To find out the mineral cycles of calcium, magnesium and sodium in dynamic grassland cosystems in a steady state condition, this investigation was conducted along the northwest side on Mt. Kwanak. The experimental results may he suromarized on the communities of a Zoysia japonica and a Miscanthus sinesis as follows. As compared with some properties of the surface soils among two semi-natural grasslands, cal- cium (Ca) was greater quantity in a Zoysia japonica, whereas, in a Miscanthus sicensis, sodium (Na)and magnesium (Mg) were greater in Mt. Kwanak. For the case of steady production and release, the ratio of annual mineral production to the amount accumulated on the top of mineral soil in a steady state provides the estimates of release constant k. The release constants of Ca, Mg and Na of the litter were 0.42, 0.25 and 0.29 in the Zoysia japonica grassland, and were 0.41, 0.54 arid 0.62 in the Miscanthus sinensis grassland, respect- ively. The half times of Ca, Mg and Na required for the release or accumulation of the litter on the grassland were 1.65, 2.77 and 2.39 in the Zoysia japonica, and were 1.69, 1.28 and 1.12 in the Miscauthus sinensis, respectively. The increasing order of the turnover parameters of the elements was Ca, Na and Mg in the Zoysia japonica grassland, and was Na, Mg and Ca in the Miscanthus Si nens is grassland. The amounts of annual cycles for Ca, Mg, Na in the grassland ecosystem under the steady-state conditions were 1.29, 0.20 and 0.12 g /m$^2$ in the Zoysia japonica grassland and 3.91, 1.04 and 0.61 g /m$^2$ in the Miscanthus sinensis grassland.
For preventing and curing the stretching mark, TECA Niosome/W/O system creams were formulated using Titrated Extract of Centella Asiatica (TECA) which is well known for its excellent wound healing effect. The lipid-water partition coefficients and the stabilities of TECA were evaluated and TECA Niosome/W/O system (TECA N/W/O) creams were prepared with different concentrations of cetyl alcohol and ceramide. TECA N/W/O cream was evaluated with respect to their rheological properties, permeation through excised skin of hairless mouse and in vitro and in vivo accumulation in the skin of hairless mouse. In addition, dermal thicknesses of hairless mouse skins were determined following the in vivo application of TECA N/W/O cream and control cream. TECA N/W/O creams showed pseudoplastic flow and hysteresis loop. The permeation of TECA from formulations through excised skin of hairless mouse did not observed. Amount of accumulated drug in the excised skin of hairless mouse was deσeased with an increase in the concentration of cetyl alcohol and showed no relationship with concentration of ceramide. Amount of accumulated drug in formulation A-3 was higher than in niosome suspension and other formulations. In in vivo experiment, amount of accumulated drug in formulation A-2 and A-3 was much higher than that of niosome suspension. Being treated with the N/W/O cream for 8 weeks, the dermal thickness of hairless mouse skin was increased 3.2 times than that of 16 weeks-control group.
저수지를 설계 및 계획함에 있어서 저수지의 설계수명 기간동안 예상되는 퇴사량을 위한 퇴사공간을 배분하는 것은 가장 중요한 절차중의 하나이다. 특히 퇴사공간의 배분과 관련된 해석 및 연구는 저수지 운영, 흐름의 수리학적 특성, 유입 유사량과 연관된 매우 다양한 변수들간의 상호작용으로 인하여 매우 복잡하고 쉽지 않은 문제이다. 퇴적물들의 공간적인 분포를 알아보기 위한 접근방법은 경험적인 방법에 의존하고 있으며 이와 같은 경험적인 방법의 적용은 대부분 실제의 퇴적현상을 엄청나게 단순화시켜야만 가능하다 본 연구에서는 소양강댐을 대상으로 하여 경험적면적감소법(Empirical Area Reduction Method)에 의해 저수지 퇴사량을 계산하고 계산된 결과를 실측자료와 비교하여 봄으로써 경험적면적감소법의 적용성을 알아보았으며 그 결과 매우 적용성이 우수한 것으로 나타났다.
Various nanomaterials may flow into the aquatic ecosystem via production, use, and treatment processes. Especially, gold nanoparticles (AuNPs) were categorized as manufactured nanomaterials presented by the Organization for Economic Cooperation and Development Working Party on Manufactured Nanomaterials (OECD WPMN) in 2010. AuNPs have been used in medical area, however, they were reported to induce cytotoxicity and oxidative DNA damage, as well as down-regulation of the DNA repair gene in mice and human cell lines. In this study, the aquatic toxicity data of AuNPs and gold ions were collected, with the specific test methods analyzed with respect to the form and size of AuNPs, test species, exposure duration, and endpoints. Currently, aquatic toxicity data of AuNPs and gold ions have been presented in 14 studies including 4 fish, 6 crustacean, 2 green algae, and 2 macrophytes studies, as well as a further 8 studies including 4 fish, 4 crustacean, 1 platyhelminthes, and 1 green algae, respectively. The AuNPs were 0.8-100 nm in size, as gold nanoparticles, gold nanorod, glycodendrimer-coated gold nanoparticles, and amine-coated gold nanoparticles. The tested endpoints were the individual toxicities, such as mortality, malformation, reproduction inhibition, growth inhibition and genetic toxicity such as oxidative stress, gene expression, and reactive oxygen species formation. The accumulation of AuNPs was also confirmed in the various receptor organs. These results are expected to be useful in understanding the aquatic toxicity of AuNPs and gold ions, as well as being applicable to future toxicity studies on AuNPs.

Objectives: The purpose of this study was to evaluate the anti-cancer activity of cisplatin by studying its effects on cell viability and identifying the mechanisms underlying the induction of cell cycle arrest and apoptosis on oral squamous cell carcinoma (OSCC) cell lines with varying p53 mutation status. Materials and Methods: Three OSCC cell lines, YD-8 (p53 point mutation), YD-9 (p53 wild type), and YD-38 (p53 deletion) were used. To determine the cytotoxic effect of cisplatin, MTS assay was performed. The cell cycle alteration and apoptosis were analyzed using flow cytometry. Western blot analysis was used to detect the expression of cell cycle alteration- or apoptosis-related proteins as well as p53. Results: Cisplatin showed a time- and dose-dependent anti-proliferative effect in all cell lines. Cisplatin induced G2/M cell accumulation in the three cell lines after treatment with 0.5 and $1.0{\mu}g/mL$ of cisplatin for 48 hours. The proportion of annexin V-FITC-stained cells increased following treatment with cisplatin. The apoptotic proportion was lower in the YD-38 cell line than in the YD-9 or YD-8 cell lines. Also, immunoblotting analysis indicated that p53 and p21 were detected only in YD-8 and YD-9 cell lines after cisplatin treatment. Conclusion: In this study, cisplatin showed anti-cancer effects via G2/M phase arrest and apoptosis, with some difference among OSCC cell lines. The mutation status of p53 might have influenced the difference observed among cell lines. Further studies on p53 mutation status are needed to understand the biological behavior and characteristics of OSCCs and to establish appropriate treatment.
Background: Rac3, a member of the Rac family of small guanosine triphosphatases (GTPases), regulates a variety of cell functions, including the organization of the cytoskeleton, cell migration, and invasion. Overexpression of Rac3 has been reported in several human cancers. However, the role of Rac3 in lung cancer (LC) has not been determined in detail. The purpose of this study was to investigate the effect of silencing of Rac3 expression in human LC cells and the consequences for cell survival. Materials and Methods: Lentivirus small hairpin RNA (shRNA) interference techniques were utilized to knock down the Rac3 gene. Gene and protein expression was quantified by quantitative real-time polymerase chain reaction (qRT-PCR) and Western blotting. LC cell apoptosis was examined by annexin V-APC /propidium iodide staining. Results: Efficient silencing of Rac3 strongly inhibited A549 cell proliferation and colony formation ability, and significantly decreased tumor growth. Moreover, flow cytometry analysis showed that knockdown of Rac3 led to G2/M phase cell cycle arrest as well as an excess accumulation of cells in the G1 and S phase. Conclusions: Thus, functional analysis using shRNAs revealed a critical role for Rac3 in the tumor growth of LC cells. shRNA silencing of Rac3 could provide an effective strategy to treat LC.
Objectives : To clarify the possible effect of Lycium chinese Mill (LC)., Morus alba L (MA)., and Lycium chinese Mill. +Morus alba L. (LC+MA), we have examined their influence on the development of pulmonary eosinophilic inflammation in the asthmatic murine model. Methods : Female Balb/c mice (5weeks) were immunized on two different days (21 days and 7 days before inhalational exposure) by intraperitonial injections of 0.2ml alum-precipitated Ag containing $100{\mu}g$ of OVA bound to 4 mg of aluminum hydroxide in PBS. Seven days after the second sensitization, mice were exposed to aerosolized ovalbumin for 30 minutes/day on 3 days/week for 8 weeks (at a flow rate of 250 L/min, 2.5% ovalbumin in normal saline) and, LC, MA, and LC+MA (500 mg/kg) were orally administered 3 times per a week for 8 weeks. Results : The suppressive effect of LC, MA, and LC+MA were demonstrated by the accumulation of eosinophills into airways, with the reduction of eosinophil, total lung leukocytes numbers. These were correlated with the marked reduction of IL-5, IL-13 and IL-4 levels in the BALF and serum. OVA-specific IgE levels were also decreased in serum and BAL from these mice. LC, MA, and LC+MA decreased eosinophil CCR3 expression and CD11b expression in lung cells. Conclusions : These results indicate that LC, MA, and LC+MA have high inhibitory effects on airway inflammation and hyper-responsiveness in the asthmatic murine model. The suppression of IL-5, IgE, eosinophil CCR3 expression and CD11b expression, and the increase of IFN-${\gamma}$ production in BALF seem to contribute to this effect. Hence, the results indicated that LC, MA, and LC+MA could act as a immuno-modulator which possesses anti-inflammatory and anti-asthmatic property by modulating the imbalance between Th1 and Th2 cytokines.
We investigated the effects of Sabaek-san (SBS) water extract on the cell proliferation of human lung carcinoma A549 cells. SBS treatment resulted in the inhibition of cell proliferation in a concentration-dependent manner. This anti-proliferative effect of A549 cells by SBS treatment was associated with morphological changes such as membrane shrinking and cell rounding up. DNA flow cytometric histograms showed that population of G1 phase of the cell cycle was increased by SBS treatment in a concentration-dependent manner. SBS treatment induced a marked accumulation of tumor suppressor p53 and a concomitant induction of cyclin-dependent kinase (Cdk) inhibitor p21WAF1/CIP, which appears to be transcriptionally upregulated and is p53 dependent. In addition, SBS treatment resulted in down-regulation of cyclooxygenase-2 (COX-2) as determined by RT-PCR analysis. The present results indicated that SBS-induced inhibition of lung cancer cell proliferation is associated with the blockage of G1/S progression the induction of apoptosis.
A 69-year-old woman was presented with progressed dysphagia, gastric soreness and weight loss during 2 months. She was performed abdomen x-ray, EGDS and abdomen CT. Abdomen x-ray demonstrated punctuate calcification on LUQ. EGDS showed an ulceroinfiltrative mass with bleeding on cardia to antrum of stomach. And CT showed diffuse gastric wall thickness with multiple calcifications. Biopsy of the stomach and esophagus during EGDS examination revealed an adenocarcinoma, with signet ring cell type, infiltrating the wall of the stomach and the distal esophagus. Then acne scan was performed a few days later. It revealed intense uptake in LUQ, corresponding to the calcium containing neoplasm seen on the abdomen x-ray, EGDS and abdomen CT. And there was no evidence of any metastatic lesion and thyroid uptake on the bone scan. There are many reports about accumulation of the tracer in extraosseous lesion, but only a few literatures were reported about gastric calcification in stomach cancer. More over, no reports showed CT images. We are performed many diagnostic examinations and found well correlation between them. The reason of gastric calcification is considered with calcium deposition within extracellular space due to hemorrhage or necrosis. Other possibility offered to explain gastric calcification have been increased blood flow and/or increased neovascularity with capillary leaks of tracer, and specific enzymatic (phosphatases) receptor binding of tracer. So, it was happened ion exchange between intracellular calcium and phosphate groups of tracer.
We investigated the catechin content in green tea leaf (GTL) and green tea seed (GTS), the antiproliferative and detoxifying phase II enzyme-inducing activities of the methanolic (80%, v/v) extracts from GTL and GTS. GTL and GTS contained $8,685{\pm}1,061$ and $108{\pm}32\;{\mu}g/g$ epigallocatechin gallate (EGCG), $11,486{\pm}506$ and $116{\pm}72\;{\mu}g/g$ epigallocatechin (EGC), $3,535{\pm}308$ and $821{\pm}95\;{\mu}g/g$ epicatechin gallate (ECG), and $1,429{\pm}177$ and $37{\pm}44\;{\mu}g/g$ epicatechin (EC), respectively. The methanolic extract of GTS showed a greater increase in quinone reductase activity and antiproliferation potential against mouse hepatoma cells than GTL extract did. GTS treatment resulted in the accumulation at sub-G1 phase of mouse hepatoma hepa1c1c7 cells as assessed by flow cytometry. Enhancement of phase II enzyme activity by GTS extract was shown to be mediated, directly or indirectly, via interaction with the antioxidant response element (ARE) sequence in the genes encoding the phase enzymes. As the catechin content in GTS was significantly lower than that in GTL, components other than catechins appear to be responsible for the anticarcinogenic activity of the seed. In summary, these results suggest that the 80% methanolic extract of GTS deserves further study to evaluate its potential as an anticarcinogenic agent and to investigate its mechanism of action.
