db/db mice에 대한 약용 식물추출 혼합물의 항당뇨 상승효과 (Antidiabetic Synergistic Effects of Medicinal Plant Extract Mixtures on db/db Mice)

  • 본 연구는 db/db mice에 대한 구아바 잎(Pg), 바나바 잎(Ls) 추출물 그리고 혼합물 A (바나바, 구아바, 뽕, 솔잎 그리고 가시오가피 뿌리 추출물)의 효과를 조사하였다. 4주간 db/db mice에 이들 추출물을 섭취시킨 결과 Pg, Ls 그리고 혼합물 A의 섭취는 당뇨 대조군에 비해 체중, 혈당, 인슐린을 감소시켰으며 이들 중에서 혼합물 A의 감소효과가 가장 높았다. 이들 추출물은 당뇨 대조군에 비해 총 콜레스테롤, 중성지방, 유리지방산을 감소시켰으며 이들 중에서 혼합물 A의 항고지혈증 효과가 가장 뛰어났다. 또한 혼합물 A는 당뇨 대조군에 비해 Langerhans' islets의 손상을 유의하게 감소시켰다. 따라서 혼합물 A는 포함된 구성물질의 상호보완적인 약리작용에 의해 체중증가의 부작용이 없이 고혈당을 개선시키는 유익한 상승효과를 발휘한 것으로 나타났다.

Synergetic effect of soluble whey protein hydrolysate and Panax ginseng berry extract on muscle atrophy in hindlimb-immobilized C57BL/6 mice

  • Background: Sarcopenia, defined as loss of muscle mass and strength with age, becomes a public health concern as the elderly population increases. This study aimed to determine whether the mixture of soluble whey protein hydrolysate (WPH) and Panax ginseng berry extract (GBE) has a synergetic effect on sarcopenia and, if so, to identify the relevant mechanisms and optimal mixing ratio. Methods: In the first experiment, C57BL/6 mice were hindlimb immobilized for one-week and then administered WPH 800 mg/kg, GBE 100 mg/kg, WPH 800 mg/kg+ GBE 100 mg/kg mixture, and Fructus Schisandrae extract (SFE) 200 mg/kg for two weeks. In the second experiment, experimental design was same, but mice were administered three different doses of WPH and GBE mixture (WPH 800 mg/kg+ GBE 100 mg/kg, WPH 800 mg/kg+ GBE 90 mg/kg, WPH 1000 mg/kg+ GBE 75 mg/kg). Results: In the first experiment, we confirmed the synergetic effect of WPH and GBE on muscle mass and identified that GBE was more effective on the protein synthesis side, and WPH tended to be slightly more effective for protein degradation. In the second experiment, among three different ratios, the WPH 800 mg/kg+ GBE 100 mg/kg was most effective for muscle mass and strength. The mixtures activated muscle protein synthesis via PI3K/Akt/mTORc1 pathway and inhibited muscle protein degradation via suppressing ubiquitin-proteasome system (UPS) and autophagy-lysosome system (ALS), and these effects were more GBE dose-dependent than WPH. Conclusion: The WPH and GBE mixture having a synergetic effect is a potential agent to prevent sarcopenia.

Effects of Natural Extract Mixtures on the Quality Characteristics of Sausages during Refrigerated Storage

  • Owing to the residual toxicity and adverse health effects of chemical preservatives, there is an increasing demand for using natural preservatives in food. Although many natural extracts have been evaluated, research on their antibacterial effects remains insufficient. Therefore, this study aimed to explore the possibility of developing Psidium guajava, Ecklonia cava, and Paeonia japonica (Makino) Miyabe & Takeda extracts as natural food preservatives. Further, the effect of mixing these extracts on microbial growth and quality was evaluated during the refrigeration of sausages. Optimal mixing ratios were determined based on the minimum inhibitory and bactericidal concentrations of each mixed extract against the Listeria monocytogenes, Salmonella spp. and Escherichia coli. D-optimal mixing design optimization tool was further used to obtain an optimum mixing ratio of Formulation 1 (F1). The antibacterial activity of F1 increased with increasing concentration, with similar activities at 0.5% and 1%. The sausages with synthetic or natural preservatives showed significantly lower lipid oxidation than those of the control and grapefruit extract-treated sausages after 4 wk of refrigeration. Total plate counts were observed only in the control and treatment groups stored for 3 wk, and no significant effect of ascorbic acid was observed. Compared to the other samples, sausages with added natural extracts showed the highest overall acceptability scores initially and after 4 wk. Therefore, similar amounts of grapefruit seed and natural extracts had the same effect on microbiological analysis and lipid rancidity during sausage storage. Hence, this mixture can serve as a potential natural preservative in meat products.

복합 생약재 열수추출물 투여에 따른 넙치 세균성 질병에 대한 질병저항성 (Disease Resistance against Bacterial Infection on Treatment of Hot-water Extract with 6 Herbal Mixtures in Olive Flounder Paralichthys olivaceus)

  • 복합 생약재 열수추출물(6종 혼합)을 농도별 기간별로 사료에 흡습시켜 12주 동안 넙치에 급여 후 비특이적 면역 및 질병저항성에 대한 효과를 조사하였다. 복합 생약재 열수추출물의 첨가는 어류 성장에는 아무런 영향을 미치지 않았으며 혈액생화학적인, 결과도 넙치의 생리에는 아무런 영향을 미치지 않았다. 라이소자임 활성은 0.01%의 복합 생약재 열수추출물 농도로 흡습시킨 사료 그룹에서 라이소자임 활성이 증가됨을 알 수 있었다. 복합 생약재 열수추출물의 투여에 따른 넙치 장기의 병리조직에 영향이 없음을 알 수 있었다. 에드와드균의 인위감염에 의한 질병저항성은 0.01% 복합 생약재 열수추출물 농도에서 상대생존율이 높게 나타났다. 이상의 결과를 종합하여 볼 때 넙치에 복합 생약재 열수추출물을 투여시에는 어류의 비특이적 면역력 증강 및 질병 저항성 증강을 나타내는 것으로 사료되었다.

사염화탄소 유발 급성 간 손상에 대한 발효 오미자박 및 헛개과병 추출물의 혼합 비율에 따른 간 보호효능 (Hepatoprotective effect of fermented Schizandrae Fructus Pomace extract and Hoveniae Semen Cum Fructus extract combination mixtures against carbon tetrachloride-induced acute liver injured mice)

  • Objectives : Present study investigated the hepatoprotective effects and the optimal mixing ratio of fermented Schizandrae Fructus Pomace (fSFP) and Hoveniae Semen Cum Fructus (HSCF) extract combination in carbon tetrachloride (CCl4)-induced acute liver injury mice. Methods : ICR mice were orally administered with 200 mg/kg of fSFP, HSCF and mixtures of fSFP and HSCF [MSH (w:w); 1:1, 1:2, 1:4, 1:6, 2:1, 4:1, 6:1, and 8:1] for 7 consecutive days. Silymarin (100 mg/kg) was administered as a reference drug. 0.5 mL/kg of CCl4 was injected intraperitoneally to induce acute liver injury. Body weight gain, relative liver weight, serum chemistry, histopathological analysis, and hepatic endogenous antioxidants capacities were observed. Results : All diverse combinations of MSH significantly reduced relative liver weight increase by CCl4. In addition, MSH administrations significantly decreased the elevation of serum alanine aminotransferase and aspartate aminotransferase activities by CCl4. Histopathological observation indicated that all MSH treatments significantly reduced the increase of degenerated hepatocytes, inflammatory cell infiltration, and histological activity index score by CCl4. Moreover, all MSH administrations reduced the elevation of malondialdehyde contents, and ameliorated the reduction of hepatic glutathione contents, superoxide dismutase activity, and catalase activity. Among the various mixing ratio of MSH combinations, MSH 1:1 and 2:1 showed the most potent anti-oxidative stress, and hepatoprotective effect. Conclusion : Present results suggest that 1:1 and 2:1 combinations of MSH is promising herbal formulation with the hepatoprotective effect against oxidative stress.

생약재 첨가 사료를 투여한 넙치( Paralichthys olivaceus )의 비특이적 면역반응, 혈액성분 및 항병력 효과 (Effects of Medicinal Herb Extract on Non - specific Immune Responses , Hematology and Disease Resistance on Olive Flounder , Paralichthys olivaceus by Oral Administration)

  • Effects of medicinal herb extract on nonspecific immune responses, hematology and disease resistance against Edwardsiella tarda in olive flounder, Paralichthys olivaceus were evaluated. Wormwood, Artemisia asiatica NAKAI and barrenwort, Epimedium koreanum NAKAI were mixed at a ratio of 7 : 3 (w/w) for 2-herbs extract and wormwood, barrenwort, Korean forsythia, Forsythia koreana NAKAI, chrysanthemum, Chrysanthemum zawadskii var. latilobum KITAMURA, peppermint, Mentha arvensis L. var, piperascens MALINV., great burnet, Snaguisorba afficinalis L., Lizard tail. Saururus chinensis BAILL., mulberry, Morus alba L., and star anise, Illicium varum HOOK, f, at the same weight for 9-herbs extract. Two-herbs of 9-herbs extract were prepared by heating after adding 10㎖ of distilled water per g of the herb mixtures. Fish (10.3$\pm$2.5g) were fed the experimental diets supplemented with the 2-herbs or 9-herbs extract at the different concentrations of 0%, 0.1%, 0.5% and 1% per kg diet for 12 weeks. Lysozyme and bactericidal activities of serum, and hematological characteristics were examined during experimental period. After feeding test period, all experimental groups were challenged with E. tarda. Lysozyme activity from the fish fed the diet supplemented with 0.1% or 0.5% of 2-herbs extract was significantly higher than the control. But there was no difference both in bactericidal activity and hematology among each group. Sixty seven % of relative percent survival values (RPS) in the group fed the diet supplemented with 0.1% of 2-herbs was higher than the other group and the control. These results suggest that supplenmentation of 0.1% of 2-herbs extract to a commercial diet may enhance disease resistance in olive flounder. Although both 0.1% and 0.5% 9-herbs extract did not improve non-specific immune reponses, they could enhance disease resistance of 53% RPS, respectively.

쑥 추출물을 이용한 향균 및 김서림 방지에 관한 연구 (Study on Antibacterial and Anti-Fogging using mugwort extract)

  • 코로나 19 이후 향균에 대한 인식이 많아 달라졌다. 외출 후의 손세정은 필수사항이 되었다. 또한 마스크 착용에 따라 안경에 김서림이 많이 발생한다. 이 문제점은 지금까지의 일상생활에 많은 불편함을 야기하고, 향균과 김서림 방지에 대한 우리들의 인식을 바꾸게 하였다. 이에 본 연구에서는 쑥 추출물을 이용하여 향균 및 김서림 방지에 효과가 있는지 연구를 하였다. 쑥 추출물을 얻기 위해 쑥의 건조과정, 알코올 추출과정, 동결건조과정을 거쳤다. 쑥 추출물과 3:7로 혼합된 징크옥사이드, 천연제올라이트 혼합물을 사용하여 향균성 테스트를 실시하였으며, 쑥 추출물과 기타 혼합물을 사용하여 김서림 방지 테스트를 실시하였다. 그 결과 향균성 테스트에서 시험균주로 사용된 E. coli와 K. pneumoniae의 경우 갯쑥추출물 1% 첨가배지부터 세균이 배양되지 않아 대표적인 그람음성균에 대한 항균력이 확인되었다. 김서림 방지 테스트에서 쑥 추출물 10wt% 혼합물이 고온부와 저온부 모두 매우 양호한 결과를 나타냈다. 우리는 쑥 추출물이 향균과 김서림 방지에 효과가 있어 코로나 19를 극복하는데 많은 도움을 줄 것으로 기대한다.

OPB, a water extract from Rehmannia glutinosa Libosch and Eleutherococcus senticosus Max, inhibits osteoclast differentiation and function

  • We performed the present study to investigate whether Rehmannia glutinosa Libosch (RG) extracts (RGX) and Eleutherococcus senticosus Max (ES) extracts (ESX) play any roles in bone metabolism. We examined cellular activities of bone cells by measurement of osteoblastic cell viability, osteoprotegerin (OPG) secretion from osteoblasts, osteoclastogenesis, and osteoclastic activity. There is no cytotoxicity from osteoblasts after treatment with RGX and ESX. The secretion of OPG from the osteoblasts showed marked increases after treatment with RGX and ESX. In addition, RGX and ESX treatment decreased the number of tartrate-resistant acid phosphatase-positive multinucleated cells and the resorption areas. RGX and ESX, when mixed at optimal ratios, induced synergic effects, in vitro. OPB, which showed synergic effects, is the extract of natural ingredients RG and ES mixed at a raw material weight ratio of 4 : 1. It can be suspected that extracts of RG and ES mixtures contains active ingredients involved in bone tissue metabolism and may be effective in improving osteoporosis.

Simultaneous HPLC Analysis of Arachidonic Acid Metabolites in Biological Samples with Simple Solid Phase Extraction

  • A reversed-phase high-performance liquid chromatogrphy (RP-HPLC) has been developed to analyze the metabolites of arachidonic acid based on the specificities of ultraviolet absorption of these various metabolites and is sensitive to the nanogram level. This procedure makes it possible to extract complex mixtures of eicosanoids efficiently with a single step and to analyze them simultaneously by RP-HPLC from biological samples using octadesylsilyl silica extraction column and $PGB_2$ as an internal standard. The cyclooxygenase products {prostaglandin $(PG)D_2,\;PGE_1,\;PGE_2,\;PGF_{1{\alpha}},\;PGF{2{\alpha}},\;6-keto-PGF_{1{\alpha}},$ and thromboxane $B_2(TXB_2)}$ and lipid peroxidation product, isoprostanes, of arachidonic acid were monitored by one isocratic HPLC system at 195 nm wavelength. The lipoxygenase products ${leukotriene(LT)B_4,\;LTC_4,\;LTD_4,$ and 5-hydroxyeicosatetraenoic acid (5-HETE), 12-HETE, 15-HETE} were measured by another isocratic HPLC system at 280 nm for LTs and 235 nm for HETEs. This method provides a simple and reliable way to extract and assess quantitatively the final arachidonic acid metabolites.

Inhibitory Effects of Ninety Nine Korean Plants on Human Immunodeficiency Virus Type 1 Pretense Activity

  • Ninety nine extracts from Korean plants were screened for their inhibitory activities on human immunodeficiency virus (HIV) type 1 pretense by an HPLC method. The pretense inhibitory activities were determined by incubating the extracts in reaction mixtures containing pretense and substrate (His-Lys-Ala-Arg-Val-Leu-(p-NO$_2$- Phe)-Glu-Ala-Nle-Ser-NH$_2$) to perform proteolytic cleavage reactions. Of the extracts tested, the water extracts of Viburnum awabuki (stem and leaves) and Distylium racemosum (leaves) had the highest pretense inhibitory activities at a concentration of 100ug/mL. Activity-guided fractionation, revealed that the n-butanol fraction of the V. awabuki extract and the ethyl acetate fraction from the D. racemosum extract had the greatest inhibitory activity on HIV-1 pretense.
