Analysis of the solution structure of the human antibiotic peptide dermcidin and its interaction with phospholipid vesicles

  • Dermcidin is a human antibiotic peptide that is secreted by the sweat glands and has no homology to other known antimicrobial peptides. As an initial step toward understanding dermcidin's mode of action at bacterial membranes, we used homonuclear and heteronuclear NMR to determine the conformation of the peptide in 50% trifluoroethanol solution. We found that dermcidin adopts a flexible amphipathic $\alpha$-helical structure with a helix-hinge-helix motif, which is a common molecular fold among antimicrobial peptides. Spin-down assays of dermcidin and several related peptides revealed that the affinity with which dermcidin binds to bacterial-mimetic membranes is primarily dependent on its amphipathic $\alpha$-helical structure and its length (>30 residues); its negative net charge and acidic pI have little effect on binding. These findings suggest that the mode of action of dermcidin is similar to that of other membrane-targeting antimicrobial peptides, though the details of its antimicrobial action remain to be determined.

  • Generally the connection of structural members is assumed as hinge, rigid and semi-rigid connections. The exact tangent stiffness matrix of a semi-rigid frame element is newly derived using the stability functions considering shear deformations. Also, linearized elastic- and geometric-stiffness matrices of shear deformable semi-rigid frame are newly proposed. For the exact stiffness matrix, an accurate displacement field is introduced by equilibrium equation for beam-column under the bending and the axial forces. Also, stability functions considering sway deformation and force-displacement relations with elastic rotational spring on ends are defined. In order to illustrate the accuracy of this study, various numerical examples are presented and compared with other researcher's results. Lastly, shear deformation and semi-rigid effects on buckling behaviors of structure are parametrically investigated.

Influence of stiffened hangers on the structural behavior of all-steel tied-arch bridges

  • In tied-arch bridges, the way the arch and the deck are connected may become crucial. The deck is usually suspended from hangers made out of steel pinned cables capable of resisting axial forces only. However, a proper structural response may be ensured by fixing and stiffening the hangers in order to resist, additionally, shear forces and bending moments. Thus, this paper studies the effect of different pinned and stiffened hanger arrangements on the structural behavior of the tied-arch bridges, with the intention of providing designers with useful tools at the early steps of design. Longitudinally and transversally stiffened hangers (and the effect of hinges at the hangers and their locations) are studied separately because the in-plane and the out-of-plane behavior of the bridge are uncoupled due to its symmetry. As a major conclusion, regarding the in-plane behavior, hangers composed of cables (either with vertical, $Nielsen-L\ddot{o}hse$ or network arrangements) are recommended due to its low cost and ease of erection. Alternatively, longitudinally stiffened hangers, fixed at both ends, can be used. Regarding the out-of-plane behavior, and in addition to three-dimensional arrangements of cables, of limited effectiveness, transversally stiffened hangers fixed at both ends are the most efficient arrangement. A configuration almost as efficient and, additionally, cheaper and easier to build can be achieved by locating a hinge at the end corresponding to the most flexible structural element (normally the arch). Its efficiency is further improved if the cross-section tapers from the fixed end to the pinned end.

  • Purpose: The authors investigated whether 35 flexible nail-rotating hinge composite reconstructions around knee joint minimize junctional osteolysis of host bone. Material and Method: The reconstructive technique was as follows: 1) en bloc tumor resection, 2) filling of the host bone marrow cavity with multiple Ender nails, 3) assembling the Ender nails and an Endo-Link type total knee component with wire and bone cement. Result: Mean follow-up was 53 months (ranged 30~79). At final follow-up, 29 patients retained a mobile joint. Resection of more than 40% of bone showed a positive relationship with junctional hypertrophy (p=0.028). Eight patients showed nail breakage and eight prostheses were removed due to early or late infection. The cumulative prosthetic survival rate was 33% at 6 year. Average functional score according to the MSTS criteria was 26.8. Conclusion: Mid-term evaluations showed that results were fair. The revision process was straightforward. Junctional hypertrophy observed appears to give some clues as to how to minimize osteolysis at the prosthesis-host junction after modular prosthesis fixation.

Investigation of rotation and shear behaviours of complex steel spherical hinged bearings subject to axial tensile load

  • Steel spherical hinged bearings have high loading capacity, reliable load transfer, flexible rotation with universal hinge and allowance of large displacement and rotation angle. However, bearings are in complex forced states subject to various load combinations, which lead to the significant influence on integral structural safety. Taking the large-tonnage complex steel spherical hinged bearings of Terminal 2 of Guangzhou Baiyun International Airport as an example, full-scale rotation and shear behaviour tests of the bearings subject to axial tensile load are carried out, and the corresponding finite element simulation analyses are conducted. The results of experiments and finite element simulations are in good agreement with the coincident development tendency of stress and deformation. In addition, the measured rotational moment is less than the calculated moment prescriptive by the code, and the relationship between horizontal displacement and horizontal shear force is linear. Finally, based on these results, the rotation and shear stiffness models of bearings subject to axial tensile load are proposed for the refinement analysis of integral structure.

Seismic Performance of Alternative Steel Structural Systems for an Equipment-Supporting Plant Structure (플랜트 설비 지지용 대안 강구조 시스템의 내진성능)

  • In this study, alternative seismic force-resisting systems for plant structure supporting equipment were designed, and the seismic performance thereof was compared using nonlinear dynamic analysis. One alternative seismic force-resisting system was designed per the requirement for ordinary moment-resisting and concentrically braced frames but with a reduced base shear. The other seismic force-resisting system was designed by accommodating seismic details of intermediate and unique moment-resisting frames and special concentrically braced frames. Different plastic hinge models were applied to ordinary and ductile systems based on the validation using existing test results. The control model obtained by code-based flexible design and/or reduction of base shear did not satisfy the seismic performance objectives, but the alternative structural system did by strengthened panel zones and a reduced effective buckling length. The seismic force to equipment calculated from the nonlinear dynamic analysis was significantly lower than the equivalent static force of KDS 41 17 00. The comparison of design alternatives showed that the seismic performance required for a plant structure could be secured economically by using performance-based design and alternative seismic-force resisting systems adopting minimally modified seismic details.

Optimal Design of a High-Agility Satellite with Composite Solar Panels

  • This paper defines mode shape function of a composite solar panel assumed as Kirchhoff-Love plate for considering a torsional mode of composite solar panel. It then goes on to define dynamic model of a high-agility satellite considering the flexibility of composite solar panel as well as stiffness of a solar panel's hinge using Lagrange's theorem, Ritz method and the mode shape function. Furthermore, this paper verifies the validity of dynamic model by comparing numerical results from the finite element analysis. In addition, this paper performs a dynamic response analysis of a rigid satellite which includes only natural modes for solar panel's hinges and a flexible satellite which includes not only natural modes of solar panel's hinges, but also structural modes of composite solar panels. According to the results, we confirm that the torsional mode of solar panel should be considered for the structural design of high-agility satellite. Finally, we performed optimization of high-agility satellite for minimizing mass with solar panel's area limit using the defined dynamic model. Consequently, we observed that the defined dynamic model for a high-agility satellite and result of the optimal design are very useful not only because of their optimal structural design but also because of the dynamic analysis of the satellite.

  • The core of IT products, electronic components, especially the MLCC, chip inductors, chip Varistors and so on. In order to test the electrical characteristics of the chip using the Reno-pin contact test method has been used. In current chips, mass production of semiconductor manufacturing processes, high-speed production test for the chip speed up, precision is required. But Vibration displacement is a very short, so in order to overcome these shortcomings, the displacement amplification to design the structure has been actively studied. In this paper, a building structure with a flexible hinge was designed amplification instrument, semiconductor chip industry in the performance test and inspection equipment to measure the electrical characteristics of high speed linear actuators Reno-Pin using system was developed.
