Objectives This study was designed to examine antimicrobial effects of Fermented Coptidis rhizoma and Lonicerae Flos against pathogens. Methods Lactobacilli MRS broth was added to 200mL glass bottle containing 20% herb powder(w/v) followed by 30 minute sonication and then shaking at 70 rpm in $70^{\circ}C$ water bath for 3 hours in order to extract fermented herb. Fermented herb extract was autoclaved at $121^{\circ}$ for 15minutes. $2{\times}10^7$ CFU/mL subcultured bacteria was inoculated and cultured for 24 hours and centrifuged at 3000 rpm for 5 minutes. After transferring to 15 mL conical tube, the viable cells were counted. Results and Conclusion Fermented Coptidis rhizoma and Lonicerae Flos both showed antimicrobial effect on pathogens especially when Fermented Coptidis rhizoma was experimented against Staphylococcus aureus.
본 연구는 홍삼 및 전분질 원료로 산야초 발효액을 첨가하여 제조한 고추장의 이화학적 특성과 항산화 및 관능적 특성 등 품질에 미치는 영향을 조사하였다. 수분함량은 일반 시판 고추장에 비해 6% 정도 낮았고 회분과 조단백의 경우 각각 3.7%, 0.9% 정도 높게 나왔다. 조지방에 경우는 두 시료 모두 별다른 차이가 나타나지 않았다. pH는 대조군에 비해 유의적으로 낮은 편이었는데 이는 다른 기능성 소재를 첨가한 고추장보다도 낮은 값으로 홍삼과 산야초 발효액 첨가가 더 많은 유기산을 생성한 것으로 생각된다. 색도의 경우 L, b값은 홍삼, 산야초 발효액 첨가 고추장이 높은 값을 띄는 반면 a값은 시판 고추장이 더 높게 나왔다. 이러한 차이는 색도의 관한 기호도 평가에서 대조군 보다 훨씬 좋은 결과를 얻을 수 있었다. 기능적인 측면을 살펴보면, DPPH 활성의 경우 전 농도에서 대조군보다 높게 나왔지만 특히 가장 높은 농도인 100 ${\mu}g$/mL에서 유의적으로 라디칼 소거능이 우수한 것으로 나타났으며, ferric reducing antioxidant power는 모든 구간에서 유의적으로 뛰어난 항산화 활성을 보였다. 지질산화를 일으키는 효소인 lipoxygenase의 저해능을 평가한 결과 100 ${\mu}g$/mL의 고농도에서 시판 고추장뿐만 아니라 양성대조군보다 2배가량 높은 저해활성을 띄는 것으로 나타났다. 관능검사에서는 맛, 색깔, 풍미, 종합적 기호도 모든 항목에서 시판 고추장보다 유의적으로 우수하다는 평가를 얻음으로 홍삼과 산야초 발효액의 첨가가 전반적인 고추장 품질을 향상시킬 수 있었음을 알 수 있었다.
Objectives : Socheongryong-Tang (小靑龍湯, SCRT) has been widely used to treat respiratory disease. In this study, we investigate the acute toxicity and safety of fermented Socheongryong-Tang extract. Methods : To evaluate the acute toxicity and safety, 0 (control group), 1250, 2500 and 5000 mg/kg of Socheongryong-Tang and fermented Socheongryong-Tang extracts were orally administered to male and female ICR mice. After single administration, we observed survival rates, general toxicity, changes of body weight for the 14 days and autopsy. Results : Compared with the control group, we could not find any toxic alteration in all treated group (1250, 2500 and 5000 mg/kg). Conclusions : $LD_{50}$ of Socheongryong-Tang and fermented Socheongryong-Tang extracts might be over 5000 mg/kg and it is safe to ICR mice.
Objectives : The aim of this study was to investigate the effects water extract of fermented new korean medicinal mixture, combinations of Mori Folium, Adenophorae Radix, Phllostachyos Folium and Citri Pericarpium (F-MAPC), on adipocyte differentiation, adipogenesis and glucose uptake using undiffernentiated 3T3-L1 adipocytes and L6 myoblasts. Methods : Each herb and those mixture were respectively fermented and then extracted with water. We carried on MTT assay for check-up on cell toxicity, Oil Red O staining for determination of cell differentiation and intracelluar adipogenesis. Western blot analysis for measurement of pAMPK and pACC, $C/EBP{\alpha}$, $PPAR{\gamma}$ and Glut-4 protein expressions were performed. Results : F-MAPC showed significant inhibitory activity on adipocyte differentiation in 3T3-L1 preadipocytes without affecting cell toxicity as assessed by measuring fat accumulation, and this effect was 2 fold higher in 0.2 mg/ml F-MAPC than that of the same dose of each fermented herbal extract alone. In addition, these effects were associated with modulation of adipogenic transcription factors, such as $C/EBP{\alpha}$, $PPAR{\gamma}$, as well as stimulated phosphorylations of AMPK and ACC. Translocation of Glut-4 was significantly increased by 10.2% in L6 cells treated with 0.2 mg/ml F-MAPC compared with that of control. Conclusions : These results demonstrate that F-MAPC may be an ideal candidate for therapy of obesity and diabetes by disturbing the differentiation into adipocytes, as well as the inducement of intramuscular glucose uptake from blood.
Enzyme fermentation is a type of food processing technique generally used to improve the biological activities of food and herbal medicines. In this study, a Syzygii Flos (clove) extract was fermented using laccase derived from Trametes versicolor (LTV). The fermented clove extract showed greater neuroprotective effects against glutamate toxicity on HT22 than the non-fermented extract did. HPLC analysis revealed that the eugenol (1) and dehydrodieugenol (2) contents had decreased and increased, respectively, after fermentation. The content of 2 peaked at 1 h after fermentation to $103.50{\pm}8.20mg/g_{ex}$ (not detected at zero time), while that of 1 decreased to $79.54{\pm}4.77mg/g_{ex}$ ($185.41{\pm}10.16mg/g_{ex}$ at zero time). Compound 2 demonstrated promising HT22 neuroprotective properties with inhibition of $Ca^{2+}$ influx, the overproduction of intracellular reactive oxygen species, and lipid peroxidation. In addition, LTV showed the best fermentation efficacy compared with laccases derived from Pleurotus ostreatus and Rhus vernicifera.

Isoflavone itself is less available in the body without the aid of intestinal bacteria. In this study, we searched for isoflavone-transforming bacteria from human fecal specimens (n = 14) using differential selection media. Isoflavone-transforming activity as the production of dihydrogenistein and dihydrodaidzein was assessed by high-performance liquid chromatography and we found Lactobacillus rhamnosus, named L. rhamnosus vitaP1, through 16S rDNA sequence analysis. Extract from Pueraria lobata (EPL) and soy hypocotyl extract were fermented with L. rhamnosus vitaP1 for 24 and 48 h at $37^{\circ}C$. Fermented EPL (FEPL) showed enhanced anti-tyrosinase activity and antioxidant capacities, important suppressors of the pigmentation process, compared with that of EPL (p < 0.05). At up to $500{\mu}g/ml$ of FEPL, there were no significant cell cytotoxicity and proliferation on B16-F10 melanoma cells. FEPL ($100{\mu}g/ml$) could highly suppress the content of melanin and melanosome formation in B16-F10 cells. In summary, Lactobacillus rhamnosus vitaP1 was found to be able to biotransform isoflavones in EPL. FEPL showed augmented anti-melanogenic potential.
The present study was designed to examine whether the fermented extract using the leaves of Pinus densiflora (FPD), has an ameliorative effect on the lipid metabolism in rats with hyperlipidemia. The body weight gain was attenuated by the adminstration of either 1-dose (2 ml/kg/day) or 2-dose (4 ml/kg/day) of FPD for 2 weeks compared with that of control or hyperlipidemia rats, while blood glucose, systolic blood pressure, sGOT, and sGPT were not altered. Serum total cholesterol, LDL-cholesterol, and athelosclerotic index were partially restored by the administration of 2-dose of FPD in rats with hyperlipidemia without changes of serum HDL-cholesterol, total lipid, and triglyceride concentration. The FPD also partially ameliorated renal functional parameters including plasma cleatinine and creatinine clearance which were attenuated in rats with hyperlipidemia. These results suggest that FPD has an ameliorative effect on the lipid metabolism and renal function in rats with hyperlipidemia.
From March 1, 2017 to May 31, 2017, subjects were allowed to apply a wrinkle-improving herbal cosmetic product containing the extract of Douchi for 8 weeks. The effectiveness of herbal cosmetics was evaluated through wrinkle improvement efficacy parameter analysis, subjective wrinkle improvement through questionnaires, and adverse reaction. The herbal cosmetic products containing the extract of Douchi showed a significant decrease in the wrinkle improvement efficacy parameter analysis result. The results of the subjective satisfaction analysis after use were also evaluated positively. We could confirm the possibility of wrinkle improvement effect of Douchi extract, and it will be possible to contribute to the expansion of korean medicine field through more studies in the future.
