MonA is a subunit of a guanine nucleotide exchange factor that is important for vacuole passing and autophagy processes in eukaryotes. In this study, we characterized the function of MonA, an orthologue of Saccharomyces cerevisiae Mon1, in the model fungus Aspergillus nidulans and a toxigenic fungus A. flavus. In A. nidulans, the absence of AnimonA led to decreased fungal growth, reduced asexual reproduction, and defective cleistothecia production. In addition, AnimonA deletion mutants exhibited decreased spore viability, had reduced trehalose contents in conidia, and were sensitive to thermal stress. In A. flavus, deletion of AflmonA caused decreased fungal growth and defective production of asexual spores and sclerotia structures. Moreover, the absence of monA affected vacuole morphology in both species. Taken together, these results indicate that MonA plays conserved roles in controlling fungal growth, development and vacuole morphology in A. nidulans and A. flavus.

To understand on-site feeding traits of Noctiluca scintillans with unprecedented high abundances around Dokdo in spring, 2014, the composition and abundance of food items in the digestive vacuole of the species were analyzed. Abundances of N. scintillans ranged from 4,328~17,791 inds.m-3 around Dokdo during the study. The incidence of preyed items in the vacuole of N. scintillans ranged by averaged 32% (24~50%) in the surface waters around Dokdo. Diverse preyed items consisted of fecal pellet, protozoans, pollen, diatoms, dinoflagellates, copepods and ichtyoplankton, while fecal pellets were dominantly fed by N. scintillans (43%) and next by protozoans (19%), pollen (18%), diatoms (7%) and dinoflagellates (4%). Fecal pellet, protozoans and pollen were relatively preferred by N. scintillans compared to diatoms, dinoflagellates and ichtyoplankton based on the incidence in the food vacuoles. Present results indicated that high abundance of N. scintillans found around Dokdo was likely associated with non-phytoplankton items rather than previously well-known diatoms as prey during the study.

Anatoxin-a, saxitoxin and neosaxitoxin are produced by Aphanizomenon flos-aquae that is a sort of the cyanobacteria phylum. Therefore, it is not permitted for food materials in Korea. Traditionally, the classification of cyanobacteria has been based on morphological characters such as trichome width, cell size, division planes, shape, and the presence of character such as gas vacuole. But, some diagnostic features, such as gas vacuole or akinetes, can show variation with different environmental or growth conditions and even be lost during cultivation. Therefore, we developed detection method for functional foods containing Aph. flos-aquae by PCR. To design the primer, 16S rRNA region of Aph. flos-aquae, Spirulina laxissima, and Spirulina spp. registered in the GeneBank (www.ncbi.nlm.nih.gov) have been used and for comparative analysis, BioEdit ver. 7.0.9.0. was used. As a result, we was design AFA-F1/AFA-R1 (363 bp) primer for the differentiation Aph. flos-aquae from chlorella, spirulina, green tea, and spinach. Also, it could be distinguished chlorella and spirulina products those are made to contain 1% Aph. flos-aquae.
This experiment was conducted to examine the changes in polysaccharide concentration and morphological variation of Fomitopsis pinicola mycelium during submerged-culture in the citrus peel medium (CP). On the 12 days culture, the yields of mycelium and alcohol insoluble substance were 40.21%(w/v) and 6.94%(w/w), respectively, which were much higher than 11.29%(w/v, wet basis) and 3.17%(w/w, wet basis) obtained from YM medium. A large amount of acid soluble polysaccharides was derived from YM medium while a larger amount of alkali soluble polysaccharide was produced from CP medium. Yields of the mycelium were higher when cultured in CP medium However, there was no significant difference in formation of membranous vesicle between mycelia cultured in CP medium and YM medium. It was also observed that the formation of vacuole was closely related to the activation of the multivescular body known as cytolysome. As a result activation of mycelium and cell wall biosynthesis were more accelerated in CP medium.
The cell membrane properties in relation to flesh browning of Fuyu persimmon fruits during CA storage were studied. Compared to intact fruits, the flesh tissue of browned fruits showed higher rate of electrolyte leakage, indicating incresed membrane permeability. It could be assumed that the increased membrane permeability results in 1eakage of phenolic compounds from vacuole and their oxidation by contacting with PPO, inducing finally the development of flesh browning. In addition, lower content of fatty acids and higher saturation rate of them were found in browned fruits. In conculusion, it was suggested that the inhibited fatty acid metabolism and fatty acid saturation during CA storage cause membrane Permeability to increase.
Saccharomyces cerevisiae는 전통적으로 알코올 음료와 bioethanol 생산에 이용되지만, 발효가 진행되는 동안 효모의 에탄올 생성은 에탄올의 축적에 의한 충격으로 세포활성에 손상을 초래한다. 본 연구는 S. cerevisiae의 에탄올 스트레스 반응과 에탄올 내성의 분자적 기초에 관해 수행되었으며, 에탄올 스트레스가 진행되는 동안 효모의 에탄올 생성 향상을 위한 유전 공학 전략의 수립에 활용될 수 있다. 이전의 연구들은 유전자 발현에 대한 에탄올 스트레스의 충격이 환경적 영향을 받기 때문에 다양한 균주와 조건들에 관해 이루어졌다. 그러나 에탄올 공격에 의해 영향을 받은 gene ontology 범주에서의 일부 공통점은 S. cerevisiae의 에탄올 스트레스 반응이 해당과정 및 미토콘드리아 기능과 관련된 유전자 발현의 증가와 에너지가 요구되는 성장과정과 관련된 유전자의 발현 감소에 따라 에너지 생산에 제약 받음을 의미한다. Genomewide screens를 이용한 연구는 vacuole function의 유지가 에탄올 내성에 대해 중요함을 암시한다. 아마도 단백질 turnover와 이온 항상성 유지에 이 세포기관의 역할이 중요하기 때문인 것으로 사료된다. 특히 에탄올 스트레스가 일어날 때 핵 내 Asr1과 Rat8의 축적은 비록 이 가설이 논란이 많은 주제로 남아있지만 S. cerevisiae가 에탄올 스트레스에 대한 특별한 반응을 가지고 있음을 의미한다.
Ethylene was treated or inhibited to investigate its effect on the physiological changes related to induction of flesh browning in Fuyu persimmon fruit. The response of fruit to ethylene was so slight, that the Fuyu fruit seemed to possess a similar characteristic to non-climacteric fruit. The flesh browning was however enhanced by ethylene treatment, although any significant increment of phenolic content or PPO activity in flesh tissue was not detected. Ethylene induced not only increasing of ion leakage from fruit tissue, but the fatty acids extracted from ethylene-treated fruit tissue were also more saturated. It was suggested that ethylene be related in the changing of membrane permeablity via saturating of fatty acid in membrane lipid. That could result in increased leakage of vacuole-stored phenolic compounds, which oxidized further by PPO to cause fruit flesh to brown.

Mon1 is a guanine nucleotide exchange factor subunit that activates the Ypt7 Rab GTPase and is essential for vacuole trafficking and autophagy in eukaryotic organisms. Here, we identified and characterized the function of Mon1, an ortholog of Saccharomyces cerevisiae Mon1, in a human fungal pathogen, Cryptococcus neoformans. Mutation in mon1 resulted in hypersensitivity to thermal stress. The mon1 deletion mutant exhibited increased sensitivity to cell wall and endoplasmic reticulum stress. However, the mon1 deletion mutant showed more resistance to the antifungal agent fluconazole. In vivo studies demonstrated that compared to the wild-type strain, the mon1 deletion mutant attenuated virulence in the Galleria mellonella insect model. Moreover, the mon1 deletion mutant was avirulent in the murine inhalation model. These results demonstrate that Mon1 plays a crucial role in stress survival and pathogenicity in C. neoformans.
This study was investigated to the structure of cell wall during maturation for the research of softening of jujube fruits. Cell was hardly combined with each other untill turning stage, but middle lamella of cell wall was splited at mature stage and was observed splited cell. The middle lamella of cell wall was not observed at green mature stage, but was observed at turning stage. Cell wall was degraded at mature stage. It was observed mitochondria, endoplasmic reticulum et. at in jujube fruit of green mature stage, but cytoplasm and organelle was attached on cell wall as vacuole was grown up after turning stage.
The mobilization of proteins in the cotyledons of germinating soybean seeds (Glycine mar [L.] Merr.) and seedlings was studied by using light microscopy and transmission electron microscopy. The cotyledon tissues of soybean. were packed with protein bodies(diameter $0.1-15{\mu}m$) where storage protein of soybean is deposited. Degradation of protein bodies started in the epidermis and vascular tissues. After swelling of the protein bodies, autolysis of storage proteins began while the external membrane remained unbroken. Hydrolysis of proteins could be internal or peripheral and fusion might begin before complete protein degradation. Possible instances of vacuolar fusion were encountered in some cells. In all cases, the result of degradation was the same; the central vacuole of the cell. At the late stages of seedling growth, breakdown of tonoplast was observed in some cells.
