환경오염에 의한 미세먼지의 증가로 피부는 산화적 손상과 노화가 가속화된다. 본 연구에서는 선발된 한약재 추출물의 항산화, hyaluronic acid, filaggrin, MMP-1, ROS 항목을 평가함으로써 PM10으로 부터의 각질형성세포 보호 효능을 확인하였다. 그 결과 1,1-diphenyl-2-picrylhydrazyl(DPPH), 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid(ABTS), FRAP assay에서 농도의존적으로 항산화능이 증가하는 것을 확인하였다. 각질형성세포에 PM10 300 ㎍/㎖을 단독으로 처리한 군에서는 hyaluronic acid 및 filaggrin이 50% 이상 감소하였으며, 고량강, 유백피, 토복령 추출물을 처리한 군에서는 증가하였다. MMP-1의 경우 PM10 단독처리군에는 55% 이상 증가하였으나, 추출물을 처리한 경우 감소하여 콜라겐, 엘라스틴의 분해를 저해하는 것으로 평가된다. 또한 제브라피쉬 배아를 이용한 ROS 측정의 경우 추출물을 처리하였을 때 감소되는 것을 확인하였다. 특히 토복령 추출물의 25 ㎍/ml에서 음성대조군과 유사한 형광의 세기를 나타내어 ROS의 생성이 유의적으로 감소한 것을 확인하였다. 본 연구를 통하여 선별된 한약재 소재인 고량강, 유백피, 토복령은 미세먼지로부터 피부를 보호하거나 개선할 수 있는 소재로서 피부 개선을 위한 안티에이징 제품으로 활용될 수 있을 것으로 사료된다.

Ursolic acid (UA) and oleanolic acid (OA) are pentacyclic triterpenoids which are widely distributed in plants, and their derivatives are aglycones of many naturally occurring saponins. It is known that pentacyclic acids may possibly enhance the mechanical barrier functions of cell membranes in plants. Recently, it has been reported that OA and UA have interesting biological activities on skin, such as anti-inflammatory and anti-wrinkle activities. Since triterpenoids are extremely insoluble and their solubility problem limits skin-care application, OA and UA were encapsulated in liposomes via micelle-to-vesicle transition to overcome poorly soluble property and enhance biological efficacy. Optimal molar ratio of OA to lecithin was found to exist for producing liposomes of small hydrodynamic size and liposomal suspensions without recrystallized precipitation of OA. From electron micrograph and dynamic light scattering studies, reconstituted OA-containing liposomes without severe mechanical treatment showed small hydrodynamic size about 150 nm. Wide-angle X-ray diffraction coupled with dynamic light scattering revealed that optimal amount of OA in liposome was 25.4 mole %. In biological evaluation, OA-containing liposome significantly increased filaggrin and transglutaminase as markers of keratinocyte differentiation in epidermal layer of hairless mouse, whereas ursolic acid-containing liposome did not show noticeable increase of filaggrin and transglutaminase compared to empty liposome. It is concluded that nano-scaled liposomes containing triterpenoids were spontaneously prepared by vesicular transition from mixed micelle and liposomal triterpenoids can enhance skin absorption of triterpenoid and biological efficacy.
Objectives The purpose of this study is to confirm the effect of alleviating inflammation through creating skin fat barrier from Scutellaria baicalensis extract. Methods Four-week-old Balb/C mice were divided into four groups: control group (Ctrl), lipid barrier eliminated group (LBEG), dexamethasone (DM) treated group after lipid barrier elimination (DMTG), and Scutellaria baicalensis (SB) treated group after lipid barrier elimination (SBTG). Scutellaria baicalensis extract were administered for 5 days after removal of the fat barrier. Changes in skin condition, improvement of the fat barrier, and relief of inflammation were observed in each group. Results Compared to LBEG and DMTG, pathological skin damage and tissue changes were less in SBTG, and transepidermal water loss (TEWL) and pH were also significantly reduced. Filaggrin was also significantly increased in SBTG. KLK7, PAR-2, and TSLP in SBTG also showed significant reduction compared to the LBEG and DMTG. Conclusions Scutellaria baicalensis extract restores skin barrier and relieves inflammation through the creation of skin fat barrier. This means that the Scutellaria baicalensis extract can regulate Th2 differentiation through the creation of the epithelial fat barrier.
사이자이지움 클래비플로룸(Syzygium claviflorum (Roxb.) Wall. ex A.M. Cowan & Cowan, S. claviflorum)의 추출물 (잎, 줄기, 열매, 꽃)의 화장품 소재로써 활용되기 위한 생리활성 능력을 검증하였다. 첫번째로, S. claviflorum 추출물은 DPPH와 ABTS assay법을 이용한 항산화 실험에서 다양한 농도로 처리한 결과, 약 80% 이상의 자유 라디칼을 제거하였다. 사람 피부 표피 각질형성세포(human epidermal keratinocytes)를 이용한 세포독성 실험에서는 S. claviflorum 추출물은 낮은 세포독성을 보였다. 또한, S. claviflorum 추출물은 각질형성세포의 분화인자 (keratin (KRT)1, KRT2, KRT9, KRT10)와 피부장벽의 기능 유지에 중요한 involucrin (IVL), loricrin (LOR), filaggrin (FLG)과 claudin1 (CLDN1) 유전자의 발현을 현저히 증가시켰다. 특히, in vitro 아토피 피부염 실험에서 interleukin (IL)-4/IL-13에 의해 억제된 FLG 단백질 발현이 S. claviflorum 추출물에 의해 회복되었다. 따라서, 뛰어난 항산화 효능과 피부장벽 개선 기능을 보유한 S. claviflorum 추출물은 향후 아토피 피부염 치료제 및 화장품 개발에 유용한 소재가 될 것이다.
본 연구에서는 머위에탄올추출물 (PJE)의 기능성 소재로서의 활용 가능성을 알아보기 위해 UVB로 유도된 Human keratinocyte (HaCaT cell)를 통해 피부장벽과 염증 개선에 대한 활성을 알아보고자 하였다. DPPH 라디칼 소거능 측정, ABTS+ 라디칼 소거능 측정, Hydrogen peroxide 소거능 측정을 통해 항산화 효과를 확인한 결과, 1 mg/ml 농도에서 대조군인 ascorbic acid과 비슷한 항산화 효과를 가지고 있음을 확인하였다. UVB로 유도된 HaCaT 세포의 filaggrin과 aquaporin-3의 생성능의 mRNA의 발현을 확인해본 결과, UVB 자극에 의해 줄어든 발현량이 머위 추출물 처리 시, 농도 의존적으로 증가하고 있음을 확인하였다. TNF-𝛼와 IL-1𝛽의 mRNA 발현은 UVB 자극에 의해 증가되었으며 머위 추출물을 처리하였을 때 감소되는 것을 확인할 수 있었다. Migration assay 결과 농도 의존적으로 피부 각질세포의 증식과 상처의 회복율을 증가시켰음을 확인하였다. 실험결과를 바탕으로 머위가 피부 보습 및 피부장벽 기능을 개선할 수 있는 기능성 소재의 화장품으로 사용될 수 있음을 시사한다.
호장은 마디풀과에 속하는 다년초로 우리나라를 포함한 동아시아에서 잘 자생한다. 호장근은 호장의 뿌리로 항염증과 진경제로 사용되어 왔으며 유효성분으로 emodin을 포함하고 있다. 피부의 표피는 자극, 해로운 물질을 차단하고 수분 증발을 방지함으로써 체내를 보호하는 중요한 역할을 한다. 본 연구에서는 호장근과 그 유효성분인 emodin이 피부 장벽과 보습능에 미치는 영향에 대해 평가하고자 하였다. 먼저 호장근은 ABTS+ radicals을 우수하게 제거함으로써 항산화 효능이 뛰어남을 확인하였다. 다음으로, 실시간 중합연쇄효소반응을 통해 각질형성세포의 분화에 중요한 역할을 하는 filaggrin의 유전자 발현을 비교한 결과, 호장근과 emodin에 의해 농도-의존적으로 filaggrin mRNA 발현이 증가하였다. 또한, 호장근과 emodin은 히알루론산 합성에 중요한 역할을 하는 HAS-2 mRNA 발현을 유의하게 증가시키는 것으로 나타났다. 종합적으로, emodin을 유효성분으로 포함하는 호장근은 피부 장벽 강화와 보습능 증강을 위한 기능성 화장품 소재로서 활용될 수 있을 것으로 기대된다.
Objectives Hwangnyeonhaedok-tang is a Korean herbal medical treatment that removes toxic heat, fever and inflammation. The purpose of this study was to investigate the effect of Hwangnyeonhaedok-tang treatment on the relief of atopic dermatitis (AD) through regeneration of skin lipid barrier. Methods Male NC/Nga mice (20 g, 6 week age) were used. Each 10 mice were allocated to the control group (Ctrl), the AD-induced with no treatment group (AE), and the group which induced AD after administering Hwangnyeonhaedok-tang extract (HT). To induce AD-like skin lesions, sodium dodecyl sulfate (SDS) (Sigma-Aldrich, USA) was rubbed on the back of each mouse to remove the lipid lamella of the stratum corneum, and Dermatophagoides (D.) farinae crude extract was applied. HT group was orally administered Hwangnyeonhaedok-tang after induction of AD. IL-4 IL-13, $p-I{\kappa}B$, iNOS, Sudan Black B (SB), loricrin, and filaggrin were observed to confirm the effect. Results In HT group, AD skin score was decreased by 46%. The cytokine IL-4 and IL-13, which can identify Th2 differentiation, was reduced by 73% and 58% each. Anti-inflammatory effects were observed in $p-I{\kappa}B$ and iNOS by 69% and 54%, respectively. Finally, SB showed that the regeneration of the lipid layer and the increase of the regeneration power of loricrin and filaggrin were increased by 437% and 464%, respectively. Conclusions From the study result, we observed that Hwangnyeonhaedok-tang treatment alleviates AD by decreasing skin score, reducing Th2 differentiation, inducing anti-inflammatory, and increasing skin lipid barrier regeneration. Thus, Hwangnyeonhaedok-tang treatment would be considered as an effective AD relieving treatment.
Objectives This study is conducted to evaluate Th2 skewed condition control through skin fat barrier formation from the treatment using Coptidis Rhizoma and Glycyrrhiza uralensis extract. Methods The 6-week-old NC/Nga mice were divided into 4 groups: Control group (Ctrl), lipid barrier eliminate treatment group (LBET), Coptidis Rhizoma and Glycyrrhiza uralensis feeding treatment after lipid barrier elimination group (CGFT), dexamethasone feeding treatment after lipid barrier elimination group (DxFT). After 3 days, differences in skin condition, improvement of skin fat barrier, and control of Th2 skewed condition of each group were observed. Results Pathologic skin damage and tissue changes were less in the CGFT group than in the LBET and DxFT groups, and Transepidermal water loss (TEWL) and pH were also significantly decreased (p < 0.05). The filaggrin intensity and positive response also increased significantly in the CGFT group (p < 0.05). Kallikrein-related peptidase (KLK) 7, Protease activated receptor (PAR)-2, Thymic stromal lymphopoietin (TSLP), Interleukin (IL)-4, and the products of the Th2 differentiation process also showed a significant decrease compared to the LBET and DxFT groups (all p < 0.05). Conclusions The Coptidis Rhizoma and Glycyrrhiza uralensis extract causes skin barrier recovery and function recovery through the formation of skin fat barrier. This leads to the conclusion that Coptidis Rhizoma and Glycyrrhiza uralensis extract can control Th2 differentiation through the formation of skin fat barrier.
Chrysin (5,7-dihydroxyflavone)은 프로폴리스, 꿀 같은 음식과 다양한 식물에 존재하는 천연 플라보노이드이다. Chrysin은 항산화, 항노화, 항염, 항암 효과 등 다양한 생물학적 효과를 가진다고 알려져 있다. 이 연구에서, 우리는 사람의 각질형성세포에서 chrysin이 VDR을 통한 transcriptional activity에 미치는 영향을 dual-luciferase assay을 통하여 살펴보았다. Chrysin은 농도 의존적으로 VDR을 통한 transcriptional activity를 증가시켰다. Quantitative real time PCR을 통해 chrysin이 사람의 각질형성세포에서 VDR mRNA의 발현을 증가시킴을 확인하였다. 또한, chrysin이 각질형성세포의 분화 마커인 keratin 10, involucrin 그리고 filaggrin의 mRNA 발현을 증가시킴을 확인하였다. 이러한 연구 결과는 chrysin이 VDR을 통한 transcriptional activity를 조절하여 각질형성세포의 분화를 촉진시킬 수 있다는 것을 시사한다.
본 연구에서는 프로바이오틱스 유래 엑소좀 유사 나노베지클을 분리하고, 피부에 대한 여러 가지 생리활성을 평가했다. 프로바이오틱스의 한 종인 Lactococcus lactis subsp. lactis (LL)를 배양하고 고압균질기와 한외여과를 통해 70 ~ 200 nm 크기를 갖는 LL 유래 엑소좀 유사 나노베지클(LVs)을 분리했다. 나노입자추적분석 결과 1.81 × 1011 particles/mL로 나타났다. LVs를 섬유아세포와 피부각질세포에 처리하여 피부 주름과 장벽 개선과 관련된 효능을 확인했다. 우선 섬유아세포에서 fibrillin (FBN1) 유전자 발현량이 23%, 피부각질세포에서 fibronectin (FN1)과 filaggrin (FGN) 유전자 발현량이 각각 65%, 400% 증가했다. 그리고 각질형성능은 대조군 대비 30% 증가함을 확인할 수 있었다. 또한, UV 조사한 피부각질세포에 LVs를 처리했을 때 collagen type I alpha 1 (COL1A1)이 대조군 대비 약 83% 증가하는 결과를 보여주었다. 이로써 프로바이오틱스 유래 엑소좀 유사 나노베지클은 장벽 개선과 관련하여 화장품 및 의약품 소재로 이용할 수 있음을 확인했다.
