of Kaempferol and Quercetin on Oxidative Stress in CPAE Cell (CPAE 세포를 이용한 Kaempferol과 Quercetin의 산화스트레스 극복효과)

  • Flavonoids are ubiquitous substances in fruits and vegetables. A main subgroup of the flavonoids are the flavonols, of which quercetin and kaempferol are the major representatives in foods. They are used in food supplements at high doses, because of their preventive effects on degenerative diseases. The aim of this study was to determine the combined and separate effects of kaempferol and quercetin on oxidative stress in cow pulmonary artery endothelium (CPAE) cells over a broad concentration range. The results demonstrate that cell viability was greatly increased in kaempferol and quercetin treated cells whether $H_{2}O_{2}$-treated or not. Cell viability also increased when treated with flavonols in the absence of oxidative stress. Both preincubation and simultaneous incubation with kaempferol and quercetin protected against the loss of cell viability induced by 1mM $H_{2}O_{2}(5h)$. Protective effects of flavonols against oxidative stress were shown to depend on the treated flavonol concentrations. No protective effect was shown under low concentration treatment and cell viability increased 1.6 times at $200{\mu}M$ relative to the control group. At the highest flavonol concentration of $300{\mu}M$, the increased cell viability by flavonol treatment was decreased to almost half of the maximum values. Combined treatments with kaempferol and quercetin showed more protective effects against oxidative stress by $H_{2}O_{2}$ than the separate effects of each flavonol. In conclusion, the protective effect of kaempferol and quercetin against oxidative stress was very pronounced but high concentrations of flavonols can also induce cell cytotoxicity.
