Bacillus subtilis K-54의 단백질 분해효소 처리에 의한 양모와 견의 품질개선효과 (The Effect of Quality Improvement for Wool and Silk Treated with Protease Produced by B. subtilis K-54)

  • For studies of fibrinolytic enzyme strain K-54 was isolated from the Korean traditional food chungkook-jang. Isolated strains K-54 was identified as Bacillus subtilis. The molecular weight of fibrinolytic enzyme from B. subtilis K-54 was 27 kDa. Optimum temperature for fibrinolytic enzyme of B. subtilis K-54 was $50-70^{\circ}C$ and optimum pH for producing the enzyme of this strain was ranging from 8 to 12. Also, it was found out enzyme activity was completely inhibited by 1mM PMSF. The result indicated this enzyme was thermo-stable alkaline serine protease with strong fibrinolytic activity. The wool and silk were treated with protease of B. subtilis K-54. As a result, the property of dyeing of wool fabrics was increased. By the increasing of treatment time became smoothened. But the change of mechanical properties were not changed.

혈전 분해효소 생산균의 탐색 및 효소생산 최적조건의 조사 (Screening of Fibrinolytic Enzyme Producing from Microorganisms and Optimum Conditions of Enzyme Production)

  • 전통식품(고추장, 된장, 쌈장, 젓갈류, 김치)으로부터 혈전 용해능이 있는 미생물을 분리한 후 그 중 fibrin용해능이 가장 우수한 균주 KJ-23을 최종 선발하였다. 최종 선별한 균주 KJ-23을 Microbial identification system(MIS)의 Sherlock 95 program을 이용하여 동정한 결과 Bacillus brevis KJ-23으로 동정하였다. Bacillus brevis KJ-23균주의 최대혈전용해 생산을 위한 최적배지화 조건을 검토한 결과 nutrient broth, D-ribose 0.5%, malt extract 0.5%, $K_2$HPO$_4$ 0.3% 첨가시 최고의 활성을 보였으며, 금속염을 첨가했을 경우에는 오히려 활성이 감소하는 경향을 보여 금속원은 최적 배지화 조건에서 첨가하지 않았다. 또한 pH7.0, 3$0^{\circ}C$에서 진탕배양했을 경우 최대활성을 보였으며, 시간별 경시효과에서는 24시간에서 혈전용해능이 최대를 나타내었다.

인공재배 버섯의 혈전분해효소 활성검색 (Screening of Fibrinolytic Activities from Cultured Mushrooms)

  • 인공재배 가능한 버섯 13종을 선정하여 균사체 배양 후 배양액 및 혈전분해활성을 검토한 결과 배양액에서는 0.00-0.16U 매우 낮았던 반면에 균사체의 혈전분해 활성은 0.00-0.48U로 배양액에 비하여 상대적으로 높은 활성을 보였다. 또한, 뽕나무 버섯의 건조 균체량은 6.50g/100mL로 다른 균체에 비하여 1.9-5.0배의 높은 생산력을 보여주었다. 인공 재배된 버섯 자실체(7종)의 혈전분해활성은 0.17-0.94U로 나타났으며 아위버섯 자실체의 혈전분해 활성이 가장 높게 나타났다.

젓갈류로부터 혈전용해 균주의 분리 및 동정 (Screening and Identification of the Fibrinolytic Bacterial Strain from Jeot-Gal, Salt-fermented Fish)

  • 우리나라 각지에서 젓갈류를 수집하여 혈전용해능을 나타내는 균주들을 분리하였다. 분리된 균주들의 활성을 측정한 결과 2.04 plasmin unit의 가장 우수한 혈전용해활성을 나타내는 균주를 선별하여 동정하였다. 균주의 형태학적, 배양학적 및 생화학적 성질을 조사한 결과 Bacillus pumilus로 나타났으나, 균체의 지방산조성을 분석하여 동정한 결과 Bacillus atropheus로 나타났다. 최종적 결론을 얻기 위해, 16S rRNA partial sequence(V3 region)를 결정하여 분석한 결과 Bacillus subtilis의 16S rRNA partial sequence와 동일한 것으로 나타나, Bacillus subtilis KJ-48로 명명하였다.

Effects of Water-soluble and Water-insoluble Fractions of Kimchi on the Alteration of Plasma Lipids and Fibrinolytic Activity in Middle-aged Healthy Subjects

  • Freeze-dried kimchi fractions, water-soluble (WSK) versus water-insoluble (WISK), on their effects on plasma lipids and for their fibrinolytic activities were studied. Nineteen middle-aged healthy subjects were assigned to WSK, WISK, or placebo during 6 weeks of kimchi supplementation. Subjects in the WSK and WISK groups were supplemented with 3 grams of kimchi pills (equivalent to 60 grams of fresh kimchi), while the placebo group was on their normal diets. LDL/HDL and atherogenic index decreased in the subjects who received kimchi pills, but not in those in the placebo group. The levels of plasma triglyceride were significantly decreased in the WSK group compared with those in the WISK group (p<0.05). However, there was no difference in the levels of total cholesterol and HDL between the two groups. Fibrinolytic activity of WISK was significantly higher than that of WSK.

칠갑산 야생버섯 추출물로부터 혈전용해효소의 활성 검색 (Screening of Fibrinolytic Activities of Extracts from Wild Mushrooms Collected in Mt. Chilgap of Korea)

  • 칠갑산에 자생하는 야생버섯 67종의 Fibrin 분해활성을 검색한 결과 Marasmius pulcherripes(종이꽃낙엽버섯)는 plasmin 0.75 units의 112%에 해당하는 fibrin 분해활성을 나타냈고, Helvella elastica(긴대안장버섯)는 60%, Psathyrella sp.는 49%, Fomitella fraxinea(장수버섯)은 40%, Leucoagaricus rubrotinctus(돌여우버섯)는 39%, Lepista sordida(자주방망이버섯아재비)와 Oudemansiella sp.는 각각 41%, 26%의 fibrin 분해활성을 나타냈다. 그러나 광대버섯과, 그물버섯과, 끈적버섯과, 난버섯과, 무당버섯과, 귀신그물버섯과, 독청버섯과, 꾀꼬리버섯과, 불로초과, 소나무비늘버섯과, 바구니버섯과, 말뚝버섯과, 어리알버섯과, 붉은목이과 및 술잔버섯과의 버섯은 모두 활성을 나타내지 않았다.

Purification and Characterization of a Major Fibrinolytic Enzyme from Bacillus amyloliquefaciens MJ5-41 Isolated from Meju

  • Meju is a traditional Korean fermented soy product used as a key element for soy sauce and doenjang. Bacilli with antimicrobial activity were isolated from meju prepared by traditional methods at Sunchang county, Jeollabukdo, Korea. Six isolates were identified as Bacillus amyloliquefaciens by recA gene sequencing and RAPD-PCR. One isolate, B. amyloliquefaciens MJ5-41, showed the strongest fibrinolytic activity. A 27 kDa active fibrinolytic enzyme, AprE5-41, was purified from the culture supernatant of MJ5-41 grown on LB by chromatographic methods. The optimum pH and temperature for purified AprE5-41 were 7.0 and $45^{\circ}C$, respectively. AprE5-41 quickly degraded $A{\alpha}$ and $B{\beta}$ chains but not the ${\gamma}$-chain of fibrinogen. AprE5-41 exhibited the highest specificity for N-succinyl-Ala-Ala-Pro-Phe p-nitroanilide, a known substrate for ${\alpha}$-chymotrypsin, cathepsin G, and subtilisin BPN'. The structural gene, aprE5-41, was cloned by PCR and successfully expressed in B. subtilis.

Heterologous Gene Expression of aprE2 Encoding a 29 kDa Fibrinolytic Enzyme from Bacillus subtilis in Bacillus licheniformis ATCC 10716

  • The aprE2 gene from Bacillus subtilis CH3-5 was expressed in Bacillus licheniformis ATCC 10716 using a Bacillus-Escherichai coli shuttle vector, pHY300PLK. The fibrinolytic activity of transformant (TF) increased significantly compared to B. licheniformis 10716 control cell. During the 100 hr incubation in Luria-Bertaini broth at $37^{\circ}C$, fibrinolytic activity of B. licheniformis TF increased rapidly at the late growth stage, after 52 hr of incubation, which was confirmed by zymography using a fibrin gel. pHY3-5 was stably maintained in B. licheniformis without tetracycline (Tc) in the media, 60.9% of cells still maintained pHY3-5 after 100 hr of cultivation.

Pheophytin Content and Fibrinolytic Activity of Silkworm Feces in the Different Larval Stages of Silkworms

  • In order to find potential anticancer agents, we extracted pheophytin in the silkworm feces from various larval stages by water, chloroform and methanol extraction. The cytotoxicity of the pheophytin extracts of various silkworm feces was measured in the CT-26 cells originated from murine metastatic colon cancer, by dye uptake assay. The cytotoxicity of those pheophytins in 2nd, 3rd and 4th instars was better than remaining larval stages. The in vitro anticoagulant and fibyinolytic activities of ethanol extract from varietal mulberry leaves, mulberry branches and silkworm feces and pheophytin extracts from silkworm feces obtained at various larval stages were evaluated in order to find effective therapeutic drugs for the treatment of myocardial and cerebral thrombosis. The fibrinolytic activity was tested using the activated partial thromboplastin time (APTT) and thrombin time (TT) was measured for blood clotting activity. With regards to the fibrinolytic system, ethanol extracts of silkworm feces were better than varietal mulberry leaves and mulberry branches. The pheophytin extracts from 7th days of 5th instar contained the highest percentage of pheophytin and good fibrinolytic activity.

Cordycepin 고함유 동충하초 수용성 추출물의 이화학적 특성과 혈전 용해활성 (Fibrinolytic Activity and Chemical Properties of Cordycepin-Enriched Cordyceps militaris JLM 0636)

  • Cordycepin 고함유 번데기 동충하초(Cordyceps militaris JLM0636, $CM{\alpha}$)와 일반 번데기 동충하초(Cordyceps militaris, CM)의 생리활성 물질 및 영양성분 분석과 혈전 용해 활성을 비교 검토하였다. Total polyphenol compound, flavonoids 및 glutathione 함량은 CM 보다 $CM{\alpha}$에서 높은 함량을 보였다. CM 및 $CM{\alpha}$의 주요 미네랄 구성 성분으로는 K, Ca, Mg, Na이고, 주요 지방산 조성은 linolenic acid, linoleic acid, oleic acid, palmitic acid으로 나타났다. 혈전 용해 활성을 비교 검토한 결과 $CM{\alpha}$이 CM보다 높은 활성을 보였다. 이상의 실험 결과 $CM{\alpha}$의 생리활성물질과 혈전 용해 활성을 이용한 제품 개발에 필요한 기초 자료를 제공하고 유용하게 사용될 수 있을 것으로 사료되어 진다.
