Contribution of the murI Gene Encoding Glutamate Racemase in the Motility and Virulence of Ralstonia solanacearum

  • Bacterial traits for virulence of Ralstonia solanacearum causing lethal wilt in plants were extensively studied but are not yet fully understood. Other than the known virulence factors of Ralstonia solanacearum, this study aimed to identify the novel gene(s) contributing to bacterial virulence of R. solanacearum. Among the transposon-inserted mutants that were previously generated, we selected mutant SL341F12 strain produced exopolysaccharide equivalent to wild type strain but showed reduced virulence compared to wild type. In this mutant, a transposon was found to disrupt the murI gene encoding glutamate racemase which converts L-glutamate to D-glutamate. SL341F12 lost its motility, and its virulence in the tomato plant was markedly diminished compared to that of the wild type. The altered phenotypes of SL341F12 were restored by introducing a full-length murI gene. The expression of genes required for flagella assembly was significantly reduced in SL341F12 compared to that of the wild type or complemented strain, indicating that the loss of bacterial motility in the mutant was due to reduced flagella assembly. A dramatic reduction of the mutant population compared to its wild type was apparent in planta (i.e., root) than its wild type but not in soil and rhizosphere. This may contribute to the impaired virulence in the mutant strain. Accordingly, we concluded that murI in R. solanacearum may be involved in controlling flagella assembly and consequently, the mutation affects bacterial motility and virulence.

  • Supercritical fluid chromatography (SFC) was considered for separating racemic ibuprofen. The chromatographic column (3.9 ${\times}$ 150 mm) was packed with Kromasil$^{\circledR}$ CHI-TBB, and the mobile phase was supercritical carbon dioxide with modifier of IPA. The experimental variables were the content of IPA, and temperature and pressure of supercritical mobile phase. To determine the separation condition, the empirical equation of retention factor and resolution was proposed. In the case of retention factor, the empirical equation was in the form, $k\;=\;a{\rho}\;+b/F\;+\;c\;({\rho}/F)\;+\;d$. The empirical equation for resolution was proposed as a linear form, $R\;=\;a{\rho}\;+\;bF\;+\;c$.

Blend Characteristics of PBT, Nylon6,12 and Preparation of PBT/Nylon6,12 Micro Fiber with Core/shell Structure and their Extrusion Conditions (PBT와 Nylon6,12의 블렌드 특성과 core/shell 구조를 갖는 PBT/Nylon6,12 미세모의 제조 및 압출조건)

  • Poly(butylene terephthalate) (PBT)/Nylon6,12 core/shell micro fiber were prepared by extrusion molding. To investigate their optimum extrusion conditions, compatibility of PBT/Nylon6,12 blend micro fiber in conformity to their weight ratio and manufacture temperature was explored with SEM morphology and DSC. The alterations in their mechanical properties by extrusion speed were compared and analyzed through a UTM. In comparison with SEM figures, the domain sizes of Nylon6,12 were gradually declined by increasing the extrusion temperature of blends. Furthermore, according to these SEM images, the phase separation between Nylon6,12 domain and PBT matrix became indistinct with increasing of weight percentage of Nylon6,12. In case of DSC, the boundaries of two peaks were almost disappeared when increasing the extrusion temperature and also intervals of each two melting peaks became narrow as increasing the Nylon6,12 ratio. The mechanical properties including tensile strength, elongation, flexural strength and flexural modulus were increased as the increase in the extrusion temperature until $260^{\circ}C$. However, the mechanical properties were actually deteriorated over $260^{\circ}C$. The tensile strength, elongation, flexural strength and flexural modulus at $260^{\circ}C$ were 560 $kg_f/cm^2$, 220%, 807 $kg_f/cm^2$ and 22,146 $kg_f/cm^2$, respectively. These values are more than intermediate values of mechanical properties of PBT and Nylon6,12. These results mean that there is compatibility between PBT and Nylon6,12. Based on the extrusion conditions that produced optimum compatibility of blend, as a result, our group obtained micro fibers with the core/shell structure.

  • In this study, we investigated the kinematical properties of the L1521F-IRS in Taurus region using HCN (J=1-0) molecular line. The high resolution mapping has carried out by $5{\times}5$ point observations covering $3.7'{\times}3.7'$ area using a 12-m telescope of Arizona Radio Observatory in Tucsan, USA. L1521F which harbors the faint infrared L1521F-IRS, displayed a strong central concentration of integrated intensity in HCN without serious molecular depletion. It showed a symmetric kinematical structure with the opposite infall motion in either side of the central cores. It is a direct evidence of bipolar outflows in the core of L1521F.

[$^{18}F$]-FDG PET/CT Images of Appendiceal Adenocarcinoma ($^{18}F$-FDG PET/CT로 진단된 충수 선암종)

  • A 53-year -old man underwent $^{18}F$-FDG whole body PET/CT because of the detected liver mass on abdominal CT. The PET/CT showed a huge liver mass ($9{\times}9cm$, SUV: 12.12 ) in the right lobe and a focally hypermetabolic lesion in the right lower quadrant of abdomen (SUV: 9.12). At first, we suspected that the focal hypermetabolic lesion in RLQ was the physiologic uptake of ureter or a metastatic lesion of small bowel. We repeated the abdominal PET/CT next day. The focally hypermetabolic lesion was identified as the appendiceal mass. He underwent right hemicolectomy and right lobectomy of the liver. It was confirmed that the lesion was appendiceal adenocarcinoma with liver metastasis. Cancer of the appendix is an uncommon disease that is rarely suspected before surgery. But, we suggest that PET/CT is useful to identify the small lesion like appendiceal malignant mass.

Sintering and Microwave Dielectric Properties of Bi18(Ca0.725Zn0.275)8Nb12O65 [BCZN] Dielectrics with V2O5 Addition (소결조제 V2O5 첨가에 따른 Bi18(Ca0.725Zn0.275)8Nb12O65 [BCZN] 유전체의 소결 및 마이크로파 유전특성)

  • For the aim of low-temperature co-fired ceramic microwave components, sintering behavior and microwave properties (dielectric constant ${\varepsilon}_r$, quality factor Q, and temperature coefficient of resonant frequency ${\tau}_f$) are investigated in $Bi_{18}O(Ca_{0.725}Zn_{0.275})_8Nb_{12}O_{65}$ [BCZN] ceramics with addition of $V_2O_5$. The specimens are prepared by conventional ceramic processing technique. As the main result, it is demonstrated that the additives ($V_2O_5$) show the effect of lowering of sintering temperature and improvement of microwave properties at the optimum additive content. The addition of 0.25 wt% $V_2O_5$ lowers the sintering temperature to $890^{\circ}C$ utilizing liquidphase sintering and show the microwave dielectric properties (dielectric constant ${\varepsilon}_r$ = 75, quality factor $Q{\times}f$ = 572 GHz, temperature coefficient of resonance frequency ${\tau}_f\;=\;-10\;ppm/^{\circ}C$). The estimated microwave dielectric properties with $V_2O_5$ addition (increase of ${\varepsilon}_r$, decrease of $Q{\times}f$, shift of ${\tau}_f$ to negative values) can be explained by the observed microstrucure (sintered density, abnormal grain structure) and possibly high-permittivity $Bi_{18}Zn_8Nb_{12}O_{65}$ (BZN) phase determined by X-ray diffraction.

  • The purpose of this study was to compare the growth-inhibiting effect of sodium fluoride and stannous fluoride on a cariogenic strain of Streptococcus mutans exposed to the different concentration of $SnF_2$ and NaF The result were as follows: 1. The growth rate of the Streptococcus mutans was unaffected by 75 ppmF, slowed by 150 ppmF, and immediately arrested by 300 or 600 ppmF where increments of NaF were added to actively growing 12-hour broth culture. 2. $SnF_2$ slowed the growth rate at 75 ppmF, was to bactericidal initially at 150 and 300 ppmF, and was to totally bactercidal at 600 ppmF. 3. $SnF_2$ has been shown to be more effective than NaF in inhibiting the growth activity of Streptococcus mutans. 4. The inhibitory effect of fluoride compounds on the growth of Streptococcus mutans was increased in the decreased pH at concentration of 600 ppmF.

Pericarp Anatomy of Fagopyrum (Polygonaceae) (메밀속 식물의 과피 해부형태)

  • Percicarp anatomy of Fagophrum was examined on the basis of 12 species and two subspecies to contribute to a better understanding of specific phylogenetic relationship within genus. Examined species have a similar mature pericarp structure, but differences among the species are found with respect to whether or not sclerotic cells are present, and what kind of is the sclerotic cell shape in theexocarp. By the comparisons with pericarp anatomical structure, they are classified into three groups. First clade is composed of F. esculentum , F. esculentum ssp. ancestralis and F. homotropicum ; second clade is consisted of F. tataricum, F. tataricum ssp. potanini and Fl cymosum ; third clade is composed of f. callianthum,F. capillatum, F. gracilipes, F. leptopodum, F.lineare, F. pleioramosum, F.statice and F.urophyllum. The phylogey based on pericarp characters was considerably consistent with ones proposed by previous authors. It also suggested that pericarp characters are useful informatino for deduceing phylogenetic phylogenetic realtionship within geneus Fagopyrum. On the other hand, morphological character evolution indicated that there are tow synapomorphies in genus. There,it was suggested that these species having selfing and homostylous characters are evolved form heterostylous and ourcrossing species.

Study on Antimicrobial Activity of Extracts from Fritillaria unibracteata Hsiao et K.C. Hsia and F. ussuriensis Maxim.

  • Antimicrobial activities of methanol, ethanol, water, and $CH_2Cl_2$ extracts from Fritillaria unibracteata Hsiao et K.C. Hsia and F. ussuriensis Maxim. were investigated by disk-agar diffusion method. The result showed comparatively strong antimicrobial activity against several microorganisms. The extracts from F. unibracteata and F. ussuriensis dosedependently increased the activity. However, water and $CH_2Cl_2$ extracts showed no antimicrobial activity against 7 microorganisms. Especially, against the most sensitive microorganism Staphylococcus epidermidis, methanol extracts at highest concentration of 20 mg/mL exhibited the largest clear zone on plate by 6-12 mm and ethanol extracts on plate by 6-10 mm.
