Callus Induction and Plant Regeneration from Mature Seeds of Timothy (티모시 성숙종자로부터 캘러스 유도 및 식물체 재분화)

  • Timothy (Phleum pratense L.) is an important grass species as forage. In order to optimize tissue culture conditions of timothy, the effects of plant growth regulators on callus induction and plant regeneration was investigated with mature seeds of colt cultivar. The optimal concentration of 2,4-D for the induction of primary callus from mature seeds was 3 mg/L. The highest embryogenic callus frequenc (25%) was observed when the mature seed were cultured on MS medium supplemented with 3 mg/L 2,4-D and 0.1 mg/L BA. The highest plant regeneration frequency was observed when type B callus was transferred to N6 medium supplemented with 1 mg/L 2,4-D and 3 mg/L BA. Regenerated plants were grown normally when shoots were transplanted to the soil. A short tissue culture period and regeneration system would be beneficial for molecular breeding of timothy by the production of transgenic plant.

Effects of Seed Mixture and Nitrogen Levels on Botanical Composition and Forage Productivity for Pasture in Jeju (제주지역 초지에서 혼파조합 및 질소수준이 식생구성 및 사초생산성에 미치는 영향)

  • A study was conducted to determine the effects of seed mixture and nitrogen application levels on botanical composition and forage productivity in grazing pasture. Legume forage have the ability to take nitrogen from the atmosphere and convert it into a form usable by plants. Including legumes in mixtures with grass lowers the amount of nitrogen fertilizer required to produce forages. Dry matter (DM) yield of grass-legume mixtures was more than that of grass mono-cultivated and grass+white clover+red clover mixtures was the highest as 17,391 kg/ha in legume mixture (P<0.01). The highest DM yield was in N-200 kg/ha, but it was similar between N-100 kg/ha(15,128) and N-200 kg/ha (16,017). The large decline in the proportion of grass during April to June in 2004 probably was due to the drought and summer depression in grass-legume mixtures. Grass proportions decreased by 8.2% after 2 year, and white clover was dominated in grass-white clover mixture by May 2005. Crud protein (CP) content tends to increase with increased nitrogen application level.

Growth Characteristics and Productivity of New Hairy Vetch(Vicia villosa Roth) Early-Maturing "Cold Green" and Medium-Late Maturing "Cheong Pa" (헤어리베치 신품종 조생종 콜드그린과 중만생종 청파의 생육특성 및 수량성)

  • The objective of this research was to develop a high yielding, early and medium-late flowering new hairy vetch(Vicia villosa Roth) varieties derived from an accession in Korea. Most vigorous early and medium-late flowering hairy vetch lines were selected and crossed by open pollination and their seeds were bulked and plants were reselected at Seongju in the Keongbuk and at Sacheon in the Keongnam. A performance trial was conducted to evaluate agronomic characteristics, forage quality and dry matter(DM) yield of new hairy vetch early-maturing 'Cold green' and medium-late maturing variety 'Cheong pa' at Seongju and Sacheon. 'Cold green' and 'Cheong pa' are hair but 'Haymaker plus' and 'Latigo' are nearly hairless. 'Cold green' has purple flowers while 'Cheong pa' has violet. The cold tolerance of 'Cold green,' 'Cheong pa' and 'Latigo' was higher than 'Haymaker plus' at Seong-ju, Keongbuk province, in inland region but there were a little differences at Sacheon, Keongnam province in southern coast region. Fifty percent-flowering dates of 'Cold green' and 'Haymaker plus' had earlier than others and 'Cheong pa' was earlier than 'Latigo'. The DM yield of 'Cold green' was higher than that of others, but 'Cheong pa' and 'Haymaker plus' had higher than 'Latigo' at Seong-ju(P<0.05). In Sacheon, The DM yield of 'Cold green' and 'Haymaker plus' was higher than others and 'Cheong pa' had higher than 'Latigo'.

Glutathione Concentration as Affected by Sulfate Supply Level and its Relationship with Sulfate Uptake and Assimilatory Enzymes Activity in Rape Plants (유채에서 황 공급수준에 따른 글루타치온 함량의 변화가 황산염 흡수 및 동화관련 효소활력에 미치는 영향)

  • The glutathione (GSH) concentration in leaves of different maturities and roots of forage rape (Brassica napus L.) supplied with four levels of external $SO_4^{2-}$ (0, 0.1, 1.0 and 2.0 mM) supply were measured. The relationships of GSH concentration with $SO_4^{2-}$ uptake, ATP sulfurylase (ATPS) and O-acteylserine (thiol) lyase (OASTL) activity were also assessed. The $SO_4^{2-}$ uptake increased in parallel with the external $SO_4^{2-}$ supply, while protein concentration was not significantly changed. The ATPS activity increased continuously with decreasing $SO_4^{2-}$ supply from 2.0 to 0 mM, while the OASTL activity decreased significantly only at S-deficient conditions (0 and 0.1 mM). The GSH concentration in the young leaves, middle leaves and roots continuously increased (except for between 1.0 and 2.0 mM in the middle leaves and roots) as the external S supply was increased, but no significant changes occurred in the old leaves. The increased endogenous GSH concentration, affected by the $SO_4^{2-}$ supply level, was significantly related with the decrease in ATPS activity in both leaves and roots, and the decrease in OASTL activity only in leaves..

Agronomic Characteristics and Productivity of Introduced Common Vetch (Vicia sativa) Cultivars and Korean Wild Common Vetch in Kyeongbuk (경북에서 도입 콤먼베치 및 자생종의 생육특성과 수량)

  • This experiment was conducted to evaluate agronomic characteristics, forage quality, and dry matter (DM) yield of hairy vetch 'Cold green', common vetch 'Maxivesa', 'Wild common', 'Rasina', 'Morava', and 'Blanchefleur', at Seongju in Kyeongbuk from 2010 to 2012. The experiment was arranged in randomized complete block design with three replications. Vetchs were seeded on the 10th of Oct. 2010 and on the 3rd of Oct. 2011. Vetch plants were harvested on the 4th of May 2011 and on the 10th of May in 2012. 'Cold green' and 'Wild common' exhibited greater cold tolerance than other cultivars. Fifty percent-flowering of 'Cold green' occurred on the 22nd of Apr., whereas that of 'Wild common' occurred on the 26th of Apr. and those of 'Rasina' and 'Blanchefleur' occurred on 28th of Apr. and that of 'Maxivesa' occurred on 17th of May. The DM yield of 'Cold green' was higher (p<0.05) than all other cultivars, whereas the DM yield of 'Wild common' was higher (p<0.05) than those of the remaining 4 cultivars. According to the results obtained from this study, it is suggested 'Wild common' vetch is be recommendable for cultivation where increased DM production is sought.

Effect of Surface Sterilization Method on Agrobacterium-mediated Transformation of Field-grown Zoysiagrass Stolon (포장생육 잔디 포복경을 이용한 잔디 형질전환에 있어서 살균방법의 영향)

  • Zoysiagrass (Zoysia japonica Steud.) is an important forage and turfgrass that spreads by stolons and rhizomes. Zoysiagrass stolon can be used directly for Agrobacterium-mediated genetic transformation by exploiting the potential of direct shoot formation. However, surface sterilization of field-grown stolons is difficult and remains to be explored. We developed an effective surface sterilization and culture method using the stolon explant for infection with Agrobacterium tumefaciens. Among various treatments, sequential disinfection in 30% bleach for 15 min followed by 0.1% mercuric chloride for 25 min resulted in the highest number of clean stolons. The efficacy of mercuric chloride was increased under vacuum conditions by incubating at 800 mbar for 5 min. The inclusion of 2.5 mg/l amphotericin B further prevents fungal growth in in vitro cultures. This protocol would speed up the development of transgenic plants by utilizing field-grown stolon nodes.

SO42- Uptake and Assimilation in Forage Rape (Brassica napus) (유채의 SO42- 흡수 및 동화에 관한 연구)

  • To investigate the sulfate utilization efficiency that has been examined in rape (Brassica napus L.) cultivars (cv. Akela and Colosse). During 72h of treatment, in two cultivars, ${SO_4}^{2-}$ uptake was significantly higher in complete S condition (2.0 mM ${SO_4}^{2-}$) than that of the S-deficient condition (2.0 mM ${SO_4}^{2-}$). In young leaves, ATP sulfurylase activity increased after S-deficient treatment. However, in old leaves, ATP sulfurylase activity was not significantly changed in Akela. Glutathione concentration in young leaves significantly increased in all cultivars examined under S-deficient condition. The rate of increase in glutathigne concentration caused by S-deficiency treatment was higher in Akela. However, in old leaves, the glutathione concentration in Akela significantly decreased. The results suggest that ${SO_4}^{2-}$ uptake and ATP sulfurylase in rape plants were species specific, and that ${SO_4}^{2-}$ uptake was highly related to its assimilation into S containing compound such as glutathione.

Sulfur Deficiency Effects on Sulfate Uptake and Assimilatory Enzymes Activity in Rape Plants (유채에서 황 결핍이 황산염 흡수 및 동화관련 효소활력에 미치는 영향)

  • To determine $SO_4^{2-}$ uptake and its assimilation in response to the exogenous $SO_4^{2-}$supply level in forage rape (Brassica napus L.), the concentration of this element in plant tissues and the activity of ATP sulfurylasc and APS reductase was measured after 25 hours of treatment (1.0 mM $SO_4^{2-}$, control; 0.1 mM $SO_4^{2-}$, S deficiency; 0 mM $SO_4^{2-}$, S deprivation). $SO_4^{2-}$ uptake and the concentration in the plant tissues significantly decreased in S-deficient and S-deprived condition, while it maintained at nearly same level in the control. The activity of ATP sulfurylase tended to increase with decreasing the exogenous $SO_4^{2-}$ supply, while that of APS reductase to decrease. A significant change in both enzymes responding to S-deprivation treatment was observed only young and middle leaves. The results indicated that $SO_4^{2-}$ assimilation in young leaf tissues would be much more sensitively responded to S-limited nutrition.

Comparison of Forage Performance of Introduced Vetch Cultivars and Korean Wild Vetch (자생 베치와 도입 베치의 생육특성과 건물수량 비교)

  • A field experiment was conducted to evaluate agronomic characteristics, and dry matter yield of Korean wild hairy vetch(Vicia villosa Roth), Korean wild woolly pod vetch(Vicia villosa ssp. dasycarpa(Ten.) Eav.), introduced hairy and woolly pod vetch cultivars from Japan, and Haymaker plus(woolly pod vetch) from Australia. Seeds were sown on 7 October 2000 at the Livestock Experiment Fm, Keimyung College, Seongju, Kyeongbuk. Vetch Plants were harvested on 19 May 2001. Cold tolerance for woolly pod vetch from Japan and Haymaker plus were lower than other cultivars. Dry matter content of the Haymaker Plus was higher than other cultivars. Fifty percent flowering was observed 16 April for Haymaker plus, 20 April fur woolly pod vetch from Japan, 22 April fur Korean wild hairy vetch( I ), 28 April fer Korean wild hairy vetch(II), 1 May for Korean wild hairy vetch(III), 5 May fur Korean wild woolly pod vetch, and 10 May for hairy vetch from Japan. Dry matter yield per hectar was significantly highest(P<0.05) for wild hairy vetch(III) and wild hairy vetch(II) was higher(P<0.05) than other cultivars. Dry matter yield Per hectar was higher(P<0.05) for Haymaker Plus and wild hairy vetch(I) than hairy vetch from Japan.

Performance Trial for Developing High - Yielding and Early Maturing Hairy Vetch (Vicia villosa Roth) Cultivar under Double Cropping System (2모작 작부체계 적응 다수성 및 조숙성 헤어리베치 품종 선발을 위한 생산력 검정)

The experiment was arranged in randomized complete block design with three replications. Hairy vetchs were seeded on Sep. 26 and Oct. 11 in 2009 and 2010. Vetch plants were harvested on 30 Apr. in 2010 and 2011. Cold tolerance for Haymaker plus and Cappello were a little lower than other cultivars. Fifty percent-flowering dates of Cold green had earlier than others and Haymaker plus and Cappello were earlier than Cheong pa, Challenger, Hungvillosa and Latigo. Cheong pa and Challenger were ealirer than Hungvillosa and Latigo. The DM yield of Cold green was the highest (p<0.05) of the cultivar except Haymaker plus. Haymaker plus was higher (p<0.05) than others and Cappello, Cheong pa, Challenger were higher (p<0.05) than Hungvillosa and Latigo in 2010. In 2011, the DM yield of Cold green was the highest (p<0.05) of the cultivar and Haymaker plus was higher (p<0.05) than others. Cappello was higher (p<0.05) than Hungvillosa and Latigo. Cheong pa and Challenger were higher (p<0.05) than Latigo. Vetch cultivar was high in crude protein (CP) and low in Acid detergent fiber (ADF). According to the results obtained from this study, it is suggested that early-maturing cultivar would be recommendable for fall sown vetch for DM production.
