다양한 페놀성 물질과 Folin-Ciocalteu 시약과의 반응성에 미치는 단백질의 영향 (Effects of Proteins on the Reactivity of Various Phenolic Compounds with the Folin-Ciocalteu Reagent)

  • 본 연구는 기능성 소재 등의 페놀성 성분 함량 분석에 널리 이용되는 Folin-Denis 정량반응에서 단백질이 미치는 영향을 조사하였다. BSA와 SMP는 Folin-Denis 페놀정량반응에서 농도의존적인 발색반응을 나타내었고 BSA가 SMP보다 더 민감한 반응성을 나타내었다. 그러나 BSA는 각종 페놀성 물질과 같이 존재할 경우 페놀성 물질 자체에 의한 발색도 이하로 반응성을 감소시켰으며, 0.25보다 0.5 mg/mL의 BSA 농도에서 더 저하된 발색도를 유도하였다. BSA에 의한 서로 다른 페놀성 물질 간의 반응성 차이는 크게 나타나지 않았으나, SiA, Ctc, 및 EGCG 등에서 물질 당량 당 흡광도 증가의 기울기가 비교적 크게 감소하였다. 이들은 BSA가 없을 때에 비해 0.25와 0.5 mg/mL BSA 존재 시 각각 80%대 및 70%대의 기울기 감소율을 나타내었다. 한편, 페놀성 화합물들의 산화를 유도하여 F-C시약과 반응시킨 결과, TA, GA 및 EGCG에서 반응성의 감소가 크게 나타났으며, BSA 존재 시 이들 산화물의 상대적 발색도는 산화시간의 증가 및 BSA 농도 증가에 따라 감소하는 경향을 나타내었다. BSA의 형광강도가 TA, EGCG에 의해 유의적으로 감소하는 것으로 보아, BSA에 의한 페놀성 물질의 F-C시약과의 반응성 감소는 페놀성 물질과 BSA의 결합특성에 의한 것으로 사료된다. 이상의 결과는 단백질이 페놀성 물질과의 상호작용을 통해 이들의 정량에 간섭할 뿐만 아니라 페놀성 물질의 다양한 생리활성에도 영향을 미칠 수 있다는 것으로 시사하며, 보다 정확한 기작검토를 위하여 관련 연구가 지속적으로 수행 되어야 할 것으로 보인다.

  • 본 연구에서는 F-C시약을 이용한 페놀성 물질의 정량방법에서 다양한 종류의 페놀성 성분들의 반응특성 및 반응영향요인들을 분석하였다. 정량방법 중 $Na_2CO_3$를 선처리는 F-C시약을 먼저 처리하는 방법에 비해 대부분 페놀성 물질들의 발색반응도 감소를 야기하였으며, 특히 -OH 밀도가 높은 galloyl group을 가진 gallic acid 및 EGCG 등의 성분이 두드러진 감소를 나타내었다. F-C시약과 $Na_2CO_3$를 동시에 처리하는 경우 F-C시약을 선처리 하는 경우에 비해 각 페놀성 물질의 반응성이 약간 감소되는 경향을 나타내었다. 그 원인은 페놀성 물질들이 산성인 F-C시약에서보다 $Na_2CO_3$ 용액의 알칼리 환경에서 화학적으로 불안정하기 때문이며, pH 7.4 및 9의 환경에서 페놀성 물질로부터 생성된 산화물들은 F-C시약과의 반응성이 약화됨을 확인하였다. 이 과정중에 같이 형성된 $H_2O_2$는 F-C와의 반응에 직접적인 영향을 미치지 않았다. FeA와 SiA와 같은 monophenol류는 방법의 차이에 따라 큰 반응성의 변화를 나타내지 않았으며, 발색도는 느리지만 꾸준히 증가하는 kinetics 패턴을 보였다. 실제 시료에의 적용을 위해 연잎 추출물을 제조하고 각 정량방법에 대한 반응도 차이를 조사한 결과, 역시 $Na_2CO_3$ 처리환경에서 현저한 발색반응도 저하를 나타내었다. 보다 정확한 페놀성 물질의 정량을 위해 적절한 표준물질의 선택 및 정량 방법 등에 대한 표준화가 필요할 것으로 사료된다.

리포좀 제제 중 렉틴-엘라지탄닌 포합체의 분석법 확립 (Assay Method for Lectin-conjugated Ellagitannin Encapsulated in Liposomal Formulations)

  • Lectin-conjugated ellagitannin (LET), a newly introduced melanoma-specific antitumor agent which has been synthesized by conjugation of wheat germ agglutinin as a lectin with praecoxin A as an ellagitannin, was encapsulated into sterically stabilized liposomes (SSL). Modified Folin phenol method was established for the quantitation of LET contents in liposomal formulations protein employing the standard calibration curve with bovine serum albumin. After removal of phospholipid by organic solvent extraction, which interferes the specific selectivity of the Folin-Ciocalteu reagent with the protein, recovery of LET was $94.5{\pm}2.3%$ and the encapsulation efficiency was revealed as $37.8{\pm}5.9%$ for 2.5 mg/ml LET solution.

Total Phenolic Content and Antioxidant Activities of Leaves and Bark Extract of Adenanthera pavonina L.

  • Adenanthera pavonina L. is an important medicinal plant for Indonesian traditional medicine ("Jamu"). The present study is to determine the phenolic content and evaluate the antioxidant activity of ethyl acetate and ethanolic extracts of leaves and bark of A. pavonina. The total phenolic content was determined using the Folin-Ciocalteu reagent methods, and the antioxidant activity was performed by phosphomolybdate assay, through EC50 values. The highest phenolic content (81.379 mg GAE/g) was found in the hydro-ethanolic extract of the leaves and (80.630 mg GAE/g) in the bark. The antioxidant activity of the two samples was 42.270 ㎍/mL (leaves) and 45.261 ㎍/mL (bark). Plants have the potential as drugs with good antioxidant potential and phenolic compound content. Further scientific studies of this plant are supported to evaluate the antioxidant activity using other methods to validate these preliminary experiments properly.

Effective Extraction of Oligomeric Proanthocyanidin (OPC) from Wild Grape Seeds

  • The Oligomeric proanthocyanidin (OPC) in green and black tea, grape seeds, grapes and wine has raised much attention but that OPC in wild grape seed remains to be intensively investigated. This study investigated the total OPC contents and total antioxidant activity of wild grape seeds and developed an efficient extraction process with various temperatures, solvent compositions and times. Also, a chromatography column packed with the Dia-ion HP-20 resin was used for further purification of the ope. The total OPC contents were determined with the Folin-Ciocalteu reagent, and the antioxidant activity using total antioxidant potential (TAP) and 1,1-dipheny|-2picrylhydrazy| (DPPH). The yield of final purified OPC was 1.78 (+)-catechin equivalent (CE) g/100 g, with $IC_{50}$ activities of TAP and DPPH of 31.60 and $15.70\;{\mu}\;g/mL$. These activi­ties of the final purified OPC were about two times higher than that of the BHA used as a refer­ence sample.

Protection of Green Leafy Vegetable Extracts Against Oxidation of Human Low Density Lipoprotein

  • Oxidation of low density lipoprotein (LDL) is regarded to play an important role in the development of atherosclerosis. In the present study, salad vegetables with a remarkable DPPH radical-scavenging activity were extracted with methanol, and the methanol extracts were evaluated for the inhibition of $Cu^{2+}$-induced oxidation of human LDL. Separately, the amount of total phenolics was determined colorimetrically using Folin-Ciocalteu reagent. The vegetable extracts, expressing a strong inhibition of LDL oxidation ($IC_{50}$ values, <$100\;{\mu}g/mL$), were from angelica, dandelion, mustard leaf, and water spinach, which contained relatively high level of polyphenol content. Noteworthy, a highly positive correlation was observed between inhibition of LDL oxidation and amount of total polyphenol (p<0.01). Based on these results, it is suggested that salad vegetables, especially angelica, dandelion, and mustard leaf, may be used as easily accessible sources of natural antioxidants, especially in anti-atherosclerosis.

Free radical scavenging property of Annona reticulata leaves

  • Annona reticulata belonging to the family Annonaceae is traditionally used as anthelmintic and it is reported to have anticancer activity. Free radicals are known to be involved in various types of diseases like cancer, diabetes, neurological disorders etc. The present study was aimed to evaluate free radical scavenging property of methanol extract of the leaves of Annona reticulata in some in vitro models e.g. DPPH, nitric oxide, superoxide and lipid peroxide radical model. The extract showed good dose-dependent free radical scavenging property in all the models. $IC_{50}$ values were found to be 3.22, 170.01, 25.12 and $140.12\;{\mu}g/ml$ respectively in DPPH, nitric oxide, superoxide and lipid peroxidation inhibition assays. Reductive ability of the extract was also tested based on potassium ferricyanide reduction where dose dependant reducing capability was observed. Measurement of total phenolic compounds by Folin-Ciocalteu's phenol reagent indicated that 1mg of the extract contained $146.20\;{\mu}g$ equivalent of pyrocatechol. The findings ascertain promising free radical scavenging property of the extract and the antioxidant property of the extract may be due to the high content of phenolic compounds.

In vitro antioxidant and free radical scavenging activities of stem extract of Euphorbia trigona Miller

  • Antioxidative and free radical scavenging properties of different stem extracts of Euphorbia trigona were evaluated and correlated with its total phenolic content. Aqueous, acetone and methanolic extracts of shade dried stem were obtained and were concentrated in vacuo. The antioxidant and free radical scavenging activities of stem extracts was determined by 2,2-Diphenyl-1-picrylhydrazyl (DPPH) scavenging assay, reducing power assay, deoxyribose degradation assay and $Fe^{2+}$ chelating assay. Total phenolic contents (TPC) were evaluated using Folin-Ciocalteu reagent. The results confirmed that the plant is a rich source of polyphenolic compounds which are invariably higher compared to other herbs. All extracts showed TPC in the range of 146.6 - 168.6 mg/g gallic acid equivalents at $300{\mu}g/ml$ of extract. Among the three extracts ME showed highest scavenging activity as evidenced by maximum scavenging of DPPH (83.2%), $OH{\bullet}$ radicals (94.81%), $Fe^{2+}$ chelating activity (88.59%) and a high reducing power 0.623 at $300{\mu}g/ml$. Our results demonstrate that Euphorbia trigona, an unexplored xerophytic plant could be potential source of natural antioxidants and phytotherapeutic agents. The plant possess invariably high amount of polyphenolic compounds with a broad spectrum of antioxidant properties and could be further used for food, feed and pharmaceutical applications.

국산 블루베리 착즙액의 안토시아닌 분석 및 RAW267.4 세포주에서의 항염효과 (Anthocyanin Analysis of Pressure-extracted Korean Blueberry Juice and in vitro Anti-inflammatory in RAW267.4 Cell line)

  • Blueberry juice possesses rich-procyanidins and - anthocyanidin, comprised a group of with numerous health benefits such as protection against coronary heart disease, detoxification, and obesity. Blueberry (Vaccinium virgatum) juice extracts were analyzed and separated by an HPLC method for the purpose of the separation and quantification in polyphenolic groups. In specific HPLC conditions, a binary mobile phase consisting of formic acid: water (10:90, v/v, solvent A) and formic acid: water: acetonitrile (10:60:30, v/v/v, solvent B) was utilized and it is detected at 546 nm wavelength. The phenolic contents of the extracts are determined using Folin-Ciocalteu phenol reagent. In order to test anti-inflammation activity assay, after producing nitric oxide (NO) in lipopolysaccharide activated RAW 264.7 cells, at concentration of $20-500{\mu}g/mL$ it reduced to NO production at a dose-dependent manner. Importantly, cytotoxicity assay with up to $500{\mu}g/mL$ of the extract from blueberry juice showed ~100% cell viability for RAW264.7 cell line. Therefore, Korean blueberry juice might have potential as anti-oxidant and antiinflammation agents.

Antioxidant Activity of Salad Vegetables Grown in Korea

  • The antioxidant activity of forty two kinds of salad vegetables grown in Korea was evaluated. Methanol extract of freeze-dried vegetable was assayed by radical scavenging activity using 1,2-diphenyl-2-picrylhydrazyl (DPPH) and Fe^{2+}$-catalyzed lipid peroxidation inhibition by TBA method. Total phenolics were determined colorimetrically using Folin-Ciocalteu reagent. The highest radical scavenging activity was expressed by perilla leaf, followed by dandelion leaf, red and green leafy lettuce, of which $IC_{50}$ was less than 0.10 mg/mL. Angelica leaf showed the highest inhibitory action for lipid peroxidation with $95\%$, and then dandelion leaf, water spinach, and perilla leaf inhibited over $80\%$. However, lettuce (Iceberg) and young Chinese cabbage exhibited the lowest antioxidant activity based on both assay methods. Highly positive correlations between antioxidative activities and total phenolics were observed (p < 0.001). The results suggested that salad vegetables, especially perilla leaf, leafy lettuce, dandelion or angelica, could be used for easily accessible sources of natural antioxidants.
