Effects of Cryoprotectants and Equilibration Time on the Viability of Frozen-thawed Porcine Oocytes (동결-융해된 돼지난포란의 생존성에 대한 항동해제와 평형시간의 영향)

  • This study was undertaken in an effort to develop a cryopreservation system of immature and mature porcine oocytes. For this aim, the experiments were designed to examine the effect of cryoprotectants and equdibration time on the viability of frozen-thawed oocytes by using trypan blue(TB) and fluorescene diacetate(FDA) test. The viability of frozen immature oocytes evaluated by TB test was slightly higher than that of frozen mature oocytes. The viability(25.O%) after IVM of frozen-thawed immature oocytes greatly decreased that(42.9%) of oocytes just after thawing, but it was higher than frozen-thawed mature oocytes(15.8%). When immature oocytes were equilibrated for 10, 20 and 30 minutes before freezing the oocyte viability was 20.0, 31.3 and 42.9%, respectively. There was a tendency for long equilibration before oocyte freezing to be more effective for the immature oocytes and a short equilibration time for mature oocytes. Although there was no difference in viability index of frozen oocytes hetween the viability test methods, the index of TB test was slightly higher than that of FDA test. The viability(FDA test) of frozen-immature oocytes with 3 different crtoprotectants was 22.2% for propylene glycol(PG), 9.3% for polyehtylene glycol(PEG) and 65.6% for PG+PEG, in which PG+PEG was more protective against freezing effect.

  • A need still exists to evaluate the durability performance of cemented gedmaterials in road constructions using various cementing binders due to seasonal changes in fields as well as structural performance. In this paper, durability characteristics of stabilized granular base and subgrade materials, which is widely used in korea, was assessed using a laboratory repeated load resilient-deformation test under various freezing-thawing and wet-drying cyclic conditions. In addition, various resilient modulus models were adopted based on the test results. As a result, the estimated model coefficients agree well with the values from the literatures.

The Evaluation of Surface Scaling and Resistance of Concrete to Frost Deterioration with Freezing-Thawing Action by Salt Water (염화물이 함유된 동결수의 동결융해 작용에 따른 콘크리트의 내동해성과 표면열화 평가)

  • This study presents the experimental results of frost durability including resistance to freezing-thawing and surface scaling of concrete. Mixing design was proportioned with the various water-binder ratio between 0.37 and 0.47 and three different binder compositions corresponding to Type I cement without any supplementary cementitious materials(OPC), Type II cement with 50% blast-furnace slag replacement(BFS50), and ternary cement with Type III cement, 15% fly ash, and 35% slag replacement (BFS35%+FA15%). Test results showed that the mixing design with BFS50% and BFS35%+FA15% exhibited higher durability factor than that made with OPC only. Finally, the use of blend cement containing slag can be used effectively in terms of frost durability of the concrete exposed to severe condition under coastal environment like as flying salt, sea water spray, etc.

  • The purpose of this study was to evaluate the effects of ground granulated blast-furnace slag on strength development and durability of ordinary portland cement concrete (OPC) with steam curing types. Main experimental variables were slag contents(0%, 10%, 30%, 50%, 70%) and curing types (standard, accelerated curing). It were performed to check the basic properties of concretes that compressive strength, rapid chloride ion permeability and chemical resistance. From the result, we have found that increasing the amount of blast-furnace slag produced concrete with increased compressive strength and permeability resistance. Rapid freezing-thawing test showed that they were good enough to protect the concrete structures and to carry out cyclic freezing and thawing. The freeze-thaw resistance of blast-furnace slag produced concretes maintained above 90% of relative dynamic modulus after 300 freezing-thawing cycles. Increasing the amount of blast-furnace slag produced concretes with increased chemical resistance.

A Study on the Effect of Sample Storage Condition on the RIA Results of Plasma renin activity Test (Plasma renin activity 검사의 검체 보관 방법이 방사면역 측정법 결과에 미치는 영향에 대한 고찰)

  • Purpose Plasma renin activity (PRA) test is important for the diagnosis of primary aldosteronism. PRA is an easily deformed substance in vitro and affected by temperature changes. Laboratory of ASAN medical center has consistently found that there was a difference between the initial and re-experimental results. We compared and analyzed the differences in PRA test results according to the sample storage status. Materials and Methods The measurement of PRA was performed by using the radioimmunoassay. From August to September 2020, 43 PRA re-test samples were tested with different sample storage condition. The first group was re-examined by freezing the plasma-separated samples at -18℃, and the second group was re-examined with refrigerated EDTA sample. Also, additional tests were conducted on 13 PRA samples to verify the effect on thawing temperature differences in plasma-separated samples. The same samples were divided into two parts and stored frozen at -18℃, respectively, and thawing samples in room temperature and those in refrigerator were were conducted. Each result was compared and analyzed based on the initial experimental results. Results The results of re-examination after frozen storing plasma separation samples showed a lower correlation than the results of re-examination with EDTA plasma samples in refrigerator. When calculating the percentage based on the initial test results, the average percentage of each was 404.9% and 133.8%. The correlation coefficient was also R=0.8501 and R=0.9966, respectively, showing a higher correlation between plasma in the refrigerated sample EDTA tube. In comparison experiments with differences in thawing temperature, average percentage of the results of initial test and room temperature thawing was 94.3% and the average percentage of the results of refrigerated thawing was 88.0%. After again freezing the sample, the average percentage of the second room temperature thawing result is 107.5%, and the second refrigerated thawing group is 112.7%. Both groups showed an increase from first thawing. Conclusion A comparative analysis of retesting according to differences in sample storage methods in PRA tests showed a higher correlation between the results of retesting of the refrigerated EDTA plasma. And repeated freezing and melting of plasma separation samples, regardless of temperature during defrosting, has been shown to affect results. Therefore, retest of PRA should re-collect plasma from original EDTA plasma to increase reproducibility.

Evaluation of Bond Strength in FRP Hybrid Bar Affected by Freezing/thawing Test and UV Rays (동결융해 및 UV 폭로시험을 거친 FRP Hybrid Bar의 인발거동특성 평가)

  • FRP Hybrid Bar, composed of an embedded steel and the coated composites with epoxy and glass fiber, is an effective construction material with tension-hardening performance and lightweight. The epoxy exposed to UV(Ultra Violet Rays) and FT(Freezing and Thawing) action easily shows a surface deterioration, which can cause degradation of bonding strength between inside-steel and outside-concrete. In the present work, surface inspection for 3 different samples of normal steel, FRP Hybrid Bar before UV, and FRP Hybrid Bar after UV test was performed, then concrete samples with 3 reinforcement types were prepared for accelerated FT test. Through visual inspection on 3 typed reinforcement, no significant deterioration like chalking was evaluated. The results from FT test to 120 and 180 cycles showed FRP Hybrid Bar exposed to UV test has higher bonding strength than normal steel by 106.3% due to enlarged bond area by silica coating. The 3 cases showed a similar bond strength tendency with increasing FT cycles, however a relatively big deviations of bond strength were evaluated in FRP Hybrid Bar after UV test due to loss of silica coating.

Influence of Osmolality of Complete Semen Extender on Motion Characteristics of Frozen-thawed Ram Spermatozoa

  • The present study was conducted to observe the effect of osmolality of glycerolated TEST-yolk glycerol extenders on post-thawing sperm kinematics of ram spermatozoa of the native Malpura breed maintained in a semi-arid tropical environment. Good quality semen obtained from adult rams was pooled, split and diluted to 1,000 million spermatozoa per ml in complete TEST-yolk-glycerol extenders of 900, 1,200, 1,500 and 1,800 mOsm/kg osmolality. Diluted semen samples were loaded in 0.25 ml straws and cooled down to $-125^{\circ}C$ freezing temperature at the rate of $-25^{\circ}C$ per minute under controlled conditions before plunging into liquid nitrogen for storage. The thawing of straws was performed at $50^{\circ}C$ in a water bath for 10 seconds and sperm kinematics of the frozen-thawed spermatozoa were assessed by a computer-assisted sperm analysis technique. Osmolality of diluent had no significant effect on post-thawing % motility, % rapid, % medium and % slow moving frozen-thawed spermatozoa but significantly (p< 0.05) affected the % linearity and % straightness. The post-thawing % motility and % rapid motile spermatozoa were highest in samples extended in diluent of 1,500 mOsm/kg osmolality and lowest in 900 mOsm/kg. The curvilinear velocity of spermatozoa was significantly (p<0.05) higher for samples extended in 1,800 mOsm/kg, compared to those in 900 and 1,200 mOsm/kg, but the effect was not significantly different to those extended in diluent of 1,500 mOsm/kg osmolality. The study indicated that ram spermatozoa could tolerate a wide osmolality range for dilution in the complete TEST-yolk-glycerol extender for their cryosurvival. The highest recovery of motile spermatozoa following thawing was achieved in samples extended in the TEST-yolk-glycerol diluent of 1,500 mOsm/kg osmolality.

  • Effects of the glycerol addition and removal medium containg 10% sucrose on the mouse embryo survival after freezing in a liquid introgen container were determined using the FDA test. The summarized results are the following. 1. The FDA score was higher (P<0.05) when embryos were frozen in the glycerol addition medium with sucrose that the one without it(3.4 vs 3.0). 2. No difference in the score was found between the glycerol removal medium containing 10% sucrose(3.2) and PBS+10% sucrose(3.3). 3. The score was higher(P<0.01) at morular stage than blastocyst stage of embryos.

  • This study was experimented on accelerated weathering test using salt and freeze-thaw to prove effects of consolidants and consolidation for stone cultural heritage. The samples used four kinds of stones (Gyeongju Namsan Granite, Iksan Granite, Yeongyang Sandstone and Jeongseon Marble) which to distributed by three type of weathering grade (Fresh, Weathered Stone and Highly Weathered Stone) added for thermal treatment. The samples were treated with three consolidants (Wacker OH 100, Remmers KSE 300 and 1T1G), and tested by 500 cycles with freezing-thawing and 50 cycles of salt weathering test. As a results of freezing-thawing test, the crack and destruction occurred from some samples. And total immersed samples maintained effect of consolidation to 200 cycles. Also, The rock particle was fall off and gradually destructed by salts weathering test. The consolidated sample relatively had fewer changes by the weathering than not treated sample. The sprayed sample had not continuous effect on weathering.

Evaluation of Tension Behavior in FRP Hybrid Bar Affected by UV Exposure and Freezing/Thawing Tests (UV 폭로 및 동결융해 시험을 거친 FRP Hybrid Bar의 인장거동 평가)

  • The present work is for an evaluation of tension behavior and surface deterioration of FRP Hybrid Bar due to UV exposure and freezing/thawing(F/T) actions. For the work, FRP Hybrid Bar is subjected to UV exposure test, then F/T test is performed successively to 180 cycles. In FRP Hybrid Bar, no significant surface deterioration is evaluated after UV exposure. Tension hardening performance, a unique engineering advantage of FRP Hybrid Bar, is still maintained after F/T test. The performance in FRP Hybrid Bar exposed to UV is still effective. FRP Hybrid Bar exposed to UV have almost similar tension behavior of FRP Hybrid Bar without UV exposure. Although F/T cycles increase to 180, steel rebar, FRP Hybrid Bar, and FRP Hybrid Bar exposed to UV show no significant changes in tension behavior. In the work, UV exposure and F/T actions are evaluated to have little negative effect on surface deterioration and tensile performance in FRP Hybrid Bar, however spalling of silica coating due to impact should be considered since it affects bonding strength to outer concrete.
