Flacherie symptom was found in the fifth instar larvae of silkworm, Bombyx mori. The bacterial pathogen was isolated from the hemolymph of the infected silkworm and identified. The isolated bacteria caused a significant flacherie pathogenicity to the fifth instar larvae of B. mori when $5{\times}10^{6}$ cfu (colony-forming unit) of the bacteria was injected into each larva. The infected larvae began to die at 6 days after injection and resulted in complete mortality at 10 days. The bacterium was identified as Staphylococcus gallinarum based on the morphological and physiological characteristics described in Bergey's manual. This identification was further supported by the characters of carbohydrate utility analyzed from a bacterial identification system ($MicroLog^{\circledR}$) and also by the molecular structure of 165-23S rDNA internal transcribed spaces. As an insecticidal action, S. gallinarum caused hemolymph septicemia by its cytotoxic effect on the hemocytes of B. mori.
The silkworms were inoculated with flacherie virus and reserved at high temperature (37$^{\circ}C$) to find out the effect of high temperature on the multiplication of the virus in silkworms. The obtained results are summarized as follows. 1. It was shown that the inhibitory effect of high temperature treatment on the virus multiplication was achieved at high level at the newly molted stages in 5th instar larvae and both in 4th and in 5th larvae. 2. Twelve hours treatment and 24 hours treatment showed high inhibitory effect on the multiplication of flacherie virus. 3. The silkworms were inoculated with flacherie virus at the newly molted stage in 2nd instar larvae and followed by have temperature (37$^{\circ}C$) treatment at the newly molted stages in 5th instar and both in 4th and in 5th instar for 12 and 24 hours respectively. The increase in the survival rate with high temperature treatment ranged from 33.8 to 39.5 percent in index number and cocoon productive was 45.8 to 54.2 percent compared to non-treatment. 5. In the inoculation of the virus to the 4th instar larvae, the high temperature treatment at the newly molted stage in 5th instar brought increases in the survial rate from 49.7 to 68.9 percent in index number and in cocoon production from 44.3 to 69.0 percent.
Flacherie virus (FV) and Densonucleosis virus (DNV) of the silkworm, Bombyx mori, which give the most severest damage to the silk production in korea, were fed on the mulberry wild silkworm, Bombyx mori mandarina, the mulberry pyralid, Gryphodes phyloalis, and the American fall webworm, Hypantria cunea, to investigate cross infectivity by serological and histopathological at observation. By the Ouchterlony's double difusion test the mulberry wild silkworm was infected with both FV and DNV type 1 (DNV-1) and the mulberry pyralid with DNV-1, so those were confirmed the cross infection. But the American fall webworm was not recognized the cross infection by the same method. The infection and multiplication of the FV in the mulberry wild silkworm was observed in the cytoplasm of the goblet cell with the appearance of the virus-specific vesicle. In DNV-1 infection to the mulberry wild silkworm and the mulberry pyralid, the nuclei of columnar cell in the midgut of both insects was hypertrophied and the nuclei of midgut cell of the mulberry pyralid positively stained with the feulgen stain. Multiplication of DNV-1 in the midgut cell of the mulberry wild silkworm was replicated in two different patterens as linear arrays and large masses, while that of DNV-1 in the muberry pyralid was multiplied as virus masses in several portion of the nuclei of the midgut cell.
The author studied the applicability of Furazolidone to the silkworm rearing industry as a useful remedy for certain silkworm diseases, at the silkworm rearing house of the college of agriculture, Seoul national university, during both the spring and the autumn silkworm rearing season of 1972. Discovering the fact that Furazolidone, when put on the mulberry leaves in a powdered form, is eaten along with the leaves by silkworms and thus the systematic administration of Furazolidone to silkworms is possible, the experimenter carried on a series of experiments (1. determining the in vitro antibacterial activity of Furazolidone to four pathogens of silkworm diseases-Bacillus thuringiensis, Aspergillus oryzae, Aspergillus flavus, and Isaria farinosa, 2. observing the prophylactic and therapeutic effect of Furazolidone against the experimental flacherie caused by inoculation of B. thuringiensis, and 3. examining the toxicity of Furazolidone to silkworm larvae). As the results of the experiments the investigator found out the fundamental fact that Furazolidone exerts a good prophylactic and therapeutic effect against flacherie which is the most common and important silkworm disease: Furazolidone, in in vitro test, inhibited completely the growth of B. thuringiensis, the pathogen of bacterial flacherie, at the concentration of 1 ${\mu}$g/mι. with the tube method and at the concentration of 5 ${\mu}$g/mι. with the plate method, and the drug showed an excellent prophylactic effect and a considerably good therapeutic effect, depending on the time of administration, on the 5th instar silkworms inoculated B. thuringiensis, at the tentative dose of 150mg. per 10 silkworms administered once a day for 2 days. For the practical administration of Furazolidone against flacherie, the dose, the time and duration of administration, and the form of preparation, will be investigated more closely.
Flacherie virus (FV) is an important pathogen in the silkworm, which often gives serious damage to farmers for cocoon production. The inbred parents and F$_1$'s from an eight-parent diallel were examined to determine the inheritance of resistance to flacherie virus in the silkworm. Three resistant (R), two intermediate (M) and three susceptible (S) inbreds were used in the diallel with no reciprocals. Mean resistance was measured by survival rates of larvae which were fed on mulberry leaves sprayed with diluted mid-gut homogenate of FV infected larvae. Broad-sense heritability was obtained according to inbreds and F$_1$ family performance. Estimation of general (GCA) and specific combining ability (SCA) was made according to Griffing's Model 1, Method 2. Mean FV resistance of F$_1$ family displayed additive effect of the major gene, while heterotic effect was not significant. Considerable variation in FV resistance within F$_1$ groups of R$\times$S and S$\times$S indicated that action of minor genes for FV resistance may have been involved. FV resistance of inbreds perse and predominant effect of the major gene over minor gene(s) satisfactorily predicted the FV resistance of the hybrids. Broadsense heritability value of FV resistance on the basis of F$_1$ family performance averaged 93%, which suggested that environmental effects might have not been important in this experiment. GCA was highly significant for FV resistant among inbreds. GCA effect of 13.1 in Jam 108 was highest and -17.7 in Gyeongchu lowest. Effective selection for high FV resistance would be possible, using inbreds with high GCA effect and low GCA variance. SCA was significant among hybrids. High SCA effect in the hybrid of Geumho$\times$Mudeung (13.7) and Hansaeng #4$\times$Jam 115 (11.6) indicated that the interaction effect of minor genes for resistance to FV in the silkworm could be exploited by standard silkworm breeding procedures.

A study has been made to know the effect of a phytoecdysteroid 'Sampoorna' on uniform maturation of silkworms during spinning and its effect on diseased silkworms infected by major silkworm disease viruses, Bombyx mori nuclear polyhedrosis virus (BmNPV) and Bombyx mori infectious flacherie virus (BmIFV). In the present investigation, the effect of the phytoecdysteroid "Sampoorna" on Grasserie disease caused by BmNPV have shown an average cocoon melting of 11.91% with a disease incidence of 5.83%. The values of 't' test for different treatments of BmNPV indicated low survival rate and cocoon traits were drastically reduced. Another major disease Flacherie caused by BmIFV has shown considerable levels of larval disease incidence (22-32%) and cocoon melting (3-7.67%) with an average melting of 12.95% and 20.24% disease incidence. There is a drastic reduction in survival rate, cocoon yield and other economic traits. The control batches were indicated negligible values for disease incidence and cocoon melting with Sapoorna application and without the inoculation of the two disease-causing viruses. The application of Sampoorna on already infected batches with major pathogens triggered high mortality and disease incidence and melting percentage was also significantly increased with reduced economic traits. Hence, it is suggested that application of Sampoorna in infected batches should be done only in the extreme conditions of rearing. Application of Sampoorna on healthy batches led to uniform maturation and improvement in productivity with the added advantage of better quality cocoons and labour saving.
These surveys were designed to know decreasing rate of cocoon production all over the country and purchased amount of cocoons at public market which had been caused by various diseases in autumn of 1975. The results can be summarized as follows; 1. Percentage of sericuitural houses which sustain disease damage was 18.5 percent, and cocoon yield per box in bad crop houses was 8.8kg smaller than that in normal houses. 2. About 838M/T of decreased cocoons and 5.6 percent of damaging rate can be estimated in this autumn throughout the country. 3. In our country, it was 40.9 per cent of Grasseril and 34.5 per cent of Flacherie of silkworm diseases.
Monoclonal antibodies were prepared against Bombyx densonucleosis virus type-II(Yamanashi isolate). Four hybridoma clones, named C4, Fl, H2, M9 were only reacted with the DNV-II, but those were not reacted with Bombyx densonucleosis virus type-I(Ina isolate) and infectious flacherie virus(IFV) by double diffusion test in 0.8% agarose gel. C4, Fl and M9 of them were reacted with 53KDa polypeptide of DNV-II, and H2 was reacted with 46. 5KDa polypeptide of the virus.

Central Sericultural Research and Training Institute, Mysore have evolved several highly productive bivoltine hybrids which can produce international grade raw silk. Among them $CSR2{\times}CSR4,\;CSR2{\times}CSR5,\;CSR3{\times}CSR6,\;CSR17{\times}CSR16,\;CSR18{\times}CSR19$ and $CSR12{\times}CSR6$ are being popularized in the field. There is a minimum difference in their economic characters but they appear to differ in survival. Though they are productive under high input management conditions, they are very susceptible to different diseases under normal rearing practices. No systematic attempts have been made to test their susceptibility status / resistance. Thus the present study is a modest attempt to screen the above six productive bivoltine hybrids to two important pathogens viz., Bombyx mori Nuclear Polyhedrosis Virus (BmNPV) and Bombyx mori Infectious Flacherie Virus (BmIFV) along with existing hybrid, $KA{\times}NB4D2$ to assess their susceptibility / resistance. The results shows that the productive hybrid $CSR2{\times}CSR4$ is the most resistant to BmNPV and it is suggested by its highest $LC_{50}$ value followed by $CSR12{\times}CSR6,\;KA{\times}NB4D2,\;CSR3{\times}CSR6,\;CSR17{\times}CSR16,\;CSR18{\times}CSR19,\;CSR2{\times}CSR5$. Based on the $LC_{50}$ value and $LT_{50}$ values for BmIFV, the hybrid $KA{\times}NB4D2$ was found to be the most resistant (1st position) one followed by $CSR3{\times}CSR6$ (2nd position) $CSR2{\times}CSR$ (3rd position) and $CSR12{\times}CSR6$ (4th position) $CSR17{\times}CSR16$, $CSR18{\times}CSR19$ (5th position) and $CSR2{\times}CSR5$ being the least. The response of 7 bivoltine hybrids to both the pathogens BmNPV and BmIFV indicates that, the hybrids $CSR2{\times}CSR4$, $CSR12{\times}CSR6$ and $KA{\times}NB4D2$ were found to be the most resistant when compared to others. Further, $KA{\times}NB4D2$ being less productive hybrid with a shell ratio of 20.08%, the other two hybrids $CSR2{\times}CSR4$ (Cocoon shell ratio, 21.44%) and $CSR12{\times}CSR6$ (cocoon shell ratio, 23.45%) can be considered to be most productive with superior quality cocoon and resistant to both BmNPV and BmIFV pathogens. The overall study indicated that the hybrid $CSR2{\times}CSR5$ is the most susceptible hybrid to both the pathogens.
The prevalence of the infectious flacherie virus (FV) disease causes a severe damage to cocoon yield and various methods to control the disease have been studied. In this regard, guanidine hydrochloride (GH), one of the guanidine derivatives known as the most inhibitory agent against the replication of picorna virus, was applied to silkworms per os with mulberry leaves and the results were as follows. 1. The application of GH below 0.01% of the chemical concentration did not give any damage to silkworm larvae. 2. The transmission of the virus disease by introducing the FV infected larvae to the healthy larvae group was proportioned to the number of infected larvae. When l% of infected larvae was introduced to the rearing tray of healthy larvae, the pupation rate was 70.7%(79) and it was 38.4% (43) to 5% of infected larvae introduced, while the control of non-mixed with infected larvae gave 89.2% (100) of pupation rate. The cocoon yield from 10,000 larvae also showed the same tendency as the pupation rate. 3. The inhibitory effect of GH against the replication of FV showed ten times in treatment of 0,01% of the chemical agent compared to the non-treatment. 4. The successive application of GH after virus inoculation to silkworm larvae led to the most effective on the inhibition of the virus replication. 5. The immediate application of GH after the virus inoculation also gave the best effect on the inhibition of the virus replication in silkworm larvae. 6. The effect of GH on the inactivation of FV in vitro was not observed.
