혈전용해활성이 우수한 효모의 탐색 (Screening of a New Fibrinolytic Substances-Producing Yeast)

  • 한국 식품연구원에서 분양받은 48종의 효모들의 PD broth 배양 농축물들의 혈전용해활성을 조사하였다. Saccharomyces cerevisiae Y99-7의 PD broth 배양 농축물이 25 mm의 용해환(clear zone)을 보여 가장 높은 혈전용해 활성을 보였다. 또한 선발 효모를 PD broth 배지로 $30^{\circ}C$에서 24 h 배양하였을 때 yeast extract-peptone dextrose 배양농축물 보다 높은 혈전용해활성을 보였다.

  • The in vitro anticoagulant and fibrinolytic activities of crude extract from insects which have been used as traditional medicines. The extracts of Formica, Huechys and Eupolyph-aga/Steleophaga prolonged activated partial thromboplastin time and thrombin time compared to the value of the control. The fibrinolytic activity of insect extracts was also tested by fibrin plate method. We found that the extracts of Cicadae Periostracum, Eupolyphaga/Steleophdga, Mantidis $O{\ddot{o}}otheca$ and Huechys directly could hydrolyze the fibrin clot without the activation of plasminogen by plasminogen activators.

  • Lumbrokinase, potent fibrinolytic enzyme purified from earthworm, was immobilized onto the total artificial heart valves using photoreaction. This valve were implanted into the lamb for three days. After experiments, thrombus was observed in the untreated valves whereas no thrombus was observed in the lumbrokinase immobilized valves. The fibrinolytic activity and proteolytic activity of the implanted valve was examined. The fibrinolytic activity of the valve was remained after the implantation. The lumbrokinase could be a suitable fibrinolytic agents in the vascular contacting devices to reduce the thrombus.

Purification and Biochemical Characterization of a Serine Protease with Fibrinolytic Activity from Maggots of Mimela splendems

  • Maggot fibrolase (MsMg-1) was purified from the maggots of Mimela splendems using ammonium sulfate fractionation, DEAE Affi-gel affinity chromatography. This protease had a molecular weight of 85 kDa as determined by SDS-polyacrylarnide gel electrophoresis under reducing conditions. It showed strong proteolytic and fibrinolytic activities. The purified enzyme was strongly inhibited by phenylmethanesulfonyl fluoride, Mn/sup 2+/, and Zn/sup 2+/ but it was not by EDTA, EGT, Mg/sup 2+/, Ca/sup 2+/, and Li/sup 2+/ ions. In these experimental results, we have speculated that MsMg-1 is a serine protease with a strong fibrinolytic activity.

약용버섯의 생리활성 분석 (Analysis of Biological Activities of Medicinal Mushrooms)

  • This study was carried out to observe the antioxidative activity, fibrinolytic activity, nitrite savenging ability and adenosine content of three medicinal mushrooms. Among water and ethanol extract of each mushroom, water extract of Inonotus obliquus (Czech) showed the highest antioxidative activity $(57.1{\mu}g/ml)$, nitrite scavenging ability (52.04%), fibrinolytic activity (86.8%) and adenosin content $(94.3{\mu}g/g)$, whereas nitrite scavenging ability of ethanol extract of Phellinus linteus showed higher than that of water extract. Apart from the above statements, water was effective than ethanol as extracting solvent in general. These results suggest that Inonotus obliquus (Czech) showed higher activities such as antioxidative activity, nitrite scavenging ability, fibrinolytic activity and adenosin content and water was effective as solvent except for nitrite scavenging ability.

새우젓 유래 Bacillus sp. S19가 생산하는 혈전용해 요소의 정제 및 특성 (Purification and Characterization of a Fibrinolytic Enzyme Produced by Bacillus sp. S19 from Shrimp)

  • A fibrinolytic enzyme was purified to homogeneity from Bacillus sp. S19 using DEAE and CM column chromatograhies, and gel filtration with a recovery yield of 13%. Its molecular mass was estimated to be 42 kDa by SDS-PAGE. The pH and temperature optima were 8.0 and $40^{\circ}C$, respectively. The enzyme was stable up to $45^{\circ}C$ and over a pH range of 6-9. The N-terminal amino acid sequence of the enzyme was determined as Alsa-Gln-Asp-Ala-Thr-Val-Asn-Ile-Ser-Ala-Glu-Arg-Gln-Val-Ile. The fibrinolytic activity was increased by $Cu^{2+}$ while it was strongly inhibited by metal ions such as $Cd^{2+}$ and $Ba^{2+}$ . In addition, the enzyme was inhibited by EDTA, but not by PMSF, suggesting that it is a metallorprotease.

Production of Carrot Pomace Fortified with Mucilage, Fibrinolytic Enzyme and Probiotics by Solid-state Fermentation Using the Mixed Culture of Bacillus subtilis and Leuconostoc mesenteroides

  • Bioactive compounds were produced from carrot pomace by solid-state fermentation using Bacillus subtilis HA and Leuconostoc mesenteroides. The carrot pomace (CP) fermented by B. subtilis HA with 3% monosodium glutamate (MSG) showed higher production of various bioactive compounds, with 1.64 Pa·sn of consistency, 2.31% of mucilage content, 16.95 unit/g of fibrinolytic enzyme activity, 35.3 unit/g of proteolytic activity and 37.5 mg% of tyrosine content. The mucilage production was greatly dependent upon the concentration of MSG added. Most MSG added in CP was converted into mucilage (2.3%) including 0.83% poly-$gamma$-glutamic acid (PGA) with 1,505 kDa of molecular weight. The CP fermented secondly by Leuc. mesenteroides showed acidic pH and lower consistency. However, the fibrinolytic and proteolytic activities were increased. The secondly fermented CP showed the viable cell counts with $2.5{\time}108$ CFU/g of B. subtilis HA and $3.7{\time}109$ CFU/g of Leuc. mesenteroides, respectively. The freeze-dried fermented CP showed 2.88 Pa·sn of consistency, 24% of mucilage content and 104.9 unit/g of fibrinolytic enzyme activity, respectively. Also, the powder of fermented CP indicated viable cell counts of $8.0{\time}107$ CFU/g of B. subtilis and $4.0{\time}108$ CFU/g of Leuc. mesenteroides. Therefore, the fermented CP that was fortified with dietary fibers, fibrinolytic enzyme and probiotics could be utilized as valuable ingredients of functional foods in food or cosmetic industries.

토양 metagenome library로부터 혈전용해효소의 탐색 (Screening of Fibrinolytic Enzymes from Soil Metagenome Library)

  • Fibrin clots of blood vessels are one of the serious factor caused cardiovascular disease. The development of a antithrombotic and thrombolysis solvent is necessary to prevent and treat these diseases. It has been reported that a strong fibrin-specific fibrinolytic enzyme was produced from a Korean fermented soybean paste similar to Japanese miso. We have been screened the known or novel fibrinolytic enzymes by activity-based and sequence-based screening from soil DNA metagenome library containing all kinds of environmental genomic DNA. The activity-based screening was determined the protease activity on 0.5% skim milk. For sequence-based screening, we designed a set of primer expanding gene sequence of fibrinolytic enzyme, performed PCR and selected clones showing the expected size of amplicons from metagenome library. Transformation of the gene encoding fibrinolytic enzyme was carried out with commercial vectors and their transformants were selected. Finally, we found 15 positive clones from metagenome library. Then each of sequences were analyzed and identified as similar or known the clones of nattokinase. We are going to perform full sequence of each clones, ligate with expression vector, transform into competent cells and then determine activity of expressed enzymes.

된장 유래 혈전분해효소 생산균주의 분리 및 최적 효소생산 조건 탐색 (Screening of Fibrinolytic Enzyme Producing from Microorganisms in Korean Fermented Soybean Paste and Optimum Conditions of Enzyme Production.)

  • 전통발효식품인 된장으로부터 높은 활성의 fibrinolytic enzyme를 생산하는 미생물을 수십 종 분리하였다. 산업적으로 우수한 균종을 선별하기 위하여 분리된 미생물중 fibrinolytic enzyme활성과 성장 속도면에서 가장 우수한 균주를 선별하여 하였으며, 형태학적, 생화학적 및 생리학적 특성을 조사한 후 Bacillus sp.로 동정되었다. Fibrinolytic enzyme생산을 위한 최적 배양조건은 초기 pH $6\~8$일 때 상대활성도가 $80\%$이상을 나타내었고, 배양 12시간째에 가장 높은 활성도를 나타내었다. Fibrin plate를 이용한 혈전용해능을 확인한 결과 높은 혈전용해능을 가지고 있었다. 탄소원으로 galactose를 $4\%$ 첨가 하였을 때 가장 우수하였으며, 질소원은 malt extract를 $4\%$ 첨가 하였을 때 가장 우수하였다. 무기염은 $K_2HPO_4$를 첨가하였을 때 가장 우수한 활성을 나타내었다.

청국장에서 분리한 세균인 Bacillus licheniformis HK-12의 혈전용해활성 및 특징 (Fibrinolytic Activity and Characterization of Bacillus licheniformis HK-12 Isolated from Chungkook-Jang)

  • 이 연구의 목적은 자연 천연한 청국장에서 분비되는 혈전용해효소를 생산하는 Bacillus Iicheniformis HK-12의 혈전용해활성과 특성을 조사하기 위하여 실시한 것이다. 먼저 균주 HK-12의 생리생화학적 특성에 대하여 조사하였다. BIOLOG GP2 MicroPlate system과 16S rRNA 염기서열 분석을 통하여 균주를 동정하였고, 그 결과 Bacillus licheniformis로 확인되었고, B. licheniformis HK-12로 명명하였으며, 이 균주는 GenBank에 [Eu288193]로 등재하였다. 16S rRNA 염기서열 분석에 근거하여, B. licheniformis HK-12의 계통수를 작성하였다. B. licheniformis HK-12의 배양기간동안 세균의 생장, 혈전용해활성, pH의 변화를 모니터링하였다. 36시간배양 후, 배양의 최대 혈전용해활성은 대조군으로서 플라스민과 비교하여 약 2.25배로 나타났다. 혈전용해활성과 관련하여, B. licheniformis HK-12의 생장에서 최적 pH와 온도는 각각 초기 pH 7.0과 40$^{\circ}C$로 나타났다.
